Patterns of peripheral steroid metabolism vary with sex, season, and tissue type in blotched blue-tongued lizards (Tiliqua nigrolutea).

We examined sexual and seasonal variation in the ability of reproductively relevant tissues (liver, skin, adrenal gland, cloaca, kidney, renal sexual segment, epididymis, oviduct, muscle, testis, and ovary) to metabolise a primary steroid [testosterone (T) or estradiol (E2)] in the scincid lizard, Tiliqua nigrolutea. We observed considerable variation between sexes and across seasons in the patterns of conjugation and derivatisation of the primary steroids by these tissues. All tissues demonstrated the ability to conjugate the relevant primary steroid. Other general trends included increased c

Influence of endomorphins along the pituitary-ovary axis in the Mozambique Tilapia Oreochromis mossambicus.

Endomorphins (EM-1 and EM-2) are the tetrapeptides involved in pain and neuroendocrine responses with a high affinity for μ-opioid receptors in vertebrates. However, their role in fish reproduction is not clear. The aim of this study was to investigate the influence of EM-1 and EM-2 on the pituitary-ovary axis in the Mozambique tilapia Oreochromis mossambicus. The experimental set-up consisted of four groups, namely, initial controls, controls, EM-1- and EM-2-treated groups (n = 10 in each group consisting of two replicates). Although the number of stage IV (vitellogenic) follicles was signifi

Effect of pinealectomy on testosterone, estradiol-17 beta, esterone, and 17 alpha-hydroxyprogesterone levels during the annual reproductive cycle in the freshwater catfish, Clarias batrachus.

The present study examines the relationship of the pineal gland to gonadal activity during the annual reproductive cycle in the freshwater catfish Clarias batrachus. The hormonal profiles of testosterone, estradiol-17 beta, estrone, and 17 alpha-hydroxyprogesterone along with the gonadosomatic index (GSI) were monitored to assess the effect of pinealectomy. Pinealectomy appeared to accelerate the gonadal development from January to May. Removal of pineal prior to the start of gonadal recrudescence (January and February), induced initiation of ovarian recrudescence earlier than scheduled in nat

Genomic insights into the Ixodes scapularis tick vector of Lyme disease.

Ticks transmit more pathogens to humans and animals than any other arthropod. We describe the 2.1 Gbp nuclear genome of the tick, Ixodes scapularis (Say), which vectors pathogens that cause Lyme disease, human granulocytic anaplasmosis, babesiosis and other diseases. The large genome reflects accumulation of repetitive DNA, new lineages of retro-transposons, and gene architecture patterns resembling ancient metazoans rather than pancrustaceans. Annotation of scaffolds representing ∼57% of the genome, reveals 20,486 protein-coding genes and expansions of gene families associated with tick-host

A Glycoprotein Hormone Orchestrates Ovarian Development in the Fall Armyworm Spodoptera Frugiperda.

Glycoprotein hormones serve as critical regulators of diverse physiological processes in both vertebrates and invertebrates, yet their functional roles in insect reproduction remain poorly understood. Here, we identified two glycoprotein hormone subunits, SfLFT1 and SfLFT2, and their cognate receptor SfLFTR (a leucine-rich-repeat-containing GPCR) in Spodoptera frugiperda. Liposome-encapsulated siRNA-mediated knockdown of these genes severely disrupted ovarian development as evidenced by suppressed vitellogenin biosynthesis and reduced mature follicle formation. Coimmunoprecipitation and molecu

Molecular characterization of three GnRH receptor paralogs in the European eel, Anguilla anguilla: tissue-distribution and changes in transcript abundance during artificially induced sexual development.

Gonadotropin-releasing hormone receptor (GnRH-R) activation stimulates synthesis and release of gonadotropins in the vertebrate pituitary and also mediates other processes both in the brain and in peripheral tissues. To better understand the differential function of multiple GnRH-R paralogs, three GnRH-R genes (gnrhr1a, 1b, and 2) were isolated and characterized in the European eel. All three gnrhr genes were expressed in the brain and pituitary of pre-pubertal eels, and also in several peripheral tissues, notably gills and kidneys. During hormonally induced sexual maturation, pituitary expres

Endocrine control of embryonic diapause in the Australian sharpnose shark Rhizoprionodon taylori.

The reproductive cycle of the Australian sharpnose shark, Rhizoprionodon taylori, includes a temporary suspension of development at the commencement of embryogenesis termed embryonic diapause. This study investigated levels of 17β-estradiol (E2), testosterone (T) and progesterone (P4) in plasma samples of mature wild female R. taylori captured throughout the reproductive cycle and correlated them with internal morphological changes. Levels of T were elevated through most of the embryonic diapause period, suggesting a role of this hormone in the maintenance of this condition. Increasing plasma

Adverse outcome pathways: a conceptual framework to support ecotoxicology research and risk assessment.

Ecological risk assessors face increasing demands to assess more chemicals, with greater speed and accuracy, and to do so using fewer resources and experimental animals. New approaches in biological and computational sciences may be able to generate mechanistic information that could help in meeting these challenges. However, to use mechanistic data to support chemical assessments, there is a need for effective translation of this information into endpoints meaningful to ecological risk-effects on survival, development, and reproduction in individual organisms and, by extension, impacts on pop

Determination of Fsh quantity and bioactivity during sex differentiation and oogenesis in European sea bass.

Follicle-stimulating hormone (FSH) is a glycoprotein hormone that plays a key role in the regulation of gonadal functions in vertebrates. The present study reports the monitoring of pituitary and plasma Fsh levels during sex differentiation and oogenesis in European sea bass (Dicentrarchus labrax) using a homologous immunoassay and an in vitro bioassay. Both assays were used complementarily for the first time in a fish species. High levels of Fsh bioactivity in plasma were found during the initial phases of sexual differentiation. Plasma and pituitary Fsh (quantity and bioactivity) levels and

The effect of a parenteral ivermectin/closantel injection on the growth and reproductive development of early immature Fasciola hepatica in cattle.

Sixteen calves approximately 6 months old were each infected with 500 metacercariae of Fasciola hepatica. Thirty-two days later they were weighed and divided into two groups, and on day 35 all calves in one of the groups were injected subcutaneously with an ivermectin/closantel combination. Both groups were sacrificed between days 70 and 72 to enable counting and examination of the flukes recovered from the bile ducts. Eggs released by the flukes were collected for incubation, hatching and estimation of egg viability. Flukes were counted, flat-fixed in 70% ethanol, stained with catechol and ca

Seasonal changes in gonadal activity and the effects of stress on reproductive hormones in the common snapping turtle, Chelydra serpentina.

The seasonal gonadal cycle (including gonadal histology, sex steroids, and gonadotropins) was studied in freshly captured common snapping turtles, Chelydra serpentina, from Wisconsin, and the effects of capture stress were evaluated. The ovarian and testicular cycles are shorter than those reported in other freshwater turtles; the cycles commence in mid-May and terminate in early September, immediately after the completion of gonadal growth and maturation. In the female, testosterone (T), 17beta-estradiol (E2), and progesterone (Pro) were highly correlated with follicular growth and vitellogen

Vitellogenetic defects in hybrids of the species pair Drosophila virilis and Drosophila texana.

In interspecific matings between the species Drosophila virilis and Drosophila texana, female sterility can be observed in F2 backcross females and in F2 hybrid females. The results presented in this report show that the female sterility, whenever it exists, is due to prevention of vitellogenin synthesis in the fat body, but other abnormalities such as defects with the hybrid ovaries are not excluded. The observation that sterility appears among females from backcrosses suggests that incompatibilities between interspecific genes may cause female sterility even in the presence of a complete hab

Xenoestrogen exposure and effects in English sole (Parophrys vetulus) from Puget Sound, WA.

Vitellogenin, a yolk protein produced in the liver of oviparous animals in response to estrogens, normally occurs only in sexually mature females with developing eggs. However, males can synthesize vitellogenin when exposed to environmental estrogens, making the abnormal production of vitellogenin in male animals a useful biomarker for xenoestrogen exposure. In 1997-2001, as part of the Washington State's Puget Sound Assessment and Monitoring Program, we surveyed English sole from a number of sites for evidence of xenoestrogen exposure, using vitellogenin production in males as an indicator. Absence of daily cycles in plasma sex steroids in male and female tuatara (Sphenodon punctatus), and the effects of acute capture stress on females.

The possible existence of daily cycles in plasma concentrations of sex steroids was examined in wild male and female tuatara (Sphenodon punctatus). Samples were collected from freshly captured animals at dusk, middle of the night, dawn, and middle of the day in January (summer) and July (winter). Males showed daily cycles in mean body temperature (Tb) in both seasons but no daily cycle in mean plasma testosterone concentration in either season. Vitellogenic female tuatara in January and females in mixed reproductive condition in July also showed significant daily variation in Tb. However, ther

Kr-h1 Encoding Juvenile Hormone Transcription Factor Impacts Reproductive Functions in Coccinella septempunctata.

The gene encoding juvenile hormone transcription factor Krüppel homolog 1 (Kr-h1) was studied for its effects on ovary development and reproduction in the ladybug beetle, Coccinella septempunctata. Kr-h1 expression in C. septempunctata was evaluated in females supplied with a juvenile hormone (JH) amended diet using quantitative PCR, and the function of Kr-h1 in female reproduction was assessed using RNAi technology. Expression of Kr-h1 in females supplied with a JH diet was significantly higher at 5 and 10 d than in females not supplied with JH. At 5 and 10 d post-injection with Kr-h1-dsRNA,

Separation of Follicular Cells and Oocytes in Ovarian Follicles of Zebrafish.

Zebrafish has become an ideal model to study the ovarian development of vertebrates. The follicle is the basic unit of the ovary, which consists of oocytes and surrounding follicular cells. It is vital to separate both follicular cells and oocytes for various research purposes such as for primary culture of follicular cells, analysis of gene expression, oocyte maturation and in vitro fertilization, etc. The conventional method uses forceps to separate both compartments, which is laborious, time consuming and has high damage to the oocyte. Here, we have established a simple method to separate b

Rapamycin inhibits pathogen transmission in mosquitoes by promoting immune activation.

Repeated blood meals provide essential nutrients for mosquito egg development and routes for pathogen transmission. The target of rapamycin, the TOR pathway, is essential for vitellogenesis. However, its influence on pathogen transmission remains to be elucidated. Here, we show that rapamycin, an inhibitor of the TOR pathway, effectively suppresses Plasmodium berghei infection in Anopheles stephensi. An. stephensi injected with rapamycin or feeding on rapamycin-treated mice showed increased resistance to P. berghei infection. Exposing An. stephensi to a rapamycin-coated surface not only decrea

Response to pinealectomy and blinding in vitellogenic female frogs (Rana perezi) subjected to high temperature in autumn.

The present experiments were carried out to investigate the effects of pinealectomy and bilateral enucleation on the ovarian activity in Rana perezi frogs maintained in 12-h light--12-h dark photoperiod and 20 +/- 1 degrees C during the vitellogenetic growth in late autumn. These environmental conditions, mainly temperature, induce a gonadal and metabolic response similar to that observed in the natural habitat in summer: a marked ovarian follicular regression, a depletion of the energetic resources from fat bodies and liver, and a minimum in oestradiol circulating levels. This response is par

Biological Functions of Growth Differentiation Factor 9 in Early Ovarian Development of Japanese Eel (Anguilla japonica).

Growth differentiation factor 9 (GDF9) is a member of the transforming growth factor-β (TGF-β) superfamily and is expressed in an oocyte-specific manner. It plays a crucial role in the early stage of ovarian development. Japanese eel (Anguilla Japonica), a spawning migration teleost, has artificial reproduction still under investigation. Aimed at developing a novel method for the successful artificial reproduction of Japanese eel, in the present study, the role of GDF9 in the regulation of early ovarian development was investigated. The Gdf9 gene in Japanese eel was 1293 bp in length, coding f

Insulin-like peptides activate egg formation in the Asian malaria mosquito Anopheles stephensi.

BACKGROUND: The mosquito family Culicidae diverged into the subfamilies Anophelinae and Culicinae approximately 179 million years ago. Most female mosquitoes are anautogenous and must blood-feed on a vertebrate to produce eggs. Regulation of egg-producing gonotrophic cycles is best understood in the culicine Aedes aegypti. Anopheline mosquitoes encode all of the hormones that regulate gonotrophic cycles in Ae. aegypti, but the processes regulating egg formation may not be fully similar. In this study, we conducted experiments that compared egg formation in Anopheles stephensi to prior findings
