Transcriptional responses to starvation stress in the hepatopancreas of oriental river prawn Macrobrachium nipponense.

Various crustaceans are farmed using aquaculture, and food deprivation or fasting can occur due to changing of environmental or management strategies. However, the molecular mechanisms underlying responses to starvation in crustaceans remain unclear. To address this, 12 hepatopancreas transcriptomes were compared for oriental river prawn (Macrobrachium nipponense) from four fasting stages (0, 7, 14 and 21 d). Gene Ontology functional annotation and Kyoto Encyclopaedia of Genes and Genomes pathway enrichment analysis of differentially expressed genes were subsequently performed. During the earl

Neuropeptide F alters expression of feeding, energy metabolism, and reproduction genes in female freshwater prawn, Macrobrachium rosenbergii.

Neuropeptide F (NPF) plays a crucial role in regulating various physiological processes in crustaceans; however, its underlying molecular mechanisms in regulating feeding, energy homeostasis, and reproduction in female prawn, Macrobrachium rosenbergii remain poorly understood. In this study, we investigated the impact of M. rosenbergii NPF (MrNPF) on the central nervous system (CNS) and digestive tract (DT) in female M. rosenbergii. A comparative transcriptome analysis was performed between MrNPF treatment and control groups for both tissues. We identified 545 differentially expressed genes (D

Differentiation among the Vibrio cholerae serotypes O1, O139, O141 and non-O1, non-O139, non-O141 using specific monoclonal antibodies with dot blotting.

Seven different monoclonal antibodies (MAbs) specific to only Vibrio cholerae were produced using a combination of five representative serotypes of V. cholerae for immunization. The first three MAbs (VC-93, VC-82 and VC-223) were specific to the V. cholerae serogroup O1 with different avidity for the serotypes O1 Inaba and O1 Ogawa. The fourth and the fifth MAbs were specific to V. cholerae O139 (VC-812) or O141 (VC-191) serogroups, respectively. The sixth MAb (VC-26) bound to all three serogroups of V. cholerae. The seventh MAb (VC-63) bound to all twenty five isolates of V. cholerae used in

A single-nucleus transcriptomic atlas of early marine shrimp embryogenesis reveals cell-type specification and eye pigment transport mechanisms.

Early embryogenesis and their underlying regulatory mechanisms in crustaceans remains unclear at the cellular level. Here, we mapped the early developmental stages of marine shrimp Palaemon carinicauda using single nucleus RNA sequencing (snRNA-seq), spanning the transitional stages of blastula to protozoea. Profiling over 60,000 nuclei, we identify six distinct embryonic cell types, including the multiple neuronal subtypes, extracellular matrix- producing cells, cuticle-forming cells, and an eye-related cells that emerged during protozoea development. Cell-type identities were validated using

Identification of a novel spätzle in Macrobrachium rosenbergii and its regulatory effects on the immune responses against Vibrio parahaemolyticus and Vibrio alginolyticus.

As a cytokine-like ligand, Spätzle activates the Toll signaling pathway through binding to Toll-like receptors (TLRs), thereby participating in the innate immune defense of crustaceans against diverse pathogens. A novel Spätzle (MrSpz5) was cloned from Macrobrachium rosenbergii in this study. The MrSpz5 cDNA is 1900 bp, comprising a 972-bp open reading frame (ORF) encoding 323 amino acids, a 662-bp 5' UTR, and a 266-bp 3' UTR. MrSpz5 was constitutively expressed in various tissues, with the highest expression in the gills. Knockdown of MrSpz5 via RNA interference in M. rosenbergii significantl

BPA disrupts crustacean reproduction: Tissue-specific accumulation, oxidative stress, and gene dysregulation in Macrobrachium nipponense.

Bisphenol A (BPA), a pervasive environmental endocrine disruptor, increasingly threatens aquatic ecosystems due to its reproductive toxicity. This study investigates how BPA disrupts ovarian development in Macrobrachium nipponense - by analyzing its tissue-specific bioaccumulation (ovaries, hepatopancreas, fertilized eggs), antioxidant responses, and developmental gene regulation. Experimental results demonstrated that after 10-day exposure to 10, 100, and 1000 μg/L BPA, significant BPA accumulation was observed in both ovaries and hepatopancreas, while fertilized eggs showed notable accumulat

Identification and characterization of the Dmrt1B gene in the oriental river prawn, Macrobrachium nipponense.

Dmrt (doublesex and mab-3 related transcription factor) is a protein family of transcription factors implicated in sexual regulation. Dmrt proteins are widely conserved and known for their involvement in sex determination and differentiation across species, from invertebrates to humans. In this study, we identified a novel gene with a DM (doublesex/Mab-3)-domain gene in the river prawn, Macrobrachium nipponense, which we named MniDmrt1B due to its similarities and close phylogenetic relationship with Dmrt1B in Macrobrachium rosenbergii. Through amino acid alignments and structural predictions,

Metal toxicity, uptake and bioaccumulation in aquatic invertebrates--modelling zinc in crustaceans.

We use published data on the different patterns of the bioaccumulation of zinc by three crustaceans, the caridean decapod Palaemon elegans, the amphipod Orchestia gammarellus and the barnacle Amphibalanus amphitrite, to construct comparative biodynamic models of the bioaccumulation of zinc into metabolically available and detoxified components of accumulated zinc in each crustacean under both field and laboratory toxicity test conditions. We then link these bioaccumulation models to the onset of toxic effects on exposure of the crustaceans to high dissolved zinc bioavailabilities, using the te

Antiviral activity of cathelicidin 5, a peptide from Alligator sinensis, against WSSV in caridean shrimp Exopalaemon modestus.

White spot disease caused by white spot syndrome virus (WSSV) is responsible for harming shrimp aquaculture industry and results in a pandemic throughout the world. Cathelicidin 5 treatment enhanced immune parameters including antioxidant enzyme activity and immune-related genes expression in shrimp Exopalaemon modestus. Shrimp treated with cathelicidin 5 and inoculated with white spot syndrome virus (WSSV) exhibited a significantly lower mortality rate and lower viral VP28 amplification and expression than control. This study addresses the role of cathelicidin 5 in immune stimulatory and anti

Characterization of systemic allergenicity of tropomyosin from shrimp (Macrobrachium nipponense) and anaphylactic reactions in digestive tract.

BACKGROUND: Tropomyosin (TM) is the major allergen of crustaceans. The allergenicity of TM from Macrobrachium nipponense (MnTM) and the anaphylactic reaction in the digestive tract are still unclear. The aim of this study was to evaluate the allergenicity of MnTM and the anaphylactic reaction in the digestive tract. RESULTS: Serum IgE and IgG1 binding ability in the TM group were significantly higher than those in the PBS and CT groups (P < 0.01) and CP group (P < 0.05), while serum IgG and IgG2a binding ability showed no obvious difference between the four groups (P > 0.05). Experimental vertical transmission of Macrobrachium rosenbergii nodavirus (MrNV) and extra small virus (XSV) from brooders to progeny in Macrobrachium rosenbergii and Artemia.

White tail disease (WTD) is a serious problem in hatcheries and nursery ponds of Macrobrachium rosenbergii in India. Experiments were carried out to determine the possibility of vertical transmission of M. rosenbergii nodavirus (MrNV) and extra small virus (XSV) in M. rosenbergii and Artemia. Prawn broodstock inoculated with MrNV and XSV by oral or immersion challenge survived without any clinical signs of WTD. The brooders spawned 5-7 days after inoculation and the eggs hatched. The survival rate of larvae gradually decreased, and 100% mortality was observed at the post-larvae (PL) stage. Whi

Cellular signalling of PCH-induced pigment aggregation in the crustacean Macrobrachium potiuna erythrophores.

The cellular system responsible for the transduction of the pigment-concentrating hormone (PCH) signal was investigated in erythrophores of the freshwater shrimp, Macrobrachium potiuna. Dose-response curves to the hormone were determined in the absence and in the presence of several drugs that affect sequential steps of the Ca(2+)-dependent signalling pathway. Additionally, the ability of forskolin to induce pigment dispersion was evaluated. Neomycin sulphate (10(-4) and 10(-3) mol.l-1), trifluoperazine (10(-5) and 10(-4) mol.l-1), 1-(5-isoquinolinesulfonlyl)-2-methylpiperazine (10(-7) and 10(

ATG12 contributes to antibacterial immunity against Aeromonas veronii in Macrobrachium rosenbergii.

Autophagy is a central component of host immune defense against various infections. Autophagy-related protein 12 (ATG12) is a key regulator of autophagy, but its role in antibacterial immunity remains unclear. In this study, Macrobrachium rosenbergii ATG12 (MrATG12) was cloned, and its role in response to Aeromonas veronii challenge was investigated. The open reading frame of MrATG12 was 360 bp in length and encoded a polypeptide of 119 amino acids. Sequence comparison revealed that MrATG12 from M. rosenbergii shared the highest sequence identity with Palaemon carinicauda. Tissue distribution

Reduced transmission of human schistosomiasis after restoration of a native river prawn that preys on the snail intermediate host.

Eliminating human parasitic disease often requires interrupting complex transmission pathways. Even when drugs to treat people are available, disease control can be difficult if the parasite can persist in nonhuman hosts. Here, we show that restoration of a natural predator of a parasite's intermediate hosts may enhance drug-based schistosomiasis control. Our study site was the Senegal River Basin, where villagers suffered a massive outbreak and persistent epidemic after the 1986 completion of the Diama Dam. The dam blocked the annual migration of native river prawns (Macrobrachium vollenhoven

Potential use of the Macrobrachium rosenbergii lectin for diagnosis of T-cell acute lymphoblastic leukemia.

T-cell acute lymphoblastic leukemia is the most common form of cancer in children. Lectins are proteins or glycoproteins from plants or animals that recognize oligossacharides on the cell surface and have been used to characterize the structural changes of oligosaccharides in leukemias. In this study, we used the lectin from the freshwater prawn Macrobrachium (M. rosenbergii), specific for acetyl groups in sialylated glycans, because increased sialylation of glycoproteins and glycolipids has been identified in lymphoblastic leukemias. We compared the specificity of the M. rosenbergii lectin fo

Regulation of AMPs expression by six Tolls in stomach during WSSV infection in Macrobrachium nipponense.

Tolls or Toll-like receptors (TLRs) are involved in innate immunity in invertebrates and vertebrates. In this study, six Toll genes (named MnToll1-6) were found in the oriental river prawn Macrobrachium nipponense, and MnToll1-4 were newly identified. All six Tolls have a transmembrane domain and a TIR domain, containing different numbers of leucine-rich repeats (LRR), LRR C-terminal, and LRR N-terminal domains. They can be classified into single cysteine cluster Toll, multiple cysteine clusters Toll, and MnToll2 (Toll without a cysteine cluster), and are located in the six branches (groups A-

Association of riverine prawns and intermediate host snails and correlation with human schistosomiasis in two river systems in south-eastern Côte d'Ivoire.

The current emphasis of schistosomiasis control is placed on preventive chemotherapy using praziquantel. However, reinfection may occur rapidly in the absence of complementary interventions. Recent studies from Senegal suggest that predatory prawns might feed on intermediate host snails and thus impact on schistosomiasis transmission. We designed a study with four repeated cross-sectional surveys pertaining to prawns and snails, coupled with a single cross-sectional parasitological survey among humans. We assessed for potential associations between the presence/density of prawns and snails and
