Peptide molecular lock-engineered nanobodies enable an oriented dual-modal immunoassay for reliable detection of Cronobacter sakazakii.

Conventional nanobody ELISAs for trace Cronobacter sakazakii in powdered infant formula suffer from random orientation and low signal output. We developed an oriented dual-modal immunoassay that combines site-specific biotinylation via a C-terminal AviTag and a peptide molecular lock, enabling controlled surface orientation while preserving nanobody structural integrity. This strategy was further integrated with phage-displayed nanobodies for multivalent amplification and both fluorescent and colorimetric readouts. The assay exhibited a broad linear range of 103-106 CFU/mL, with limits of dete

Meningitis and brain abscess formation caused by Cronobacter malonaticus sequence type 440 in a full-term neonate.

Cronobacter spp. cause serious diseases, such as necrotizing enterocolitis, bacteremia, and meningitis in neonates and infants. Most Cronobacter-associated meningitis is reportedly due to C. sakazakii and the majority of infections caused by C. malonaticus occur in adults and are less severe. We report the case of meningitis and brain abscess caused by C. malonaticus Sequence Type (ST) 440 in a healthy full-term neonate. We should consider the possibility that full-term neonates may develop meningitis due to C. malonaticus and treat appropriately because its mortality rate is very high, and su

Molecular and functional characterization of cold-adaptive phage LPCS39 for effective control of Cronobacter sakazakii in refrigerated foods.

Cronobacter sakazakii (C. sakazakii) is a foodborne pathogen with the ability to survive and proliferate under cold-chain conditions, thereby posing serious risks to infants and public health. However, most reported phages targeting this pathogen demonstrate limited bacteriolytic activity at low temperatures. In this study, the lytic phage LPCS39 was isolated, characterized, and tested in food matrices including milk, reconstituted powdered infant formula (RPIF), and lettuce. LPCS39 exhibited a broad host range against multiple Cronobacter species, and Enterobacter cloacae, along with high sta

Effects and molecular mechanism of sugar transporter ESA_RS15745 on desiccation resistance, motility, and biofilm formation of Cronobacter sakazakii.

Cronobacter sakazakii, an important Gram-negative foodborne pathogen, can cause neonatal meningitis and sepsis with high rates of infection and death. Gene ESA_RS15745 encodes a sugar transporter protein, which is not only essential for osmotic pressure maintenance during bacterial growth and reproduction but also associated with their desiccation tolerance, motility, and biofilm formation. Here, a mutant strain of ESA_RS15745 (ΔESA_RS15745) and the complementation strain (cpESA_RS15745) were constructed using a suicide vector knockout and gene complementation. ΔESA_RS15745 was found to have a

Inactivation kinetics of Cronobacter sakazakii and Salmonella enterica in reconstituted powdered infant formula and survival during subsequent storage.

Powdered infant formula (PIF) has been implicated as a potential vehicle for foodborne pathogens, such as Cronobacter sakazakii and Salmonella enterica, which can cause life-threatening infections in infants. For high-risk infants, public health agencies recommend using hot water to reconstitute PIF for safety. This study evaluated the influence of reconstitution water temperature on pathogen inactivation and subsequent survival and growth during storage. Milk-based (MB) and soy-based (SB) PIF were inoculated with C. sakazakii or S. enterica via spray atomization and left to stabilize for 1 mo

Roles and mechanisms of gapA and gap-encoded proteins in cronobacter sakazakii adhesion to and invasion of intestinal cells and neonatal rats.

Cronobacter sakazakii (C. sakazakii) is a foodborne pathogen whose initial process involves intestinal cell adhesion mediated by numerous virulence factors encoded in various genes. The key metabolic enzyme, glyceraldehyde-3-phosphate dehydrogenase (GAPDH, also known as GapA), is encoded by gapA in the glycolysis pathway and acts as an adhesin in some bacteria. In C. sakazakii, there is also a key enzyme in the gluconeogenesis pathway, Gap, with type I GAPDH and erythrose-4-phosphate dehydrogenase activities and encoded by gap. This study aimed to investigate the virulence properties of GapA a

Comparative proteomics reveals the antibiotic resistance and virulence of Cronobacter isolated from powdered infant formula and its processing environment.

Cronobacter species are opportunistic foodborne pathogens that can cause neonatal meningitis, sepsis, and necrotizing enterocolitis. In this genus, certain level strains have high mortality to infant (Cronobacter sakazakii and Cronobacter malonaticus) and antibiotic tolerance. Cronobacter has strong environmental tolerance (acid resistance, high temperature resistance, UV resistance, antibiotic resistance, etc.) and can survive in a variety of environments. It has been isolated in various production environments and products in several countries. However, the relationships between Cronobacter

The diguanylate cyclase CrdA contributes to cell adhesion and gut colonization by switching motility and sessile state of Cronobacter malonaticus.

Cronobacter malonaticus is an emerging foodborne pathogen posing severe threats to vulnerable populations. Despite the established role of the ubiquitous second messenger cyclic diguanylate (c-di-GMP) in bacterial virulence, the specific diguanylate cyclases (DGCs) that orchestrate pathogenicity in C. malonaticus are unknown. Here, we systematically characterized 13 putative DGCs in C. malonaticus and identified CrdA as a pivotal regulator. We demonstrated that CrdA reciprocally inhibits swimming motility and promotes cellulose-dependent biofilm formation by modulating intracellular c-di-GMP l
