Psilocybin and Eugenol Reduce Inflammation in Human 3D EpiIntestinal Tissue.

Inflammation plays a pivotal role in the development and progression of inflammatory bowel disease (IBD), by contributing to tissue damage and exacerbating the immune response. The investigation of serotonin receptor 2A (5-HT2A) ligands and transient receptor potential (TRP) channel ligands is of significant interest due to their potential to modulate key inflammatory pathways, mitigate the pathological effects of inflammation, and offer new avenues for therapeutic interventions in IBD. This study investigates the anti-inflammatory effects of 5-HT2A ligands, including psilocybin, 4-AcO-DMT, an

Psilocybin facilitates fear extinction in mice by promoting hippocampal neuroplasticity.

BACKGROUND: Posttraumatic stress disorder (PTSD) and depression are highly comorbid. Psilocybin exerts substantial therapeutic effects on depression by promoting neuroplasticity. Fear extinction is a key process in the mechanism of first-line exposure-based therapies for PTSD. We hypothesized that psilocybin would facilitate fear extinction by promoting hippocampal neuroplasticity. METHODS: First, we assessed the effects of psilocybin on percentage of freezing time in an auditory cued fear conditioning (FC) and fear extinction paradigm in mice. Psilocybin was administered 30 min before extinc

Special considerations for evaluating psilocybin-facilitated psychotherapy in vulnerable populations.

Psilocybin-facilitated psychotherapy shows potential transdiagnostic efficacy for a range of mental health conditions. Though vulnerable populations bear disproportionate mental health burden, they have been largely neglected in the clinical psilocybin literature. However, if the field is to best respond to the diverse needs of individuals from vulnerable populations, care must be taken to ensure these individuals are represented in the empirical research. This report calls attention to this concern by detailing the challenges and opportunities associated with evaluating psilocybin-facilitated

Body mass index (BMI) does not predict responses to psilocybin.

BACKGROUND: Psilocybin is a serotonin type 2A (5-HT2A) receptor agonist and naturally occurring psychedelic. 5-HT2A receptor density is known to be associated with body mass index (BMI), however, the impact of this on psilocybin therapy has not been explored. While body weight-adjusted dosing is widely used, this imposes a practical and financial strain on the scalability of psychedelic therapy. This gap between evidence and practice is caused by the absence of studies clarifying the relationship between BMI, the acute psychedelic experience and long-term psychological outcomes. METHOD: Data

Psilocybin impairs high-level but not low-level motion perception.

The hallucinogenic serotonin(1A&2A) agonist psilocybin is known for its ability to induce illusions of motion in otherwise stationary objects or textured surfaces. This study investigated the effect of psilocybin on local and global motion processing in nine human volunteers. Using a forced choice direction of motion discrimination task we show that psilocybin selectively impairs coherence sensitivity for random dot patterns, likely mediated by high-level global motion detectors, but not contrast sensitivity for drifting gratings, believed to be mediated by low-level detectors. These results a

Psilocybin therapy for adult females with anorexia nervosa: pilot study.

BACKGROUND: Anorexia nervosa is a debilitating eating disorder with high mortality and chronicity rates owing to the paucity of effective existing treatments. Several clinical trials using psilocybin therapy have demonstrated therapeutic efficacy and safety in psychiatric conditions, including anorexia nervosa. AIMS: This study aimed to further assess the safety, feasibility and potential efficacy of psilocybin therapy in anorexia nervosa. METHOD: This single-blind, within-individual pilot study recruited 21 females with anorexia nervosa, who underwent three dosing sessions with oral psilocy

Psilocybin for psychiatric disorders: History, clinical trials, neuroimaging, and regulations.

Psilocybin, a classic psychedelic compound, has garnered renewed interest as a potential treatment for various psychiatric disorders. This review provides a comprehensive overview of psilocybin's history, recent clinical evidence, ongoing clinical trials, neuroimaging findings, and regulations. Historically used in spiritual and healing rituals, psilocybin was in the early 1970s subjected to strict legal restrictions that stalled research for decades. However, renewed scientific interest began in the 1990s, with studies demonstrating psilocybin's therapeutic potential for psychiatric disorders

Psilocybin prevents chemotherapy-induced peripheral neuropathy through mitochondrial trafficking preservation.

Chemotherapy-induced peripheral neuropathy (CIPN) is a disabling, often irreversible toxicity that affects millions of patients, limits life-saving cancer therapy, and lacks proven treatment. In this work, we show that as little as two doses of psilocybin before chemotherapy durably prevented the onset of CIPN across platinum- and taxane-based models, including repeated chemotherapy cycles, without impairing antitumor efficacy. Peripherally, psilocybin maintained tactile sensitivity and intraepidermal nerve fiber endings through axonal mitochondrial trafficking and distribution preservation, t

Synergistic behavioral and neuroplastic effects of psilocybin-NMDAR modulator administration.

The full therapeutic potential of serotonergic psychedelics (SP) in treating neuropsychiatric disorders, such as depression and schizophrenia, is limited by possible adverse effects, including perceptual disturbances and psychosis, which require administration in controlled clinical environments. This study investigates the synergistic benefits of combining psilocybin (PSIL) with N-methyl-D-aspartate receptor (NMDAR) modulators D-serine (DSER) and D-cycloserine (DCS) to enhance both efficacy and safety. Using ICR male mice, we examined head twitch response (HTR), MK-801-induced hyperlocomotion

Endocrinology-informed neuroimaging in eating disorders: GLP1, orexins, and psilocybin.

The neurobiology of eating disorders [EDs; anorexia nervosa (AN), bulimia nervosa (BN), and binge eating disorder (BED)] remains poorly understood. Here, I describe how neuroimaging, accompanied by peripheral endocrine measures, can provide insights into the neurobiological drivers of eating disorders. Orexins/hypocretins, glucagon-like peptide-1 receptor (GLP1R) agonists, and psilocybin are highlighted as avenues for investigation.

Microdosing with psilocybin mushrooms: a double-blind placebo-controlled study.

The use of low sub-perceptual doses of psychedelics ("microdosing") has gained popularity in recent years. Although anecdotal reports claim multiple benefits associated with this practice, the lack of placebo-controlled studies severely limits our knowledge of microdosing and its effects. Moreover, research conducted in standard laboratory settings could fail to capture the motivation of individuals engaged or planning to engage in microdosing protocols, thus underestimating the likelihood of positive effects on creativity and cognitive function. We recruited 34 individuals starting to microdo

Psilocybin: A brief overview for psychiatric mental health nurse practitioners.

The use of psychedelics, such as psilocybin, has emerged in recent literature as a novel therapeutic treatment for various psychiatric disorders, including substance use, depression, and anxiety. While international and domestic trials are currently underway, there is data demonstrating both the relative safety and potential efficacy of psilocybin. Psychiatric mental health nurse practitioners are essential mental health providers that may be at the forefront of delivering these new treatment modalities. Therefore, they must be aware of the psychopharmacological and psychotherapeutic tenets of

Psychedelic perceptions: mental health service user attitudes to psilocybin therapy.

INTRODUCTION: Despite the rapid advance of psychedelic science and possible translation of psychedelic therapy into the psychiatric clinic, very little is known about mental health service user attitudes. OBJECTIVES: To explore mental health service user attitudes to psychedelics and psilocybin therapy. METHODS: A questionnaire capturing demographics, diagnoses, previous psychedelic and other drug use, and attitudes to psychedelics and psilocybin therapy was distributed to mental health service users. RESULTS: Ninety-nine participants completed the survey (52% female, mean age 42 years). Th

Psilocybin as serotonergic therapy in epilepsy: Narrative review of potentials and limitations.

Psilocybin shows potential in neuropsychiatric disorders but has a complex relationship with epilepsy and seizures. We reviewed literature up to November 2025 to identify studies relevant to psilocybin's effects on seizures and epilepsy, including our research on serotonergic modulation. Core studies addressing the research question and supplementary mechanistic evidence were included for synthesis. Risk of bias (RoB) was assessed by study type; independent reviewers achieved strong agreement (unweighted Cohen's κ = 0.80, SE = 0.185; 99% CI 0.006-0.011; per cent agreement = 88.9%). Experimenta

Psilocybin in the Treatment of Cocaine Use Disorder: A Randomized Clinical Trial.

IMPORTANCE: Cocaine use disorder is a serious public health problem and no medications have been proven effective for its treatment. OBJECTIVE: To evaluate psilocybin in the treatment of cocaine use disorder. It was hypothesized that psilocybin, compared with placebo, would yield a higher percentage of cocaine abstinent days, a greater likelihood of complete abstinence from cocaine, and a greater latency to first cocaine lapse through 180 days after end of treatment. Prior trauma shapes the effects of psilocybin on stress sensitization in mice.

Exposure to traumatic events induces persistent alterations in defensive behavior that extend beyond associative fear memories, including heightened responses to novel stimuli (stress sensitivity) and sustained defensive states in neutral contexts (stress sensitization). While serotonergic psychedelics such as psilocybin have been shown to facilitate fear extinction in associative paradigms, their effects on trauma-induced non-associative stress adaptations remain poorly understood. Here, we developed a behavioral paradigm in mice that dissociates contextual fear recall from stress sensitivity

Rebuttal to "Questioning the recovery of dissociated traumatic memories under psilocybin".

In our original case report we provide detailed accounts of two research participants who reported the emergence of spontaneously recovered, previously forgotten traumatic memories of sexual assaults during psilocybin treatment. In their commentary of this article, Kangaslampi et al. argue that we preemptively label the experiences as dissociated traumatic memories in the absence of corroborating evidence and consideration of alternative explanations. Here we further address potential therapeutic effects associated with the classification of experiences and provide a rebuttal to the authors' c
