Research Note: The infectious bursal disease (Gumboro disease) vaccination scheme affects the quantitative and qualitative carcass characteristics and the immune response of Ross 308 broiler chickens.

After different vaccinations, the study aimed to assess carcass composition, meat quality, and the level of the infectious bursal disease antibody titers in broiler chickens. In the experiment, 4 (30 chicks per group, 120 total) groups of Ross 308 broiler chickens (males and females) were included. Then, they were vaccinated at the hatchery. Group I was immunized against infectious avian bronchitis (IB) and Newcastle disease (ND) using vaccine A (20 mL/100 birds) and vaccine B (20 mL/100 birds), respectively, with injectable vaccine C (0.2 mL/each) targeting Infectious bursal' and Marek's dise

Immuno-protective effect of commercial IBD vaccines against emerging novel variant infectious bursal disease virus in specific-pathogen-free chickens.

IMPORTANCE: Infectious bursal disease (IBD) is an important viral poultry disease that vaccination can control. OBJECTIVE: This study examined the immune protection of immune-complex (Vaccine A) and attenuated live (Vaccine B) IBD vaccines in specific-pathogen-free (SPF) chickens against a novel Malaysian variant IBD virus (vaIBDV) challenge. METHODS: One-day-old (n =75) SPF chickens were divided randomly into the following three groups of 25 chicks each: Control, Vaccine A, and Vaccine B groups. The vaIBDV strain, UPM1432/2019, was used for the challenge at 21 and 28days post-vaccination (d

Protective effects of rabbit sacculus-derived antimicrobial peptides on SPF chicken against infection with very virulent infectious bursal disease virus.

Previous studies here have demonstrated that the rabbit sacculus rotundus-derived antimicrobial peptides (RSRP) could alter the intestinal mucosal immune responses in specific-pathogen-free (SPF) chickens, however, the protective effects of RSRP on chickens against infection remain questionable. In the present study, eighty SPF chickens were randomly divided into five groups and challenged with very virulent infectious bursal disease virus (vvIBDV) to determine the protective effects and its underlying mechanism of RSRP. Histopathology examination found that vvIBDV-infection caused severe dama

Monitoring serologic response to single in ovo vaccination with an immune complex vaccine against infectious bursal disease in broilers.

The infectious bursal disease (IBD) virus is one of the most resistant and prevalent virus worldwide in the poultry industry, being vaccination the main tool to control the disease. For this reason, consistent and uniform immunization of broiler flocks against IBD is necessary to avoid the disease spreading. The aim of this study was to apply and assess an epidemiologic mapping tool focused on the immunization by in ovo single broiler vaccination using an immune complex IBD vaccine. With this regard, 7,576 serum samples were collected from 603 broiler flocks raised in 354 Spanish farms. To do

Infectious bursal disease virus suppresses H9N2 avian influenza viral shedding in broiler chickens.

1. Infectious bursal disease virus (IBDV) causes immunosuppression in chickens, increasing their susceptibility to other infectious diseases and resulting in vaccination failure. Here, we investigated the immune-depressing effect of IBDV on H9N2 avian influenza viral infection in the broiler chickens. 2. For this purpose, chickens were divided into four groups. In group A, chickens were inoculated with IBDV at 21 days of age and H9N2 avian influenza virus (AIV) 5 days later. Groups B and C only received AIV at 26 days of age and IBDV at 21 days, respectively. The control group (D) were inocula

Field and laboratory findings following the large-scale use of intermediate type infectious bursal disease vaccines in Denmark.

Following a period of clinical outbreaks of very virulent infectious bursal disease virus (vvIBDV) in Denmark, the histological bursal lesion score (HBLS) was used on a national scale to screen broiler flocks vaccinated with intermediate IBD vaccines for lesions indicative of IBDV challenge. High lesion scores were detected in a high percentage of healthy and well performing flocks despite the lack of other indications of the presence of vvIBDV. RT-PCR and subsequent sequencing showed the frequent presence of H253Q and H253N IBDV strains that were genetically close to the sequence of the inter

Development and Evaluation of the Protective Efficacy of Novel Marek's Disease Virus Rispens Vector Vaccines Against Infectious Bursal Disease.

Infectious bursal disease (IBD) is a major disease affecting the poultry industry and is caused by infection with IBD virus (IBDV). To develop a novel vaccine to prevent IBD in chickens, recombinant Marek's disease virus Rispens viruses carrying the VP2 gene of IBDV driven by five different promoters (Rispens/IBD) were constructed using homologous recombination and a bacterial artificial chromosome (BAC). Rispens/IBD driven by the chicken beta-actin (Bac) promoter (Rispens/Bac-IBD), Rous sarcoma virus promoter, or simian virus 40 promoter were administered to 1-day-old SPF chicks, and the prot

Diversity of insect-specific viruses in Culex mosquitoes from Papua New Guinea.

Insect-specific viruses (ISVs) are integral components of mosquito viromes, influencing virus evolution, vector competence, and arbovirus ecology. Papua New Guinea (PNG) is a region of exceptional biodiversity, yet ISVs in local mosquito populations remain poorly characterized. Here, we present the first comprehensive metagenomic survey of ISVs in Culex mosquitoes collected across five provinces of PNG between 2019 and 2021. Using unbiased shotgun metagenomic sequencing, 13,871 female Culex mosquitoes representing five species were analyzed across 86 samples. Approximately 1.6 billion paired-e

Molecular characterization of infectious bursal disease virus (IBDV) strains of genogroup A2B1 circulating in Delaware, Maryland, and Virginia from 2018 to 2023.

Infectious bursal disease virus (IBDV) causes a major immunosuppressive disease in chickens. In this study, we characterized the IBDV strains circulating in Delaware, Maryland, and Virginia (Delmarva) from 2018 to 2023. The sequence of the hypervariable region (HVR) of the VP2 capsid encoded by segment A, and a region of the VP1 polymerase gene encoded by segment B was obtained from 53 and 34 bursal samples, respectively. Phylogenetic analysis revealed that all the sequences belonged to genogroups A2 and B1, typical of US variant strains. An amino acid signature was identified in the HVR conse

Contrasted modifications of IgM and IgT repertoires induced by high- and low-virulent infectious pancreatic necrosis virus strains in rainbow trout (Oncorhynchus mykiss).

INTRODUCTION: Infectious pancreatic necrosis virus (IPNV) is a significant pathogen in salmonid aquaculture, with virulence determined mainly by amino acid residues in the VP2 capsid protein. METHODS: This study investigates the modifications of rainbow trout (Oncorhynchus mykiss) IgM and IgT repertoire induced by two IPNV strains that differ at VP2 positions 217 and 221, specifically IPNV-TA (T217A221) and IPNV-PT (P217T221). For these studies, doubled-haploid isogenic trout were immunised with IPNV-TA or IPNV-PT, and IgHμ and IgHτ repertoires were analysed by high-throughput sequencing. RE

Erythrocyte-targeted recombinant subunit vaccine: A novel approach to improve viral vaccines in fish.

Teleost erythrocytes or red blood cells (RBCs) are nucleated cells with an increasingly recognized role in antiviral defense. To explore the potential of RBCs as immune delivery-competent cells, we explored their response to nanostructured subunit vaccines (nanopellets [NPs]) based on viral antigens from viral hemorrhagic septicemia virus (VHSV) or infectious pancreatic necrosis virus (IPNV) functionalized with interferon-gamma (IFNγ). Additionally, we developed a synthetic RBC-targeting ligand added to the VHSV vaccine for cell-targeting. In ex vivo and in vivo assays, IFNγ enhanced NP uptake

Molecular characterisation of IBDV isolates in Turkey revealed reassortant strains.

1. Infectious bursal disease (IBD) is an acute, highly contagious viral disease of chickens caused by a virus (IBDV) which has a bi-segmented, double-stranded RNA genome. It has five viral proteins in its structure; the VP1 gene is encoded in segment B and the other four are in segment A.2. In this study, bursae of Fabricius and spleen samples taken from chickens suspected of having clinical or subclinical IBD from a total of 50 chicken flocks located in different geographical regions of Turkey were examined.3. The RT-PCR analysis of the VP2 gene showed that 30 of the 50 samples (60%) tested p

Efficacy, Safety, and Interactions of a Live Infectious Bursal Disease Virus Vaccine for Chickens Based on Strain IBD V877.

Infectious bursal disease (IBD) is a highly contagious disease in young chickens which can result in high morbidity and mortality and also in great economic losses. The main target for the virus is the lymphoid tissue with a special predilection for the bursa of Fabricius. Several vaccines are available to control the disease. Intermediate plus vaccines are used in chickens with high maternal antibody titers which face high infection pressure. An example of an intermediate plus vaccine is a live vaccine based on IBD strain V877. The results of an efficacy study in commercial broilers with diff

Molecular characterization of the VP2 gene of infectious pancreatic necrosis virus (IPNV) isolates from Mexico.

Infectious pancreatic necrosis virus (IPNV) is one of the most important viruses in the Pacific salmon Oncorhynchus spp., Atlantic Salmon Salmo salar, and Rainbow Trout O. mykiss industry. This virus has been shown to produce high mortality among salmonid fry and juveniles, and survivors might become carriers. Since 2000, IPNV has affected Mexican Rainbow Trout culture, resulting in considerable economic losses. In the current study, molecular characterization of the VP2 gene of a number of Mexican IPNV isolates was done and the virus's phylogenetic relationships to IPNV reference strains were

Rab1b-GBF1-ARF1 Secretory Pathway Axis Is Required for Birnavirus Replication.

Birnaviruses are members of the Birnaviridae family, responsible for major economic losses to poultry and aquaculture. The family is composed of nonenveloped viruses with a segmented double-stranded RNA (dsRNA) genome. Infectious bursal disease virus (IBDV), the prototypic family member, is the etiological agent of Gumboro disease, a highly contagious immunosuppressive disease in the poultry industry worldwide. We previously demonstrated that IBDV hijacks the endocytic pathway for establishing the viral replication complexes on endosomes associated with the Golgi complex (GC). Here, we report

Epidemiological Insights Into Infectious Pancreatic Necrosis Virus: Detection, Distribution and Persistence in Chilean Salmon Farms.

This study presents a comprehensive molecular and epidemiological characterisation of infectious pancreatic necrosis virus (IPNV) in Chilean salmon aquaculture. Between 2015 and 2016, 26 sampling events were conducted in freshwater and seawater farms spanning a broad latitudinal gradient (30.3° S to 52.6° S). Three diagnostic methods were used: Quantitative real-time Polymerase Chain Reaction (PCR) (qRT-PCR) (targeting VP1 and VP2 protein coding genes), nested PCR and virus isolation in CHSE-214 cells. In addition, a 523-bp VP2 gene fragment was sequenced directly from samples. These approache

Development and Characterization of Monoclonal Antibodies Against VP3 Protein of Infectious Bursal Disease Virus.

Infectious bursal disease virus (IBDV) causes an acute, highly contagious and immunosuppressive disease in 3-5-week-old chicken, called infectious bursal disease (IBD). Current vaccines targeting the hypervariable VP2 gene fail to provide cross-protection against different IBDV strains, necessitating the development of novel diagnostic and preventive strategies that explore other candidate genes to ensure immune efficacy. Here, VP3, a conserved nucleocapsid protein of IBDV, was selected for further analysis. A prokaryotic expression vector, pET-32a-IBDV-VP3, was constructed, followed by expres

Novel Antigenic Variant Infectious Bursal Disease Virus Outbreaks in Japan from 2014 to 2023 and Characterization of an Isolate from Chicken.

Novel antigenic variant strains of the infectious bursal disease virus (IBDV) classified into genogroup A2d have been found in the western part of Japan since 2017. Novel antigenic variant IBDVs now occur in higher frequencies in poultry houses and have been detected in the eastern part of Japan, indicating the spread of IBDVs despite the usual IBDV vaccination. We isolated a novel antigenic variant IBDV, designated as the B2977CE2C3 strain. The B2977CE2C3 strain had two genogroup A2d specific amino acids-lysine and isoleucine, at 221 and 252 aa-along with the other genogroup A2 common amino a
