The effects of dantrolene on the contraction, relaxation, and energetics of the diaphragm muscle.

UNLABELLED: Dantrolene is used in patients with muscle spasticity and is the only known effective treatment for malignant hyperthermia. However, its effects on muscle relaxation and energetics are unknown and may have important consequences in diaphragmatic function. We studied the effects of dantrolene (10(-8) to 10(-4) M) on diaphragm muscle strips (n = 12) in the hamster in vitro (Krebs-Henseleit solution, 29 degrees C, 95% oxygen/5% carbon dioxide) in response to tetanic stimulation (50 Hz). We studied contraction and relaxation under isotonic and isometric conditions, as well as energetic

[Effect of neurotoxins on the electrical activity and contraction of the heart muscle].

The goal of the present review is to report the effect of the main neurotoxins known on the electrical and mechanical activity of heart muscle. Toxins which block the sodium channel (tetrodotoxin, saxitoxin) shorten the action potential (AP) duration and decrease the initial depolarizing phase of the AP. Toxins which occupy different sites in the channel and alter the gating mechanisms of the Na channel (aconitine, batrachotoxin, veratridine, sea anemone and scorpion toxins, brevetoxin and ciguatoxin) depolarize, lengthen the AP duration, increase the contraction and cause arrhythmias. Ca chan

Paradoxical contraction of pelvic floor muscles: clinical significance.

Paradoxical contraction of pelvic floor muscles during defecation straining has been said to be a cause of constipation and difficulty in passing feces. Nevertheless, controversies about its clinical significance still remained. Twenty patients with constipation and electromyographic evidence of paradoxical contraction of both puborectalis and external anal sphincter were investigated. An anorectal manometry performed in 17 confirmed the paradoxical contraction in 13 (76%). Electromyography revealed neurogenic signs in 11. Defecography demonstrated the paradoxical contraction in 6 only, but, t

Sonographic imaging of muscle contraction and fasciculations: a correlation with electromyography.

Precise quantitation of fasciculations with EMG is difficult because of their random location and discharge frequency in muscle. We studied the clinical value of real-time ultrasound in the study of normal voluntary muscle contraction and in the identification of fasciculations in 22 patients. Sonography effectively imaged fasciculations, demonstrating them in both resting and actively contracting extremity muscles and in less accessible muscles such as the tongue. In two instances ultrasound identified fasciculations not apparent on EMG. Analysis of the video images generated quantitative dat

[Effect of calmodulin inhibitors on the contraction and relaxation of the heart muscle].

The calmodulin inhibitor trifluoroperazine (5 microM) diminished the velocity of contraction and relaxation of the guinea-pig papillary muscle by 31 and 53%, respectively. Methophenazine showed approximately the same affinity to both calmodulin and trifluoroperazine and nearly 40-fold less affinity to troponin. Methophenazine (5 microM) did not change the contraction velocity and diminished the relaxation velocity ty 21%. It is suggested that calmodulin participates mainly in the control of myocardial relaxation.

Effect of tubocurarine and decamethonium on voluntary muscle contractions in man.

Six healthy, young male subjects performed repeated brisk maximal voluntary muscle contractions (MVC) with the knee and hip extensors. Three MVCs were performed every minute. On separate days decamethonium 0.03 mg X kg-1 and tubocurarine 0.01 mg X kg-1 were administered intravenously during repeated MVCs. While ordinary MVCs showed a slow rate of rise of tension over approximately 1 s, brisk MVCs showed a steep rate of rise of tension and a biphasic configuration appeared, as a notch was seen 370-480 ms after the initiation of the contraction curve. An arbitrary straight line was drawn connect

The force of contraction of the human ciliary muscle during accommodation.

1. Apparatus has been designed to alter the shape of the human lens by tensile forces applied to the zonular fibres indirectly through the ciliary body. The changes in dioptric power of the lens for monochromatic sodium light were measured at the same time. Simultaneous serial photography, and direct measurement enabled one to relate a change in shape of the lens to the change in dioptric power. Subsequently, the same lens was isolated and spun around its antero-posterior polar axis and high speed photography recorded its changing profile.2. By comparing the changes in lens profile due to zonu

β-Hydroxybutyrate attenuates bronchial smooth muscle proinflammatory cytokine production and contraction.

Asthma is a chronic respiratory condition characterized by airway inflammation, remodeling, and hyperresponsiveness to triggers that lead to airway constriction and impaired airflow. Bronchial smooth muscle (BSM) plays a central role in these processes by constricting the airways and producing proinflammatory cytokines in response to environmental triggers, allergens, and cytokines. Although current therapies, including bronchodilators, corticosteroids, and biologics, effectively treat many patients, additional strategies are needed for difficult-to-treat asthma. Emerging evidence suggests tha

Respiratory Muscle Contraction Characteristics and Potential Mechanisms in Severely Burned Rats.

Postburn hypermetabolism remains an important clinical problem. During this phase, there is a significant loss of diaphragmatic proteins. Better understanding of respiratory muscle dynamics and potential mechanisms affecting respiratory muscle function is necessary for the development of effective therapeutic approaches. Male Wistar rats were subjected to 50% TBSA burns and sham injuries, and respiratory muscle function was assessed with 0, 1, 4, 7, and 14 days postinjury, including pulmonary function, blood gas analysis, transdiaphragmatic pressure, diaphragm ultrasonography, isolated diaphra

Dynamic magnetic resonance imaging of muscle contraction in facioscapulohumeral muscular dystrophy.

Quantitative muscle MRI (water-T2 and fat mapping) is being increasingly used to assess disease involvement in muscle disorders, while imaging techniques for assessment of the dynamic and elastic muscle properties have not yet been translated into clinics. In this exploratory study, we quantitatively characterized muscle deformation (strain) in patients affected by facioscapulohumeral muscular dystrophy (FSHD), a prevalent muscular dystrophy, by applying dynamic MRI synchronized with neuromuscular electrical stimulation (NMES). We evaluated the quadriceps muscles in 34 ambulatory patients and

Verbal Instruction for Pelvic Floor Muscle Contraction among Healthy Young Males.

Teaching Pelvic Floor Muscle (PFM) contraction is a challenging task for clinicians and patients, as these muscles cannot be directly visualized. Thus, this study's objective is to compare the effectiveness of six verbal instructions for contracting the PFM among young men, as observed with transabdominal ultrasound imaging. Thirty-five male physiotherapy students, mean age 25.9 ± 1.9 years, participated in the study. A 6 MHz 35-mm curved linear array ultrasound transducer (Mindray M5) was placed in the transverse plane, supra-pubically, and angled 15-30° from the vertical plane. During crook

Excitation-Contraction Coupling in Ureteric Smooth Muscle: Mechanisms Driving Ureteric Peristalsis.

The ureter acts as a functional syncytium and is controlled by a propagating plateau-type action potential (AP) which gives rise to a wave of contraction (ureteral peristalsis) via a process called excitation-contraction (E-C)coupling. The second messenger Ca2+ activates Ca2+/calmodulin-dependent myosin light chain kinase-dependent phosphorylation of 20-kDa regulatory light chains of myosin which leads to ureteric contraction. Ca2+ entry from the extracellular space via voltage-gated L-type Ca2+ channels (VGCCs) provides the major source of activator Ca2+, responsible for generation of both th

Nerve growth factor-induced muscle hyperalgesia facilitates ischaemic contraction-evoked pain.

BACKGROUND: Intramuscular injection of Nerve Growth Factor (NGF) may influence the responsiveness of active chemo-sensitive channels affecting muscle pain sensitivity. This double-blinded crossover study in healthy humans assessed contraction-evoked pain responses and pain sensitivity during acute ischaemia in the tibialis anterior (TA) muscle before and 24 hr after five distributed NGF injections (1 µg, 4 cm interval) compared with control injections (isotonic-saline). METHODS: Twenty-one subjects participated in two experimental phases, each including five sessions over 7 days, with a gap o

Hypertonic saline inhibits airway smooth muscle contraction by inhibiting Ca2+ sensitization.

The effects of hypertonic solution on airway smooth muscle (ASM) contraction and the underlying mechanisms are largely unknown. We found that hypertonic saline (HS) inhibited acetylcholine (ACh)-induced contraction of ASM from the mouse trachea and human bronchi. In single mouse ASM cells (ASMCs), ACh induced an increase in intracellular Ca2+ that was further enhanced by 5% NaCl, indicating that the HS-induced inhibition of ASM contraction was not mediated by a decrease in cytosolic Ca2+ . The Rho-associated kinase (ROCK) inhibitor Y-27632 relaxed ACh-induced precontraction of mouse tracheal r

Influence of phasic muscle contraction upon the quadripulse stimulation (QPS) aftereffects.

OBJECTIVE: Contractions of the target muscle influence the aftereffects of repetitive transcranial magnetic stimulation (rTMS). The aim of this paper is to investigate whether or not voluntary hand movement influences the aftereffects of quadripulse stimulation (QPS) on the hand motor area. METHODS: Thirteen healthy volunteers participated in this study. After QPS-5 or QPS-50 intervention over the motor hot spot for the right first dorsal interosseous muscle (FDI), the subjects performed voluntary motor tasks (opening-closing right hand movements at 1 Hz for 1 min). We compared the time cours

Muscle activation varies with contraction mode in human spinal cord injury.

INTRODUCTION: To better understand volitional force generation after chronic incomplete spinal cord injury (SCI), we examined muscle activation during single and repeated isometric, concentric, and eccentric knee extensor (KE) maximal voluntary contractions (MVCs). METHODS: Torque and electromyographic (EMG) activity were recorded during single and repeated isometric and dynamic KE MVCs in 11 SCI subjects. Central activation ratios (CARs) were calculated for all contraction modes in SCI subjects and 11 healthy controls. RESULTS: SCI subjects generated greater torque, KE EMG, and CARs during

Influence of menstrual cycle on indices of contraction-induced muscle damage.

Limited evidence suggests women exhibit a dampened response to contraction-induced muscle damage (CIMD). The purpose of this study was to examine if differences in symptoms of CIMD exist when induced in the menstrual cycle follicular or luteal phase. Sixteen resistance exercise trained women between the ages of 18-37 completed 75 eccentric-biased extension exercises with their nondominant arm. Creatine kinase (CK), elbow joint angles, arm volume, strength, and soreness were measured over 7 days. Estrogen was higher (p < 0.001) in the luteal group. The high estrogen group (luteal) had an overal

[Forskolin inhibits spontaneous contraction of gastric antral smooth muscle in rats].

The aim of the present study was to investigate the effects of cyclic adenosine monophosphate (cAMP) on rat gastric antral circular smooth muscle function. Forskolin, a direct activator of adenylyl cyclase (AC), was used to observe the influences of cAMP. Multi-channel physiological recorder was used to record spontaneous contraction activity of gastric antral circular muscle from Wistar rats. And ELISA method was used to detect the change of cAMP production in perfusate. The results showed that forskolin concentration-dependently suppressed the amplitude and frequency of the spontaneous contr

FKBP12 deficiency reduces strength deficits after eccentric contraction-induced muscle injury.

Strength deficits associated with eccentric contraction-induced muscle injury stem, in part, from excitation-contraction uncoupling. FKBP12 is a 12-kDa binding protein known to bind to the skeletal muscle sarcoplasmic reticulum Ca2+ release channel [ryanodine receptor (RyR1)] and plays an important role in excitation-contraction coupling. To assess the effects of FKBP12 deficiency on muscle injury and recovery, we measured anterior crural muscle (tibialis anterior and extensor digitorum longus muscles) strength in skeletal muscle-specific FKBP12-deficient and wild-type (WT) mice before and aft
