Immune checkpoint inhibitors (ICIs) have revolutionized cancer immunotherapy by enhancing T-cell-mediated anti-tumor responses in solid malignancies. However, their efficacy is often limited by tumor-specific factors, T-cell dysfunction in cold tumors, or in the presence of lymph node metastasis (LNM). Moreover, clinical trials indicate no significant survival advantage of sentinel lymph node biopsy (SLNB) over no lymph node surgery in early-stage cancers, highlighting the need for novel combinatorial approaches to improve treatment outcomes. Tumor electrochemotherapy (ECT) can overcome immuno

The study was undertaken to compare antitumor efficacy of electrochemotherapy (ECT) with cold plasma therapy (CP) in a melanoma mouse model. After melanoma implantation into the flank of C57BL/6N mice, CP by two different plasma sources (APPJ and DBD) was applied directly to the tumor surface. ECT was performed with bleomycin intravenously at a field strength of 1000 V/cm without or combined with CP. Primary endpoints were tumor growth acceleration (TGA), daily volume progression (DVP) and survival after treatment. Both plasma sources as single treatment showed a significant TGA delay, which p

Electrochemotherapy, a tumour ablation modality, which facilitates intracellular delivery of poorly-permeable cytotoxic drugs, such as bleomycin, has shown promising results in the treatment of cutaneous and subcutaneous melanomatous and non-melanomatous metastases. We report the case of a 52-year-old Caucasian gentleman with a keloid scar to his left earlobe that developed following a piercing. Despite multiple intralesional steroidal injections, five intralesional excisions and a course of superficial radiotherapy the keloid scar remained over nine years later. For 15 years the patient had a

BACKGROUND: Surgery and/or radiotherapy are the elective therapies for most primary skin cancers. Nevertheless, some patients develop recurrences, and chemotherapy has resulted in poor complete responses. Permeabilization of the cell membrane by electric pulses allows bleomycin to enter into the cell, increasing possibility of cytotoxicity.
METHODS: From November 1998 through November 1999, 15 patients with 38 skin lesions participated in a phase II prospective clinical trial, using intralesional bleomycin plus electric pulses delivered 10 min after bleomycin injection, which lasted 100 micro

OBJECTIVE: Tumors in elephants present substantial therapeutic challenges because of their large size, anatomical complexity, and the risks associated with surgical intervention. Targeted electrochemotherapy (ECT) has emerged as an effective local treatment modality in both human and veterinary oncology. However, its application in exotic species has been rarely reported.
ANIMAL: An adult female Asian elephant (Elephas maximus) housed at the El Paso Zoo in Texas.
CLINICAL PRESENTATION, PROGRESSION, AND PROCEDURES: The elephant was diagnosed with a large solid tumor considered unsuitable for

Electrochemotherapy (ECT) is a local treatment combining chemotherapy with electroporation. This prospective multicentre study aimed to evaluate the efficacy of ECT in the treatment of patients with skin metastases from breast cancer and confirm whether "luminal A-like" tumors are more responsive to treatment. One-hundred and ninety-five patients were included in the analysis. 55% achieved complete response, 27% partial response (objective response OR 82%); 12% stable disease and 5% experienced progressive disease. The analysis by tumor phenotype showed a significant better response rate in Lu

BACKGROUND: Cutaneous mast cell tumor (MCT) is one of the most common skin neoplasms in cats and typically exhibits clinically benign behavior. Multiple MCTs, recurrent tumors, or infiltrated lymph nodes are associated with a more guarded prognosis. Surgery is the treatment of choice. However, the presence of multiple or large tumors can make the procedure challenging. Electrochemotherapy (ECT) is an effective local treatment, suitable for managing cutaneous or subcutaneous malignant tumors. This case series describes the first report of the use of ECT in the treatment of four cats with MCT.

Feline histiocytic diseases are uncommon and rarely reported. Feline progressive histiocytosis (FPH) is the most common histiocytic disease in cats, predominantly affecting middle-aged animals. The most common presentation is the cutaneous form with solitary or multiple cutaneous nodules. A female, mixed-breed 6-year-old cat was presented with a 9-month history of a nodule in the nasal planum and was diagnosed by histopathology with histiocytic proliferation. At the time of diagnosis, new nodules were discovered on the lower lip, digit, and two lesions in the tail region, with the largest meas

Electrochemotherapy is an anticancer treatment based on applying electric field pulses that reduce cell membrane selectivity, allowing chemotherapy drugs to enter the cells. In parallel to electrochemotherapy clinical tests, in silico experiments have helped scientists and clinicians to understand the electric field distribution through anatomically complex regions of the body. In particular, these in silico experiments allow clinicians to predict problems that may arise in treatment effectiveness. The current work presents a metastatic case of a mast cell tumor in a dog. In this specific trea

In cats, the high incidence of malignant mammary neoplasms with elevated rates of metastasis and recurrence underscores the need for more effective therapies to prevent disease progression. This preliminary study evaluated the impact of carboplatin or bleomycin electrochemotherapy on recurrence rates, adverse effects, and overall survival in female cats with mammary carcinoma undergoing mastectomy. Thirty-four female cats with mammary carcinoma were assigned to four groups: Group 1 (mastectomy), Group 2 (mastectomy plus chemotherapy), Group 3 (mastectomy plus electrochemotherapy with carboplat

We describe a novel lab-on-a-chip microsystem for cells electroporation, which can be used in studies comparing the effectiveness of two anti-cancer procedures. Our device in which we can simultaneously test the effect of a drug and electrically stimulate cells can be used to assess the effectiveness of chemo- and electrochemotherapy. The microsystem comprised two pairs of indium tin oxide electrodes that were stacked parallel to the culture microchambers. It enables the observation and performance of tests on adherent cells at the same time for 4 different test samples in 6 repetitions (contr

Cutaneous metastases from breast carcinoma represent a debilitating complication of advanced disease progression, significantly impacting patients' quality of life. Electrochemotherapy (ECT), which combines cytotoxic drugs such as bleomycin or cisplatin with electric pulses to enhance cellular permeability, has shown promising efficacy as a local treatment for these lesions. Objectives: This literature review examines the principles of ECT, its mechanisms of action and the clinical outcomes in patients with primary breast cancer. Across clinical series, patient-level ORR typically ranges from

Electrochemotherapy has been proven to be an efficient treatment for cutaneous metastases of various cancers. Data on breast cancer (BC) patients with cutaneous metastases were retrieved from the INSPECT database. Patients were divided by their receptor status: HER2+, HR+ (ER/PgR+), and TN (triple negative). Groups were similar for histological subtype and location of the nodules. Most patients were previously treated with surgery/systemic therapy/radiotherapy. We found no differences in the three groups in terms of response ratio (OR per patient 86% HER2+, 80% HR+, 76% TN, p = 0.8664). The on

BACKGROUND: The effectiveness of electrochemotherapy of tumors (ECT) and of irreversible electroporation ablation (IRE) depends on different mechanisms and delivery protocols. Both therapies exploit the phenomenon of electroporation of the cell membrane achieved by the exposure of the cells to a series of high-voltage electric pulses. Electroporation can be fine-tuned to be either reversible or irreversible, causing the cells to either survive the exposure (in ECT) or not (in IRE), respectively. For treatment of tissues located close to the heart (e.g., in the liver), the safety of electropora

BACKGROUND: Radiotherapy-associated angiosarcoma (RAA) of the breast is a rare complication of radiotherapy, which is often difficult to identify and has poor prognosis. It usually presents as violaceous skin, erythema or rapidly growing palpable firm mass that can be confused with other benign skin lesions.
PATIENTS AND METHODS: After reviewing the literature, we found only four cases with RAA after mastectomy and autologous breast reconstruction. The presented case is the first that was treated by electrochemotherapy. The patient presented with secondary angiosarcoma of the breast five year

BACKGROUND: Approximately 200,000 new cases of melanoma are diagnosed worldwide each year. Skin metastases are a frequent event, occurring in 18.2% of cases. This can be distressing for the patient, as the number and size of cutaneous lesions increases, often worsened by ulceration, bleeding and pain. Electrochemotherapy (ECT) is a local modality for the treatment of cutaneous or subcutaneous tumors that allows delivery of low- and non-permeant drugs into cells. ECT has been used in palliative management of metastatic melanoma to improve patients' quality of life. This is, to our knowledge, th

OBJECTIVES: Electrochemotherapy (ECT) is a novel therapeutic option for the treatment of cutaneous and subcutaneous metastases of malignant melanoma. During the treatment, electric pulses are applied to tumor nodules to deliver nonpermeant or poorly permeant chemotherapeutic agents into the cells, increasing local cytotoxicity of anticancer drugs. We compared the clinical effectiveness of ECT as an alternative palliative treatment option for unresectable metastatic lesions of malignant melanoma with a systematic review of reported outcomes.

BACKGROUND: Electroporation is a process that causes a transient increase in the permeability of cell membranes. It can be used to increase the intracellular concentration of chemotherapeutic agents in tumor cells (electrochemotherapy; ECT). A clinical study was initiated to determine if this mode of treatment would be effective against certain primary and metastatic cutaneous malignancies. A group of six patients, three with malignant melanoma, two with basal cell carcinoma, and one with metastatic adenocarcinoma, were enrolled in the study. the treatment was administered in a two-step proces
