Mercury bioavailability, transformations, and effects on freshwater biofilms.

Mercury (Hg) compounds represent an important risk to aquatic ecosystems because of their persistence, bioaccumulation, and biomagnification potential. In the present review, we critically examine state-of-the-art studies on the interactions of Hg compounds with freshwater biofilms, with an emphasis on Hg accumulation, transformations, and effects. Freshwater biofilms contain both primary producers (e.g., algae) and decomposers (e.g., bacteria and fungi), which contribute to both aquatic food webs and the microbial loop. Hence they play a central role in shallow water and streams, and also con

Transcriptomic analysis of Propionibacterium acnes biofilms in vitro.

Propionibacterium acnes is a well-known commensal of the human skin connected to acne vulgaris and joint infections. It is extensively studied in planktonic cultures in the laboratory settings but occurs naturally in biofilms. In this study we have developed an in vitro biofilm model of P. acnes and studied growth features, matrix composition, matrix penetration by fluorescent-labeled antibiotics as well as gene expression. Antibiotic susceptibility of biofilms was studied and could be enhanced by increased glucose concentrations. Biofilm cells were characterized by up-regulated stress-induced

Combinatorial drug approaches to tackle Candida albicans biofilms.

The human fungal opportunistic pathogen Candida albicans resides in the human gut, genitourinary tract and on the skin. The majority of infections caused by C. albicans are biofilm-related. In the first part of this review, we discuss new insights into C. albicans biofilm characteristics, concentrating on the extracellular matrix, phenotypic switching, efflux pumps and persister cells. It is widely accepted that this multicellular lifestyle is more resistant to traditional antifungal treatment compared to free-living cells. Therefore, much effort is put in the search for combinations of drugs

Oral epithelial cell responses to multispecies microbial biofilms.

This report describes the use of a novel model of multispecies biofilms to stimulate profiles of cytokines/chemokines from oral epithelial cells that contribute to local inflammation in the periodontium. Streptococcus gordonii (Sg)/S. oralis (So)/S. sanguinis (Ss) and Sg/Fusobacterium nucleatum (Fn)/Porphyromonas gingivalis (Pg) biofilms elicited significantly elevated levels of IL-1α and showed synergistic stimulatory activity compared with an additive effect of the 3 individual bacteria. Only the Sg/Actinomyces naeslundii (An)/Fn multispecies biofilms elicited IL-6 levels above those of cont

Infection through structured polymicrobial Gardnerella biofilms (StPM-GB).

BACKGROUND: We analysed data on bacterial vaginosis (BV) contradicting the paradigm of mono-infection. METHODOLOGY: Tissues and epithelial cells of vagina, uterus, fallopian tubes and perianal region were investigated using fluorescence in situ hybridization (FISH) in women with BV and controls. RESULTS: Healthy vagina was free of biofilms. Prolific structured polymicrobial (StPM) Gardnerella-dominated biofilm characterised BV. The intact StPM-Gardnerella-biofilm enveloped desquamated vaginal/prepuce epithelial cells and was secreted with urine and sperma. The disease involved both genders a

The characteristics of biofilms in peri-implant disease.

AIM: To describe the microbiota associated with peri-implant disease, with a specific emphasis on the differential diagnosis of the condition. MATERIAL AND METHODS: The potentially relevant literature was preliminarily assessed via scoping searches to find the most appropriate search terms and the most efficient Boolean search algorithm. We identified 29 reports on subjects with osseointegrated implants, with a pathological condition compatible with the definition of "peri-implant disease", and reporting microbiological data from samples taken in affected sites. RESULTS AND CONCLUSIONS: In m

Quantitative analysis of in vivo mucosal bacterial biofilms.

BACKGROUND: Quantitative assays of mucosal biofilms on ex vivo samples are challenging using the currently applied specialized microscopic techniques to identify them. The COMSTAT2 computer program has been applied to in vitro biofilm models for quantifying biofilm structures seen on confocal scanning laser microscopy (CSLM). The aim of this study was to quantify Staphylococcus aureus (S. aureus) biofilms seen via CSLM on ex situ samples of sinonasal mucosa, using the COMSTAT2 program. METHODS: S. aureus biofilms were grown in frontal sinuses of 4 merino sheep as per a previously standardized

Hacking into bacterial biofilms: a new therapeutic challenge.

Microbiologists have extensively worked during the past decade on a particular phase of the bacterial cell cycle known as biofilm, in which single-celled individuals gather together to form a sedentary but dynamic community within a complex structure, displaying spatial and functional heterogeneity. In response to the perception of environmental signals by sensing systems, appropriate responses are triggered, leading to biofilm formation. This process involves various molecular systems that enable bacteria to identify appropriate surfaces on which to anchor themselves, to stick to those surfac

Gentamicin promotes Staphylococcus aureus biofilms on silk suture.

BACKGROUND: Communities of bacteria, termed biofilms, develop on biotic and abiotic surfaces, including medical devices and surgical suture. Biofilm-associated bacteria are typically recalcitrant to antibiotic therapy, and the effects of antibiotics on microbial biofilms are not clearly understood. There is emerging evidence that under specific conditions, aminoglycosides may actually promote biofilm development. Experiments were designed to study the effects of gentamicin on suture-associated Staphylococcus aureus biofilms. MATERIALS AND METHODS: S. aureus biofilms were formed after 24 h inc

Biofilm is a integrated multucellular organism with own cycle of development, cooperative behavior of units forming it, which coordinated by QS-system based on production of signal molecules or autoinductors and ability of bacteria to receive these signals. Presence ofbacteria attached to surface of biomedical devices and formation of bacterial biofilms in the microorganism could lead to chronic inflammation, which characterized by presence of macrophages and lymphocytes in the focus of inflammation as well as proliferation of connective tissue, accumulation of matrix proteins and stimulation

Time-dependent effects of rifampicin on staphylococcal biofilms.

OBJECTIVE: To test the time-dependent effects of rifampicin on established biofilms of Staphylococcus epidermidis isolated from patients with cardiac implant infections and catheter-related bacteremia. METHODS: Biofilms were grown in microtiter plates for 24 hours, dyed and stained with crystal violet. The mean optical density (OD) was used for quantification. The OD ratio (ODr = OD of the treated biofilm/OD of the untreated biofilm) was used to measure changes in the thickness of the biofilms over the time. Biofilms were incubated with rifampicin (0.6 mg/mL) for 1, 5, 15, 30 and 60 minutes.

Clinical and laboratory evidence for Neisseria meningitidis biofilms.

Neisseria meningitidis is the etiologic agent of meningococcal meningitis. Carriage of the organism is approximately 10% while active disease occurs at a rate of 1:100,000. Recent publications demonstrate that N. meningitidis has the ability to form biofilms on glass, plastic or cultured human bronchial epithelial cells. Microcolony-like structures are also observed in histological sections from patients with active meningococcal disease. This review investigates the possible role of meningococcal biofilms in carriage and active disease, based on the laboratory and clinical aspects of the dise

The reproducibility of curet sampling of subgingival biofilms.

BACKGROUND: Because few studies have examined the critical issue of sampling reproducibility, the purpose of the present study was to examine the reproducibility of curet sampling of subgingival biofilms. METHODS: Seven subgingival biofilm samples were taken successively, using a curet, from each of 80 sites and individually analyzed for their content of 40 bacterial species using checkerboard DNA-DNA hybridization. One healthy site was sampled in each of 20 periodontally healthy subjects, and one sulcus/pocket of < or =3, 4 to 5, and > or =6 mm was sampled in each of 20 subjects with chronic

Role of bacterial biofilms in urinary tract infections.

Bacterial urinary tract infections represent the most common type of nosocomial infection. In many cases, the ability of bacteria to both establish and maintain these infections is directly related to biofilm formation on indwelling devices or within the urinary tract itself. This chapter will focus on the role of biofilm formation in urinary tract infections with an emphasis on Gram-negative bacteria. The clinical implications of biofilm formation will be presented along with potential strategies for prevention. In addition, the role of specific pathogen-encoded functions in biofilm developme

Bacterial biofilms in patients with indwelling urinary catheters.

Bacteria have a basic survival strategy: to colonize surfaces and grow as biofilm communities embedded in a gel-like polysaccharide matrix. The catheterized urinary tract provides ideal conditions for the development of enormous biofilm populations. Many bacterial species colonize indwelling catheters as biofilms, inducing complications in patients' care. The most troublesome complications are the crystalline biofilms that can occlude the catheter lumen and trigger episodes of pyelonephritis and septicemia. The crystalline biofilms result from infection by urease-producing bacteria, particular

Utilization of microbial biofilms as monitors of bioremediation.

A down-well aquifer microbial sampling system was developed using glass wool or Bio-Sep beads as a solid-phase support matrix. Here we describe the use of these devices to monitor the groundwater microbial community dynamics during field bioremediation experiments at the U.S. Department of Energy Natural and Accelerated Bioremediation Research Program's Field Research Center at the Oak Ridge National Laboratory. During the 6-week deployment, microbial biofilms colonized glass wool and bead internal surfaces. Changes in viable biomass, community composition, metabolic status, and respiratory st

Nanoparticle-based approaches to tackling urinary tract biofilms.

Urinary tract infections represent a substantial global clinical and economic burden, exacerbated by recurrence, indwelling medical devices, and the emergence of multidrug-resistant uropathogens. Biofilm formation contributes to treatment failure due to its resistance to antimicrobial agents, influenced by the urinary tract's unique properties. Therefore, this review examines urinary tract biofilm pathophysiology and evaluates nanoparticle-based strategies, including metallic, polymeric, lipid-based, hybrid, and stimuli-responsive nanosystems. Mechanisms of action, applications in medical devi

The role of extracellular DNA in Neisseria biofilms.

The genus Neisseria includes two human pathogens, Neisseria gonorrhoeae and Neisseria meningitidis, in addition to commensal species that also inhabit human and animal hosts. One of the features that occurs during neisserial host colonization is biofilm formation. In other bacteria, biofilm formation enables persistence as well as evasion of the host immune system. Extracellular DNA (eDNA) forms a major component of Neisseria biofilms and contributes to initial steps in their formation, as well as the structure and stability of the biofilm extracellular matrix. Here, we review the potential ro
