Systematic engineering of Micromonospora echinospora cell factory for gentamicin C1a overproduction.

The clinical importance of gentamicin C1a as a broad-spectrum aminoglycoside antibiotic underscores the need for efficient biomanufacturing strategies. In this study, we developed a systematic engineering framework to enhance gentamicin C1a production. First, a genome-scale metabolic model (iFX1172) was reconstructed to pinpoint critical bottlenecks in both regulatory and biosynthetic pathways. Guided by model predictions and experimental validation, we identified genC, metK, and BldD as synergistic targets. Coordinated overexpression of these genes increased gentamicin C1a titers to 198.1 mg/

Cell Therapy and Graft Engineering for Chronic Graft-versus-host Disease.

Chronic graft-versus-host disease (cGVHD) is one of the most serious long-term complications of allogeneic haematopoietic stem cell transplantation (allo-HSCT), substantially impairing quality of life and increasing non-relapse mortality. This narrative review summarises current evidence on cellular therapies for cGVHD, focusing on mesenchymal stromal cells (MSCs) and regulatory T cells (Tregs), with emphasis on graft source differences, GVHD prevention and immune reconstitution, cell source, manufacturing, dosing, concomitant immunosuppression, and endpoint definition, alongside emerging appr

Engineering biocompatible polylactic acid (PLA) microcarriers for enhanced 3D cell culture.

This study analyses the performance of polylactic acid (PLA)-collagen microcarriers engineered under laboratory-scale conditions with scalable potential for three-dimensional (3D) cell culture applications. The microcarriers were evaluated for their ability to support cell adhesion, viability, proliferation and conglomerate formation using Caco-2 cells. Results show that PLA-collagen microcarriers significantly improve cell density, with notable proliferation by day 7. Scanning Electron Microscopy (SEM) revealed a rough, porous surface topology that fosters cellular aggregation and conglomerat

In vivo CAR T cell engineering: design principles and open questions.

Chimeric antigen receptor (CAR) T cell therapy has transformed the treatment of hematologic malignancies, leading to seven FDA-approved products, but widespread adoption has been limited by costly and complex manufacturing and administration. Several methods have emerged to directly reprogram T cells within patients, with the potential to simultaneously improve potency, reduce costs, and broaden accessibility. While early clinical studies demonstrate safety and therapeutic activity, critical questions remain regarding long-term efficacy, the interplay between means of delivery and host biology

Recent Advances in Engineering Microbial Cell Factories for Tryptophan-Derived Chemicals.

Tryptophan-derived metabolites are indole-containing compounds with applications in medicine, functional foods, and agriculture. Conventional production via plant extraction or chemical synthesis is often inefficient and unsustainable, prompting the development of microbial biosynthesis. In this review, these metabolites are classified based on reaction sites and the extent of l-tryptophan scaffold remodeling into side-chain-modified compounds, indole ring-functionalized compounds, oxidative coupling products, and complex indole alkaloids, providing a framework to compare biosynthetic logic ac

CAR T cell engineering impacts antigen-independent activation and co-inhibition.

Viral vectors have successfully modified T cells to express chimeric antigen receptors (CARs), leading to clinical approvals. However, their high cost and regulatory challenges hinder rapid clinical translation. Here, we demonstrate that our lentivirally (LV) manufactured R110-CAR T cells, targeting a leukemia neoepitope, can also be engineered using the non-viral sleeping beauty (SB) transposition with minimal-sized DNA vectors. Flow cytometry and single-cell sequencing were used to compare the two production modes using healthy donor and chronic lymphocytic leukemia (CLL)-patient-derived T c

Engineering robust Saccharomyces cerevisiae cell factory for improving fatty alcohol biosynthesis.

Engineering yeast cell factories is a feasible approach to produce value chemicals from renewable feedstocks. However, during the production process, reprogramming of the internal metabolic pathways of yeast cells and environmental stress always compromises its production performance. Here, we engineered the robust Saccharomyces cerevisiae to enhance the production of fatty alcohols by downregulating the expression of target of rapamycin gene TOR1 and deleting histone deacetylase gene HDA1 in S. cerevisiae. The enhanced cellular robustness resulted in the extended chronological lifespan (CLS)

Human and mouse iNKT cell expansion and engineering with viral vectors.

Invariant natural killer T (iNKT) cells are a non-conventional T-cell population characterized by the expression of a conserved semi-invariant T-cell receptor (TCR) with specificity towards self or microbial lipid antigens, presented by the non-polymorphic MHC class I-related molecule CD1d. iNKT cells play a pivotal role in tumor immunosurveillance and serve as a potent tool for anti-cancer therapies. Notably, iNKT cells can be effectively redirected against both hematological and solid malignancies through genetic engineering using either Chimeric Antigen Receptors (CARs) or TCRs targeted to

Microfluidic Engineering of Crater-Terrain Hydrogel Microparticles: Toward Novel Cell Carriers.

Fabrication and application of novel anisotropic microparticles are of wide interest. Herein, a new method for producing novel crater-terrain hydrogel microparticles is presented using a concept of droplet-aerosol impact and regional polymerization. The surface pattern of microparticles is similar to the widespread "crater" texture on the lunar surface and can be regulated by the impact morphology of aerosols on the droplet surface. Methodological applicability was demonstrated by producing ionic-cross-linked (alginate) and photo-cross-linked (poly(ethylene glycol) diacrylate, PEGDA) micropart

Surface Engineering of FLT4-Targeted Nanocarriers Enhances Cell-Softening Glaucoma Therapy.

Primary open-angle glaucoma is associated with elevated intraocular pressure (IOP) that damages the optic nerve and leads to gradual vision loss. Several agents that reduce the stiffness of pressure-regulating Schlemm's canal (SC) endothelial cells, in the conventional outflow pathway of the eye, lower IOP in glaucoma patients and are approved for clinical use. However, poor drug penetration and uncontrolled biodistribution limit their efficacy and produce local adverse effects. Compared to other ocular endothelia, FLT4/VEGFR3 is expressed at elevated levels by SC endothelial cells and can be

Tissue engineering using a combined cell sheet technology and scaffolding approach.

Cell sheet technology has remained quite popular among tissue engineering techniques over the last several years. Meanwhile, there is an apparent trend in modern scientific research towards combining different approaches and strategies. Accordingly, a large body of work has arisen where cell sheets are used not as separate structures, but in combination with scaffolds as supporting constructions. The aim of this review is to analyze the intersection of these two vast areas of tissue engineering described in the literature mainly within the last five years. Some practical and technical details

Cell Sources for Tissue Engineering Strategies to Treat Calcific Valve Disease.

Cardiovascular calcification is an independent risk factor and an established predictor of adverse cardiovascular events. Despite concomitant factors leading to atherosclerosis and heart valve disease (VHD), the latter has been identified as an independent pathological entity. Calcific aortic valve stenosis is the most common form of VDH resulting of either congenital malformations or senile "degeneration." About 2% of the population over 65 years is affected by aortic valve stenosis which represents a major cause of morbidity and mortality in the elderly. A multifactorial, complex and active

Tissue Engineering and Cell-Based Therapies for Fractures and Bone Defects.

Bone fractures and segmental bone defects are a significant source of patient morbidity and place a staggering economic burden on the healthcare system. The annual cost of treating bone defects in the US has been estimated to be $5 billion, while enormous costs are spent on bone grafts for bone injuries, tumors, and other pathologies associated with defective fracture healing. Autologous bone grafts represent the gold standard for the treatment of bone defects. However, they are associated with variable clinical outcomes, postsurgical morbidity, especially at the donor site, and increased surg

Cell biology of sarcomeric protein engineering: disease modeling and therapeutic potential.

The cardiac sarcomere is the functional unit for myocyte contraction. Ordered arrays of sarcomeric proteins, held in stoichiometric balance with each other, respond to calcium to coordinate contraction and relaxation of the heart. Altered sarcomeric structure-function underlies the primary basis of disease in multiple acquired and inherited heart disease states. Hypertrophic and restrictive cardiomyopathies are caused by inherited mutations in sarcomeric genes and result in altered contractility. Ischemia-mediated acidosis directly alters sarcomere function resulting in decreased contractility

Engineering cells with intracellular agent-loaded microparticles to control cell phenotype.

Cell therapies enable unprecedented treatment options to replace tissues, destroy tumors and facilitate regeneration. The greatest challenge facing cell therapy is the inability to control the fate and function of cells after transplantation. We have developed an approach to control cell phenotype in vitro and after transplantation by engineering cells with intracellular depots that continuously release phenotype-altering agents for days to weeks. The platform enables control of cells' secretome, viability, proliferation and differentiation, and the platform can be used to deliver drugs or oth

Treatment of bladder dysfunction using stem cell or tissue engineering technique.

Tissue engineering and stem cell transplantation are two important options that may help overcome limitations in the current treatment strategy for bladder dysfunction. Stem cell therapy holds great promise for treating pathophysiology, as well as for urological tissue engineering and regeneration. To date, stem cell therapy in urology has mainly focused on oncology and erectile dysfunction. The therapeutic potency of stem cells (SCs) was originally thought to derive from their ability to differentiate into various cell types including smooth muscle. The main mechanisms of SCs in reconstitutin

Cell-based tissue engineering therapies: the influence of whole body physiology.

A technology has been developed to isolate a developmentally and phenotypically homogeneous population of pluripotent human mesenchymal stem cells (hMSCs) from adult bone marrow and mitotically expand these cells in culture. These hMSCs have osteoblasts as one of their potential developmental end-stage phenotypes, and, in addition to their osteogenic potential, these hMSCs synthesize and secrete a variety of macromolecules that are known regulators of osteoclast differentiation and activity. In this review, data are presented that demonstrate the phenotypic and developmental homogeneity of the

Engineering Intracellular Protein Sensors in Mammalian Cells.

Proteins can function as biomarkers of pathological conditions, such as neurodegenerative diseases, infections or metabolic syndromes. Engineering cells to sense and respond to these biomarkers may help the understanding of molecular mechanisms underlying pathologies, as well as to develop new cell-based therapies. While several systems that detect extracellular proteins have been developed, a modular framework that can be easily re-engineered to sense different intracellular proteins was missing. Here, we describe a protocol to implement a modular genetic platform that senses intracellular pr

Engineering universal cells that evade immune detection.

The prospect of transplanting cells and tissues without the risk of immune rejection or the need for powerful immunosuppressive drugs is the 'holy grail' of transplantation medicine. Now, with the advent of pluripotent stem cells, CRISPR-Cas9 and other gene-editing technologies, the race to create 'off-the-shelf' donor cells that are invisible to the immune system ('universal cells') has started. One important approach for creating such cells involves the manipulation of genes required for immune recognition, in particular HLA class I and II proteins. Other approaches leverage knowledge of imm
