Design of beam shaping assembly for 2.4 MeV electrostatic tandem accelerator-based neutron source for boron neutron capture therapy: A Monte Carlo study.

In this study, the lithium target thickness and a beam-shaping assembly (BSA) system were designed using Monte Carlo N-Particle version 6.2 for a 2.4 MeV, 15 mA proton accelerator-based neutron source. The BSA components consist of lead (Pb), magnesium fluoride (MgF2), boron carbide (B4C), and tungsten (W) used for the reflector, moderator, and collimator, respectively. The specifications (area and thickness) of each BSA part were varied to analyze the characteristics of neutrons passing through the BSA. The system's performance was evaluated based on various indicators, including the epitherm

Whole-brain spatial metabolomics reveals metabolic gradient shifts in a murine glioma model following boron neutron capture therapy.

BACKGROUND: Glioblastoma (GBM) is the most aggressive and lethal primary brain tumor, characterized by poor prognosis and resistance to current therapies. Boron neutron capture therapy (BNCT) is a tumor-selective particle radiation therapy in which high-linear energy transfer (LET) α-particles and recoil 7Li nuclei are generated through the capture reaction between 10B and thermal neutrons. Although BNCT has been shown to prolong survival in GBM, its effects on global metabolomic alterations of the whole brain, particularly in relation to the tumor microenvironment (TME), remain incompletely u

Modification Technology for Exosome Membranes using Membrane-Curvature-Sensing Peptides and Its Application to Boron Neutron Capture Therapy.

Extracellular vesicles such as exosomes are expected to be used as drug delivery vehicles, particularly in personalized medicine, due to their pharmacological advantages. However, the development of functionalization techniques for isolated extracellular vesicles-including improving their intracellular uptake efficacy from the perspective of a drug delivery system and targeting disease-associated cells to reduce side effects-has become an urgent priority. The aim of this study was to develop a technology for modifying the surface of exosome membranes with functional molecules using membrane-cu

L-Phenylalanine restriction enhances L-boronophenylalanine uptake and improves boron neutron capture therapy efficacy in tumor cell lines.

Boron neutron capture therapy (BNCT) relies on the selective uptake of boron-10 compounds by tumor cells. L-boronophenylalanine (BPA) serves as a key carrier, and enhancing its accumulation is critical for improving BNCT efficacy. In this study, we examined how L-phenylalanine (Phe) restriction affects BPA uptake and related cellular responses in the tumor cell lines SAS, U87-MG, PANC-1, and A375, as well as in the immortalized keratinocyte line HaCaT. Quantitative analysis using inductively coupled plasma atomic emission spectroscopy (ICP-AES) showed that 24 h Phe restriction increased BPA up

Evaluation of a CdTe semiconductor-based gamma camera for real-time dose dosimetry in boron neutron capture therapy.

BACKGROUND: Boron neutron capture therapy (BNCT) is a cancer treatment that leverages the nuclear reaction between boron-10 ( 10 B $^{10}{\rm B}$ ) and thermal neutrons to generate high-energy α $\alpha$ particles and 7 Li $^{7}{\rm Li}$ nuclei that selectively destroy cancer cells while sparing healthy tissues. However, BNCT is limited by current dosimetry methods that are incapable of monitoring boron distribution during therapy. An imaging system capable of real-time dosimetry is essential for optimizing treatment efficacy and minimizing collateral damage. PURPOSE: This study aime

From radiation-induced chromosome damage to cell death: modelling basic mechanisms and applications to boron neutron capture therapy.

Cell death is a crucial endpoint in radiation-induced biological damage: on one side, cell death is a reference endpoint to characterise the action of radiation in biological targets; on the other side, any cancer therapy aims to kill tumour cells. Starting from Lea's target theory, many models have been proposed to interpret radiation-induced cell killing; after briefly discussing some of these models, in this paper, a mechanistic approach based on an experimentally observed link between chromosome aberrations and cell death was presented. More specifically, a model and a Monte Carlo code ori

Synthesis and biological evaluation of boron-containing polyamines as potential agents for neutron capture therapy of brain tumors.

New boron-containing spermidine/spermine (SPD/SPM) analogues have been synthesized: N5-[4-(2-aminoethyl-o-carboranyl)butyl] and N5-{4-[(2,3-dihydroxypropyl)-o-carboranyl]butyl} SPD/SPM derivatives (ASPD-5, ASPM-5, DHSPD-5, and DHSPM-5) as well as N5-{[4-(dihydroxyboryl)phenyl]methyl}spermidine (BBSPD-5). These boronated polyamines retain their ability to displace ethidium bromide from calf thymus DNA and are rapidly taken up in vitro by F98 rat glioma cells. The in vitro toxicities of ASPD-5, ASPM-5, DHSPD-5, and DHSPM-5 are lower than those previously reported for N5-[4-(o-carboranyl)butyl] S

Boron neutron capture therapy delays the decline in neurological function in a mouse model of metastatic spinal tumors.

Metastatic spinal tumors are increasingly prevalent due to advancements in cancer treatment, leading to prolonged survival rates. This rising prevalence highlights the need for developing more effective therapeutic approaches to address this malignancy. Boron neutron capture therapy (BNCT) offers a promising solution by delivering targeted doses to tumors while minimizing damage to normal tissue. In this study, we evaluated the efficacy and safety of BNCT as a potential therapeutic option for spine metastases in mouse models induced by A549 human lung adenocarcinoma cells. The animal models we

Cell-cycle dependence on the biological effects of boron neutron capture therapy and its modification by polyvinyl alcohol.

Boron neutron capture therapy (BNCT) is a unique radiotherapy of selectively eradicating tumor cells using boron compounds (e.g., 4-borono-L-phenylalanine [BPA]) that are heterogeneously taken up at the cellular level. Such heterogenicity potentially reduces the curative efficiency. However, the effects of temporospatial heterogenicity on cell killing remain unclear. With the technical combination of radiation track detector and biophysical simulations, this study revealed the cell cycle-dependent heterogenicity of BPA uptake and subsequent biological effects of BNCT on HeLa cells expressing f

Therapeutic Effect of Boron Neutron Capture Therapy on Boronophenylalanine Administration via Cerebrospinal Fluid Circulation in Glioma Rat Models.

In recent years, various drug delivery systems circumventing the blood-brain barrier have emerged for treating brain tumors. This study aimed to improve the efficacy of brain tumor treatment in boron neutron capture therapy (BNCT) using cerebrospinal fluid (CSF) circulation to deliver boronophenylalanine (BPA) to targeted tumors. Previous experiments have demonstrated that boron accumulation in the brain cells of normal rats remains comparable to that after intravenous (IV) administration, despite BPA being administered via CSF at significantly lower doses (approximately 1/90 of IV doses). Boron neutron capture therapy as a larynx-preserving treatment for locally recurrent laryngeal carcinoma after conventional radiation therapy: A preliminary report.

OBJECTIVE: Laryngeal preservation and a radical cure are the treatment goals for laryngeal carcinoma, and larynx-preserving therapy is generally preferred for early-stage laryngeal carcinoma. When laryngeal carcinoma recurs locally, patients are often forced to undergo total laryngectomy, resulting in loss of vocal function. However, many patients with laryngeal carcinoma who have residual or recurrent disease after radiotherapy wish to preserve their voice. The purpose of this study was to investigate the possibility of using BNCT as a larynx-preserving treatment for residual or recurrent lar

Current role of boron neutron capture therapy in head and neck cancer: insights from 4 years of clinical experience.

PURPOSE: Boron neutron capture therapy (BNCT) was approved in Japan in June 2020 as a curative option for unresectable, locally advanced, or recurrent head and neck cancers. Relevantly, its efficacy and safety in clinical practice should be regularly monitored, particularly regarding its optimal positioning. Therefore, to advance its clinical application, we assessed clinical data on BNCT for head and neck cancers. METHODS: We retrospectively analyzed 285 patients who received initial BNCT between June 2020 and May 2024 with available ≥1 month follow-up data. The primary endpoint was local co

Development of a Theranostic Agent for Bone Metastases: Integration of Nuclear Medicine Imaging and Boron Neutron Capture Therapy (BNCT).

Boron neutron capture therapy (BNCT) is one of the promising cancer treatment methods with 10B-labeled compounds and neutron irradiation. The 10B(n,α)7Li reaction produces a 7Li nucleus and an α-particle with high linear energy transfer, which are responsible for the therapeutic effects. We hypothesized that BNCT could effectively treat bone metastases by selectively accumulating 10B at metastatic lesion sites. In this study, we designed, synthesized, and evaluated [nat/67Ga]Ga-DOTA-K(ε-closo-dodecaborate)D11 ([nat/67Ga]6), a compound integrating closo-dodecaborate for BNCT, a [nat/67Ga]Ga-DOT

Development of Monte Carlo based real-time treatment planning system with fast calculation algorithm for boron neutron capture therapy.

PURPOSE: We simulated the effect of patient displacement on organ doses in boron neutron capture therapy (BNCT). In addition, we developed a faster calculation algorithm (NCT high-speed) to simulate irradiation more efficiently. METHODS: We simulated dose evaluation for the standard irradiation position (reference position) using a head phantom. Cases were assumed where the patient body is shifted in lateral directions compared to the reference position, as well as in the direction away from the irradiation aperture. For three groups of neutron (thermal, epithermal, and fast), flux distributi
