Serum Levels of Dihomo-Gamma (γ)-Linolenic Acid (DGLA) Are Inversely Associated with Linoleic Acid and Total Death in Elderly Patients with a Recent Myocardial Infarction.

Dihomo-gamma-linolenic acid (DGLA) is an n-6 polyunsaturated fatty acid (PUFA) derived from linoleic acid (LA). The LA:DGLA ratio reflects conversion from LA to DGLA. Low levels of DGLA in serum have been related to poor outcome in myocardial infarction (MI) patients. Aims: To assess the association of DGLA and LA:DGLA with total death as a primary aim and incident cardiovascular events as a secondary objective. Methods: Baseline samples from 1002 patients, aged 70 to 82 years, included 2-8 weeks after an MI and followed for 2 years, were used. Major adverse clinical events (MACE) consisted of

Identification and validation of γ-Linolenic acid as a natural FABP5 inhibitor in hepatocellular carcinoma through deep learning and experimental approaches.

BACKGROUND: Fatty acid binding protein 5 (FABP5) is implicated in hepatocellular carcinoma (HCC) progression and represents a potential therapeutic target. METHODS: We integrated machine learning-based virtual screening, molecular docking and molecular dynamics simulations to identify natural compounds with high binding affinity to FABP5. Candidate compounds were further validated by in-vitro assays in HCC cell lines, including proliferation, migration/invasion, apoptosis/ferroptosis-related readouts, and mechanistic validation. RESULTS: The optimized models enabled efficient screening of na

Alpha-Linolenic Acid and Tarantula Cubensis Extract Attenuate Bladder Inflammation in Cyclophosphamide-Induced Cystitis: A Comparative Experimental Study.

INTRODUCTION AND HYPOTHESIS: Primary bladder pain syndrome (PBPS) is a chronic inflammatory bladder disorder characterized by pelvic pain, urinary urgency, and frequency. Current intravesical therapies such as hyaluronic acid (HA) mainly restore the urothelial barrier but may not adequately address inflammatory and oxidative mechanisms underlying bladder injury. We investigated whether alpha-linolenic acid (ALA) and Tarantula cubensis extract (TCAE) provide greater protection than intravesical HA in an experimental cystitis model. METHODS: Forty female Wistar rats were randomly assigned to fi

Flaxseed cyclolinopeptide-mediated regulation of α-linolenic acid metabolism alleviates high-fat diet-induced vascular inflammation.

Methionine-containing cyclolinopeptide ([1-9-NαC]-linusorb B2, CLB) and α-linolenic acid (ALA), two anti-inflammatory flaxseed constituents, were investigated for their combined vascular benefits. CLB-enriched orbitides were prepared for long-term high-fat diet (HFD)-fed mouse studies, while high-purity CLB monomer (>97%) was isolated via preparative chromatography for acute in vivo and cell-based assays. Structural confirmation and purity validation were achieved via mass spectrometry and liquid chromatography. CLB-enriched orbitides co-administered with ALA in HFD-fed mice reduced body weigh

Gut microbiota-related metabolite alpha-linolenic acid mitigates intestinal inflammation induced by oral infection with Toxoplasma gondii.

BACKGROUND: Oral infection with cysts is the main transmission route of Toxoplasma gondii (T. gondii), which leads to lethal intestinal inflammation. It has been widely recognized that T. gondii infection alters the composition and metabolism of the gut microbiota, thereby affecting the progression of toxoplasmosis. However, the potential mechanisms remain unclear. In our previous study, there was a decrease in the severity of toxoplasmosis after T. gondii α-amylase (α-AMY) was knocked out. Here, we established mouse models of ME49 and Δα-amy cyst infection and then took advantage of 16S rRNA

Effect of alpha linolenic acid supplementation on serum prostate specific antigen (PSA): results from the alpha omega trial.

BACKGROUND: Alpha linolenic acid (ALA) is the major omega-3 fatty acid in the diet. Evidence on health effects of ALA is not conclusive, but some observational studies found an increased risk of prostate cancer with higher intake of ALA. We examined the effect of ALA supplementation on serum concentrations of prostate-specific antigen (PSA), a biomarker for prostate cancer. METHODS: The Alpha Omega Trial (ClinicalTrials.gov Identifier: NCT00127452) was a double-blind, placebo-controlled trial of ALA and the fish fatty acids eicosapentanoic acid (EPA) and docosahexanoic acid (DHA) on the recur

An alternative solution for α-linolenic acid supplements: in vitro digestive properties of silkworm pupae oil in a pH-stat system.

α-Linolenic acid (ALA) is recognised to have a regulatory effect on cardiovascular diseases. Due to the low bioavailability of linseed oil (LINO), which is the most common ALA supplement, it is necessary to find a replacement for ALA supplements that is more easily accepted by the human body. The content of ALA in silkworm pupae oil (SPO) is 32.60 ± 0.67%, and SPO can be substituted as a dietary lipid to meet the demand of the human body. In the present study, a pH-stat system was used to investigate the release degree of free fatty acids (FFAs) from SPO and construct a first-order kinetic mod

The effect of γ-linolenic acid on Polycystic Ovary Syndrome associated Focal Segmental Glomerulosclerosis via TGF-β pathway.

BACKGROUND: In recent years, female infertility from Polycystic Ovary Syndrome (PCOS) has gained scientific interest. PCOS alters the metabolic and endocrine functioning in females. The elevation in androgens can damage the androgen receptors present on the kidney giving rise to renal disorders like Focal Segmental Glomerulosclerosis (FSGS). Transforming Growth Factor Beta (TGF-β) in the ovary is activated by activin for Follicle Stimulating Hormone (FSH) secretion and in the kidney by thrombospondin 1 (TSP1) for cell growth and apoptosis. Studies show that gamma-linolenic acid (GLA) effective

Parental LC-PUFA biosynthesis capacity and nutritional intervention with alpha-linolenic acid affect performance of Sparus aurata progeny.

Environmental factors such as nutritional interventions during early developmental stages affect and establish long-term metabolic changes in all animals. Diet during the spawning period has a nutritional programming effect in offspring of gilthead seabream and affects long-term metabolism. Studies showed modulation of genes such as fads2, which is considered to be a rate-limiting step in the synthesis of n-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA). However, it is still unknown whether this adaptation is related to the presence of precursors or to limitations in the pre-formed pro

Subchronic alpha-linolenic acid treatment enhances brain plasticity and exerts an antidepressant effect: a versatile potential therapy for stroke.

Omega-3 polyunsaturated fatty acids are known to have therapeutic potential in several neurological and psychiatric disorders. However, the molecular mechanisms of action underlying these effects are not well elucidated. We previously showed that alpha-linolenic acid (ALA) reduced ischemic brain damage after a single treatment. To follow-up this finding, we investigated whether subchronic ALA treatment promoted neuronal plasticity. Three sequential injections with a neuroprotective dose of ALA increased neurogenesis and expression of key proteins involved in synaptic functions, namely, synapto

Stereotactic radiosurgery XII. Large AVM and the failure of the radiation response modifier gamma linolenic acid to improve the therapeutic ratio.

Large arteriovenous malformations (AVM) are less likely to be 'cured' by single fraction stereotactically delivered radiation therapy (radiosurgery) and such treatment is attended by higher complication risks. Out of 200 successive AVM patients accepted for curative radiosurgery at St. Bartholomew's Hospital (and all treated by the same 6MV x-ray technique to 17.5 Gy marginal dose) were 62 patients whose target volume exceeded 10 ml. Laboratory data demonstrating that gamma linolenic [omega-6-] acid (GLA) protected rat spinal cord from the damaging effect of single shot radiation prompted this

Effect of intravenous lipid emulsions enriched with gamma-linolenic acid on plasma n-6 fatty acids and prostaglandin biosynthesis after burn and endotoxin injury in rats.

OBJECTIVE: To study the effect of intravenous lipid emulsions enriched with gamma-linolenic acid on plasma fatty acids and series-2 prostaglandins to determine if the slow conversion of linoleic acid by delta-6-desaturase to gamma-linolenic acid could be bypassed to provide substrate for the formation of dihomo-gamma-linolenic acid, the immediate precursor for series-1 prostaglandins, in control and injured rats. Dihomo-gamma-linolenic acid can also compete with arachidonic acid for oxidative metabolism by cyclooxygenase to modulate series-2 prostaglandin biosynthesis. DESIGN: Prospective, ra

Isolation and identification of alpha-(4-pyridyl-1-oxide)-N-tert-butylnitrone radical adducts formed by the decomposition of the hydroperoxides of linoleic acid, linolenic acid, and arachidonic acid by soybean lipoxygenase.

alpha-(4-Pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN) radical adducts, which are formed in the reactions of soybean lipoxygenase with linoleic acid, arachidonic acid, and linolenic acid, were isolated using HPLC-ESR spectroscopy. Both linoleic acid and arachidonic acid gave one radical adduct, whereas in the case of linolenic acid, two radical adducts were isolated. These radical adducts all showed virtually identical uv spectra with lambda max at 292 and 220 nm in hexane. The absence of absorbance with lambda max at 234 nm indicates that a conjugated diene structure is not contained in these

Impact of perinatal α-linolenic acid deficiency combined with high-fat diet consumption on tissue-specific fatty acid profiles in rats: Maternal and offspring analysis.

BACKGROUND: An imbalance in the dietary ratio of omega-6 (n-6) to omega-3 (n-3) polyunsaturated fatty acids (PUFAs) and/or a high-fat diet (HFD) may disrupt the metabolic conversion of essential PUFAs, such as linoleic acid (LA) and α-linolenic acid (ALA), into their long-chain derivatives (LC-PUFAs), thereby altering their accumulation in various tissues. To determine the effects of low maternal intake of ALA and/or an HFD on FA composition in maternal tissues (liver, adipose tissue, and erythrocytes) and offspring tissues (liver, erythrocytes, adipose tissue, heart, kidney, intestine, muscle
