Neurotoxicity aims to understand how xenobiotics interfere with the function of the nervous system and to unravel their mechanisms of action. Neuronal activity is the primary functional output of the nervous system and deviations from its resting level may indicate toxicity. Consequently, the monitoring of electrophysiological activity in complex cell culture systems appears particularly promising for neurotoxicity assessment. To detect acute neurotoxic effects of chemicals we developed a test system based on the electrophysiological recordings from neural networks in re-aggregating brain cell

Organophosphorus (OP) and N-methylcarbamate pesticides inhibit acetylcholinesterase (AChE), but differences in metabolism and detoxication can influence potency of these pesticides across and within species. Carboxylesterase (CaE) and A-esterase (paraoxonase, PON1) are considered factors underlying age-related sensitivity differences. We used an in vitro system to measure detoxication of AChE-inhibiting pesticides mediated via these esterases. Recombinant human AChE was used as a bioassay of inhibitor concentration following incubation with detoxifying tissue: liver plus Ca(+2) (to stimulate P

Susceptibility to organophosphorus (OP) insecticides and nerve agents is strongly influenced by genetic and developmental factors. A number of organophosphorothioate insecticides are detoxified in part via a two-step pathway involving bioactivation of the parent compound by the cytochrome P450 systems, then hydrolysis of the resulting oxygenated metabolite (oxon) by serum and liver paraoxonases (PON1). Serum PON1 has been shown to be polymorphic in human populations. The Arg192 isoform (PON1R192) of this HDL-associated protein hydrolyzes paraoxon (POX) at a high rate, while the Gln192 isoform

Serum paraoxonase (aryldialkylphosphatase, EC 3.1.8.1., PON1) is an esterase protein synthesised by the liver and released into the serum, where it is associated with HDL lipoproteins. In this study, we have determined the in vitro effects of the following antibiotics: sodium ampicillin, ciprofloxacin, Rifamycin SV and clindamiycin phosphate, on human hepatoma (HepG2) cells (liver hPON1). All the antibiotics caused a dose-dependent and time-dependent decrease in the paraoxonase activity while Rifamycin SV was the most effective antibiotic due to its low 50% inhibition concentration (IC(50)) va

Experiments were carried out on mice between the 14th and the 19th day following the infection with Trichinella larvae. E 600 was given in the lowest lethal dose (0-8 mg/kg) and Toxobidin in a curative dose of 15 mg/kg. The activities of AChE and PChE were examined with the use of the substrates of acetylthiocholine and butrylthiocholine iodides (Koelle-Friedenwald's method in Gomori's modification). Toxobidin was found to influence significantly restitution of AChE activity. The mice which had received E 600 under the protection of Toxobidin exhibited but a slight degree of inhibition of AChE

Immobilized enzymes offer several advantages for biosensing, including high specificity and reusability, but their practical implementation is often hindered by a significant loss in substrate binding or other catalytic activity upon immobilization. In this work, we have demonstrated a simple and robust method for the detection of organophosphate pesticides (OPs) using acetylcholinesterase (AChE) directly adsorbed on bare Au surfaces. Our sensor exhibits a remarkably low limit of detection (149 pM) for paraoxon (POx), a model organophosphate, retains ∼70% of its activity after multiple cycles

The M2 subtype of muscarinic receptor is predominant in heart, and such receptors were reported to be located in muscles as well as in presynaptic cholinergic and adrenergic nerve terminals. Muscarinic receptors of rat heart were identified by the high affinity binding of the agonist (+)-[3H]cis-methyldioxolane ([3H]CD), which has been used to label a high affinity population of M2 receptors. A single population of sites (KD 2.74 nM; Bmax of 82 fmol/mg protein) was detected and [3H]CD binding was sensitive to the M2 antagonist himbacine but much less so to pirenzepine, the M1 antagonist. These

Paraoxonase 1 (PON1) is a hydrolyzing paraoxon enzyme; it prevents the development of oxidative stress damage. PON1 primarily binds to high-density lipoprotein (HDL). Although PON1 is not directly expressed in the brain, it crosses the blood-brain barrier (BBB) via HDL, potentially exerting neuroprotective effects. It has been shown that the reduction of PON1 activity is associated with the development and progression of Alzheimer's disease (AD) and other neurodegenerative diseases by inducing oxidative stress and neuroinflammation. Despite these findings, the exact role of PON1 in AD and othe

Metabolic disorders are characterized by an overall state of inflammation and oxidative stress, which highlight the importance of a functional antioxidant system and normal activity of some endogenous enzymes, namely paraoxonase-1 (PON1). PON1 is an antioxidant and anti-inflammatory glycoprotein from the paraoxonases family. It is mainly expressed in the liver and secreted to the bloodstream, where it binds to HDL. Although it was first discovered due to its ability to hydrolyze paraoxon, it is now known to have an antiatherogenic role. Recent studies have shown that PON1 plays a protective ro

The early and late effect of three anticholinesterase agents (physostigmine, paraoxon and soman) on core temperature and brain acetylcholinesterase (AcChE) inhibition are compared. The study was performed in adult male rats using sublethal doses of all drugs. AcChE activity was determined by hypothalamus, striatum, hippocampus, medulla oblongata-pons and rest of the brain. The co-existence of intoxication symptoms, hypothermia and AcChE inhibitions was clearly shown by the early effects. However, no relationship was found between the degree of hypothermia and that of inhibition whatever brain

Intoxication by organophosphate (OP) nerve agents and pesticides should be addressed by efficient, quickly deployable countermeasures such as antidotes reactivating acetylcholinesterase or scavenging the parent OP. We present here synthesis and initial in vitro characterization of 14 imidazole aldoximes and their structural refinement into three efficient reactivators of human butyrylcholinesterase (hBChE) inhibited covalently by nerve agent OPs, sarin, cyclosarin, VX, and the OP pesticide metabolite, paraoxon. Rapid reactivation of OP-hBChE conjugates by uncharged and nonprotonated tertiary i

1. An organism that utilizes acetophenone as sole source of carbon and energy was isolated in pure culture and tentatively identified as an Arthrobacter sp. 2. Cell-free extracts of the acetophenone-grown organism contained an enzyme, acetophenone oxygenase, that catalysed an NADPH-dependent consumption of O(2) in the presence of the growth substrate; approx. 1mol of O(2) and 1mol of NADPH were consumed per mol of acetophenone oxidized. 3. Cell-free extracts also contained an enzyme capable of the hydrolysis of phenyl acetate to phenol and acetate. The amount of this esterase was increased mar

Lightweight and wearable fabrics with rapid self-detoxification functions are highly desired to resist chemical warfare agents (CWAs). Metal organic frameworks (MOFs) with high specific surface area and customizability are singularly attractive because of their ability to effectively capture and catalytically degrade CWAs. Herein, photothermal graphene-based nanocomposite fabrics are designed by wet-spinning and chemical reduction of graphene oxide fibers followed by in situ growth of UiO-66-NH2. The flexible graphene fabrics decorated with UiO-66-NH2 nanoparticles exhibit an ultrafast phototh

Pesticides play a crucial role in crop production. However, they also pose significant pollution and health risks. This study presents a novel dual-mode cascade strategy for detecting pesticide residues, addressing the growing demand for improved safety control. The proposed method utilizes a bifunctional metal-organic framework nanozyme (BMFN) with both fluorescence and biomimetic capabilities, enabling efficient analysis of pesticide residues. The bifunctionalities of BMFN is achieved by modulating Ce3+/Ce4+ valence states and adjusting reaction conditions. The dual functionality is activate

The optical transducer of CdTe semiconductor quantum dots (QDs) has been integrated with acetylcholinesterase enzyme (AChE) by the layer-by-layer (LbL) assembly technique, resulting in a highly sensitive biosensor for detection of organophosphorus pesticides (OPs) in vegetables and fruits based on enzyme inhibition mechanism. The detection limits of the proposed biosensors are as low as 1.05 × 10(-11) M for paraoxon and 4.47 × 10(-12) M for parathion, which are significantly better than those of the conventional GC/MS methods or amperometric biosensors (0.5 nM). These biosensors are used for q

Butyryl cholinesterase of different origin along with variations of the time of enzyme immobilization on the potentiometric transducer surface is offered to control the ion sensitive field effect transistor (ISFET)-based biosensor sensitivity. Because butyryl cholinesterase has been already used to develop the sensors for heavy metals, organophosphorus/carbamate pesticides, and steroidal glycoalkaloids analysis, the present study has been focused on the investigation and adjustment of the ISFET-based biosensor specificity exclusively to the glycoalkaloids. More recently, electrochemical sensors developed for the universal detection of organophosphorus pesticide (OPs) usually rely on the bioenzyme-inhibition principle and intricate catalytic cascade reactions. In this work, we developed an efficient direct electrochemical sensor of OPs by using carbon nanotubes coated with NH2-UiO-66 MOF (CNTs@NH2-UiO-66) as multifunctional electrode material, which could effectively eliminate the need for bioenzymes and cascade reactions to achieve universal detection. In this proposal, the NH2-UiO-66 MOF exhibits intrinsic electroactivity as signal probe and sp

The single residue mutation of butyrylcholinesterase (BChEG117H) hydrolyzes a number of organophosphosphorus (OP) anticholinesterases. Whereas other BChE active site/proximal mutations have been investigated, none are sufficiently active to be prophylactically useful. In a fundamentally different computer simulations driven strategy, we identified a surface peptide loop (residues 278-285) exhibiting dynamic motions during catalysis and modified it via residue insertions. We evaluated these loop mutants using computer simulations, substrate kinetics, resistance to inhibition, and enzyme reactiv
