In vitro bioassays for measuring monoamine transporter function.

Monoamine transporters (MATs) - including the dopamine transporter, norepinephrine transporter, and serotonin transporter - are plasma membrane proteins that mediate the uptake of previously released neurotransmitters from the synaptic cleft, thereby terminating neurotransmitter signaling. MATs are implicated in the pathophysiology of numerous neurological and psychiatric disorders, making them key targets for a wide range of psychotherapeutic agents. Furthermore, stimulant drugs, such as cocaine, methamphetamine, and synthetic cathinones, exert their effects by disrupting normal MAT function.

Hierarchical Membrane for Photoelectrostatically Gated Monovalent-Divalent Ion Separation.

Monovalent-divalent ion separation remains a significant challenge for the selective recovery of ions from aqueous solutions. Here, we report a hierarchical membrane composed of a poly(ionic liquid) photo-grafted onto a carbon nitride thin film supported on porous anodic aluminum oxide. Under concentration gradient-driven natural diffusion, the membrane establishes a baseline level of monovalent-divalent selectivity. However, upon application of light and bias as external stimuli, the hierarchical membrane enables light-assisted modulation of ion transport, resulting in a significant increase

Degradable porous PLGA/PCL membrane enable a lung alveoli-on-a-chip for modeling particulate-induced alveolar injury.

Understanding how airborne particulates disrupt the alveolar barrier requiresin vitrosystems that recapitulate both the structure and transport properties of the lung air-blood interface. Here, we report a biodegradable lung alveoli-on-a-chip enabled by porous poly(lactic-co-glycolic acid)/polycaprolactone (PLGA/PCL) membranes with an interconnected porous architecture generated via porogen-assisted phase separation process. The membrane exhibits tunable degradation behavior, allowing progressive increases in surface porosity (∼40%) and reduction in thickness (∼3µm) during culture, while PCL m

Radixin knockdown improves the accumulation and efficiency of methotrexate in tumor cells.

The plasma membrane localization of efflux transporters, such as multidrug resistance‑associated protein (MRP/ABCC), is governed by scaffold proteins, such as the ezrin/radixin/moesin (ERM) proteins. Reduction of the anchor function of ERM proteins in tumor cells may decrease the transport activity of efflux transporters, possibly leading to increased intracellular accumulation and efficiency of anticancer drugs as substrates of efflux transporters. The effect of the knockdown of ERM proteins was examined in multiple carcinoma cell lines on the transport activity of efflux transporters and on

Uptake of arachidonic acid into membrane phospholipids: effect on chloride transport across cornea.

We demonstrate that arachidonic acid (AA) stimulation of chloride transport across frog cornea is mediated via two independent pathways: (1) stimulation of prostaglandins and cAMP synthesis, and (2) a direct physical change in the membrane produced by substitution of different phospholipid acyl chains. AA is well known as a precursor in the synthesis of prostaglandins, which have been shown to stimulate cAMP synthesis and chloride transport in frog cornea. We show that frog cornea can convert exogenous AA to PGE2, but that in the presence of 10(-5) M indomethacin both the conversion to PGE2 an

Novel fluorescence-based approaches for the study of biogenic amine transporter localization, activity, and regulation.

Pre-synaptic norepinephrine (NE) and dopamine (DA) transporters (NET and DAT) terminate catecholamine synaptic transmission through reuptake of released neurotransmitter. Recent studies reveal that NET and DAT are tightly regulated by receptor and second messenger-linked signaling pathways. Common approaches for studying these transporters involve use of radiolabeled substrates or antagonists, methods possessing limited spatial resolution and that bear limited opportunities for repeated monitoring of living preparations. To circumvent these issues, we have explored two novel assay platforms th

Influence of purinergic signaling on glucose transporters: A possible mechanism against insulin resistance?

Metabolic disorders, such as insulin resistance, affect many people worldwide due to the prevalence of obesity and type 2 diabetes, which are pathologies that impair glycemic metabolism. Glucose is the primary energetic substrate of the body and is essential for cellular function. As the cell membrane is not permeable to glucose molecules, there are two distinct groups of glucose transporters: sodium-glucose-linked transporters (SGLTs) and the glucose transporter (GLUT) family. These transporters facilitate the entry of glucose into the bloodstream or cytoplasm where it functions in the produc

A Brucella effector modulates the Arf6-Rab8a GTPase cascade to promote intravacuolar replication.

Remodeling of host cellular membrane transport pathways is a common pathogenic trait of many intracellular microbes that is essential to their intravacuolar life cycle and proliferation. The bacterium Brucella abortus generates a host endoplasmic reticulum-derived vacuole (rBCV) that supports its intracellular growth, via VirB Type IV secretion system-mediated delivery of effector proteins, whose functions and mode of action are mostly unknown. Here, we show that the effector BspF specifically promotes Brucella replication within rBCVs by interfering with vesicular transport between the trans-

Transporter Proteins as Therapeutic Drug Targets-With a Focus on SGLT2 Inhibitors.

Membrane transporters interact not only with endogenous substrates but are also engaged in the transport of xenobiotics, including drugs. While the coordinated function of uptake (solute carrier family-SLC and SLCO) and efflux (ATP-binding cassette family-ABC, multidrug and toxic compound extrusion family-MATE) transporter system allows vectorial drug transport, efflux carriers alone achieve barrier functions. The modulation of transport functions was proved to be effective in the treatment strategies of various pathological states. Sodium-glucose cotransporter-2 (SGLT2) inhibitors are the dru

Effects of Cd2+ on the epithelial Na+ channel (ENaC) investigated by experimental and modeling studies.

The function of the epithelial Na+ channel from the apical membrane of many Na+ transporting epithelia is modulated by various chemical compounds from the extracellular space, such as heavy metals, protons or chloride ions. We have studied the effect of extracellular Cd2+ on the function of the epithelial Na+ channel (ENaC) in heterologously expressed Xenopus laevis oocytes and Na+-transporting epithelia. We assayed channel function as the amiloride-sensitive sodium current (I(Na)). Cd2+ rapidly and voltage-independently inhibited INa in oocytes expressing αβγ Xenopus ENaC (xENaC). The extrace

Integrating plant lipid signaling with its membrane environment.

Plant cells generate lipid messengers such as phosphatidic acid, diacylglycerol, and phosphoinositides through rapid phospholipid remodeling, enabling the regulation of stress responses, ion transport, cytoskeletal organization, and membrane trafficking. Although the enzymatic pathways responsible for lipid interconversion are well characterized, lipid signaling is not merely a linear biochemical process but is profoundly shaped by the membrane environment in which it occurs. Accumulating evidence indicates that membrane composition, electrostatic properties, lipid packing, nanoscale and micro

Differential effects of lysophospholipid headgroups, acyl chain length and saturation on vacuole acidification, Ca2+ transport, membrane fluidity, and fusion.

SNARE-mediated membrane fusion is regulated by the lipid composition of the engaged bilayers. Lipids impact fusion through direct protein-lipid interactions or through modulating the physical properties of membranes to affect protein function. Lysophospholipids (LPLs) can affect membrane curvature, fluidity and energy of deformation. Their effects are due to their head group, and the length and saturation of their single acyl chains. Here we examined how the properties of LPLs affect yeast vacuole fusion, membrane fluidity and ion transport. We found that lysophosphatidylcholine (LPC) with 14-

Vehicle-dependent in situ modification of membrane-controlled drug release.

The possibility of utilizing the interaction between the drug vehicle and the polymeric control membrane of reservoir-type drug delivery systems to modulate the release rate of these systems was investigated. Lipid vehicles common in dermal formulations, with varying composition and viscosity and the Celgard polypropylene membrane were used to study the release of a model drug (salicylic acid). The release kinetics were investigated taking into account two consecutive transport processes, diffusion within the donor reservoir and permeation through the membrane. Membrane permeation was found to

Structural Insight on the Selectivity of Calyx[4]Arene-Based Inhibitors of Mg2+-Dependent Atp-Hydrolases.

Located in plasma membranes, ATP hydrolases are involved in several dynamic transport processes, helping to control the movement of ions across cell membranes. ATP hydrolase acts as a transport protein, converting energy from ATP hydrolysis into transport molecules against their concentration gradients. In addition to energy metabolism and active transport, ATP hydrolase is essential for maintaining cellular homeostasis and cell function. This study focused on the domain architecture model of P-type ATPases, which participate in the reaction cycles of ATP hydrolysis carried out by membrane tra

Cross-Linked Self-Standing Graphene Oxide Membranes: A Pathway to Scalable Applications in Separation Technologies.

The large-scale implementation of 2D material-based membranes is hindered by mechanical stability and mass transport control challenges. This work describes the fabrication, characterisation, and testing of self-standing graphene oxide (GO) membranes cross-linked with oxides such as Fe2O3, Al2O3, CaSO4, Nb2O5, and a carbide, SiC. These cross-linking agents enhance the mechanical stability of the membranes and modulate their mass transport properties. The membranes were prepared by casting aqueous suspensions of GO and SiC or oxide powders onto substrates, followed by drying and detachment to y

Quantification of osmotic water transport in vivo using fluorescent albumin.

Osmotic water transport across the peritoneal membrane is applied during peritoneal dialysis to remove the excess water accumulated in patients with end-stage renal disease. The discovery of aquaporin water channels and the generation of transgenic animals have stressed the need for novel and accurate methods to unravel molecular mechanisms of water permeability in vivo. Here, we describe the use of fluorescently labeled albumin as a reliable indicator of osmotic water transport across the peritoneal membrane in a well-established mouse model of peritoneal dialysis. Neisseria gonorrhoeae requires expression of TonB and the putative transporter TdfF to replicate within cervical epithelial cells.

Neisseria gonorrhoeae has evolved a repertoire of iron acquisition systems that facilitate essential iron uptake in the human host. Acquisition of iron requires both the energy-harnessing cytoplasmic membrane protein, TonB, as well as specific outer membrane TonB-dependent transporters (TdTs.) Survival within host epithelial cells is important to the pathogenesis of gonococcal disease and may contribute to the persistence of infection. However, the mechanisms by which gonococci acquire iron within this intracellular niche are not currently understood. In this study, we investigated the surviva

Architecture of a complete Bce-type antimicrobial peptide resistance module.

Gram-positive bacteria synthesize and secrete antimicrobial peptides that target the essential process of peptidoglycan synthesis. These antimicrobial peptides not only regulate the dynamics of microbial communities but are also of clinical importance as exemplified by peptides such as bacitracin, vancomycin, and daptomycin. Many gram-positive species have evolved specialized antimicrobial peptide sensing and resistance machinery known as Bce modules. These modules are membrane protein complexes formed by an unusual Bce-type ABC transporter interacting with a two-component system sensor histid

Pioglitazone Is a Mild Carrier-Dependent Uncoupler of Oxidative Phosphorylation and a Modulator of Mitochondrial Permeability Transition.

Pioglitazone (PIO) is an insulin-sensitizing antidiabetic drug, which normalizes glucose and lipid metabolism but may provoke heart and liver failure and chronic kidney diseases. Both therapeutic and adverse effects of PIO can be accomplished through mitochondrial targets. Here, we explored the capability of PIO to modulate the mitochondrial membrane potential (ΔΨm) and the permeability transition pore (mPTP) opening in different models in vitro. ΔΨm was measured using tetraphenylphosphonium and the fluorescent dye rhodamine 123. The coupling of oxidative phosphorylation was estimated polarogr
