Severe Toxic Skin Reaction Caused by a Common Anemone and Identification of the Culprit Organism.

In a marine envenomation, identification of the culprit organism can be difficult. In this case report, we present our method to identify snakelocks anemone (Anemonia viridis or formerly Anemonia sulcata) as the culprit of a severe toxic skin reaction. A. viridis is one of the most common anemones of the Mediterranean Sea and the North Atlantic Ocean. It lives at a depth of up to 10 m. It is a member of the phylum Cnidaria, which includes jellyfish, anemones, hydroids, and corals. They have toxic organelles called cnidocysts that have the capacity to inject venom with microscopic harpoon-like

Venom-Inspired Medicine: Ancient Chemicals Offer Novel Solutions.

There are an estimated 150,000 animal species that have evolved venom. Some are familiar, such as snakes, bees, scorpions, and spiders. But there are also venomous lizards, sea anemones, cone snails, and even a few mammals, like the duck-billed platypus with its venom-bearing ankle spurs [1].

Renal effects of Bunodosoma caissarum crude extract: Prostaglandin and endothelin involvement.

Sea anemones contain a variety of interesting biologically active compounds, including some potent toxins. PLA2 from Bunodosoma caissarum, a sea anemone endemic in the Brazilian southern coast, has shown renal alterations on isolated kidney. The aim of this study was to evaluate the renal and vascular effects of B. caissarum crude extract (BcE) on isolated perfused kidney and arteriolar mesenteric bed, as well the involvement of prostaglandins and endothelin. BcE did not show any effect on arteriolar mesenteric bed, but increased perfusion pressure, renal vascular resistance, urinary flow, glo

Site-3 toxins and cardiac sodium channels.

Site-3 toxins are small polypeptide venoms from scorpions, sea anemones, and spiders that bind with a high specificity to the extracellular surface of voltage-gated Na channels. After binding to a site near the S4 segment in domain IV the toxin causes disruption of the normal fast inactivation transition resulting in a marked prolongation of the action potentials of excitable tissues including those of cardiac and skeletal muscle and nerve. In this review we discuss the specific binding interactions between residues of the toxin and those of the Na channel, and the specific modification of Na

Interaction of polypeptide neurotoxins with a receptor site associated with voltage-sensitive sodium channels.

Anthopleurin A, a polypeptide toxin from the Pacific sea anemone Anthopleura xanthogrammica, enhances persistent activation of voltage-sensitive sodium channels by the alkaloid toxins veratridine and batrachotoxin with K0.5 = 20 nM. This effect is inhibited by depolarization. There is a close correlation between enhancement of sodium channel activation and block of [125I]scorpion toxin binding by unlabeled scorpion toxin, sea anemone toxin II from Anemonia sulcata, and Anthopleurin A, indicating that these three polypeptide toxins interact with a common receptor site in modifying sodium channe

Ghost anemone dermatitis.

BACKGROUND: Haloclava producta, the "ghost anemone", is a burrowing sea anemone in estuarine sediments of the US East Coast and Gulf of Mexico. It has never been identified as harmful to human beings; however, residents of Long Island, New York develop a pruritic, erythematous, vesiculopapular dermatitis on areas of the body that contact these organisms. Neither the condition nor its cause has been described in the medical literature. METHODS: We reviewed information of all water-related dermatitis reported by beach personnel, health providers, and affected swimmers to the Office of Marine Ec

[Corneal melting after moving a tropical aquarium].

BACKGROUND Keeping exotic pets can lead to dangerous situations. We describe an ophthalmological injury caused by Palythoa, an anemone that produces palytoxin when irritated. Palytoxin is one of the most poisonous marine toxins ever described. CASE This case concerns a healthy, 44-year-old man who had rubbed his eyes after setting up a tropical aquarium, which contained sea-anemones among other species. This resulted in bilateral corneal melting, or keratolysis. The injury turned out to be caused by palytoxin. As a consequence, this patient was permanently visually impaired. CONCLUSION The ane

Identification of a tetrodotoxin-sensitive Na+ channel in a variety in fibroblast lines.

The action potential Na+ ionophore of excitable cells can be activated either by alkaloid compounds such as veratridine, or by small polypeptide toxins extracted from scorpion venom or sea anemone. One of the main features of this Na+ channel is that it is blocked by tetrodotoxin (TTX). However, we report here that during analysis of Na+ influx in resting fibroblasts, we found that a variety of fibroblast lines also possess a TTX receptor. Veratridine and sea anemone toxin act synergistically to stimulate Na+ influx 7 to 10-fold in hamster and rat fibroblasts. As in excitable cells, this toxin

Chilling Injury to Algal Symbionts Induces Host Starvation and Metabolic Reorganization in a Temperate Cnidarian.

Global temperature anomalies increasingly disrupt the cnidarian-algal symbiosis resulting in a phenomenon termed bleaching. In contrast to heat stress, the mechanisms underlying symbiotic breakdown under cold stress remain largely unknown. Combining physiological and metabolomic measurements, we investigated the response of the photosymbiotic sea anemone holobiont Aiptasia couchii to an experiment mimicking a cold spell in the Mediterranean Sea. Within 4 weeks, we observed the onset of symbiotic breakdown reflected in reduced algal endosymbiont density and chlorophyll a content. While photosyn

The Nematostella synaptonemal complex mediates divergent and sex-specific meiotic programs.

Throughout eukaryotes, the synaptonemal complex (SC) is a supramolecular structure essential for meiotic chromosome dynamics and sexual reproduction. While metazoan SC proteins display substantial sequence divergence, the lack of functional analyses beyond bilaterian models has obscured a mechanistic understanding of SC evolution. Here, we report that the sea anemone Nematostella vectensis exhibits synapsis of homologous chromosomes in a classic zipper-like manner and expresses at least eight orthologs of 10 known vertebrate SC proteins. Unexpectedly, mutagenesis of the core SC components sycp

Kv1.3 Channel as a Key Therapeutic Target for Neuroinflammatory Diseases: State of the Art and Beyond.

It remains a challenge for the effective treatment of neuroinflammatory disease, including multiple sclerosis (MS), stroke, epilepsy, and Alzheimer's and Parkinson's disease. The voltage-gated potassium Kv1.3 channel is of interest, which is considered as a novel therapeutic target for treating neuroinflammatory disorders due to its crucial role in subsets of T lymphocytes as well as microglial cells. Toxic animals, such as sea anemones, scorpions, spiders, snakes, and cone snails, can produce a variety of toxins that act on the Kv1.3 channel. The Stichodactyla helianthus K+ channel blocking t

Genomic structure of a potassium channel toxin from Heteractis magnifica.

We provide information on the gene encoding the K+ channel toxin, HmK, of the sea anemone Heteractis magnifica. A series of DNA amplifications by PCR, which included the amplification of the 5'-untranslated region of the gene, showed that an intron of 402 nucleotides separated the sequence that encodes the matured toxin from the signal peptide sequence. A second 264 nucleotide intron interrupted the 5'-untranslated region of the previously reported HmK cDNA. Two possible transcription-initiation sites were identified by primer extension analysis. Corresponding TATA-box consensus sequences, cha

Nematostella vectensis exhibits an enhanced molecular stress response upon co-exposure to highly weathered oil and surface UV radiation.

Crude oil released into the environment undergoes weathering processes that gradually change its composition and toxicity. Co-exposure to petroleum mixtures and other stressors, including ultraviolet (UV) radiation, may lead to synergistic effects and increased toxicity. Laboratory studies should consider these factors when testing the effects of oil exposure on aquatic organisms. Here, we study transcriptomic responses of the estuarine sea anemone Nematostella vectensis to naturally weathered oil, with or without co-exposure to environmental levels of UV radiation. We find that co-exposure gr

Structure of the BgK-Kv1.1 complex based on distance restraints identified by double mutant cycles. Molecular basis for convergent evolution of Kv1 channel blockers.

A structural model of BgK, a sea anemone toxin, complexed with the S5-S6 region of Kv1.1, a voltage-gated potassium channel, was determined by flexible docking under distance restraints identified by a double mutant cycles approach. This structure provides the molecular basis for identifying the major determinants of the BgK-Kv1.1 channel interactions involving the BgK dyad residues Lys(25) and Tyr(26). These interactions are (i) electrostatic interactions between the extremity of Lys(25) side chain and carbonyl oxygen atoms of residues from the channel selectivity filter that may be strengthe

Detection of transient capacitance increase associated with channel formation in lipid bilayers.

The insertion of alpha- and beta-latrotoxins and sea anemone (Radianthus macrodactilus) toxin into bilayer lipid membranes (BLMs) was investigated using the method of simultaneous conductance/capacitance measurement. All the toxins investigated induced capacitance changes which preceded toxin-induced conductance increases. The processes that may underlie the observed effect are discussed.

Cnidarian Venoms in the Omics Era: Toxin Discovery and Bioactive Potential.

Cnidaria is a phylum of aquatic invertebrates that includes jellyfish, sea anemones and corals. Their venoms are among the oldest in the animal kingdom and an important source of bioactive molecules. Advances in omics technologies have revolutionized toxin discovery, yet knowledge of cnidarian toxin diversity remains fragmented across databases and individual studies. To address this gap, this review integrates information from 342 toxins curated in the ToxProt database, 76 additional toxins reported in the literature, and evidence from 81 proteomic and 31 transcriptomic studies. Toxins have b

Expression patterns of sterol transporters NPC1 and NPC2 in the cnidarian-dinoflagellate symbiosis.

The symbiotic interaction between cnidarians (e.g., corals and sea anemones) and photosynthetic dinoflagellates of the genus Symbiodinium is triggered by both host-symbiont recognition processes and metabolic exchange between the 2 partners. The molecular communication is crucial for homeostatic regulation of the symbiosis, both under normal conditions and during stresses that further lead to symbiosis collapse. It is therefore important to identify and fully characterise the key players of this intimate interaction at the symbiotic interface. In this study, we determined the cellular and subc

Ultraviolet radiation significantly enhances the molecular response to dispersant and sweet crude oil exposure in Nematostella vectensis.

Estuarine organisms are subjected to combinations of anthropogenic and natural stressors, which together can reduce an organisms' ability to respond to either stress or can potentiate or synergize the cellular impacts for individual stressors. Nematostella vectensis (starlet sea anemone) is a useful model for investigating novel and evolutionarily conserved cellular and molecular responses to environmental stress. Using RNA-seq, we assessed global changes in gene expression in Nematostella in response to dispersant and/or sweet crude oil exposure alone or combined with ultraviolet radiation (U
