Histamine H1 receptor antagonists selectively kill cisplatin-resistant human cancer cells.

Cancer therapy is often hampered by the disease's development of resistance to anticancer drugs. We previously showed that the autonomously upregulated product of fibroblast growth factor 13 gene (FGF13; also known as FGF homologous factor 2 (FHF2)) is responsible for the cisplatin resistance of HeLa cisR cells and that it is likely responsible for the poor prognosis of cervical cancer patients treated with cisplatin. Here we show that cloperastine and two other histamine H1 receptor antagonists selectively kill HeLa cisR cells at concentrations that little affect parental HeLa S cells. The se

Pyrrolidin-3-yl-N-methylbenzamides as potent histamine 3 receptor antagonists.

On the basis of the previously reported benzimidazole 1,3'-bipyrrolidine benzamides (1), a series of related pyrrolidin-3-yl-N-methylbenzamides were synthesized and evaluated as H(3) receptor antagonists. In particular, compound 32 exhibits potent H(3) receptor binding affinity, improved pharmaceutical properties and a favorable in vivo profile.

Bismuth increases hydroxyl radical-scavenging activity of histamine H2-receptor antagonists.

The effects of histamine H2-receptor antagonists, alone or in a combination with bismuth, on *OH-provoked degradation of deoxyribose were studied. The histamine H2-receptor antagonists (cimetidine, ranitidine and roxatidine), themselves decreased the deoxyribose damage in Fenton-type systems. In combinations with bismuth, their inhibitory effect in Fenton system (Fe(III)/ascorbic acid + H2O2 was stronger. Moreover, unlike F(III) and Cu(II), which in the presence of ascorbic acid + H2O2 led to an increase in the *OH formation (deoxyribose damage), Bi(III) showed an opposite effect. The present

Radioprotective properties of histamine H2 receptor antagonists: present and future prospects.

Various chemical agents were examined for their radioprotective capability to provide partial protection against radiation injury over the past 50 years. However, no suitable drug has yet been introduced for routine clinical use. In the present study, the radioprotective potential of H2 receptor antagonists was examined in in vivo and in vitro conditions. For this purpose, an in vivo micronucleus assay and an in vitro metaphase analysis were used to test the effects of cimetidine, ranitidine, and famotidine on radiation-induced clastogenic effects. For micronuclei assay, BALB/c mice were irrad

Synthesis and assessment of formamidines as new histamine H2-receptor antagonists.

Four series of compounds whose substructure contains a formamidine functionalized as a novel group in the chemistry of histamine H2-receptors have been synthesized. Series design, synthesis and pharmacological data including inhibition of histamine-stimulated acid secretion, inhibition of acid secretion p.o. and pA2 are reported. N-[(E)-[[2-[[[2](Diaminomethylene)amino]-4-thiazolyl] methyl]thio]ethyl]amino]methylene]-4-bromo-benzenesulfonamide (ebrotidine, CAS 100981-43-9, FI-3542) was selected for further research.

Synthesis and activity of pyridine analogues of histamine H2-receptor antagonists.

Derivatives of 2-amino-3-nitropyridine (4; see Table I for structures) and 2-amino-3-nitro-1H-pyrid-6-one (5), conceived as cyclic 'urea equivalents' of histamine H2-receptor antagonists, were prepared. Pharmacological testing, using the histamine-induced guinea pig atrial chronotropic response, indicated that the R1-pyridyl substituent was a determinant of activity, since the relative activities within an isomeric series were 4-pyridyl greater than 3- and 2-pyridyl. In comparison with analogous 2-amino-3-nitropyridines. (4), 2-amino-3-nitro-1H-pyrid-6-ones (5) had a substantially less histami

Effects of intraventricular histamine and H2 receptor antagonists on intraocular pressure.

Severe ocular hypertension has been reported in a chronic glaucoma patient following use of histamine H2 receptor antagonists for treatment of peptic ulcer. Subsequent studies, however, have failed to demonstrate a significant action of topical or intravenously administered H2 blockers on intraocular pressure (IOP) in humans. In this study, cimetidine and ranitidine were administered into the cerebral ventricles of unanesthetized New Zealand White rabbits. Both drugs caused prolonged increases in IOP at a dose of 1 umol. Maximal elevations of IOP occurred approximately 20 min after drug inject

Role of histamine (H2)-receptor antagonists in total parenteral nutrition patients.

Hospitalized patients, especially those who are critically ill, are often at risk for malnutrition and may require nutritional support. Although the enteral approach is generally preferred when providing nutritional support, parenteral nutrition is frequently required. These patients are also likely to receive therapeutic or prophylactic parenteral histamine (H2)-receptor antagonists. Patients receiving both total parenteral nutrition and parenteral H2-receptor antagonists may benefit from receiving the drug in combination with the total parenteral nutrition admixture. Parenteral cimetidine an

Recent advances in pharmaceutical chemistry--review II. Histamine H2-receptor antagonists.

The concept of histamine receptors is outlined and the rationale for the synthesis of H2-antagonists presented. Structure-activity relationships among these compounds are described and aspects of absorption, distribution and elimination discussed with particular reference to cimetidine and ranitidine. Oxmetidine, lupitidine and loxtidine are also considered. Methods for the analysis of these drugs in body fluids are presented followed by a discussion of their toxicology. Volunteer and patient studies are also surveyed.

Pseudosymmetry and bioisosterism in biaryl pyridyl competitive histamine H2-receptor antagonists.

A process of drug design has previously been described that led to the synthesis of 3-amino-5-[2-(ethylamino)-4-pyridyl]-1,2,4-triazole (4), a competitive histamine H2-receptor antagonist structurally unrelated to, but more potent than, cimetidine. A QSAR study on a subset of analogues closely related to 4 showed that gastric acid antisecretory activity increased with decreasing lipophilicity. An SAR study about 4 focused on (1) pyridine substitution compatible with both unidentate and bidentate hydrogen bonding, (2) exploration of the pseudosymmetry of 4, and (3) examination of triazole and i

The effect of H1 and H2 histamine antagonists on symptomatic dermographism.

In ten patients suffering from symptomatic dermographism the combined administration of chlorpheniramine + cimetidine produced a greater reduction in the weal and flare response provoked by a standardized scratch than the administration of chlorpheniramine alone. There was a statistically significant improvement in the overall assessment of the patient's skin condition with the combined administration of chlorpheniramine + cimetidine. Chlorpheniramine given alone produced no significant benefit whilst cimetidine alone produced a marked exacerbation in itching in nearly half the patients who in

The efficacy of histamine antagonists as antipruritics in experimentally induced pruritus.

1. The antipruritic ability of histamine (H1 and H2) antagonists alone and in combination in experimentally induced pruritus has been investigated in 12 normal human volunteers. 2. A combination of cimetidine (H2 anatagonist) and chlorpheniramine (H1 antagonist) proved effective in suppressing itch that was artificially induced by the application of papain and histamine. 3. The combination was more effective than chlorpheniramine or cimetidine alone or placebo. 4. The results suggest that both H1 and H2 receptors are involved in the mediation of pruritus.

Characterization and development of cimetidine as a histamine H2-receptor antagonist.

The concept of two classes of histamine receptor, H1 and H2, is introduced and the chemical derivation of histamine H2-receptor antagonists is outlined briefly. Starting from the structure of histamine, chemical modification led eventually to burimamide, the first described histamine H2-receptor antagonist. Further stepwise modifications ultimately afforded metiamide and cimetidine. In vitro studies show that cimetidine is a specific competitive histamine H2-receptor antagonist. In vivo, it is a potent inhibitor of histamine-stimulated gastric acid secretion in rats and dogs after both intrave
