Restoration of the Topological Organization of the Trigeminal System Following Trigeminal Nerve Root Injury in the Lamprey.

Two issues were examined regarding the trigeminal system in larval lampreys: (1) for normal animals, double labeling was used to confirm that the trigeminal system has a topological organization; (2) following trigeminal nerve root transections, double labeling was used to test whether the topological organization of the trigeminal system is restored. First, for normal animals, Alexa 488 dextran amine applied to the medial oral hood (anterior head) labeled trigeminal motoneurons (MNs) in the ventromedial part of the trigeminal motor nuclei (nVm) and axons of trigeminal sensory neurons (SNs) in

Molecular cloning and pharmacological characterization of two novel GnRH receptors in the lamprey (Petromyzon marinus).

This paper reports the identification, expression, binding kinetics, and functional studies of two novel type III lamprey GnRH receptors (lGnRH-R-2 and lGnRH-R-3) in the sea lamprey, a basal vertebrate. These novel GnRH receptors share the structural features and amino acid motifs common to other known gnathostome GnRH receptors. The ligand specificity and activation of intracellular signaling studies showed ligands lGnRH-II and -III induced an inositol phosphate (IP) response at lGnRH-R-2 and lGnRH-R-3, whereas the ligand lGnRH-I did not stimulate an IP response. lGnRH-II was a more potent ac

Validation and insights of anesthetic action in an early vertebrate network: the isolated lamprey spinal cord.

BACKGROUND: The lamprey spinal cord is a well-characterized vertebrate network that could facilitate our understanding of anesthetic action. We tested several hypotheses concerning the lamprey's clinical application to anesthesia, and the sites/mechanisms of anesthetic action. METHODS: In isolated lamprey spinal cords, minimum immobilizing concentrations (MICs) were determined for halothane, isoflurane, sevoflurane, desflurane, propofol, or the nonimmobilizer F6 (1,2-dichlorohexafluorocyclobutane), applied during D-glutamate-induced fictive swimming or noxious tail stimulation. Isoflurane and

Influence of synthetic lamprey GnRH-III on gonadotropin release and steroid hormone levels in gilts.

Based on the supposition that lamprey GnRH-III (lGnRH-III) elicits FSH releasing activity in swine, synthetic lGnRH-III (peforelin, Maprelin® XP10) was used in puberal estrus synchronized gilts. The secretion of reproductive hormones FSH, LH, estradiol and progesterone was analyzed, and follicle growth and ovulation recorded. Altogether, 24 German Landrace gilts were treated after an 18-day long synchronization of the estrus cycle with Regumate® as follows: 48 h after the last Regumate® feeding they received im either 150 μg Maprelin® XP10 (lGnRH-III, group Maprelin, n = 6), 50 μg Gonavet Veyx

Light- and temperature-dependence of the melatonin secretion rhythm in the pineal organ of the lamprey, Lampetra japonica.

To identify the characteristics of the oscillator located in the pineal organ, we examined the effects of temperature and light on melatonin secretion rhythm using pineal organs in cultures. At 20 degrees C, the melatonin rhythm was obvious: low secretion during the daytime and high during the nighttime. When the temperature was lowered from 20 to 10 degrees C, the melatonin rhythm disappeared. When the temperature was returned from 10 to 20 degrees C, the rhythm quickly reappeared. The plasma melatonin level was measured in living lampreys kept at 7 degrees C to establish the melatonin profil

The origin of trochlear motoneurons in the larval sea lamprey, Petromyzon marinus L. An HRP study.

The origin of trochlear motoneurons in larval lampreys was studied by injection of HRP into the orbit. Immature motoneurons had ventricular attachments, the position of which with respect to the ventricular sulci was used to study their regions of origin. Motoneurons originate from the region between the sulcus intermedius dorsalis and the sulcus intermedius ventralis, which in other parts of the brain were identified as the visceromotor and viscerosensory columns. Both ipsilateral and contralateral immature motoneurons were found. The significance of these findings is discussed in relation to

Morphology of the exocrine pancreas of the southern hemisphere lamprey, Geotria australis, and changes during metamorphosis.

The distribution and histology of zymogen cells and the activity of digestive enzymes have been examined in the alimentary canal of larval, metamorphosing (stages 1-7), and adult Geotria australis (Geotriidae). Comparisons of the arrangement of the larval and adult zymogen cells are made with those observed in Mordacia mordax, a representative of the other Southern Hemisphere lamprey family (Mordaciidae), and with those reported elsewhere for holarctic lampreys (Petromyzontidae). In larval G. australis, epithelial zymogen cells are mainly restricted to the prominent pair of tubular diverticula

Single sensory neurons activate excitatory amino acid receptors in the lamprey spinal cord.

The effects of excitatory amino acid (EAA) antagonists were tested on sensory afferent excitation in the lamprey spinal cord. Paired intracellular recordings were made from mechanosensory neurons (dorsal cells) and second order sensory neurons (giant interneurons). Stimulation of individual dorsal cells evoked mono- and/or polysynaptic excitatory postsynaptic potentials (EPSPs) in giant interneurons. These EPSPs were depressed by the EAA antagonists cis-2,3-piperidine dicarboxylate and kynurenic acid. Small components of the synaptic potentials were due to electrical coupling since they were n

LAMPrey: a standardised method for analysing quantitative LAMP reactions using the inflection cycle threshold.

Quantitative loop-mediated isothermal amplification (qLAMP) is a gene expression quantification method that has gained popularity in recent years, particularly in disease identification, including during the recent SARS-CoV-2 pandemic. Unlike conventional quantitative PCR (qPCR), qLAMP features reaction kinetics that may diverge from sigmoidal expectation, and may not include ROX dye in commercial kits. Determining cycle threshold (Ct or Cq) values through automatic thresholding may therefore produce inaccurate results, and the nature of these thresholds complicates comparability between studi

Lamprey 3D single-cell transcriptomics reveals ancestral and specialized features of the vertebrate brain.

The lamprey occupies a pivotal position for elucidating vertebrate brain evolution. Using spatial transcriptomics and single-nucleus RNA sequencing, we generated a three-dimensional molecular atlas of the lamprey brain, identifying 209 distinct cell clusters across 14 regions. Cross-species comparisons revealed broad conservation of regional spatial architecture, defining an ancestral organizational blueprint. Within this conserved framework, however, marked lineage-specific divergence emerged. We observed extensive neuronal specialization across vertebrate lineages, accompanied by regulatory

Dynamic microglia/macrophage infiltration during spinal cord regeneration in larval sea lamprey.

Both mammals and non-mammalian vertebrates display neuroimmune interactions after spinal cord injury (SCI). However, the impact of the immune response on neural regeneration remains unclear as it includes both proregenerative and inhibitory processes. To begin to understand how neuroimmune interactions influence central nervous system (CNS) regeneration, we examined the distribution of microglia/macrophages in relation to regenerating axons in larval sea lamprey (Petromyzon marinus), a non-mammalian vertebrate that exhibits robust axon and synapse regeneration after SCI. The relationship betwe

Selective expression of CSPG receptors PTPσ and LAR in poorly regenerating reticulospinal neurons of lamprey.

Disability following spinal cord injury is due to failure of axon regeneration, which has been ascribed to environmental factors in the central nervous system and a developmental loss of intrinsic growth capacity in neurons. Recently, the receptor-like protein tyrosine phosphatases, protein tyrosine phosphatase σ (PTPσ) and leukocyte common antigen-related phosphatase (LAR), have been identified as specific receptors for the regeneration-inhibiting matrix molecules chondroitin sulfate proteoglycans (CSPGs). After spinal cord transection in lampreys, axons of the large, identified reticulospina

Intestinal synthesis and secretion of bile salts as an adaptation to developmental biliary atresia in the sea lamprey.

Bile salt synthesis is a specialized liver function in vertebrates. Bile salts play diverse roles in digestion and signaling, and their homeostasis is maintained by controlling input (biosynthesis) and intestinal conservation. Patients with biliary atresia (i.e., obliteration of the biliary tree) suffer liver fibrosis and cirrhosis. In contrast, sea lamprey thrives despite developmental biliary atresia. We discovered that the sea lamprey adapts to biliary atresia through a unique mechanism of de novo synthesis and secretion of bile salts in intestine after developmental biliary atresia, in add

Activation of the newly discovered cyclostome renin-angiotensin system in the river lamprey Lampetra fluviatilis.

This study describes the first investigations of the physiological signals involved in activating the newly discovered cyclostome renin-angiotensin system (RAS) and its role in the river lamprey Lampetra fluviatilis. Experimental manipulation showed that volume depletion (removal of 40% blood volume) rapidly activated the RAS of lampreys acclimated to water at 576 mOsm kg(-1) (21 p.p.t.), significantly increasing plasma angiotensin concentrations after 30 min and 60 min. In agreement with these results, a rapid change in environmental salinity (758 mOsm kg(-1) to freshwater (FW) and FW to 605

Effects of cGMP and cAMP on light responses of the photosensory pineal neurons in the lamprey, Lampetra japonica.

We examined in this study how external cyclic nucleotides affect the light response mechanism of the pineal photoreceptors and explored the existence of parietal eye type of photoreceptor of which the internal cGMP concentration increased during the light response. Pineal organs of river lampreys, Lampetra japonica, were treated with 8-bromo guanosine 3',5'-cyclic monophosphate (8Br-cGMP) or 8-bromo adenosine 3',5'-cyclic monophosphate (8Br-cAMP) before light stimuli, and the light responses were recorded from the second order neurons, chromatic or achromatic-type neurons. Serotonergic nerve fibers in the primary olfactory pathway of the larval sea lamprey, Petromyzon marinus.

In this study, serotonin (5-hydroxytryptamine; 5HT)-immunoreactive (5HT-IR) neuronal fibers were identified in the primary olfactory pathway of the sea lamprey. These neurons are likely part of a nonolfactory neural system that innervates the olfactory sac. Cell bodies with 5HT immunoreactivity predominated in the lamina propria of the rostral portion of the nasal cavity and were less prevalent adjacent to the olfactory epithelium. The 5HT-IR fibers were parallel to axons of the olfactory receptor neurons in the lamina propria of the olfactory mucosa and in the olfactory nerve. Serotonergic fi

Cloning and characterization of the pineal gland-specific opsin gene of marine lamprey (Petromyzon marinus).

We have cloned and sequenced the pineal gland-specific opsin (P opsin) gene, p(Pm), of marine lamprey, P. marinus. The deduced P opsin, P(Pm), consisting of 444 aa, is about 90 aa longer than the orthologous opsins of chicken, pigeon, and American chameleon. The evolution of P(Pm) is characterized not only by a significantly higher rate of aa replacement than the other P opsins but also by a larger number of unique aa replacements at highly conserved sites in the nonTM region. These changes in P(Pm) might have been important in achieving marine lamprey-specific interaction between P(Pm) and ot

Biphasic role of L-TGF-β2 in modulating lamprey inflammation: Insights into vertebrate immune evolution.

The regulation of inflammatory balance is central to immune homeostasis, with disruptions contributing to autoimmune diseases. As representatives of early vertebrates, lampreys offer a valuable model for investigating the evolutionary foundations of immune regulation. This study examines the dual role of L-TGF-β2 in modulating inflammation in lampreys, providing insights into the evolutionary origins of immune homeostasis mechanisms. Using lipopolysaccharide (LPS)-induced inflammation models, recombinant L-TGF-β2 was found to enhance leukocyte chemotaxis in quiescent states while inhibiting ex

A Novel Vav3 Homolog Identified in Lamprey, Lampetra japonica, with Roles in Lipopolysaccharide-Mediated Immune Response.

Vav guanine nucleotide exchange factor 3 (Vav3), a Rho family GTPase, regulates multiple cell signaling pathways including those of T- and B-cell receptors in vertebrates through mediating the activities of the Rho family members. Whether the lamprey possesses Vav3 homolog and what role it plays in immune response remain unknown. Gene cloning, recombinant expression, antibody production and expression pattern analyses were performed to characterize the lamprey Vav3 in the current study. The lamprey Vav3 is closer to jawed vertebrates' Vav3 molecules (about 53% identities in general) than to Va

Roles for FGF in lamprey pharyngeal pouch formation and skeletogenesis highlight ancestral functions in the vertebrate head.

A defining feature of vertebrates (craniates) is a pronounced head supported and protected by a cellularized endoskeleton. In jawed vertebrates (gnathostomes), the head skeleton is made of rigid three-dimensional elements connected by joints. By contrast, the head skeleton of modern jawless vertebrates (agnathans) consists of thin rods of flexible cellular cartilage, a condition thought to reflect the ancestral vertebrate state. To better understand the origin and evolution of the gnathostome head skeleton, we have been analyzing head skeleton development in the agnathan, lamprey. The fibrobla
