Guillain-Barré syndrome (GBS) and Miller Fisher syndrome (MFS) are acute immune-mediated neuropathies, often preceded by an infection. Anti-glycolipid antibodies are frequently detected in patients' sera in the acute-phase. In particular, IgG anti-GQ1b antibodies are positive in as high as 90% of MFS cases. Anti-glycolipid antibodies are useful for the diagnosis of GBS and MFS. Protected trisaccharides containing N-acetylglucosamine can adopt unexpected conformations through the formation of hydrogen bonds involving the amide group. This conformational behavior was observed by NMR spectroscopy when three protected trisaccharides were dissolved in deuterated chloroform and to a lesser extent in deuterated dichloromethane. In contrast, NMR spectra of the same analogues acquired in the hydrogen bond-accepting solvents deuterated acetonitrile and dimethylsulfoxide showed that the N-acetylglucosamine residues adopted the expected 4C1 conformation.

Five disialylated oligosaccharides, of which three were new compounds, were isolated from pooled human milk. The isolation procedure included gel filtration and ion-exchange chromatography and the final purification was accomplished by HPLC using a mobile phase containing triethylamine as an ion-pairing reagent. The O-glycosidically linked carbohydrate units of glycophorin from bovine erythrocyte membranes were released as reduced oligosaccharides by alkaline borohydride treatment. These oligosaccharides were separated by ion-exchange chromatography followed by gel filtration.
