Emerging role of miRNAs in the regulation of ferroptosis.

Ferroptosis is a kind of cell death which has distinctive features differentiating it from autophagy, necrosis and apoptosis. This iron-dependent form of cell death is described by an increase in lipid reactive oxygen species, shrinkage of mitochondria and decrease in mitochondrial cristae. Ferroptosis is involved in the initiation and progression of many diseases and is regarded as a hotspot of investigations on treatment of disorders. Recent studies have shown that microRNAs partake in the regulation of ferroptosis. The impact of microRNAs on this process has been verified in different cance

Ferroptosis: a new strategy for cardiovascular disease.

Cardiovascular disease (CVD) is currently one of the prevalent causes of human death. Iron is one of the essential trace elements in the human body and a vital component of living tissues. All organ systems require iron for various metabolic processes, including myocardial and skeletal muscle metabolism, erythropoiesis, mitochondrial function, and oxygen transport. Its deficiency or excess in the human body remains one of the nutritional problems worldwide. The total amount of iron in a normal human body is about 3-5 g. Iron deficiency may cause symptoms such as general fatigue, pica, and nerv

Ferroptosis as a promising therapeutic strategy for melanoma.

Malignant melanoma (MM) is the most common and deadliest type of skin cancer and is associated with high mortality rates across all races and ethnicities. Although present treatment options combined with surgery provide short-term clinical benefit in patients and early diagnosis of non-metastatic MM significantly increases the probability of survival, no efficacious treatments are available for MM. The etiology and pathogenesis of MM are complex. Acquired drug resistance is associated with a pool prognosis in patients with advanced-stage MM. Thus, these patients require new therapeutic strateg

Multifaceted role of ferroptosis in cardiovascular disease.

Ferroptosis is a newly identified form of non-apoptotic cell death characterised primarily by iron-dependent lipid peroxidation. It differs morphologically, biochemically, and genetically from other forms of cell death, such as apoptosis, autophagy, and necrosis. Although the molecular mechanism underlying ferroptosis remains unclear, multiple biological processes, such as iron metabolism, lipid peroxides, and systems, such as the glutathione system and the tetrahydrobiopterin/coenzyme Q10 system, appear to be involved. While the contribution of ferroptotic mechanisms to human diseases is not

Novel Insight into Ferroptosis in Kidney Diseases.

BACKGROUND: Various kidney diseases such as acute kidney injury, chronic kidney disease, polycystic kidney disease, renal cancer, and kidney stones, are an important part of the global burden, bringing a huge economic burden to people around the world. Ferroptosis is a type of nonapoptotic iron-dependent cell death caused by the excess of iron-dependent lipid peroxides and accompanied by abnormal iron metabolism and oxidative stress. Over the past few decades, several studies have shown that ferroptosis is associated with many types of kidney diseases. Studying the mechanism of ferroptosis and

Ferroptosis and its Role in Gastric Cancer.

Gastric cancer (GC) is the fifth most common cancer and the third leading cause of cancer-related deaths worldwide. Currently, surgery is the treatment of choice for GC. However, the associated expenses and post-surgical pain impose a huge burden on these patients. Furthermore, disease recurrence is also very common in GC patients, thus necessitating the discovery and development of other potential treatment options. A growing body of knowledge about ferroptosis in different cancer types provides a new perspective in cancer therapeutics. Ferroptosis is an iron-dependent form of cell death. It

Review of the Role of Ferroptosis in Testicular Function.

Iron is an important metal element involved in the regulation of male reproductive functions and has dual effects on testicular tissue. A moderate iron content is necessary to maintain testosterone synthesis and spermatogenesis. Iron overload can lead to male reproductive dysfunction by triggering testicular oxidative stress, lipid peroxidation, and even testicular ferroptosis. Ferroptosis is an iron-dependent form of cell death that is characterized by iron overload, lipid peroxidation, mitochondrial damage, and glutathione peroxidase depletion. This review summarizes the regulatory mechanism

Ferritinophagy and ferroptosis in the management of metabolic diseases.

Ferroptosis is a form of regulated cell death modality associated with disturbed iron-homeostasis and unrestricted lipid peroxidation. Ample evidence has depicted an essential role for ferroptosis as either the cause or consequence for human diseases, denoting the likely therapeutic promises for targeting ferroptosis in the preservation of human health. Ferritinophagy, a selective form of autophagy, contributes to the initiation of ferroptosis through degradation of ferritin, which triggers labile iron overload (IO), lipid peroxidation, membrane damage, and cell death. In this review, we will

Emerging roles of ferroptosis in liver pathophysiology.

Ferroptosis is a widely recognized process of regulated cell death linking redox state, metabolism, and human health. It is considered a defense mechanism against extensive lipid peroxidation, a complex process that may disrupt the membrane integrity, eventually leading to toxic cellular injury. Ferroptosis is controlled by iron, reactive oxygen species, and polyunsaturated fatty acids. Accumulating evidence has addressed that ferroptosis plays an unneglectable role in regulating the development and progression of multiple pathologies of the liver, including hepatocellular carcinoma, liver fib

Ferroptosis: An emerging therapeutic opportunity for cancer.

Ferroptosis, a new form of non-apoptotic, regulated cell death characterized by iron dependency and lipid peroxidation, is involved in many pathological conditions such as neurodegenerative diseases, heart ischemia/reperfusion injury, acute renal failure, and cancer. While metabolic dysfunctions can lead to excessive lipid peroxidation culminating in ferroptotic cell death, glutathione peroxidase 4 (GPX4) resides in the center of a network that functions to prevent lipid hydroperoxides from accumulation, thereby suppressing ferroptosis. Indeed, RSL3 and other small-molecule GPX4 inhibitors can

Ferroptosis as a target for protection against cardiomyopathy.

Heart disease is the leading cause of death worldwide. A key pathogenic factor in the development of lethal heart failure is loss of terminally differentiated cardiomyocytes. However, mechanisms of cardiomyocyte death remain unclear. Here, we discovered and demonstrated that ferroptosis, a programmed iron-dependent cell death, as a mechanism in murine models of doxorubicin (DOX)- and ischemia/reperfusion (I/R)-induced cardiomyopathy. In canonical apoptosis and/or necroptosis-defective Ripk3-/-, Mlkl-/-, or Fadd-/-Mlkl-/- mice, DOX-treated cardiomyocytes showed features of typical ferroptotic c

Ferroptosis-inflammasome crosstalk contributes to hyperglycemia-induced neuronal injury and is modulated by the ferroptosis inhibitor UAMC-3203.

BACKGROUND: Hyperglycemia associated with diabetes and its neurological complications is strongly linked to oxidative stress and chronic inflammation; however, the contribution of ferroptosis to this process remains incompletely understood. Ferroptosis is an iron-dependent form of regulated cell death characterized by lipid peroxidation and glutathione depletion and has recently been implicated in neurodegenerative disorders and diabetic neuropathy. Nevertheless, the interaction between ferroptosis and inflammasome signaling in hyperglycemia-induced neuronal injury remains unclear. This study

A ferroptosis-related lncRNAs risk score model identifies LINC02178 as a potential regulator of ferroptosis sensitivity in cervical cancer.

BACKGROUND: Cervical cancer remains a major global health burden among women. Persistent infection with high-risk human papillomavirus (HR-HPV) is the primary driver of cervical carcinogenesis, yet additional cellular regulatory processes are required for tumor progression. Ferroptosis, an iron-dependent form of regulated cell death, has been implicated in cancer metabolism and therapeutic response. However, the involvement of ferroptosis-related long non-coding RNAs (FRLs) in human papillomavirus (HPV)-associated cervical cancer remains insufficiently understood. METHODS: Transcriptomic and

Deeper insight into ferroptosis: association with Alzheimer's, Parkinson's disease, and brain tumors and their possible treatment by nanomaterials induced ferroptosis.

Ferroptosis is an emerging and novel type of iron-dependent programmed cell death which is mainly caused by the excessive deposition of free intracellular iron in the brain cells. This deposited free iron exerts a ferroptosis pathway, resulting in lipid peroxidation (LiPr). There are mainly three ferroptosis pathways viz. iron metabolism-mediated cysteine/glutamate, and LiPr-mediated. Iron is required by the brain as a redox metal for several physiological activities. Due to the iron homeostasis balance disruption, the brain gets adversely affected which further causes neurodegenerative diseas

Fusobacterium nucleatum-induced endothelial ferroptosis promotes atherosclerosis.

OBJECTIVE: This study aimed to investigate whether Fusobacterium nucleatum (Fn), a key periodontal pathogen, induces ferroptosis in vascular endothelial cells and to evaluate its role in Fn-associated atherosclerosis progression. DESIGN: Human umbilical vein endothelial cells (HUVECs) were exposed to Fn in the presence or absence of ferroptosis inhibitors. Oxidative stress, mitochondrial function, ferroptosis-related markers, and endothelial function were systematically assessed. An apolipoprotein E knockout (ApoE⁻/⁻) mouse model was used to evaluate the effects of Fn infection and Fer-1 trea

Leukocyte-Hitchhiking Nanomedicine for Sensitized Ferroptosis Therapy.

Effective drug aggregation at tumor sites is crucial for improving outcomes in triple-negative breast cancer. However, multiple biological barriers within the tumor microenvironment limit the clinical utility, efficacy, and safety of current therapeutics, leading to treatment failure and cancer recurrence. In this study, we develop an in vivo strategy to construct a cell-mediated nanomedicine in which E-selectin-modified liposomes are intravenously injected and selectively adhere to leukocyte surfaces. These leukocytes then transport the liposome aggregates to tumor sites via inflammatory trop

Stress granules restrain ferroptosis by sequestering ferritin.

Glioblastoma stem cells (GSCs) are refractory to first-line treatment in the clinic, which includes irradiation (IR) and temozolomide (TMZ). Here we find that disrupting stress granules (SGs) sensitizes GSCs to IR/TMZ through ferroptosis. The profiling of SG proteins reveals the recruitment of iron-related proteins including ferritin. Mechanistically, G3BP1, an SG core protein, directly interacts with ferritin light chain in an IR/TMZ-induced G3BP1 methionine-333 oxidation-dependent manner. This interaction facilitates recruiting and sequestering ferritin into SGs, thereby restricting ferropto

Turning perfusion into repair through ferroptosis blockade.

By identifying graft ferroptosis as a druggable vulnerability, Veeckmans et al. show that FXT-001 intercepts lipid-radical injury during machine perfusion and improves liver and lung graft function. This work supports rather than proves machine perfusion as a therapeutic window for preimplantation graft repair, a concept that now requires postimplantation validation.
