Heat stress drives increases in phytoplankton cell size in future oligotrophic oceans.

Warming and nutrient limitation are major stressors that affect primary production in the ocean, with cascading impacts on the food web. Yet, we lack a mechanistic understanding of how phytoplankton manage multiple stressors and the implications of these responses for phytoplankton biogeography. By combining theory, proteome allocation modeling, and climate projections, we identified two potential competing strategies for multi-stressor growth: (1) nutrient efficient smaller cells or (2) heat-tolerant larger cells. We found that Prochlorococcus are more vulnerable in warmer, "heat-stressed", t

Optimization of isopropanol production by engineered cyanobacteria with a synthetic metabolic pathway.

Cyanobacterium is an attractive host for the production of various chemicals and alternative fuels using solar energy and carbon dioxide. In previous study, we succeeded to produce isopropanol using engineered Synechococcus elongatus PCC 7942 under dark and anaerobic conditions (0.43 mM, 26.5 mg/l). In the present study, we report the further optimization of this isopropanol producing condition. We then optimized growth conditions for production of isopropanol by the engineered cyanobacteria, including the use of cells in early stationary phase and buffering of the production medium to neutral

An inhalable microbe-oncolytic virus consortium for lung cancer treatment.

Oncolytic viruses (OVs) represent a promising cancer therapy due to their selective cytotoxicity, yet clinical success in lung cancer is hindered by biological barriers and immunosuppressive microenvironments. To address these barriers, an inhalable microbe-OVs consortium is developed by conjugating tumor-responsive PEGylated adenoviruses (Ads) to motile algae (Synechococcus WH8102) via click chemistry. Driven by algal motility, the consortium penetrates respiratory mucus and epithelial barriers to reach lung tumor sites. Intratumoral calcium ion deprivation by the algae disrupts cellular tigh

An innovative biologic system for photon-powered myocardium in the ischemic heart.

Coronary artery disease is one of the most common causes of death and disability, afflicting more than 15 million Americans. Although pharmacological advances and revascularization techniques have decreased mortality, many survivors will eventually succumb to heart failure secondary to the residual microvascular perfusion deficit that remains after revascularization. We present a novel system that rescues the myocardium from acute ischemia, using photosynthesis through intramyocardial delivery of the cyanobacterium Synechococcus elongatus. By using light rather than blood flow as a source of e

Sustained algal-bacterial interactions promote accumulation of recalcitrant particulate carbon with accelerated radiocarbon aging signatures.

Algal-bacterial interactions regulate the production and fate of marine particulate organic carbon (POC), yet their capacity to generate long-lived carbon sinks remains unclear. Using a two-year, nutrient-self-sustaining co-culture of Synechococcus and its mutualistic microbiota, we demonstrate that sustained algal-bacterial interactions promote the progressive accumulation of recalcitrant POC. Continuous release of algal-derived organic substrates, coupled with repeated microbial enzymatic reworking, produced inert organic particles, with ~26% resisting remineralization and forming a stable r

Removing Pb2+ ions from aqueous solution with freshwater cyanobacteria: Combined adsorption and intracellular precipitation.

This study investigated the removal of lead ions (Pb2+) from aqueous solutions by two freshwater cyanobacteria with distinct cellular inclusions: Synechococcus elongatus FACHB-410, which is rich in polyphosphate (polyP), and Cyanothece sp. PCC 7425, containing both polyP and amorphous calcium carbonate (iACC). Both strains were exposed to Pb2+ concentrations ranging from 1 to 50 mg/L and demonstrated high removal efficiencies exceeding 92%. The removal mechanism involved not only adsorption onto cell surfaces but also significant intracellular precipitation. The proportion of intracellular pre

Role for the cyanobacterial HtpG in protection from oxidative stress.

The heat shock protein HtpG, which is a homolog of HSP90, is essential for basal and acquired thermotolerances in cyanobacteria. Recently we demonstrated that HtpG was involved in the acclimation to low temperatures in cyanobacteria. In this study, we elucidated a role of HtpG in the cyanobacterium Synechococcus sp. PCC 7942, in the acclimation to oxidative stress that was caused by high irradiance and/or methyl viologen. The inactivation of the htpG gene resulted in a decrease in the survival rate and an increase in the photoinhibition of photosystem II when cells in a liquid medium were incu

Cytotoxic and antimitotic activities in aqueous extracts of eight cyanobacterial strains isolated from the marine sponge Petrosia ficiformis.

Marine cyanobacteria are photosynthetic prokaryotes of significant ecological interest, living free or in association with invertebrates. They are also considered as excellent sources of antineoplastic, antibacterial, antiviral and antifungal compounds. In this work, aqueous extracts from eight cyanobacterial strains isolated from the Mediterranean sponge Petrosia ficiformis have been investigated for their bioactive properties. Bioassays with human erythrocytes, Artemia salina nauplii, and Paracentrotus lividus gametes and embryos were performed. Some aqueous extracts exhibited citolytic effe

Very high CO2 concentrations inhibit photosynthesis and trigger transcriptomic stress responses in cyanobacteria.

CO[Formula: see text] concentrations above air level (0.04%) are beneficial for the growth of cyanobacteria. However, very high CO[Formula: see text] levels inhibit growth, limiting the usability of cyanobacteria for carbon capture and biotechnological applications. The transcriptomic changes in the cyanobacterium Synechococcus sp. PCC 7002 were investigated at varying growth rates governed by different gas-phase CO[Formula: see text] concentrations, ranging from limiting (0.04%) to optimal (4% and 8%) to inhibitory high (30%). Compared to optimal CO[Formula: see text] concentrations, large di

Action at a distance: The remarkable coupling of CO2 uptake to electron transfer in specialized cyanobacterial NDH-1 complexes.

Cyanobacteria achieve highly efficient photosynthesis using a CO2-concentrating mechanism relying on specialized Type I (NDH-1) complexes. Among these, NDH-13 and NDH-14 catalyze redox-coupled hydration of CO2 to bicarbonate, supporting carbon fixation in carboxysomes. The mechanism of coupling electron transfer to CO2-hydration by these variant NDH-1 complexes remains unknown. We engineered a Synechococcus PCC7942 strain that expresses exclusively the high flux/low affinity NDH-14 complex, enabling the observation of the coupling of CO2 hydration to cyclic electron flow in isolation from the

Native metabolomics identifies pteridines as CutA ligands and modulators of copper binding.

CutA, a conserved protein across all domains of life, has been suggested to be involved in copper tolerance in bacteria, though recent studies have questioned this association, leaving its biological role unknown. To clarify its function, we studied cutA knockout mutants from two phylogenetically distant species, Synechococcus elongatus PCC 7942 and Escherichia coli, using phenotyping combined with metabolomics. To investigate the interaction of CutA with potential substrates and cofactors, we performed a series of native metabolomics experiments with CutA and cell extracts from which we ident

PBP-A, a cyanobacterial DD-peptidase with high specificity for amidated muropeptides, exhibits pH-dependent promiscuous activity harmful to Escherichia coli.

Penicillin binding proteins (PBPs) are involved in biosynthesis, remodeling and recycling of peptidoglycan (PG) in bacteria. PBP-A from Thermosynechococcus elongatus belongs to a cyanobacterial family of enzymes sharing close structural and phylogenetic proximity to class A β-lactamases. With the long-term aim of converting PBP-A into a β-lactamase by directed evolution, we simulated what may happen when an organism like Escherichia coli acquires such a new PBP and observed growth defect associated with the enzyme activity. To further explore the molecular origins of this harmful effect, we de

Mechanisms of the novel pesticide sodium dodecyl benzene sulfonate in the mitigation of protozoan ciliated pathogens during microalgal cultivation.

Protozoan ciliates represent a common biological contaminant during microalgae cultivation, which will lead to a decline in microalgae productivity. This study investigated the effectiveness of sodium dodecyl benzene sulfonate (SDBS) in controlling ciliate populations within microalgae cultures. SDBS concentrations of 160 mg/L and 100 mg/L were found to effectively manage the representative species of ciliates contamination by Euplotes vannus and Uronema marinum during the cultivation of Synechococcus and Chlorella, and the growth vitality of microalgae has been restored. Additionally, SDBS at

Newer systems for bacterial resistances to toxic heavy metals.

Bacterial plasmids contain specific genes for resistances to toxic heavy metal ions including Ag+, AsO2-, AsO4(3-), Cd2+, Co2+, CrO4(2-), Cu2+, Hg2+, Ni2+, Pb2+, Sb3+, and Zn2+. Recent progress with plasmid copper-resistance systems in Escherichia coli and Pseudomonas syringae show a system of four gene products, an inner membrane protein (PcoD), an outer membrane protein (PcoB), and two periplasmic Cu(2+)-binding proteins (PcoA and PcoC). Synthesis of this system is governed by two regulatory proteins (the membrane sensor PcoS and the soluble responder PcoR, probably a DNA-binding protein), h

Survival of cyanobacteria and mitigation of Fe(II) toxicity effects in a silica-rich Archean ocean.

Banded iron formations (BIF) were deposited abundantly between 2.7-2.4 Ga from iron- and silica-rich oceans, with cyanobacterial oxygen (O2) as a possible oxidant for Fe(II)(aq) oxidation and Fe(III) oxyhydroxide precipitation. However, toxic reactive oxygen species (ROS) from Fe(II)/O2 interactions might have inhibited cyanobacterial growth, contributing to the delay between cyanobacterial evolution (>3.0 Ga) and the Great Oxidation Event (2.5 Ga). Here, we explored the impact of Fe(II)(aq) and SiO2(aq) on Synechococcus sp. PCC 7002. High Fe(II)(aq) ( > 500 µM) increased ROS formation, but el

Cyanosomes: A pilot study.

Cyanobacteria are the most primitive, oxygen-evolving prokaryotic organism. Several, conspicuous investigations have been done around lipids from cyanobacteria. However, to date and to the best of our knowledge, no study was conducted to formulate and evaluate liposomal vesicles prepared from cyanobacterial lipid. In the present study, cyanosomes were formulated using natural lipids extracted from the cyanobacterium Synechococcus elongatus PCC7942 which were further loaded with thymoquinone. The results showed that the prepared cyanosomes were homogenous with an overall negative charge of part

Synthetic microbial communities of heterotrophs and phototrophs facilitate sustainable growth.

Microbial communities comprised of phototrophs and heterotrophs hold great promise for sustainable biotechnology. Successful application of these communities relies on the selection of appropriate partners. Here we construct four community metabolic models to guide strain selection, pairing phototrophic, sucrose-secreting Synechococcus elongatus with heterotrophic Escherichia coli K-12, Escherichia coli W, Yarrowia lipolytica, or Bacillus subtilis. Model simulations reveae metabolic exchanges that sustain the heterotrophs in minimal media devoid of any organic carbon source, pointing to S. elo

The small helix-loop-helix protein NsrC is a regulator of phycobilisome biosynthesis.

Nitrogen is a key nutrient that influences primary productivity in both aquatic and terrestrial ecosystems. In cyanobacteria, its deficiency affects growth and phycobiliprotein biosynthesis and leads to degradation of phycobilisomes (PBSs). While the biosynthesis genes of phycobiliprotein are regulated by strong promoters, the molecular mechanisms underlying their regulation in response to nitrogen deficiency remain largely unknown. Using nondiazotrophic cyanobacterium Synechococcus elongatus PCC 7942 as a model system, we identified a novel PBS biosynthesis regulator, a small helix-loop-helix

Programming Bio-Bio Electronic Interfaces for Light-Driven Interspecies Electron Transfer.

Living systems organize electron flow through continuous, spatially and energetically structured redox networks, whereas most synthetic light-driven bioelectronic platforms rely on abiotic materials to generate and inject electrons into cells, limiting selective coupling between living partners. Here, we report programmable living electronic interfaces that enable direct, light-driven interspecies electron transfer (IET) between two living microorganisms.
