BACKGROUND: We investigated the application of boron neutron capture therapy (BNCT) to suitable cancers other than glioblastoma and melanoma. Head and neck malignancies were consequently selected as adaptable cancers. We reported the clinical results of our first case treated and discussed several advantages to the application of BNCT to head and neck tumors.
METHODS: The patient was a 48-year-old woman with recurrent submandibular gland cancer. We confirmed the p-boronophenylalanine (BPA)-accumulating capacity of the tumor by fluorine-18-labeled p-boronophenylalanine positron emission tomogr

Metastatic spinal tumors are increasingly prevalent due to advancements in cancer treatment, leading to prolonged survival rates. This rising prevalence highlights the need for developing more effective therapeutic approaches to address this malignancy. Boron neutron capture therapy (BNCT) offers a promising solution by delivering targeted doses to tumors while minimizing damage to normal tissue. In this study, we evaluated the efficacy and safety of BNCT as a potential therapeutic option for spine metastases in mouse models induced by A549 human lung adenocarcinoma cells. The animal models we

This study introduces the MKM_B model, an approach derived from the MKM model, designed to evaluate the biological effectiveness of Boron Neutron Capture Therapy (BNCT) in the face of challenges from varying microscopic boron distributions. The model introduces a boron compensation factor, allowing for the assessment of compound Biological Effectiveness (CBE) values for different boron distributions. Utilizing the TOPAS simulation platform, the lineal energy spectrum of particles in BNCT was simulated, and the sensitivity of the MKM_B model to parameter variations and the influence of cell siz

Boron neutron capture therapy (BNCT) is a unique radiotherapy of selectively eradicating tumor cells using boron compounds (e.g., 4-borono-L-phenylalanine [BPA]) that are heterogeneously taken up at the cellular level. Such heterogenicity potentially reduces the curative efficiency. However, the effects of temporospatial heterogenicity on cell killing remain unclear. With the technical combination of radiation track detector and biophysical simulations, this study revealed the cell cycle-dependent heterogenicity of BPA uptake and subsequent biological effects of BNCT on HeLa cells expressing f

In recent years, various drug delivery systems circumventing the blood-brain barrier have emerged for treating brain tumors. This study aimed to improve the efficacy of brain tumor treatment in boron neutron capture therapy (BNCT) using cerebrospinal fluid (CSF) circulation to deliver boronophenylalanine (BPA) to targeted tumors. Previous experiments have demonstrated that boron accumulation in the brain cells of normal rats remains comparable to that after intravenous (IV) administration, despite BPA being administered via CSF at significantly lower doses (approximately 1/90 of IV doses). Three series of new boron-containing spermidine/spermine (SPD/SPM) analogues have been synthesized: N1- and N5-(4-carboranylbutyl) SPD/SPM derivatives (SPD-1, SPD-5, SPM-1, SPM-5); N1,N10-diethyl-N5-(4-carboranylbutyl)spermidine (DESPD-5), N1,N14-diethyl-N5-(4-carboranylbutyl)spermine (DESPM-5); and N5,N10-bis(4-carboranylbutyl)spermine (SPM-5,10). In vitro studies using rat F98 glioma cells have shown that these polyamines retain the ability to displace ethidium bromide from calf thymus DNA and are rapidly taken up by F98 glioma cells. However, their cytotoxicities, especially those with term

OBJECTIVE: Laryngeal preservation and a radical cure are the treatment goals for laryngeal carcinoma, and larynx-preserving therapy is generally preferred for early-stage laryngeal carcinoma. When laryngeal carcinoma recurs locally, patients are often forced to undergo total laryngectomy, resulting in loss of vocal function. However, many patients with laryngeal carcinoma who have residual or recurrent disease after radiotherapy wish to preserve their voice. The purpose of this study was to investigate the possibility of using BNCT as a larynx-preserving treatment for residual or recurrent lar

Boron neutron capture therapy requires sustained intracellular 10B accumulation, yet the clinical standard, L-boronophenylalanine (BPA, 1), rapidly effluxes from cells. We explored an intracellular retention strategy by converting borocaptate (BSH, 2) into a nuclear receptor ligand-based conjugate to improve intracellular retention of boron delivery agents. Retinoid X receptor (RXR) ligand scaffolds were selected as a proof-of-concept platform because RXRs are broadly expressed and predominantly nuclear. The lead conjugate CBTF-BSH (3), designed from fluorescent RXR antagonist 5, showed greate

INTRODUCTION: Boron neutron capture therapy (BNCT) is a targeted radiotherapy that represents a promising treatment for glioblastoma multiforme. BNCT efficacy depends on selective boron carriers capable of crossing the blood-brain barrier (BBB). Therefore, we propose an original strategy for boron delivery in BNCT, based on the cellular carriers. This study assessed the ability of bone marrow-derived macrophages (BMDMs) to accumulate boron carbide (B4C) nanoparticles, transport them across an in vitro BBB model toward glioblastoma cell-conditioned medium, and interact with tumor cells.

BACKGROUND: Glioblastoma (GBM) is the most aggressive and lethal primary brain tumor, characterized by poor prognosis and resistance to current therapies. Boron neutron capture therapy (BNCT) is a tumor-selective particle radiation therapy in which high-linear energy transfer (LET) α-particles and recoil 7Li nuclei are generated through the capture reaction between 10B and thermal neutrons. Although BNCT has been shown to prolong survival in GBM, its effects on global metabolomic alterations of the whole brain, particularly in relation to the tumor microenvironment (TME), remain incompletely u

Extracellular vesicles such as exosomes are expected to be used as drug delivery vehicles, particularly in personalized medicine, due to their pharmacological advantages. However, the development of functionalization techniques for isolated extracellular vesicles-including improving their intracellular uptake efficacy from the perspective of a drug delivery system and targeting disease-associated cells to reduce side effects-has become an urgent priority. The aim of this study was to develop a technology for modifying the surface of exosome membranes with functional molecules using membrane-cu

Boron neutron capture therapy (BNCT) relies on the selective uptake of boron-10 compounds by tumor cells. L-boronophenylalanine (BPA) serves as a key carrier, and enhancing its accumulation is critical for improving BNCT efficacy. In this study, we examined how L-phenylalanine (Phe) restriction affects BPA uptake and related cellular responses in the tumor cell lines SAS, U87-MG, PANC-1, and A375, as well as in the immortalized keratinocyte line HaCaT. Quantitative analysis using inductively coupled plasma atomic emission spectroscopy (ICP-AES) showed that 24 h Phe restriction increased BPA up

BACKGROUND: Boron neutron capture therapy (BNCT) is a cancer treatment that leverages the nuclear reaction between boron-10 ( 10 B $^{10}{\rm B}$ ) and thermal neutrons to generate high-energy α $\alpha$ particles and 7 Li $^{7}{\rm Li}$ nuclei that selectively destroy cancer cells while sparing healthy tissues. However, BNCT is limited by current dosimetry methods that are incapable of monitoring boron distribution during therapy. An imaging system capable of real-time dosimetry is essential for optimizing treatment efficacy and minimizing collateral damage.

BACKGROUND: Boron neutron capture therapy (BNCT) is a radiotherapeutic approach that can destroy cancer cells while sparing the surrounding normal cells. Currently, boronophenylalanine (BPA) is the most common boron delivery agent used in BNCT for treating recurrent cancers of the head and neck, gliomas, and melanomas. On the other hand, valproic acid (VPA) is one of the representative class I histone deacetylase inhibitors (HDACi), which is a promising sensitizer for cancer therapies. In this study, we aimed to verify whether VPA could induce an enhanced effect in destroying melanoma cells in

Cell death is a crucial endpoint in radiation-induced biological damage: on one side, cell death is a reference endpoint to characterise the action of radiation in biological targets; on the other side, any cancer therapy aims to kill tumour cells. Starting from Lea's target theory, many models have been proposed to interpret radiation-induced cell killing; after briefly discussing some of these models, in this paper, a mechanistic approach based on an experimentally observed link between chromosome aberrations and cell death was presented. More specifically, a model and a Monte Carlo code ori

Undifferentiated thyroid carcinoma (UTC) is a rapidly growing, highly invasive malignant tumor that currently lacks any effective treatment. Boron neutron capture therapy (BNCT) has been investigated recently for some types of tumors including glioblastoma multiforme and malignant melanoma. In previous studies we have shown the selective uptake of p-boronophenylalanine (BPA) by undifferentiated thyroid cancer cells in vitro and in vivo, as well as the histologic cure of 50% of the nude mice transplanted with human UTC cells when treated with BPA and an appropriate neutron beam. The present stu

Successful treatment of cancer by boron neutron capture therapy (BNCT) requires the selective delivery of (10)B to constituent cells within a tumor. The expression of the folate receptor is amplified in a variety of human tumors and potentially might serve as a molecular target for BNCT. In the present study we have investigated the possibility of targeting the folate receptor on cancer cells using folic acid conjugates of boronated poly(ethylene glycol) (PEG) containing 3rd generation polyamidoamine dendrimers to obtain (10)B concentrations necessary for BNCT by reducing the uptake of these c

The feasibility and safety of a second course of boron neutron capture therapy (BNCT) are unknown. In this study, we retrospectively analyzed the treatment outcomes and toxicities in recurrent head and neck cancer (RHNC) patients who underwent BNCT twice at Southern TOHOKU BNCT Research Center. In total, 33 patients who underwent BNCT twice for RHNC at Southern TOHOKU BNCT Research Center between June 2020 and March 2024 were included. The indications for second BNCT were local recurrence or residual tumor after the first course. Eligible patients either declined salvage surgery or had no cura
