Divalent aptamer-mediated clustering for extracellular vesicle separation.

Extracellular vesicles (EVs) are important mediators of intercellular communication and promising sources of diagnostic and therapeutic biomarkers, yet effective EV isolation remains challenging due to trade-offs among yield, purity, and adaptability across biofluids. Here, we introduce divalent aptamer-mediated clustering (DAC), a streamlined affinity-based EV isolation strategy that induces controllable vesicle clustering and enables recovery by standard filtration. By exploiting multivalent aptamer binding to EV surface markers, DAC converts nanoscale vesicles into micron-scale clusters whi

Analysis of Purity and Quality of Small Extracellular Vesicles Isolated from Blood Plasma by Proteomics and Raman Spectroscopy.

Ultracentrifugation (UC) has long been considered the "gold standard" for extracellular vesicle (EV) isolation. However, due to its drawbacks such as high cost of an ultracentrifuge and rotors, time-consuming and labor-intensive protocol, low yield considering initial biofluid volume and low throughput, development of new EV isolation approaches is still ongoing. Here we compare three methods for isolating the most studied EV subtype, small extracellular vesicles (sEVs), from human plasma: ultracentrifugation (UC), express asymmetric depth filtration (ExADFi), and anti-CD9 immunoaffinity captu

The role of link-protein in the structure of cartilage proteoglycan aggregates.

Proteoglycan fractions were prepared from pig laryngeal cartilage. The effect of link-protein on the properties of proteoglycan-hyaluronate aggregates was examined by viscometry and analytical ultracentrifugation. Aggregates containing link-protein were more stable than link-free aggregates at neutral pH, at temperatures up to 50 degrees C and in urea (up to 4.0M). Oligosaccharides of hyaluronate were able to displace proteoglycans from link-free aggregates, but not from the link-stabilized aggregates. Both types of aggregate were observed in the ultracentrifuge, but at the concentration inves

Isolation and Proteomic Characterization of Outer Membrane Vesicles in Fusobacterium nucleatum.

Fusobacterium nucleatum is an anaerobic Gram-negative bacterium associated with periodontal disease and colorectal cancer. Outer membrane vesicles (OMVs) produced by F. nucleatum are nanoscale lipid vesicles enriched with immunogenic and virulence-associated proteins, making them promising candidates for vaccine development. This protocol describes a comprehensive workflow for culturing F. nucleatum, isolating and purifying OMVs via ultracentrifugation and density gradient ultracentrifugation, and characterizing their biophysical properties using transmission electron microscopy (TEM) and dyna

Hyperlipoproteinaemia associated with atherosclerosis and cutaneous xanthomatosis in a cat.

Bilateral uveitis and multiple xanthomas (fat deposits in the skin) are described in a 3.5-year-old ovariohysterectomized female Persian cat. The cat had been treated for 2 years with corticosteroids. Examinations included a routine blood chemistry profile, radiographic examination of the thorax and abdomen, histopathological examination of multiple skin punch biopsies, and analysis of blood lipid components by cellulose-acetate electrophoresis and by preparative ultracentrifugation studies. Total lipid values were 23 g/l. Ultracentrifugation studies indicated strongly elevated VLDL and LDL fr

Surface glycosylation profiles of urine extracellular vesicles.

Urinary extracellular vesicles (uEVs) are released by cells throughout the nephron and contain biomolecules from their cells of origin. Although uEV-associated proteins and RNA have been studied in detail, little information exists regarding uEV glycosylation characteristics. Surface glycosylation profiling by flow cytometry and lectin microarray was applied to uEVs enriched from urine of healthy adults by ultracentrifugation and centrifugal filtration. The carbohydrate specificity of lectin microarray profiles was confirmed by competitive sugar inhibition and carbohydrate-specific enzyme hydr

Optimizing plasma-derived small extracellular vesicle purification to enhance downstream applications.

Small extracellular vesicles (sEVs) are functional carriers of biomolecules from host cells, making them highly suitable for early disease diagnosis and monitoring disease progression. However, the purification of sEVs from plasma is complicated by the presence of high levels of proteins, particularly lipoproteins. We have developed an original pretreatment method that effectively removes lipoproteins and fibrinogen, thereby facilitating subsequent sEV purification. Sequential application of ultracentrifugation and optical density gradient on pretreated plasma significantly enhances the purity

Proteomic profiling of mesenchymal stem cell-derived extracellular vesicles: Impact of isolation methods on protein cargo.

Extracellular vesicles (EVs) are nanosized vesicles with a lipid bilayer that are secreted by cells and play a critical role in cell-to-cell communication. Despite the promising reports regarding their diagnostic and therapeutic potential, the utilization of EVs in the clinical setting is limited due to insufficient information about their cargo and a lack of standardization in isolation and analysis methods. Considering protein cargos in EVs as key contributors to their therapeutic potency, we conducted a tandem mass tag (TMT) quantitative proteomics analysis of three subpopulations of mesenc

Improved specificity of a new homogeneous assay for LDL-cholesterol in serum with abnormal lipoproteins.

BACKGROUND: Although a homogeneous assay for serum LDL-cholesterol (LDL-C) has become a routine clinical procedure, problems remain in assay performance characteristics. METHODS: We examined the performance of a recently developed automated homogeneous assay (New-Daiichi assay) for serum LDL-C and compared the results with those obtained by the current homogeneous method (Denka-Seiken assay) or by ultracentrifugation as a control. RESULTS: The New-Daiichi assay showed satisfactory basic performance characteristics such as reproducibility, linearity, and stability. There was no interference i

Mammalian class I myosin, Myo1b, is monomeric and cross-links actin filaments as determined by hydrodynamic studies and electron microscopy.

The class I myosin, Myo1b, is a calmodulin- and actin-associated molecular motor widely expressed in mammalian tissues. Analytical ultracentrifugation studies indicate that Myo1b purified from rat liver has a Stokes radius of 6.7 nm and a sedimentation coefficient, s(20,w), of 7.0 S with a predicted molar mass of 213 kg/mol. These results indicate that Myo1b is monomeric and consists primarily of a splice variant having five associated calmodulins. Molecular modeling based on the analytical ultracentrifugation studies are supported by electron microscopy studies that depict Myo1b as a single-h

An Improved Method to Enrich Large Extracellular Vesicles Derived from Giardia intestinalis through Differential Centrifugation.

Giardia intestinalis is a flagellated unicellular protozoan that colonizes the small intestine, causing the diarrheal disease called giardiasis. The production of extracellular vesicles (EVs) by G. intestinalis and the role of these EVs in the parasite's interaction with the host have been described. According to biogenesis, EVs are grouped mainly into large (microvesicles-derived from the plasma membrane) and small (exosomes-derived from multivesicular bodies). Populations of EVs are heterogeneous, and improved methods to separate and study them are needed to understand their roles in cell ph

Interaction of Mycotoxins with α1-Acid Glycoprotein (AGP) and Bovine Milk Proteins: Zearalenone, Zearalenols, and Sterigmatocystin Form Highly Stable Complexes with AGP.

Mycotoxins are frequent food contaminants posing health risk to humans and animals. Since these interactions have been barely studied yet, we examined the potential complex formation of mycotoxins with human α1-acid glycoprotein (AGP) and with bovine milk proteins (including casein (CSN), β-lactoglobulin (LG), and α-lactalbumin (LA)) based on fluorescence spectroscopic and ultracentrifugation techniques. Only weak interactions (logK = 2.7 to 3.5) of certain mycotoxins were observed with CSN, LG, and/or LA. Ultracentrifugation experiments demonstrated that aflatoxin M1, zearalenone, and α-zeara

FrozONE: quick cell nucleus enrichment for comprehensive proteomics analysis of frozen tissues.

Subcellular fractionation allows for the investigation of compartmentalized processes in individual cellular organelles. Nuclear enrichment methods commonly employ the use of density gradients combined with ultracentrifugation for freshly isolated tissues. Although it is broadly used in combination with proteomics, this approach poses several challenges when it comes to scalability and applicability for frozen material. To overcome these limitations, we developed FrozONE (Frozen Organ Nucleus Enrichment), a nucleus enrichment and proteomics workflow for frozen tissues. By extensively benchmark

Isolation and Characterization of Small Extracellular Vesicles from Infrapatellar Fat Pads of Osteoarthritis Patients.

Extracellular vesicles (EVs), particularly small extracellular vesicles (sEVs) defined as vesicles smaller than 200 nm, serve as essential mediators of intercellular communication and released by nearly all cell types into various biological fluids. The infrapatellar fat pad (IPFP) is closely associated with the development and progression of knee osteoarthritis (OA). However, standardized methods for isolating sEVs directly from IPFP explants remain limited. We present a reproducible protocol for the isolation and characterization of sEVs derived from IPFP tissues obtained from OA patients. C

Isolation of Proteins and Extracellular Vesicles Secreted by Pollen of Actinidia chinensis L. During In Vitro Germination.

Extracellular vesicles (EVs) are membrane-bound particles secreted into the extracellular space by cells of any living kingdom. Plant-derived EVs (PDEVs) in particular are regarded as promising nutraceutical and drug carriers, but their purification is hindered by the contamination from intracellular and intramural vesicles, which is inherently associated with the existing PDEVs isolation protocols. In the last decade, pollen has been proven to release EVs, called pollensomes, during in vitro germination. When secreted in a liquid culture medium, pollensomes can be easily isolated with minimal

Human aqueous humor exosomes.

Aqueous humor (AH) is a dynamic intraocular fluid that supports the vitality of tissues that regulate intraocular pressure. We recently discovered that extracellular nanovesicles called exosomes are a major constituent of AH. Exosomes function in extracellular communication and contain proteins and small RNA. Our goal was to characterize the physical properties of AH exosomes and their exosomal RNA (esRNA) content. We isolated exosomes from human AH collected during cataract surgery from five patients using serial ultracentrifugation. We measured the size and concentration of AH exosomes in so

Methodological insights into the isolation efficiency of human milk-derived extracellular vesicles.

Human milk-derived extracellular vesicles (HMEV) are promising supplements for infant formula, as they contribute to the health benefits of breast milk. Efficient isolation of pure, intact extracellular vesicles (EV) remains challenging due to the complex milk matrix. This study compared common HMEV isolation methods-polyethylene glycol (PEG)-based precipitation, ultracentrifugation (70,000 × g, 100,000 × g, and 100,000 × g, repeated 3 times), and 0.45-µm filtration-to identify protocols optimizing yield, integrity, and purity. Before isolation, samples underwent preliminary purification to re

Structural characterization of β-propiolactone inactivated severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) particles.

The severe COVID-19 pandemic drives the research toward the SARS-CoV-2 virion structure and the possible therapies against it. Here, we characterized the β-propiolactone inactivated SARS-CoV-2 virions using transmission electron microscopy (TEM) and atomic force microscopy (AFM). We compared the SARS-CoV-2 samples purified by two consecutive chromatographic procedures (size exclusion chromatography [SEC], followed by ion-exchange chromatography [IEC]) with samples purified by ultracentrifugation. The samples prepared using SEC and IEC retained more spikes on the surface than the ones prepared

Characterization of high density lipoproteins in human cholestasis.

High density lipoproteins (HDL) of plasma from patients with long-standing cholestasis were studied by several methods including crossed immunoelectrophoresis, gel filtration, and zonal ultracentrifugation. The crossed immunoelectrophoretic pattern against anti-HDL was characterized by the presence of several immunoprecipitates, in striking contrast to the pattern formed by normal HDL. One of these precipitates corresponded to a lipoprotein species dominated by apoprotein A-II (apo A-II). Zonal ultracentrifugation of cholestatic plasma showed a decrease of HDL and a disappearance of the normal
