Cerebellar-Motor Cortex Connectivity: One or Two Different Networks?

Anterior-posterior (AP) and posterior-anterior (PA) pulses of transcranial magnetic stimulation (TMS) over the primary motor cortex (M1) appear to activate distinct interneuron networks that contribute differently to two varieties of physiological plasticity and motor behaviors (Hamada et al., 2014). The AP network is thought to be more sensitive to online manipulation of cerebellar (CB) activity using transcranial direct current stimulation. Here we probed CB-M1 interactions using cerebellar brain inhibition (CBI) in young healthy female and male individuals. TMS over the cerebellum produced

Impaired motor cortex plasticity in patients with Noonan syndrome.

OBJECTIVE: Noonan syndrome (NS; OMIM 163950) is a developmental disorder caused by activating mutations in various components of the RAS-MAPK pathway. Recent in vitro studies demonstrated impairment of synaptic plasticity caused by RAS-MAPK pathway hyperactivity. Induction of synaptic plasticity critically depends on the level of attention. We therefore studied the induction of synaptic plasticity in patients with NS and healthy volunteers under different conditions of attention using transcranial magnetic stimulation. METHODS: We investigated 10 patients with NS and healthy controls (HC) usi

Functional properties of human primary motor cortex gamma oscillations.

Gamma oscillations in human primary motor cortex (M1) have been described in human electrocorticographic and noninvasive magnetoencephalographic (MEG)/electroencephalographic recordings, yet their functional significance within the sensorimotor system remains unknown. In a set of four MEG experiments described here a number of properties of these oscillations are elucidated. First, gamma oscillations were reliably localized by MEG in M1 and reached peak amplitude 137 ms after electromyographic onset and were not affected by whether movements were cued or self-paced. Gamma oscillations were fou

Walking the talk--speech activates the leg motor cortex.

Speech may have evolved from earlier modes of communication based on gestures. Consistent with such a motor theory of speech, cortical orofacial and hand motor areas are activated by both speech production and speech perception. However, the extent of speech-related activation of the motor cortex remains unclear. Therefore, we examined if reading and listening to continuous prose also activates non-brachiofacial motor representations like the leg motor cortex. We found corticospinal excitability of bilateral leg muscle representations to be enhanced by speech production and silent reading. Con

Excitability of human motor cortex during hyperventilation and hypercapnia.

We tested the hypothesis that the excitability of corticospinal neurons was altered by changes in PCO2. Magnetic stimulation was used to excite the neurons in the human motor cortex that give rise to the fast-conducting corticospinal pathway. The characteristics of the composite excitatory postsynaptic potentials (EPSPs) produced in individual spinal motoneurons by cortical stimulation were derived from changes in the firing probability of voluntarily activated motor units. The amplitudes of these composite EPSPs in response to a constant cortical stimulus were assumed to reflect the excitabil

Motor cortex stimulation as treatment of trigeminal neuropathic pain.

A report is given on first experiences with motor cortex stimulation in 10 patients with different forms of neuropathic pain. Three of them had central pain as sequelae of cerebrovascular disease. In none of them did the stimulation provide pain relief. Two patients had pain from peripheral nerve injuries. One did not respond, but the other obtained about 50% pain relief. The remaining 5 patients with trigeminal neuropathy experienced definite pain relief varying between 60 and 90%. During test stimulation most patients had one or two short-lasting generalized seizures. But no one had any moto

Timing-dependent modulation of the posterior parietal cortex-primary motor cortex pathway by sensorimotor training.

Interplay between posterior parietal cortex (PPC) and ipsilateral primary motor cortex (M1) is crucial during execution of movements. The purpose of the study was to determine whether functional PPC-M1 connectivity in humans can be modulated by sensorimotor training. Seventeen participants performed a sensorimotor training task that involved tapping the index finger in synchrony to a rhythmic sequence. To explore differences in training modality, one group (n = 8) learned by visual and the other (n = 9) by auditory stimuli. Transcranial magnetic stimulation (TMS) was used to assess PPC-M1 conn

Macaque ventral premotor cortex exerts powerful facilitation of motor cortex outputs to upper limb motoneurons.

The ventral premotor area (F5) is part of the cortical circuit controlling visuomotor grasp. F5 could influence hand motor function through at least two pathways: corticospinal projections and corticocortical projections to primary motor cortex (M1). We found that stimulation of macaque F5, which by itself evoked little or no detectable corticospinal output, could produce a robust modulation of motor outputs from M1. Arrays of fine microwires were implanted in F5 and M1. During terminal experiments under chloralose anesthesia, single stimuli delivered to M1 electrodes evoked direct (D) and ind

Targeted stimulation of motor cortex ensembles drives learned movements.

During the execution of learned motor skills, the neural population in layer 2/3 (L2/3) of primary motor cortex (M1) expresses a reproducible spatiotemporal activity pattern1. It is debated whether M1 actively participates in generating this learned pattern and whether this pattern causally drives the learned movement2-5. Here we combined in vivo two-photon calcium imaging and holographic optogenetic stimulation of functionally defined M1 L2/3 ensembles in mice performing a skilled lever-pressing task. Brief synchronous stimulation of about 20 neurons whose activity onset precedes voluntary mo

Early premotor-somatosensory remodeling after primary motor cortex injury.

Focal ischemic injury to the primary motor cortex (M1) disrupts distributed sensorimotor networks and initiates structural remodeling in spared cortical regions. Although previous studies have demonstrated axonal sprouting from ventral premotor cortex (PMv) to somatosensory area 1/2 at later post-injury time points, the onset of this reorganization remains unknown. We examined whether PMv-to-area 1/2 structural remodeling is detectable four weeks after focal infarction of the M1 distal forelimb representation in adult squirrel monkeys. Animals received focal ischemic lesions of the M1 distal f

Treatment selection and functional outcomes in motor cortex metastases.

BACKGROUND: Brain metastases are associated with a poor prognosis. While systemic therapy primarily determines overall survival, surgical cytoreduction may improve local disease control. Preserving neurological function and independence is particularly important in patients with limited life expectancy. However, surgery is often avoided for metastases located in eloquent brain regions. This study evaluates treatment selection and functional outcomes in patients with metastases involving the primary motor cortex. METHODS: We conducted a retrospective single-center analysis of patients treated

Connectivity underlying motor cortex activity during goal-directed behaviour.

Neural representations of information are shaped by long-range input and local network interactions. Previous studies linking neural coding and cortical connectivity have focused on input-driven activity in the sensory cortex1-3. Here we studied neural activity in the motor cortex while mice gathered rewards with multidirectional tongue reaching. This behaviour does not require training, allowing us to probe neural coding and connectivity before activity is shaped by extended learning. Motor cortex neurons were tuned to target location and reward outcome, and typically responded during and aft

Speech motor cortex enables BCI cursor control and click.

Objective.Decoding neural activity from ventral (speech) motor cortex is known to enable high-performance speech brain-computer interface (BCI) control. It was previously unknown whether this brain area could also enable computer control via neural cursor and click, as is typically associated with dorsal (arm and hand) motor cortex.Approach.We recruited a clinical trial participant with amyotrophic lateral sclerosis and implanted intracortical microelectrode arrays in ventral precentral gyrus (vPCG), which the participant used to operate a speech BCI in a prior study. We developed a cursor BCI

Influences of current direction on 1 Hz motor cortex rTMS.

OBJECTIVE: Neuromodulatory changes induced by repetitive transcranial magnetic stimulation (rTMS) are highly variable and lack replicability. Amongst many influencing factors, the current direction of the stimulation is known to affect rTMS aftereffects. In this study, we investigated the influence of current direction on cortical and peripheral markers of motor cortex plasticity as induced by 1 Hz rTMS as well as the correlation of cortical and peripheral markers. Additionally, we investigated possible confounding variables. METHODS: Twenty-five healthy subjects received 2000 pulses of 1 Hz

Prefrontal and somatosensory-motor cortex effective connectivity in humans.

Effective connectivity, functional connectivity, and tractography were measured between 57 cortical frontal and somatosensory regions and the 360 cortical regions in the Human Connectome Project (HCP) multimodal parcellation atlas for 171 HCP participants. A ventral somatosensory stream connects from 3b and 3a via 1 and 2 and then via opercular and frontal opercular regions to the insula, which then connects to inferior parietal PF regions. This stream is implicated in "what"-related somatosensory processing of objects and of the body and in combining with visual inputs in PF. A dorsal "action

Disruption of somatosensory cortex impairs motor learning and retention.

This study tests for a function of the somatosensory cortex, that, in addition to its role in processing somatic afferent information, somatosensory cortex contributes both to motor learning and the stabilization of motor memory. Continuous theta-burst magnetic stimulation (cTBS) was applied, before force-field training to disrupt activity in either the primary somatosensory cortex, primary motor cortex, or a control zone over the occipital lobe. Tests for retention and relearning were conducted after a 24 h delay. Analysis of movement kinematic measures and force-channel trials found that cTB

Reduced motor cortex GABABR function following chronic alcohol exposure.

The GABAB receptor (GABABR) agonist baclofen has been used to treat alcohol and several other substance use disorders (AUD/SUD), yet its underlying neural mechanism remains unclear. The present study aimed to investigate cortical GABABR dynamics following chronic alcohol exposure. Ex vivo brain slice recordings from mice chronically exposed to alcohol revealed a reduction in GABABR-mediated currents, as well as a decrease of GABAB1/2R and G-protein-coupled inwardly rectifying potassium channel 2 (GIRK2) activities in the motor cortex. Moreover, our data indicated that these alterations could b

The human motor cortex microcircuit: insights for neurodegenerative disease.

The human motor cortex comprises a microcircuit of five interconnected layers with different cell types. In this Review, we use a layer-specific and cell-specific approach to integrate physiological accounts of this motor cortex microcircuit with the pathophysiology of neurodegenerative diseases affecting motor functions. In doing so we can begin to link motor microcircuit pathology to specific disease stages and clinical phenotypes. Based on microcircuit physiology, we can make future predictions of axonal loss and microcircuit dysfunction. With recent advances in high-resolution neuroimaging

The posterior parietal cortex (PPC) mediates anticipatory motor control.

BACKGROUND: Flexible and precisely timed motor control is based on functional interaction within a cortico-subcortical network. The left posterior parietal cortex (PPC) is supposed to be crucial for anticipatory motor control by sensorimotor feedback matching. OBJECTIVE: Intention of the present study was to disentangle the specific relevance of the left PPC for anticipatory motor control using transcranial direct current stimulation (tDCS) since a causal link remains to be established. METHODS: Anodal vs. cathodal tDCS was applied for 10 min over the left PPC in 16 right-handed subjects in
