Axenic cultures of Phytomonas sp. were obtained from naturally infected tomatoes and from Phthia picta, a predator of tomato plants, by using a biphasic medium with Roitman's complex medium overlaying rabbit blood-agar slants. Light and electron microscopy of both isolates showed a similarity of morphological characteristics among the flagellates in fresh material or after cultivation. Other properties, including their agglutinability with the haemolymph of Phthia picta, suggest that these isolates are virtually identical.

Synthetic insect repellents, IR3535 and KBR 3023 (also known as picaridin, or by the trade name Bayrepel, were tested in Burkina Faso against mosquito vectors of disease to compare their relative efficacy and persistence profiles to those of the 'gold standard' DEET. Collection of >49000 mosquitoes (approximately 95% belonging to the Anopheles gambiae complex) showed that after an exposure of 10h, KBR 3023 produced the highest protection against anophelines, followed by DEET, then IR3535. The response of aedines was more variable. By fitting a logistic plane model we estimated 95% effective do

This systematic review and meta-analysis aims to: (1) characterize the distribution of published inhibition of emergence (IE50, IE90, and IE95) reference values for pyriproxyfen and methoprene in Culex pipiens [L.], Aedes aegypti [L.], and Aedes albopictus [Skuse]; (2) generate combined-effect IE values using a DerSimonian and Laird (DL) random-effects model to establish benchmarks for future resistance assessments; and (3) compare these combined-effect IE values with previously published literature. A systematic search was conducted in PubMed, SciELO, J-STAGE, and Google Scholar up to 10 Febr

The causative agent for Chagas disease, Trypanosoma cruzi, is transmitted to a human host in the urine/feces of the kissing bug, Rhodnius prolixus, following blood feeding. Kinins are important chemical messengers in the overall control of blood feeding physiology in R. prolixus, including hindgut contractions and excretion. Thus, disruption in kinin signaling would have damaging consequences to the insect but also interfere with the transmission of Chagas Disease. Here, a heterologous functional receptor assay was used to confirm the validity of the previously cloned putative kinin G-protein-

BACKGROUND: Human African trypanosomiasis (HAT) is a neglected tropical disease caused by Trypanosoma brucei gambiense transmitted by tsetse flies in sub-Saharan West Africa. In southern Chad the most active and persistent focus is the Mandoul focus, with 98% of the reported human cases, and where African animal trypanosomosis (AAT) is also present. Recently, a control project to eliminate tsetse flies (Glossina fuscipes fuscipes) in this focus using the sterile insect technique (SIT) was initiated. However, the release of large numbers of sterile males of G. f. fuscipes might result in a pote

We established a laboratory colony of Culex pipiens mosquitoes from eggs collected in Colorado and discovered that mosquitoes in the colony are naturally infected with Culex flavivirus (CxFV), an insect-specific flavivirus. In this study we examined transmission dynamics of CxFV and effects of persistent CxFV infection on vector competence for West Nile virus (WNV). We found that vertical transmission is the primary mechanism for persistence of CxFV in Cx. pipiens, with venereal transmission potentially playing a minor role. Vector competence experiments indicated possible early suppression of

Vector control programmes in many countries face the dual problems of parasite drug resistance and insecticide resistance in the insect vectors of the disease. Here we report for the first time a new esterase-based insecticide resistance mechanism in the filariasis vector Culex quinquefasciatus. The field collected COL strain of C. quinquefasciatus from Columbia was heterogeneous for organophosphorus insecticide resistance. On native polyacrylamide gels it had an elevated beta-naphthyl acetate specific esterase with the same Rf as that for the Est beta 1s involved in insecticide resistance in

Lipophorin is the main lipoprotein in the hemolymph of insects. During vitellogenesis, lipophorin delivers its hydrophobic cargo to developing oocytes by its binding to non-endocytic receptors at the plasma membrane of the cells. In some species however, lipophorin may also be internalized to some extent, thus maximizing the storage of lipid resources in growing oocytes. The ectopic β chain of ATP synthase (β-ATPase) was recently described as a putative non-endocytic lipophorin receptor in the anterior midgut of the hematophagous insect Panstrongylus megistus. In the present work, females of t

Homalodisca vitripennis (Germar), a vector of Xylella fastidiosa, is associated with citrus plantings in California. Infested citrus orchards act as a source of vectors to adjacent vineyards where X. fastidiosa causes Pierce's disease. An analysis of the pattern and rate of movement of H. vitripennis and its egg parasitoid, Gonatocerus ashmeadi Girault, was conducted in a citrus orchard by using a protein mark-capture technique to quantify movement and net dispersal rates in the experimental areas. Treatments included irrigation at 100% of the crop evapotranspiration rate (ET(c)), 80, and 60%

Previous studies (1982, 1987) have emphasized the superiority of sylvatic vector species over domestic species as xenodiagnostic agents in testing hosts with acute or chronic infections by T. cruzi "Y" stock. The present study, which is unique in that it contains data on both infectivity rates produced by the same stock in 11 different vector species and also the reaction of the same vector species to seven different parasite stocks, establishes the general validity of linking efficiency of xenodiagnosis to the biotope of its agent. For example, infectivity rates produced by "São Felipe" stock

Curly top disease (CTD) of sugar beet is characterized by leaf curling and stunted growth. Historic CTD emerged independently in western North America (~1900), South America (~1925), and the Middle East (~1955), following intensive cultivation of this Old World crop. In these regions, CTD is caused by genetically distinct monopartite geminiviruses transmitted by different leafhopper species. In North America, CTD is caused by 11 strains of beet curly top virus (BCTV) and transmitted by the beet leafhopper (BLH, Circulifer tenellus). Here, the emergence and evolution of BCTV and the role of agr

Insects act as vectors to carry a wide range of bacteria and viruses that can cause multiple vector-borne diseases in humans. Diseases such as dengue fever, epidemic encephalitis B, and epidemic typhus, which pose serious risks to humans, can be transmitted by insects. Due to the absence of effective vaccines for most arbovirus, insect control was the main strategy for vector-borne diseases control. However, the rise of drug resistance in the vectors brings a great challenge to the prevention and control of vector-borne diseases. Therefore, finding an eco-friendly method for vector control is

Blood-sucking insects are important vectors of disease, with biting Diptera (flies) alone transmitting diseases that cause an estimated 700 000 human deaths a year. Insect vectors also bite nonhuman hosts, linking them into host-biting networks. While the major vectors of prominent diseases, such as malaria, yellow fever, dengue, and Zika, are intensively studied, there has been limited focus on the wider interactions of biting insects with nonhuman hosts. Drawing on network analysis and visualisation approaches from food-web ecology, we discuss the value of a network perspective for understan

Plant viruses depend on insect vectors for dissemination in complex agroecosystems and often exploit plant and insect physiology to enhance transmission. This review synthesizes recent advances in understanding cross-kingdom exchange of non-coding RNAs (ncRNAs) as a central mechanism shaping plant-virus-vector interactions and broader agroecosystem dynamics. ncRNAs are exchanged among plants, insect vectors, and viruses via the phloem, insect saliva, and external plant surfaces through exudates. These molecules can suppress plant antiviral and anti-insect plant responses, modulate vector biolo

The re-emergence of arboviral diseases such as Dengue Fever and La Crosse encephalitis is primarily due to the failure of insect vector control strategies. The development of a procedure capable of producing stable germ-line transformants in the insect vectors of these diseases would bridge the gap between gene expression systems being developed to curb vector transmission and the identification of important genes and regulatory sequences and their reintroduction back into the insect genome in the form of vector control strategies. The transposable element piggyBac is capable of transposition

In 1999 and 2006, two viral diseases emerged massively and unexpectedly in the United States (West Nile disease) and northern Europe (bluetongue disease). Control of infectious diseases transmitted by insect vectors is based on a variety of approaches (including sanitary measures), but primarily on vaccination. Vaccination is more efficient and less expensive than monitoring of insect vectors. The dynamics and epidemiology of two arboviral diseases (West Nile and bluetongue) are described, together with the different vaccines and vaccination methods.

Engineered underdominance is one of a number of different gene drive strategies that have been proposed for the genetic control of insect vectors of disease. Here we model a two-locus engineered underdominance based gene drive system that is based on the concept of mutually suppressing lethals. In such a system two genetic constructs are introduced, each possessing a lethal element and a suppressor of the lethal at the other locus. Specifically, we formulate and analyse a population genetics model of this system to assess when different combinations of release strategies (i.e. single or multip

Phloem-inhabiting unculturable bacterial pathogens are persistently transmitted by insect vectors. However, how they evade insect immune responses to ensure persistent transmission remains unknown. The important melanization immune response in insects is triggered by cleavage of prophenoloxidase (PPO) into active phenoloxidase (PO) via clip-domain serine proteases (CLIPs). Here, we demonstrate that infection of Candidatus Liberibacter asiaticus (CLas) in psyllid vectors activates the peptidoglycan recognition protein (PGRP)-CLIP1-CLIP4-PPO-PO signaling cascade to induce a mild melanization res

Vector-borne diseases continue to impose a major health burden on Peru and neighboring countries. The challenge of addressing vector-borne disease is compounded by changing social, economic, and climatic conditions. Peri-urban Arequipa is an important region to study insect infestations because of ongoing challenges with disease vectors such as triatomines and a variety of other insects. We conducted surveys (N = 1,182) and seven focus groups (average seven participants) in peri-urban Arequipa to explore knowledge of and perception toward various insects that infest the region. Focus group par
