Enhanced cargo of plasma alpha B crystallin in oligodendrocyte-derived exosomes accompanied by elevated expression of inflammatory factors in the temporal cortex of patients with refractory epilepsy.

PURPOSE: Plasma exosomes have emerged as key mediators of communication between neural cells and the periphery, playing pivotal roles in various neurological diseases. However, their involvement in the pathogenesis of epilepsy remains unclear. This study aimed to investigate the expression of alpha B crystallin (CRYAB) in plasma exosomes and the temporal cortex as a potential contributor to epileptogenesis. METHODS: The study included 112 participants, comprising patients with refractory medial temporal lobe epilepsy, glioma patients with and without pre-operative epilepsy, and age- and sex-m

Immune responses to Mycobacterium tuberculosis membrane-associated antigens including alpha crystallin can potentially discriminate between latent infection and active tuberculosis disease.

Changes in expression of membrane antigens may accompany the transition of Mycobacterium tuberculosis (Mtb) from 'dormant' to 'active' states. We have determined whether antibody and T cell responses to Mtb membrane (MtM)-associated antigens, especially the latency-induced protein alpha crystallin (Acr), can discriminate between latent tuberculosis infection (LTBI) and active TB (ATB) disease. Study subjects comprised a previously described cohort of healthcare workers (HCWs, n = 43) and smear-positive ATB patients (n = 10). HCWs were further categorized as occupational contacts (OC, n = 30),

Presbyopia and heat: changes associated with aging of the human lens suggest a functional role for the small heat shock protein, alpha-crystallin, in maintaining lens flexibility.

Presbyopia, the inability to focus up close, affects everyone by age 50 and is the most common eye condition. It is thought to result from changes to the lens over time making it less flexible. We present evidence that presbyopia may be the result of age-related changes to the proteins of the lens fibre cells. Specifically, we show that there is a progressive decrease in the concentration of the chaperone, alpha-crystallin, in human lens nuclei with age, as it becomes incorporated into high molecular weight aggregates and insoluble protein. This is accompanied by a large increase in lens stiff

[Action of mutant genes on crystallin synthesis in the developing mouse lens. III. The aphakia gene].

The synthesis of crystallins was studied by means of indirect immunofluorescence in 10-18 days old embryos and newborn ak/ak mice. Immune gamma-globulins to alpha-, beta- and gamma-crystallins of adult mice were used. A small amount of alpha-crystallins was found in an abnormal lens vesicle of 14 days old ak/ak embryo. The initiation of synthesis of these proteins is, hence, delayed by 4 days, as compared with that in the normal mice. In 15 days old mutant embryos the amount of alpha-crystallins increased markedly and was kept at about the same level till the end of embryogenesis. In the newbo

Assessment of structure, stability and aggregation of soluble lens proteins and alpha-crystallin upon non-enzymatic glycation: The pathomechanisms underlying cataract development in diabetic patients.

Total soluble lens proteins (TSPs) and α-crystallin (α-Cry) were individually subjected to the long-term glycation in the presence of d-glucose. The glycated and non-glycated protein counterparts were incubated under different stress conditions and compared according to their structure, stability and aggregation propensity by various spectroscopic techniques and gel mobility shift analyses. Extensive glycation of the lens proteins was accompanied with structural alteration, reduction in their surface hydrophobicity and increment of their surface tension. Our results suggest that glycation caus

Cell mediated immunity in relation to cataract and cataract surgery.

Two groups of patients suffering from cataract were investigated by the lymphocyte stimulation test. One group was tested before and the other after cataract surgery. Human (total) lens crystallins and bovine alpha crystallin were used as the antigens. Lymphocytes of healthy persons showed practically no positive stimulation with either antigen. Of the patients tested before operation 38% reacted positively with lens crystallins and 15% with alpha crystallin. This difference in sensitisation suggests a leakage of lenticular antigens other than alpha crystallin out of the lens during cataract p

Glycation of crystallins in lenses from aging and diabetic individuals.

Water-soluble crystallins were obtained from clear human lenses of different age (4-81-year-olds) and lenses of individuals showing senile or diabetic cataracts. Levels of early glycation products were high in the high molecular weight material (HM) and the alpha-crystallin fractions, compared with beta- and gamma-crystallins. This difference becomes more prominent upon aging. The content of total early glycation products in HM and alpha-crystallin increases clearly with age, whereas levels remain relatively constant in the beta- and gamma-crystallins. There is an elevation of early products i

T cell response to alpha crystallin and Mycobacterium tuberculosis specific antigens using ex-vivo elispot assay for detecting latent tuberculosis infection in Addis Ababa, Ethiopia.

BACKGROUND: Mycobacterium tuberculosis (Mtb) persists in a state of non-replication or stationary phase, but resulting in active tuberculosis (TB) when the immune system is suppressed. Alpha-crystallin (ACR) is one of the bacterial antigens characterized known to be related to shifting of the bacilli from growth to a non-replicating persistent state. OBJECTIVE: To compare the ex-vivo responsiveness of active TB patients, close household contacts and healthy controls to specific Mtb antigens. METHODOLOGY: Antigen-specific interferon-gamma (IFN-g) responses were measured to a 16kDa-alpha cryst

Biochemical comparison of lens crystallins from three reptilian species.

Lens crystallins were isolated from the homogenates of reptilian eye lenses derived from three different species by gel-permeation chromatography and characterized by gel electrophoresis, amino-acid analysis, N-terminal sequence analysis and circular dichroism. Four fractions corresponding to alpha-, delta/epsilon/beta-, beta- and gamma-crystallins were obtained for the crystallins from caiman lenses, whereas delta- and gamma-crystallin fraction were present in lesser amounts or missing in the turtle and snake lenses, respectively. The native molecular masses for these purified fractions and t

Acidic lens-protein degrading activity in bovine ciliary body. III. Lens crystallin degradation by fractions A and B.

We studied the in vitro digestion of lens crystallin by two fractions (Fractions A and B) of acidic lens-protein degrading activity from the bovine ciliary body. With whole lens crystallin as substrate, Fraction A degraded beta L-crystallin most significantly, and beta H-, gamma-, and alpha-crystallins moderately. Fraction B degraded alpha-crystallin most significantly, beta H- and beta L-crystallins moderately, and gamma-crystallin slightly. Insoluble protein was most resistant to digestion by Fractions A and B. Crystallins from the lens cortex and nucleus were degraded in the same manner as

In vitro glycation and acetylation (by aspirin) of rat crystallins.

In vitro studies with rat lens crystallins were conducted to explore the mechanism by which aspirin (ASA-acetylsalicylic acid) could inhibit cataractogenesis. The purpose of the present study is to show whether gamma-crystallin is the primary target for glycation by glucose and acetylation by ASA. Lens soluble fractions from one and seven month old Sprague-Dawley rats were incubated with 5 mM [14C]glucose with and without 10 mM ASA. alpha, beta, and gamma-crystallins were separated by molecular sieve HPLC and specific activities of each crystallin determined. In vitro acetylation was also stud

epsilon-Crystallin, a novel avian and reptilian eye lens protein.

Gel filtration of Peking duck eye lens proteins reveals a component eluting just behind delta-crystallin and comprising approximately 10% of the total soluble protein. The native Mr of this additional component is estimated to be 120000; it appears to be composed of three identical chains of Mr 38000 and pI 7.5. Circular dichroic spectroscopy showed a relatively high alpha-helical content. No immunological cross-reactivity is found with alpha-, beta-, gamma- or delta-crystallins, and partial amino acid sequence determinations likewise failed to reveal any similarity with other known crystallin

Glycation by ascorbic acid oxidation products leads to the aggregation of lens proteins.

Previous studies from this laboratory have shown that there are striking similarities between the yellow chromophores, fluorophores and modified amino acids released by proteolytic digestion from calf lens proteins ascorbylated in vitro and their counterparts isolated from aged and cataractous lens proteins. The studies reported in this communication were conducted to further investigate whether ascorbic acid-mediated modification of lens proteins could lead to the formation of lens protein aggregates capable of scattering visible light, similar to the high molecular aggregates found in aged h

Immunity to ocular and collagen antigens in childhood arthritis and uveitis.

Humoral and cellular immunity to ocular antigens (S antigen and alpha, beta heavy, beta light, and gamma crystallins) and connective tissue antigens (type I, II, III, and IV collagens) were studied in children with juvenile rheumatoid arthritis (JRA) with or without uveitis and in controls. There was no association between the presence of uveitis and antibody to any collagen type or of lymphocyte transformation to type II collagen. Antibodies to all crystallins were more common in children with JRA than in controls, and antibodies to BH crystallins were more frequent in children with JRA and u

[Immunochemical study of the water-soluble lens proteins in the embryo of Xenopus laevis with the mutation of periodic albinism].

The crystallins of ap mutants of Xenopus laevis have been studied in comparison with those of normal embryos and adults using the complex of immunochemical methods (immunoelectrophoresis, immunodiffusion, immunoadsorption, immunofluorescence, isoelectrofocusing with immunoidentification). The analysis was carried out with antisera to electrophoretic fractions of the mutant lens. 11 organ-specific antigens were found in the lens of both the normal and mutant animals. These proteins are heterogenous by electrophoretic mobility, isoelectrical point, antigenic and species specificity. Each class o

Conformational changes in soluble lens proteins during the development of senile nuclear cataract.

Soluble crystallins were isolated, by gel filtration, from the nucleus and cortex of types I-IV cataractous lenses and neonatal lenses. Extinction coefficients were determined and used to calculate the proportion of the crystallins. alpha Crystallins were the major proteins in the cortex of the cataractous lens whereas beta crystallins predominate in the nucleus and in the neonatal lens. Electrophoretic and immunological data, indicated that there was very little soluble gamma crystallin in the cataractous tissue. Highly specific competitive radioimmunoassays were used to estimate the crystall

Triose phosphate isomerase, a novel enzyme-crystallin, and tau-crystallin in crocodile cornea. High accumulation of both proteins during late embryonic development.

Several enzymes are known to accumulate in the cornea in unusually high concentrations. Based on the analogy with lens crystallins, these enzymes are called corneal crystallins, which are diverse and species-specific. Examining crystallins in lens and cornea in multiple species provides great insight into their evolution. We report data on major proteins present in the crocodile cornea, an evolutionarily distant taxon. We demonstrate that tau-crystallin/alpha-enolase and triose phosphate isomerase (TIM) are among the major proteins expressed in the crocodile cornea as resolved by 2D gel electr
