β-Hydroxy- and β-Aminophosphonate Acyclonucleosides as Potent Inhibitors of Plasmodium falciparum Growth.

Malaria is an infectious disease caused by a parasite of the genus Plasmodium, and the emergence of parasites resistant to all current antimalarial drugs highlights the urgency of having new classes of molecules. We developed an effective method for the synthesis of a series of β-modified acyclonucleoside phosphonate (ANP) derivatives, using commercially available and inexpensive materials (i.e., aspartic acid and purine heterocycles). Their biological evaluation in cell culture experiments and SAR revealed that the compounds' effectiveness depends on the presence of a hydroxyl group, the chai

Epidermal growth factor receptor inhibitors: a review of cutaneous adverse events and management.

Epidermal growth factor inhibitors (EGFRI), the first targeted cancer therapy, are currently an essential treatment for many advance-stage epithelial cancers. These agents have the superior ability to target cancers cells and better safety profile compared to conventional chemotherapies. However, cutaneous adverse events are common due to the interference of epidermal growth factor receptor (EGFR) signaling in the skin. Cutaneous toxicities lead to poor compliance, drug cessation, and psychosocial discomfort. This paper summarizes the current knowledge concerning the presentation and managemen

[Therapy with epidermal growth factor receptor inhibitors. Clinical spectrum of cutaneous adverse effects].

Recently, inhibitors of the epidermal growth factor receptor (EGFR), such as erlotinib, gefitinib, cetuximab or panitumumab, have been successfully established in the therapy of a variety of solid tumors. Cutaneous adverse effects are the most frequent side-effects of these so-called targeted cancer drugs and occur in 45-100% of patients. In addition to a characteristic papulo-pustular rash, adverse effects include painful paronychia, xerosis cutis, pruritus, alopecia or alterations of the hair structure. These often stigmatizing side-effects represent a serious threat to the patients' quality

Influence of cardiometabolic medications on abdominal aortic aneurysm growth in the UK Aneurysm Growth Study: metformin and angiotensin-converting enzyme inhibitors associated with slower aneurysm growth.

BACKGROUND: There is a clinical need for treatments that can slow or prevent the growth of an abdominal aortic aneurysm, not only to reduce the need for surgery, but to provide a means to treat those who cannot undergo surgery. METHODS: Analysis of the UK Aneurysm Growth Study (UKAGS) prospective cohort was conducted to test for an association between cardiometabolic medications and the growth of an abdominal aortic aneurysm above 30 mm in diameter, using linear mixed-effect models. RESULTS: A total of 3670 male participants with data available on abdominal aortic aneurysm growth, smoking st

Direct thrombin inhibitors as alternatives to heparin to preserve lung growth and function in a murine model of compensatory lung growth.

Infants with congenital diaphragmatic hernia (CDH) may require cardiopulmonary bypass and systemic anticoagulation. Expeditious lung growth while on bypass is essential for survival. Previously, we demonstrated that heparin impairs lung growth and function in a murine model of compensatory lung growth (CLG). We investigated the effects of the direct thrombin inhibitors (DTIs) bivalirudin and argatroban. In vitro assays of lung endothelial cell proliferation and apoptosis were performed. C57BL/6 J mice underwent left pneumonectomy and subcutaneous implantation of osmotic pumps. Pumps were pre-l

Determination of Chemical Inhibitor Efficiency against Intracellular Toxoplasma Gondii Growth Using a Luciferase-Based Growth Assay.

Toxoplasma gondii is a protozoan pathogen that widely affects the human population. The current antibiotics used for treating clinical toxoplasmosis are limited. In addition, they exhibit adverse side effects in certain groups of people. Therefore, discovery of novel therapeutics for clinical toxoplasmosis is imperative. The first step of novel antibiotic development is to identify chemical compounds showing high efficacy in inhibition of parasite growth using a high throughput screening strategy. As an obligate intracellular pathogen, Toxoplasma can only replicate within host cells, which pro

KRN633, an inhibitor of vascular endothelial growth factor receptor tyrosine kinase, induces intrauterine growth restriction in mice.

We previously reported that treatment with KRN633, a vascular endothelial growth factor receptor tyrosine kinase inhibitor, during mid-pregnancy caused intrauterine growth restriction resulting from impairment of blood vessel growth in the labyrinthine zone of the placenta and fetal organs. However, the relative sensitivities of blood vessels in the placenta and fetal organs to vascular endothelial growth factor (VEGF) inhibitors have not been determined. In this study, we aimed to examine the effects of KRN633 on the vasculatures of organs in mother mice and their newborn pups by immunohistoc

Alternatively spliced vascular endothelial growth factor receptor-2 is an essential endogenous inhibitor of lymphatic vessel growth.

Disruption of the precise balance of positive and negative molecular regulators of blood and lymphatic vessel growth can lead to myriad diseases. Although dozens of natural inhibitors of hemangiogenesis have been identified, an endogenous selective inhibitor of lymphatic vessel growth has not to our knowledge been previously described. We report the existence of a splice variant of the gene encoding vascular endothelial growth factor receptor-2 (Vegfr-2) that encodes a secreted form of the protein, designated soluble Vegfr-2 (sVegfr-2), that inhibits developmental and reparative lymphangiogene

Cutaneous adverse effects of combination epidermal growth factor receptor inhibitor and immune checkpoint inhibitor cancer therapy.

PURPOSE: Epidermal growth factor receptor inhibitors (EGFRIs) and immune checkpoint inhibitors (ICIs) show promising efficacy in colorectal cancer and head and neck squamous cell carcinoma therapy, especially in combination. However, given the many cutaneous adverse effects (CAE) associated with each, there is concern for overlap and exacerbation of toxicities. EGFRIs are associated with acneiform rashes and paronychia, while ICIs are associated with lichenoid dermatitis and psoriasis. Xerosis, pruritus, and eczema are seen with both drugs. Understanding the CAEs of EGFRI-ICI combination thera

A selective NVL inhibitor suppresses tumor growth by targeting ribosome biogenesis.

Ribosome biogenesis is highly upregulated in cancer and essential for malignant growth, yet efforts to selectively target this process pharmacologically have been unsuccessful. Here we identify nuclear valosin-containing protein-like (NVL), a hexameric AAA+ ATPase required for large ribosomal subunit (60S) assembly, as a druggable target in this pathway. Using forward genetics, we discovered that the cytotoxic dibenzothiazepinone MM17 inhibits NVL. Mutations conferring resistance to MM17 in cancer cells also rendered recombinant NVL refractory to enzymatic inhibition in vitro, providing conver

Synthesis and mechanistic studies of a novel homoisoflavanone inhibitor of endothelial cell growth.

Preventing pathological ocular angiogenesis is key to treating retinopathy of prematurity, diabetic retinopathy and age-related macular degeneration. At present there is no small molecule drug on the market to target this process and hence there is a pressing need for developing novel small molecules that can replace or complement the present surgical and biologic therapies for these neovascular eye diseases. Previously, an antiangiogenic homoisoflavanone was isolated from the bulb of a medicinal orchid, Cremastra appendiculata. In this study, we present the synthesis of a novel homoisoflavano

Chemical synthesis and biological evaluation of an antimicrobial peptide gonococcal growth inhibitor.

Gonococcal growth inhibitor 1 (GGI-1) is a 44-residue peptide with potent anti-Legionella activity. It has been isolated from Staphylococcus haemolyticus but, to date, its chemical synthesis has not been reported. Acquisition of this peptide via this means would enable a more detailed examination of its antimicrobial properties. However, its synthesis represents a significant challenge because of two predicted "difficult sequences" within the peptide. Its successful solid-phase assembly is reported in this paper, and was accomplished by use of simple palliative measures including the introduct

The AG1478 tyrosine kinase inhibitor is an effective suppressor of leiomyoma cell growth.

BACKGROUND: Uterine leiomyomas are the most common benign smooth muscle cell tumours in women. Formation of leiomyomas, still not completely understood, is viewed as a multistep process, with involvement of ovarian steroid hormones, cytokines and growth factors. Our study aimed to identify tyrosine kinase inhibitors as potential 'signal transduction therapeutics' for leiomyomas, underlying the effect of ovarian steroidal hormones. METHODS: The selective epidermal growth factor (EGF) receptor blocker AG1478 was evaluated as a potential target, since EGF has been shown to mediate estrogen actio

The anthocyanidins cyanidin and delphinidin are potent inhibitors of the epidermal growth-factor receptor.

The aglycons of the most abundant anthocyanins in food, cyanidin (cy) and delphinidin (del), were found to inhibit the growth of human tumor cells in vitro in the micromolar range, whereas malvidin (mv), a typical anthocyanidin in grapes, was less active. The aglycons preferentially inhibited the growth of the human vulva carcinoma cell line A431, overexpressing the epidermal growth-factor receptor (EGFR). The glycosides cyanidin-3-beta-D-galactoside (cy-3-gal, idaein) and malvidin-3-beta-D-glucoside (mv-3-glc, oenin) did not affect tumor cell growth up to 100 microM. The tyrosine kinase activ

Adenine nucleotide levels in Neurospora, as influenced by conditions of growth and by metabolic inhibitors.

By a combination of luciferase and fluorescence methods adenine nucleotide pools in Neurospora crassa have been examined under various conditions of growth and metabolic inhibition. During sustained exponential growth (25 C, shaking liquid cultures), the intracellular adenosine 5'-triphosphate (ATP) concentration, [ATP](i), rises slowly from the conidial level near 1 mM (1 mmol/kg of cell water) to a maximum of 2.0 to 2.5 mM at 14 h, after which it slowly declines. The adenosine 5'-diphosphate and adenosine 5'-monophosphate (AMP) curves show two peaks, at 8 and 20 h, with a minimum at 16 h. Th

Novel NOTCH1-unmutated T-ALL Cell Line With Suppressed Growth by Gamma-secretase Inhibitors.

BACKGROUND/AIM: T-cell acute lymphoblastic leukemia (T-ALL) frequently involves the activation of NOTCH1 signaling. However, effective NOTCH-directed therapies remain elusive. We established and characterized a novel T-ALL cell line, TMD11, lacking NOTCH mutations but sensitive to γ-secretase inhibitors (GSIs), providing a tool for discovering novel molecular targeted therapies. MATERIALS AND METHODS: TMD11 cells were established by long-term proliferation of leukemia cells from a patient with T-ALL without adding growth factors. Immunoblotting, whole-exome sequencing, and RNA sequencing were

Epidermal Growth Factor Receptor and Tyrosine Kinase Inhibitors: Current and Emerging Strategies in Glioblastoma.

Epidermal growth factor receptor (EGFR) is the most frequent alteration in isocitrate dehydrogenase - wild-type glioblastoma and has been considered a promising therapeutic target. Despite extensive investigation, EGFR-targeted therapies have not improved overall survival because of limited central nervous system drug delivery, tumor heterogeneity, adaptive resistance, and an evolving tumor microenvironment. This review summarizes the molecular biology of EGFR, clinical experience with EGFR-targeted therapies, mechanisms underlying therapeutic resistance, and emerging strategies that may impro
