GWAS for serum galactose-deficient IgA1 implicates critical genes of the O-glycosylation pathway.

Aberrant O-glycosylation of serum immunoglobulin A1 (IgA1) represents a heritable pathogenic defect in IgA nephropathy, the most common form of glomerulonephritis worldwide, but specific genetic factors involved in its determination are not known. We performed a quantitative GWAS for serum levels of galactose-deficient IgA1 (Gd-IgA1) in 2,633 subjects of European and East Asian ancestry and discovered two genome-wide significant loci, in C1GALT1 (rs13226913, P = 3.2 x 10-11) and C1GALT1C1 (rs5910940, P = 2.7 x 10-8). These genes encode molecular partners essential for enzymatic O-glycosylation

The mammalian target of rapamycin pathway in the basolateral amygdala is critical for nicotine-induced behavioural sensitization.

Repeated exposure to nicotine increases psychomotor activity. Long-lasting neural plasticity changes that contribute to the nicotine-induced development of locomotor sensitization have been identified. The mammalian target of rapamycin complex 1 (mTORC1) signalling pathway is involved in regulating the neuroplasticity of the central nervous system. In this study, we examined the role of mTORC1 in the amygdala in nicotine-induced locomotor sensitization. Rapamycin, an inhibitor of mTORC1, was infused into the basolateral amygdala (BLA) and central amygdala (CeA) or systemically administered to

The association of the kynurenine pathway of tryptophan metabolism with acute brain dysfunction during critical illness*.

OBJECTIVES: Plasma tryptophan levels are associated with delirium in critically ill patients. Although tryptophan has been linked to the pathogenesis of other neurocognitive diseases through metabolism to neurotoxins via the kynurenine pathway, a role for kynurenine pathway activity in intensive care unit brain dysfunction (delirium and coma) remains unknown. This study examined the association between kynurenine pathway activity as determined by plasma kynurenine concentrations and kynurenine/tryptophan ratios and presence or absence of acute brain dysfunction (defined as delirium/coma-free d

Critical pathway and hospital-hospital cooperation in acute stroke. Reduction of the length of hospital stay.

Stroke patients tend to stay longer in one hospital compared to patients with other neurological disease. After the introduction of 3 types of critical pathway dedicated for various severity of acute ischemic stroke in 1995, the average length of inhospital days declined from 30.0 days (1993) to 15.3 days (1998), ie 49% reduction. This reduction was achieved by the use of critical pathway and the hospital-hospital cooperation.

Critical frequency of photic driving: which part of the visual pathway do we examine?

Studies on critical frequency of photic driving were performed on patients with optic atrophy, moderately expressed suprachiasmal lesions and normal subjects. A reduction was present only in optic atrophy patients. The critical frequency of photic driving showed significant correlations with visual acuity and differential light sensitivity of the areas located within 15 degrees from the fixation point. The correlation proved higher for foveal sensitivity expressed in non-logarithmic rather than in logarithmic units, the opposite being the case for the sensitivity of parafoveal areas. It may be

Hippo Signaling Pathway Has a Critical Role in Zika Virus Replication and in the Pathogenesis of Neuroinflammation.

Zika virus (ZIKV) is a reemerging human pathogen that causes congenital abnormalities, including microcephaly and eye disease. The cellular/molecular basis of ZIKV and host interactions inducing ocular and neuronal pathogenesis are unclear. Herein, we noted that the Hippo/Salvador-Warts-Hippo signaling pathway, which controls organ size through progenitor cell proliferation and differentiation, is dysregulated after ZIKV infection. In human fetal retinal pigment epithelial cells, there is an early induction of transcriptional coactivator, Yes-associated protein (YAP), which is later degraded w

The alternative pathway of complement activation is critical for blister induction in experimental epidermolysis bullosa acquisita.

Epidermolysis bullosa acquisita is a subepidermal blistering disease associated with tissue-bound and circulating autoantibodies against type VII collagen, a major constituent of the dermal-epidermal junction. The passive transfer of Abs against type VII collagen into mice induces a subepidermal blistering disease dependent upon activation of terminal complement components. To further dissect the role of the different complement activation pathways in this model, we injected C1q-deficient, mannan-binding lectin-deficient, and factor B-deficient mice with rabbit Abs against murine type VII coll

Per- and polyfluoroalkyl substances in the environment and human tissue: A critical review of distribution, exposure pathways, and health risks.

Per- and polyfluoroalkyl substances (PFAS) represent a substantial group comprising over 14,000 synthetic chemicals that are widely utilized in industrial and consumer applications. These compounds pose significant environmental concerns owing to their persistence, bioaccumulation potential, and associated toxicity. Despite the extensive research conducted on PFAS, notable knowledge gaps persist regarding their global distribution, human exposure pathways, and comprehensive health effects, particularly concerning emerging PFAS alternatives. This review is structured according to environmental

Whole Lung Irradiation in Ewing Sarcoma with Pulmonary Metastases: Critical Appraisal of Evolution, Evidence, and Future Pathways in Precision Oncology.

OPINION STATEMENT: Based on the cumulative evidence, we advocate for the continued use of whole lung irradiation (WLI) as an integral component of multimodal therapy for Ewing sarcoma patients with pulmonary metastases, particularly in those with isolated lung involvement. Modern cardiac-sparing techniques, such as intensity-modulated radiotherapy and proton therapy, should be prioritized to mitigate acute cardiopulmonary toxicity, despite the unknown long-term implications of the low-dose bath. Dose prescription should be individualized according to age, risk stratification, and response to c

Multi-genomics data mining identified critical transcription factors and immune response pathways in traumatic optic nerve injury and retinal injury.

BACKGROUND: Traumatic optic nerve and retinal injuries can lead to severe visual impairment, with undetermined underlying molecular mechanism and few reliable molecular biomarkers. This study aims to identify dysregulated genes and signaling pathways potentially involved in these injuries. METHODS: We first identified differentially expressed genes (DEGs) and their enriched functions using two time-course gene expression profile datasets after retinal injury of rats. The hub genes were then obtained from DEGs by protein-protein interaction network, and then assessed by receiver operating char

Comparison the impact of IFN-β alone or in combination with vitamin D on critical pathways involved in AML progression.

BACKGROUND: Acute myeloid leukemia (AML) is a malignant disorder characterized by the accumulation of immature myeloid cells which can be developed and exacerbated through inflammation. Interferon-β (IFN-β) and vitamin D (Vit D) are known for their immunomodulatory and anti-proliferative effects. Key molecules like IL-1β, Gal-9, β-catenin, and NF-κB play important roles in AML progression. This study examines the effects of IFN-β alone and in combination with Vit D on U937 cell proliferation and the regulation of these key molecules. METHODS: Cell counting kit-8 (CCK-8) was applied to assess

Identification of Potential Genes and Critical Pathways in Postoperative Recurrence of Crohn's Disease by Machine Learning And WGCNA Network Analysis.

BACKGROUND: Crohn's disease (CD) is a chronic idiopathic inflammatory bowel disease affecting the entire gastrointestinal tract from the mouth to the anus. These patients often experience a period of symptomatic relapse and remission. A 20 - 30% symptomatic recurrence rate is reported in the first year after surgery, with a 10% increase each subsequent year. Thus, surgery is done only to relieve symptoms and not for the complete cure of the disease. The determinants and the genetic factors of this disease recurrence are also not well-defined. Therefore, enhanced diagnostic efficiency and progn

Parental incarceration and child physical health outcomes from infancy to adulthood: A critical review and multilevel model of potential pathways.

BACKGROUND: There are currently 2.2 million people incarcerated in U.S. prisons and jails, representing a 500% increase over the past 40 years. An emerging literature suggests the impact of mass incarceration extends beyond the prison, jail, or detention center to the families of incarcerated individuals. Less scholarship has considered consequences of parental incarceration for their children's physical health. METHODS: We conduct a critical review of the literature investigating an association between parental incarceration and children's physical health outcomes from infancy to adulthood.

Identification of critical molecular pathways involved in exosome-mediated improvement of cardiac function in a mouse model of muscular dystrophy.

Duchenne muscular dystrophy (DMD) is a progressive disease characterized by skeletal muscle atrophy, respiratory failure, and cardiomyopathy. Our previous studies have shown that transplantation with allogeneic myogenic progenitor-derived exosomes (MPC-Exo) can improve cardiac function in X-linked muscular dystrophy (Mdx) mice. In the present study we explored the molecular mechanisms underlying this beneficial effect. We quantified gene expression in the hearts of two strains of Mdx mice (D2.B10-DmdMdx/J and Utrntm1Ked-DmdMdx/J). Two days after MPC-Exo or control treatment, we performed unbia

Critical roles for both STAT1-dependent and STAT1-independent pathways in the control of primary dengue virus infection in mice.

Dengue virus (DEN), a flavivirus, causes dengue fever and dengue hemorrhagic fever/dengue shock syndrome, the most common mosquito-borne viral illnesses in humans worldwide. In this study, using STAT1(-/-) mice bearing two different mutant stat1 alleles in the 129/Sv/Ev background, we demonstrate that IFNR-dependent control of primary DEN infection involves both STAT1-dependent and STAT1-independent mechanisms. The STAT1 pathway is necessary for clearing the initial viral load, whereas the STAT1-independent pathway controls later viral burden and prevents DEN disease in mice. The STAT1-indepen

Low-temperature autooxidation of cyclopentanone and 3-pentanone: the critical role of competing radical chain branching and chain inhibiting pathways.

The oxidation of two archetypical ketones, cyclopentanone and 3-pentanone, is explored experimentally at 4500 torr (6 bar) and 400-650 K, and computationally, using theory-based master equation calculations. We use H abstraction by photolytic Cl atoms in a high-pressure flow reactor to form a mixture of radicals that are resonantly stabilized by the carbonyl group and other, non-stabilized radicals. Subsequent coupled O2 addition, isomerization, and decomposition pathways are elucidated by time-resolved synchrotron VUV photoionization mass spectrometry. Reaction intermediates - peroxy radicals

Gene co-expression network identifies critical genes, pathways and regulatory motifs mediating the progression of rift valley fever in Bostaurus.

Rift Valley Fever (RVF) is a mosquito-borne viral disease caused by the Rift Valley Fever Virus. The disease is a zoonosis that largely affects domestic animals, including sheep, goats, and cattle, resulting in severe morbidity and mortality marked by massive storm abortions. To halt human and livestock deaths due to RVF, the development of efficacious vaccines and therapeutics is a compelling and urgent priority. We sought to identify potential key modules (gene clusters), hub genes, and regulatory motifs involved in the pathogenesis of RVF in Bos taurus that are amenable to inhibition. We an

Two apicoplast dwelling glycolytic enzymes provide key substrates for metabolic pathways in the apicoplast and are critical for Toxoplasma growth.

Many apicomplexan parasites harbor a non-photosynthetic plastid called the apicoplast, which hosts important metabolic pathways like the methylerythritol 4-phosphate (MEP) pathway that synthesizes isoprenoid precursors. Yet many details in apicoplast metabolism are not well understood. In this study, we examined the physiological roles of four glycolytic enzymes in the apicoplast of Toxoplasma gondii. Many glycolytic enzymes in T. gondii have two or more isoforms. Endogenous tagging each of these enzymes found that four of them were localized to the apicoplast, including pyruvate kinase2 (PYK2
