Plants employ receptor-like kinases (RLKs) and receptor-like proteins for rapid recognition of invading pathogens, and RLKs then transmit signals to receptor-like cytoplasmic kinases (RLCKs) to activate immune responses. RLKs are under fine regulation mediated by subcellular trafficking, which contributes to proper activation of plant immunity. In this study, we show that Arabidopsis thaliana RECEPTOR-LIKE KINASE 902 (RLK902) plays important roles in resistance to the bacterial pathogen Pseudomonas syringae, but not to the fungal powdery mildew pathogen Golovinomyces cichoracearum. RLK902 loca

Pathogen-associated molecular pattern (PAMP)-trigged immunity (PTI) is the first defensive line of plant innate immunity and is mediated by pattern recognition receptors. Here, we show that a mutation in BR-SIGNALING KINASE1 (BSK1), a substrate of the brassinosteroid (BR) receptor BRASSINOSTEROID INSENSITIVE1, suppressed the powdery mildew resistance caused by a mutation in ENHANCED DISEASE RESISTANCE2, which negatively regulates powdery mildew resistance and programmed cell death, in Arabidopsis thaliana. A loss-of-function bsk1 mutant displayed enhanced susceptibility to virulent and avirule

Given the dearth of effective antiviral drugs, the exogenous delivery of dsRNA for RNAi against plant viral diseases holds great promise. Here, we present an effective delivery approach of dsRNA utilizing graphene oxide nanosheets (GONs) on mature plant leaves via a spray. Our method achieves rapid and sustained gene knockdown, reducing the level of the target gene to 46% by day 2, and continuously releases dsRNA for at least 6 days. The coupling of GONs with specific fragments of coat protein and replicase gene dsRNA exhibited a superior antiviral effect compared to specific fragments of RNA-

Plants pass transgenerational inheritance through genetic and epigenetic information; however, the soil's role as the host environment in this memory transfer remains largely unexplored. Research within this field has largely been domain-specific, with only 117 publications in 24 years integrating fields of soil persistent metabolites (soil chemistry), stress-induced microbial consortia (soil microbiology), and plant molecular science (plant epigenetics). In this perspective article, we synthesize evidence and propose a testable hypothesis, the Hologenomic Stress Legacy (HSL) hypothesis. It st

Extracellular Adenosine 5'-triphosphate (eATP) is a damage-associated molecular pattern in both plants and animals, playing an important role as a danger signal in response to various environmental stresses. eATP works both independently and in coordination with other signaling pathways. Here, we identify a previously uncharacterized proline-rich extensin-like receptor kinase 15 (PERK15), which interacts with the plant eATP receptor P2K1 and integrates eATP and salicylic acid (SA)-mediated pathogen defense responses. perk15 mutant plants display reduced eATP responses and compromised pathogen

Plants deploy a two-layered immune system, comprising pattern-triggered immunity (PTI) and effector-triggered immunity (ETI), to defend against pathogens. Although PTI and ETI pathways converge on similar downstream responses, they are activated by distinct molecules at the cell surface and in the cytoplasm, respectively. Here, we report that the harpin-like effector RipW from Ralstonia solanacearum localizes to both the apoplastic and intracellular compartments, where it acts as a dual elicitor capable of activating PTI-like and ETI-like responses. We show that RipW triggers PTI-like response

Ulcerative colitis (UC) is a condition with complex immune dysregulation and chronic intestinal inflammation. Currently, there are still a lack of natural and low-toxicity therapeutic approaches for UC. Emerging research has underscored the potential of plant-derived exosomes for UC treatment. In this study, it is found that Centella Asiatica-derived exosomes (CAEs) have anti-colitis effects. CAEs selectively accumulate in inflamed colons, enabling targeted delivery of RNA cargos. MicroRNA (miRNA)-enriched CAEs not only mitigate inflammation but also facilitate the reconstitution of a balanced

Reactive nitrogen species (RNS) play an important role in plant immunity as signaling factors. We previously developed a plasma technology to partially convert air molecules into dinitrogen pentoxide (N2O5), an RNS whose physiological action is poorly understood. To reveal the function of N2O5 gas in plant immunity, Arabidopsis thaliana was exposed to plasma-generated N2O5 gas once (20 s) per day for 3 days, and inoculated with Botrytis cinerea, Pseudomonas syringae pv. tomato DC3000 (Pst), or cucumber mosaic virus strain yellow (CMV(Y)) at 24 h after the final N2O5 gas exposure. The apicomplexan parasite Toxoplasma gondii produces the plant hormone abscisic acid, but it is unclear if phytohormones are produced by the malaria parasite Plasmodium spp., the most important parasite of this phylum. Here, we report detection of salicylic acid, an immune-related phytohormone of land plants, in P. berghei ANKA and T. gondii cell lysates. However, addition of salicylic acid to P. falciparum and T. gondii culture had no effect. We transfected P. falciparum 3D7 with the nahG gene, which encodes a salicylic acid-degrading enzyme isolated from plant-infecting Pseudomonas sp., and

Light and chloroplast function is known to affect the plant immune response; however, the underlying mechanism remains elusive. We previously demonstrated that two light signaling factors, FAR-RED ELONGATED HYPOCOTYL 3 (FHY3) and FAR-RED IMPAIRED RESPONSE 1 (FAR1), regulate chlorophyll biosynthesis and seedling growth via controlling HEMB1 expression in Arabidopsis thaliana. In this study, we reveal that FHY3 and FAR1 are involved in modulating plant immunity. We showed that the fhy3 far1 double null mutant displayed high levels of reactive oxygen species and salicylic acid (SA) and increased

BACKGROUND: Pathogen infection triggers a large-scale transcriptional reprogramming in plants, and the speed of this reprogramming affects the outcome of the infection. Our understanding of this process has significantly benefited from mutants that display either delayed or accelerated defense gene induction. In our previous work we demonstrated that the Arabidopsis Elongator complex subunit 2 (AtELP2) plays an important role in both basal immunity and effector-triggered immunity (ETI), and more recently showed that AtELP2 is involved in dynamic changes in histone acetylation and DNA methylati

Sclerotinia sclerotiorum is a necrotrophic fungus that causes severe plant diseases but can also colonize wheat as a symptomless endophyte. However, the mechanism by which its effector proteins regulate this dual lifestyle remains unclear. This study aimed to identify key LysM effectors involved in S. sclerotiorum-plant interactions and elucidate their specific functions. Among seven SsLysM genes, SsLysM6 and SsLysM7 were transcriptionally upregulated during both the pathogenic infection of rapeseed and the endophytic colonization of wheat. The ΔSsLysM6 and ΔSsLysM7 mutants exhibited significa

Sensing of pathogen-associated molecular patterns (PAMPs) by pattern recognition receptors at the cell surface initiates the first layer of host immunity against invading microbial pathogens. In plants, the immune receptor Flagellin Sensing 2 (FLS2) perceives bacterial flagellin to trigger downstream signaling that culminates in PAMP-triggered immunity (PTI). FLS2 protein levels are critical for effective PTI, yet the regulatory mechanisms governing its stability remain to be fully elucidated. Our previous work showed that the Arabidopsis Really Interesting Gene (RING) type ubiquitin ligase XB

Colletotrichum asianum, the cause of mango anthracnose, is a major threat to mango production worldwide. However, the mechanisms underlying plant invasion by C. asianum are poorly understood. Mini-chromosomes are increasingly recognised as indispensable components in the virulence of plant-pathogenic fungi. Here, we identify CaMutA, a secreted glycoside hydrolase 71 effector encoded on a mini-chromosome, as a critical pathogenicity factor that suppresses plant immune responses and is essential for infection by C. asianum. CaMutA directly interacts with the mango chitinase MiChi1. When heterolo

Salicylic acid (SA) plays important roles in plant local and systemic resistance. Isochorismate synthase 1 (ICS1) is a key enzyme in SA synthesis. Pathogens infection triggered the ICS1 expression and induced SA production. However, the molecular regulation mechanism of ICS1 against virus infection remains unclear. Here, we employed molecular genetics and physiobiochemical approaches to confirm a transcription factor NbNAC1 from Nicotiana benthamiana is a positive regulator of resistance against tobacco mosaic virus (TMV). The pathways NbNAC1 and NbICS1 can be triggered by TMV infection. Silen

Zika virus infection causes multiple clinical issues, including Guillain-Barré syndrome and neonatal malformation. Vaccination is considered as the only strategy for the prevention of ZIKV-induced clinical issues. This study developed a plant-based recombinant vaccine that transiently expressed the ZIKV envelope protein (ZikaEnv:aghFc) in Nicotiana benthamiana and evaluated the protective immunity afforded by it in immunocompetent mice. ZikaEnv:aghFc induced both humoral and cellular immunity at a low dose (1-5 μg). This immune-inducing potential was enhanced further when adjuvanted CIA09A. In

Tobacco mosaic virus (TMV) is the most widely spread and harmful virus in the world, causing serious economic losses annually. However, the low anti-erosion ability of the pesticides for TMV management make it easy to be washed by the rain, which makes the effective duration of the pesticides shorter. In this paper, a new bio-based nanogel with superior antiviral activity was reported, and its slow-release behavior, rain erosion resistance and the antiviral mechanism was systematically studied. The results determined that the nanogels (Zn2+@ALGNP and Zn2+@ALGNP@PL) exhibited sustained releasin

Jijie Chai earned his Ph.D. in analytical chemistry from the Institute of Materia Medica, Chinese Academy of Medical Sciences, and conducted postdoctoral research at Princeton University. In 2004, he established his independent research group at National Institute of Biological Science (China), focusing on plant immune receptors, particularly nucleotide-binding leucine-rich repeat (NLR) proteins. NLRs are the largest family immune receptors that enable plants to detect a wide array of pathogen effectors and activate effector-triggered immunity (ETI). Despite their recognition diversity, they e

Bacterial quorum-sensing (QS) molecules are one of the primary means allowing communication between bacterial cells or populations. Plants also evolved to perceive and respond to those molecules. N-acyl homoserine lactones (AHL) are QS molecules, of which impact has been extensively studied in different plants. Most studies, however, assessed the interactions in a bilateral manner, a nature of interactions, which occurs rarely, if at all, in nature. Here, we investigated how Arabidopsis thaliana responds to the presence of different single AHL molecules and their combinations. We assumed that

Gram-negative bacteria continuously pinch off portions of their outer membrane, releasing membrane vesicles. These outer membrane vesicles (OMVs) are involved in multiple processes including cell-to-cell communication, biofilm formation, stress tolerance, horizontal gene transfer, and virulence. OMVs are also known modulators of the mammalian immune response. Despite the well-documented role of OMVs in mammalian-bacterial communication, their interaction with plants is not well studied. To examine whether OMVs of plant pathogens modulate the plant immune response, we purified OMVs from four di
