Fluorescence behavior of cysteine-mediated Ag@CdS nanocolloids.

Ag@CdS quantum dots were synthesized by a colloidal method. CdS nanoparticles stabilized by L-cysteine were doped with varying silver concentrations. Ag@CdS nanoparticles were characterized by UV-visible spectroscopy, steady-state and time-resolved fluorescence, X-ray diffraction, Fourier transform infrared resonance (FTIR), and transmission electron microscopy. The effect of dopant concentration and effect of aging of Ag@CdS colloids were studied. At low silver content, emission was found to enhance, while at high silver content, emission quenching was observed after aging. Ag-CdS nanocolloid

Mercury(II) cysteine complexes in alkaline aqueous solution.

Mercury(II) complexes with l-cysteine (H(2)Cys) in alkaline aqueous solutions have been structurally characterized by means of extended X-ray absorption fine structure (EXAFS) spectroscopy. The distribution of [Hg(Cys)(n)] (n = 2, 3, and 4) species in approximately 0.09 mol dm(-3) mercury(II) solutions with H(2)Cys/Hg(II) ratios varying from 2.2 to 10.1 has been evaluated by fitting linear combinations of simulated EXAFS functions for the separate complexes to the experimental EXAFS data, aided by (199)Hg NMR and Raman results. For the [Hg(Cys)(2)](2-) and [Hg(Cys)(3)](4-) complexes and the no

Oxidative deamination of carboxyethyl-cysteine and carboxymethyl-homocysteine.

Snake venom L-aminoacid oxidase, while inactive on 4C and 5C dicarboxylic alpha-amino acids, may oxidize their 6C and 7C homologues alpha-aminoadipic and alpha-aminopimelic acid. It has been demonstrated that the enzyme also oxidizes S-carboxyethyl-L-cysteine and S-carboxymethyl-L-homocysteine, two analogues of alpha-aminopimelic acid with the gamma or delta methylene groups substituted by a sulfur atom. As oxidation products the corresponding ketoacids were obtained. Thus substrate specificity of L-aminoacid oxidase for dicarboxylic alpha-aminoacids seems highly dependent on the carbon chain

Cysteine stimulation of glucose oxidation in thyroid tissue.

Oxidation of D-glucose-C(14) by bovine thyroid slices was stimulated by cysteine, 2-aminoethanethiol, and cystamine. Amino acids tested, other than cysteine, were not stimulatory, except tyrosine and arginine to a slight degree. The pattern of stimulation resembled that brought about by thyroid-stimulating hormone. Stimulation of labeled CO(2) production was greater when glucose-1-C(14) rather than glucose-6-C(14) was the substrate.

Unravelling the Therapeutic Potential of Cysteine for Cardiovascular Health.

Cardiovascular diseases (CVDs) are a major global health concern, representing a leading cause of death and imposing a significant financial burden on public healthcare systems worldwide. They raise national healthcare expenditure into trillions of dollars globally. The development and progression of CVDs are heavily driven by oxidative stress, inflammation, lipid dysregulation, and endothelial dysfunction. Recent studies have highlighted the potent antioxidant and anti-inflammatory properties of cysteine-containing compounds, suggesting their role in mitigating cardiac diseases. This review e

Introducing Cysteines into Nanobodies for Site-Specific Labeling.

We have developed a generally applicable methodology for cysteine mutagenesis of nanobody (Nb) framework region serine residues. This strategy allows for subsequent labeling with thiol-reactive compounds without disrupting Nb antigen binding. We provide a protocol for production, labeling, and affinity determination of cysteine-engineered Nbs (cys-Nbs) with Alexa Fluor 488-maleimide and the mercury compound para-chloromercuribenzoic acid (PCMB). Alexa Fluor 488- and PCMB-labeled cys-Nbs can be used for immunofluorescence microscopy and experimental phasing in crystallography, respectively.

Heterogeneous adaptation of cysteine reactivity to a covalent oncometabolite.

An important context in which metabolism influences tumorigenesis is the genetic cancer syndrome hereditary leiomyomatosis and renal cell carcinoma (HLRCC), a disease in which mutation of the tricarboxylic acid cycle enzyme fumarate hydratase (FH) causes hyperaccumulation of fumarate. This electrophilic oncometabolite can alter gene activity at the level of transcription, via reversible inhibition of epigenetic dioxygenases, as well as posttranslationally, via covalent modification of cysteine residues. To better understand the potential for metabolites to influence posttranslational modificat

Selective and reversible modification of kinase cysteines with chlorofluoroacetamides.

Irreversible inhibition of disease-associated proteins with small molecules is a powerful approach for achieving increased and sustained pharmacological potency. Here, we introduce α-chlorofluoroacetamide (CFA) as a novel warhead of targeted covalent inhibitor (TCI). Despite weak intrinsic reactivity, CFA-appended quinazoline showed high reactivity toward Cys797 of epidermal growth factor receptor (EGFR). In cells, CFA-quinazoline showed higher target specificity for EGFR than the corresponding Michael acceptors in a wide concentration range (0.1-10 μM). The cysteine adduct of the CFA derivati

Cysteine Cathepsins and their Extracellular Roles: Shaping the Microenvironment.

: For a long time, cysteine cathepsins were considered primarily as proteases crucial for nonspecific bulk proteolysis in the endolysosomal system. However, this view has dramatically changed, and cathepsins are now considered key players in many important physiological processes, including in diseases like cancer, rheumatoid arthritis, and various inflammatory diseases. Cathepsins are emerging as important players in the extracellular space, and the paradigm is shifting from the degrading enzymes to the enzymes that can also specifically modify extracellular proteins. In pathological conditio

[Availability of D-cysteine as a protectant for cerebellar neurons].

Hydrogen sulfide (H2S) has been focused as a biological mediator, which modulates signal transduction and protects cells and tissues from oxidative stress. H2S is also expected as a neuroprotectant because it has a neuroprotective activity. Endogenous H2S is mainly generated from L-cysteine. However, it is difficult to use L-cysteine as a neuroprotectant because of its neurotoxicity. In 2013, a novel biogenesis pathway of H2S from D-cysteine has been identified. In this pathway, D-amino acid oxidase (DAO) converts D-cysteine to 3-mercaptopyruvate (3MP), followed by the generation of H2S from 3

The functional role of UBA1 cysteine-278 in ubiquitination.

Although total UBA1 levels were unchanged, after oxidation for 60 min, we observed dramatic changes in the levels of BIAM-labeled UBA1 in both the membrane and cytosol fractions that suggested oxidative stress induces translocation of UBA1 from the cytosol to the membrane. Notably, in PrdxII(-/-) oxRBCs, ubiquitination levels were reduced about 75% in the membrane fraction after 90 min, even though UBA1 levels were increased. These results suggest ubiquitination levels are determined by UBA1 activity, not the level of UBA1 protein. Levels of ubiquitin conjugate (denoted ∼Ub) in HEK293T and CMT

Cure of hookworm infection with a cysteine protease inhibitor.

BACKGROUND: Hookworm disease is a major global health problem and principal among a number of soil-transmitted helminthiases (STHs) for the chronic disability inflicted that impacts both personal and societal productivity. Mass drug administration most often employs single-dose therapy with just two drugs of the same chemical class to which resistance is a growing concern. New chemical entities with the appropriate single-dose efficacy are needed. METHODS AND FINDINGS: Using various life-cycle stages of the hookworm Ancylostoma ceylanicum in vitro and a hamster model of infection, we report t

Cysteine conjugate of methazolamide is metabolized by beta-lyase.

Bovine kidney and liver homogenates degraded a cysteine conjugate of methazolamide, S-(5-acetylimino-4-methyl-Delta2-1,3,4-thiadiazolin-2-yl)cysteine. We isolated the degradation product following incubation with kidney homogenate by high-performance liquid chromatography on reversed-phase columns. The chemical structure was confirmed by proton and carbon-13 nuclear magnetic resonance spectroscopy (1H NMR and 13C NMR, respectively), and elemental analysis by high-resolution mass spectrometry to be N-(3-methyl-5-mercapto-Delta4-1,3,4-thiadiazol-2-yl)acetamide, a thiol compound. In vivo and in vitro nephrotoxicity of the cysteine conjugate of hexachlorobutadiene.

Hexachlorobutadiene (HCBD), a renal toxin and carcinogen, is thought to require bioactivation to exert toxicity. The chemically synthesized cysteine conjugate of structurally similar halogenated hydrocarbons, trichloroethylene, chlorotrifluoroethylene, and chlorodifluoroethylene, have been shown to be nephrotoxic. Hence the cysteine conjugate of HCBD, S-pentachlorobuta-1,3-dienyl cysteine (PCBC), was assessed for potential nephrotoxicity. Active acid and base transport in isolated rabbit renal tubules was used to screen nephrotoxicity. A dose-dependent decrease in acid and base transport was o

Versatile roles of cysteine persulfides in tumor biology.

Rewiring the transsulfuration pathway is recognized as a rapid adaptive metabolic response to environmental conditions in cancer cells to support their increased cysteine demand and to produce Reactive Sulfur Species (RSS) including hydrogen sulfide (H2S) and cysteine persulfide. This can directly (via RSS) or indirectly (by supplying Cys) trigger chemical or enzyme catalyzed persulfidation on critical protein cysteine residues to protect them from oxidative damage and to orchestrate protein functions, and thereby contribute to cancer cell plasticity. In this review key aspects of persulfide-m

Predicting cysteine reactivity changes upon phosphorylation using XGBoost.

Cysteine reactivity serves as a significant indicator of protein function and can be affected by phosphorylation events. Experimental approaches have been developed to investigate this effect, but the scale is still relatively limited. Machine-learning approaches promise to accelerate the investigation of these phenomena. In this study, protein sequence information, distances to the closest phosphorylation sites, and the membership score of the intrinsically disordered region were used to represent the cysteine. Following the feature selection using an elastic net model, two groups of binary c

A Cysteine-Mediated Synthesis of Red Phosphorus Nanosheets.

Among phosphorus-based nanomaterials, layered black phosphorus and violet phosphorus have been actively explored in the past decade. However, methods for the synthesis of red phosphorus nanosheets (RPNSs) is lacking, even though red phosphorus (RP) is commercially available at low cost and has excellent chemical stability at room temperature. We report an efficient strategy for fabrication of RPNSs and doped RPNSs using cysteine as a reducing reagent. Data from in vitro and in vivo studies suggested that RPNSs can trigger production of reactive oxygen species, DNA damage, and subsequent autoph
