Protein corona composition and precise physiological understanding of differentially expressed proteins are key for identifying disease biomarkers. In this report, we presented a distinctive quantitative proteomics table of molecular cell signaling differentially expressed proteins of corona that formed on iron carbide nanoparticles (NPs). High-performance liquid chromatography/electrospray ionization coupled with ion trap mass analyzer (HPLC/ESI-Orbitrap) and MASCOT helped quantify 142 differentially expressed proteins. Among these proteins, 104 proteins showed upregulated behavior and 38 pro

Alzheimer's disease (AD) is the most severe form of neurological disorder, characterized by the presence of extracellular amyloid-β (Aβ) plaques and intracellular tau tangles. For decades, therapeutic strategies against the pathological symptoms of AD have often relied on the delivery of monoclonal antibodies to target specifically Aβ amyloid or oligomers, largely to no avail. Aβ can be traced in the brain as well as in cerebrospinal fluid and the circulation, giving rise to abundant opportunities to interact with their environmental proteins. Using liquid chromatography tandem-mass spectromet

One of the main diseases contributing to human death are malignant tumors. Phototherapy is a promising approach for cancer therapy, and functional nanoparticles with targeting ligands are commonly used to improve the therapeutic efficiency. However, recent studies have shown that nanoparticles in contact with a biological fluid can rapidly form a "protein corona" on their surface, which will remarkably decrease the targeting ability. Here, we describe the preparation of hybrid nanomaterials with Bi2S3 nanorods as the core, and fluorescein-isothiocyanate and folic acid-modified human serum albu

Ulcerative colitis (UC) is a complex inflammatory bowel disease characterized by multiple factors. Alleviating inflammation is the primary therapeutic approach. However, currently employed anti-inflammatory treatments have limited efficacy and cause side effects. Safer, more effective therapies are needed. Selenium nanosheets (SeNSs) are biocompatible, anti-inflammatory, and low-toxicity nanomaterials with high surface areas and abundant active sites, making them potential therapeutic agents for UC. This study indicates that SeNSs can interact with macrophages and adhere to their cell membrane

Engineered nanoparticles are widely used in biological imaging and drug delivery because of their excellent physical and chemical properties, but almost all the original functions of engineered nanoparticles suffer from a complex matrix. Herein, we proposed a strategy of preparing nanoparticle protein corona antigens (NPCAgs) through exposing a magnetic core silicon shell (Fe3O4@SiO2) fluorescent probe to an antigen protein solution, which could reduce the adsorption of nanoparticles (NPs) with other proteins in serum. In the presence of target anti-BSA IgG, a competitive-type displacement rea

Nanomaterials are interesting candidates for applications in medicine as drug delivery or diagnostic agents. For safe application, they have to be evaluated in in vitro and in vivo models to finally be translated to human clinical trials. However, often those transfer processes fail, and it is not completely understood whether in vitro models leading to these animal models can reliably be compared to the situation in humans. In particular, the interaction of nanomaterials with components from different blood plasma sources is difficult to compare, and the outcomes of those interactions with re

We report the synthesis of micellar phosphatidylcholine-coated superparamagnetic iron oxide nanoparticles as a new long circulation contrast agents for magnetic resonance imaging. Oleic acid-coated Fe3 O4 nanoparticles were first prepared through thermal degradation and then encapsulated into small clusters with a phosphatidylcholine coating to obtain hydrophilic nanomicelles. A thorough characterization confirmed the chemical nature of the coating and the excellent colloidal stability of these nanomicelles in aqueous media. Magnetization and relaxivity properties proved their suitability as m

Polymer coatings are widely used to suppress nonspecific protein adsorption and modulate nanoparticle-cell interactions. However, the biological outcomes of polymer-functionalized nanoparticles remain difficult to predict in complex biological environments because protein corona formation rapidly alters nanoparticle identity. This challenge is particularly pronounced when hydrophobic moieties are incorporated for imaging or therapeutic applications, as they can promote nonspecific interactions at the nano-bio interface. Herein, we establish a modular grafting-to platform based on cellulose nan

The "biological identity" of a nanoparticle is determined not only by its surface but by the complex protein corona (PC) it forms when entering a biological system. There have been significant advances in nanoparticle (NP) design that overcome various barriers, however, the protein corona remains a major challenge in the field of nanotechnology. Despite the use of various analytical techniques and a wide range of experimental conditions in protein corona research, there is a critical need for standardized, high-resolution methods to achieve the most accurate and comprehensive characterization

This study describes synthesis of N‑acetyl‑l‑cysteine-capped CdTe quantum dots (QDs) and investigates their interaction with plasma protein fibrinogen (FIB) and the structural changes of FIB. It is shown that the interaction of QDs with FIB is a spontaneous process and the major driving forces are van der Waals forces and hydrogen bonds. Multi-spectroscopic measurements show that the intrinsic fluorescence of FIB was quenched and secondary and tertiary structures were altered due to the interaction with QDs. In addition, the aggregation state of FIB was altered in the presence of QDs. Furtherm

Nanotechnology has been an important tool for the production of nanoparticles with controlled release of drugs for therapeutic applications. Here, we produced solid lipid nanoparticles (SLN) loaded with curcumin and capsaicin (NCC) following the overarching goals of green chemistry. Currently, besides evaluating the composition, and size of these, it is necessary to understand the interactions between nanoparticles and the biomolecules present in the biological medium. For this, assays were conducted in order to evaluate the potential formation of the protein 'corona', and to better understand

Extensive exploitation of titanium dioxide nanoparticles (TiO2NPs) augments rapid release into the marine environment. When in contact with the body fluids of marine invertebrates, TiO2NPs undergo a transformation and adhere various organic molecules that shape a complex protein corona prior to contacting cells and tissues. To elucidate the potential extracellular signals that may be involved in the particle recognition by immune cells of the sea urchin Paracentrotus lividus, we investigated the behavior of TiO2NPs in contact with extracellular proteins in vitro. Our findings indicate that TiO

Silica nanoparticles (SiNPs), widely used in diverse industrial and biomedical applications, are ubiquitous environmental pollutants of emerging concern, posing significant risks to human respiratory health via inhalation exposure. Substantial evidence demonstrates that inhaled SiNPs induce pulmonary fibrosis. Nevertheless, the underlying pathogenic mechanisms remain elusive. While nanomaterials adsorb proteins to form a protein corona that alters their biological activity and contributes to nanoparticle-induced tissue damage, whether this process contributes to SiNPs-induced pulmonary fibrosi

The wide applications of proteinosomes as protocells and nanoreactors benefit from their capsule-like structures and protein functions. In regular proteinosomes, the hydrophilic protein corona can be denatured in unfavorable conditions, such as extreme pH and protease. Herein, proteinosomes with a PEG protein mixed corona are prepared to protect the protein's biofunction. The block copolymer polyethylene glycol-block-poly(di(ethylene glycol) methyl ether methacrylate) (PEG-b-PDEGMA) bearing an adamantyl group at the junction undergoes molecular recognition with β-cyclodextrin-modified proteins

Parkinson's disease (PD) is characterized by the loss of dopaminergic neurons along with the accumulation of amyloid plaques with alpha-synuclein (αS) as the major constituent. αS is an intrinsically disordered protein with the potential to undergo a cascade of structural transitions from a soluble disordered conformation to ordered cross-β-sheet-rich insoluble amyloid fibrils. Small molecules like polyphenols and peptides with anti-amyloidogenic potential can mitigate fibrillation in vitro but fail in vivo owing to poor bioavailability. To overcome this problem, a platform that simultaneously

Numerous nanocarriers of small molecules depend on either non-specific physical encapsulation or direct covalent linkage. In contrast, this manuscript explores an alternative encapsulation strategy based on high-specificity avidity between a small molecule drug and its cognate protein target fused to the corona of protein polymer nanoparticles. With the new strategy, the drug associates tightly to the carrier and releases slowly, which may decrease toxicity and promote tumor accumulation via the enhanced permeability and retention effect. To test this hypothesis, the drug Rapamycin (Rapa) was

Multi-walled carbon nanotubes (MWCNTs) have emerged as promising candidates for bone tissue engineering due to their excellent biocompatibility, mechanical strength, and chemical stability. However, disparities often exist between their osteogenic performance in vitro and in vivo. The formation of protein corona on biomaterials is a key factor that may govern these biological outcomes. This study investigated how the distinct protein corona compositions in mouse serum (MS) and fetal bovine serum (FBS) modulate the immune recognition and osteogenic capacity of MWCNTs, thereby mimicking in vitro

Polymersomes (PS) are a class of hollow polymeric nanoparticle vesicles made of amphiphilic block co-polymers that self-assemble via hydrophobic interactions. One of the significant unsung challenges for their translation is the uncontrolled formation of the protein corona, which can influence PS biodistribution, cellular uptake, and immune recognition. Despite the major benefits associated with PS, no studies have yet explored engineering their protein corona. Evidence suggests that the confirmation of polyethylene glycol (PEG) chains, which can vary in response to Hofmeister series salts, ca

OBJECTIVE: Drug delivery strategies for joint diseases need to overcome the negatively charged cartilage matrix. Previous studies have extensively investigated particle approaches to increase uptake efficiency by harnessing the anionic charge of the cartilage but have neglected to address potential interactions with the protein-rich biological environment of the joint space. We aimed to evaluate the effects of hard protein coronas derived from osteoarthritis (OA) and rheumatoid arthritis (RA) patient synovial fluids as well as the commonly used fetal calf serum (FCS) on nanoparticle (NP) uptak
