C-phycocyanin protects against ethanol-induced gastric ulcers in rats: Role of HMGB1/NLRP3/NF-κB pathway.

Gastric ulcer is a widespread inflammatory disease with high socio-economic burden. C-phycocyanin is one of the active constituents of Spirulina microalgae, and although it is well known for its antioxidant and anti-inflammatory properties, its protective effects against gastric ulcer have not yet been identified. High-mobility group box 1 (HMGB1) is a nuclear protein that, once secreted extracellularly, initiates several inflammatory reactions, and it is involved in the pathogenesis of gastric ulcer. The aim of the present study was to investigate the anti-inflammatory and anti-ulcerogenic ef

Dynamic expression of intra- and extra-cellular proteome and the influence of epiphytic bacteria for Nostoc flagelliforme in response to rehydration.

Nostoc flagelliforme is well known for its strong ecological adaptability in inhabiting desert biological soil crusts. However, the mechanism of its recovery from quiescent to active state after prolonged dormancy remains poorly characterized. Especially how exoproteome be related to the adaptive strategies and participate in the microalgae-bacteria interaction. In the present work, we analysed the intra- and extra-cellular proteome of N. flagelliforme over a complete rehydration period both in sterilization and in natural condition for the first time. The protein expression profile for N. fla

Current genetic engineering strategies for the production of antihypertensive ACEI peptides.

Hypertension is a major and highly prevalent risk factor for various diseases. Among the most frequently prescribed antihypertensive first-line drugs are synthetic angiotensin I-converting enzyme inhibitors (ACEI). However, since  their use in hypertension therapy has been linked to various side effects, interest in the application of food-derived ACEI peptides (ACEIp) as antihypertensive agents is rapidly growing. Although promising, the industrial production of ACEIp through conventional methods such as chemical synthesis or enzymatic hydrolysis of food proteins has been proven troublesome.

Heat shock protein genes in the green alga Tetraselmis suecica and their role against redox and non-redox active metals.

Microalgae are capable of tolerating variations in water temperature and sudden exposures to toxic substances, and cellular heat shock proteins (HSPs) help to protect cells from such stress. Here, we determined the complete open reading frames (ORF) of small TsHSP20 and large TsHSP70 and 100 in the chlorophyte Tetraselmis suecica, and examined the expression levels of these genes after exposure to thermal stressors, redox-active metals, and non-redox-active metals. Putative TsHSP20, TsHSP70, and TsHSP100 proteins had conserved HSP-family motifs with different C-terminus motifs. Phylogenetic an

In vivo functionalization of diatom biosilica with sodium alendronate as osteoactive material.

Bisphosphonates are a class of drugs widely used in the clinical treatment of disorders of bone metabolism, such as osteoporosis, fibrous dysplasia, myeloma and bone metastases. Because of the negative side effects caused by oral administration of bisphosphonates, various silica mesoporous materials have been investigated for a confined and controlled release of these drugs. Here, we propose biosilica from diatoms as suitable substrate for alendronate local activation of bone cells. Following a novel strategy, sodium alendronate can be in vivo incorporated into biosilica shells of cultured Tha

The acclimation of Chlorella to high-level nitrite for potential application in biological NOx removal from industrial flue gases.

Nitrogen oxides (NOx) are the components of fossil flue gas that give rise to the greatest environmental concerns. This study evaluated the ability of the green algae Chlorella to acclimate to high level of NOx and the potential utilization of Chlorella strains in biological NOx removal (DeNOx) from industrial flue gases. Fifteen Chlorella strains were subject to high-level of nitrite (HN, 176.5 mmolL(-1) nitrite) to simulate exposure to high NOx. These strains were subsequently divided into four groups with respect to their ability to tolerate nitrite (excellent, good, fair, and poor). One st

Detection of marine microalgal biotoxins using bioassays based on functional expression of tunicate xenobiotic receptors in yeast.

Marine microalgae can produce biotoxins that cause widespread poisoning in marine ecosystems and may also affect human health. While established microalgal biotoxins are detectable using chemical methods, a need remains for robust, inexpensive bioassays. Ligand-binding domains (LBDs) from a tunicate nuclear receptor, VDR/PXRα, which is orthologous to both the vertebrate pregnane X receptor (PXR) and the vitamin D receptor (VDR), can be activated by microalgal biotoxins when expressed in mammalian cell lines. Building on this observation, we developed a generic recombinant yeast bioassay platfo

Safety evaluation of Whole Algalin Protein (WAP) from Chlorella protothecoides.

Microalgae such as Chlorella spp., were once consumed as traditional human foods; now they are being developed as ingredients for modern diets. Whole Algalin Protein (WAP) from dried milled Chlorella protothecoides was evaluated for dietary safety in a 13-week feeding trial in rodents with genotoxic potential evaluated using in vitro and in vivo assays and the likelihood of food allergy potential evaluated via human repeat-insult patch test (HRIPT). In the subchronic study, rats consumed feed containing 0, 25,000, 50,000 or 100,000 ppm WAP for 92-93 days. No treatment-related mortalities or ef

High plasticity in the production of diatom-derived polyunsaturated aldehydes under nutrient limitation: physiological and ecological implications.

Diatoms have evolved a silicified cell wall that provides an efficient barrier against herbivores. These microalgae also produce chemical compounds such as polyunsaturated aldehydes (PUAs) that can potentially impair recruitment and cause malformations in the offspring of such grazers. We measured silica content as an indication of cell wall thickness, organic nutrient cell quotas, PUAs and polyunsaturated fatty acid cell content in Skeletonema marinoi grown under N-, P- and Si-limitation in continuous cultures. A 7.5 fold increase in PUA production was observed (27.5 fmol cell(-1)) in Si-limi

Cell architecture regulates species-specific micro-nanoplastic toxicity through targeted organelle disruption and energetic pathway rewiring in freshwater algae: Insights from deuterium isotope tracing, ultrastructural profiling, and multi-omics.

Micro-nanoplastics (MNPs) are prevalent in aquatic environments, yet the species-specific toxicological mechanisms associated with inherent cellular architecture remain poorly understood. Here we utilized synthesized deuterium (2H) stable isotope-labeled polystyrene MNPs (2H-PSMNPs; 0.1 μm and 2 μm) to quantify cellular uptake in two microalgae with contrasting cell architectures: Chlamydomonas reinhardtii (rigid cell wall) and Euglena gracilis (cell-wall deficient). Exposure at 1 mg/L (environmentally relevant) and 5 mg/L (a mechanistic dose) showed that 2H-PSMNPs inhibited growth in both alg

Molecular Evaluation of Deep Eutectic Solvents for Wet Algae Lipid Extraction and Biofuel Production.

Microalgae represent promising feedstocks for biofuel production, yet cell-wall disruption remains a critical bottleneck for scaling up lipid extraction. This study evaluates four cholinium-based ionic liquids (ILs) and deep eutectic solvents (DESs) as green, energy-efficient pretreatment agents to facilitate the lipid release from Chlorella vulgaris under wet conditions. A geranic acid-based DES (CG 1-2 DES) was identified as the optimal agent, demonstrating direct wet biomass extraction with substantial process energy savings at comparable overall lipid yields. To minimize the energetic cost

Multiomics studies reveal how ambient temperature changes govern cellular responses of Chlamydomonas.

Photosynthetic protists, known as microalgae, face increasing temperatures due to climate change. The green biflagellate alga Chlamydomonas reinhardtii (Chlamydomonas) serves as a model for thermoregulation. While responses to thermal stress are well characterized, much less is known about the impact of ambient temperature shifts. Understanding microalgal responses to environmental temperature changes is critical, as these primary producers drive ecosystem productivity and food web dynamics. Here, Chlamydomonas grew mixotrophically at ambient temperatures from 18 °C to 33 °C. Transcriptomic pr

Effects of Orange Peel-Derived Carbon Sources on Nannochloropsis salina Mixotrophic Cultivation.

Microalgae are recognized as sustainable biofactories for metabolites relevant to nutraceutical, pharmaceutical, and marine drug applications. Nannochloropsis salina is notable for its production of bioactive lipids, including the pharmaceutically relevant omega-3 eicosapentaenoic acid (EPA). Here, we evaluated orange peel extract (OPE), a citrus by-product, as a substrate for mixotrophic cultivation of N. salina within a bioeconomy framework. OPE supplementation triggered dose-dependent physiological responses. Among the tested OPE concentrations, 5% supplementation resulted in the highest to

Chlorella-enriched hydrogels protect against myocardial damage and reactive oxygen species production in anin vitroischemia/reperfusion model using cardiac spheroids.

Microalgae have emerged as promising photosynthetic microorganisms for biofabricating advanced tissue constructs, with improved oxygenation and reduced reactive oxygen species (ROS) production. However, their use in the engineering of human tissues has been limited due to their intrinsic growth requirements, which are not compatible with human cells. In this study, we first formulated alginate-gelatin (AlgGel) hydrogels with increasing densities ofChlorella vulgaris. Then, we characterised their mechanical properties and pore size. Finally, we evaluated their effects on cardiac spheroid (CS) p

The Potential of Spirulina platensis to Ameliorate the Adverse Effects of Highly Active Antiretroviral Therapy (HAART).

The human immunodeficiency virus (HIV) is one of the most prevalent diseases globally. It is estimated that 37.7 million people are infected with HIV globally, and 8.2 million persons are infected with the virus in South Africa. The highly active antiretroviral therapy (HAART) involves combining various types of antiretroviral drugs that are dependent on the infected person's viral load. HAART helps regulate the viral load and prevents its associated symptoms from progressing into acquired immune deficiency syndrome (AIDS). Despite its success in prolonging HIV-infected patients' lifespans, th

In vitro intestinal digestion of lipids from the marine diatom Porosira glacialis compared to commercial LC n-3 PUFA products.

Marine sources of long chain omega-3 polyunsaturated fatty acids (LC n-3 PUFA) are in high demand for use in health supplements. Mass cultivated marine microalgae is a promising and sustainable source of LC n-3 PUFA, which relieves pressure on natural fish stocks. The lipid class profile from cultivated photosynthetic algae differ from the marine organisms currently used for the production of LC n-3 PUFA. The objective of this study was to compare in vitro intestinal digestion of oil extracted from the cold-adapted marine diatom Porosira glacialis with commercially available LC n-3 PUFA supple

Molecularly imprinted nanoparticle-based assay (MINA): Potential application for the detection of the neurotoxin domoic acid.

Domoic acid (DA) is a natural amino acid and water-soluble neurotoxic biotoxin primarily produced by the microalgae Pseudo-nitzschia. DA can cause poisoning in humans and a wide variety of marine species. In this work, a molecularly imprinted nanoparticle-based assay (MINA) was developed as an alternative to enzyme-linked immunosorbent assay (ELISA) for selective detection of DA. In contrast with ELISA, MINA uses molecularly imprinted polymer nanoparticles (nanoMIPs) as plastic antibodies due to its higher stability and lower production costs. In this work, dihydrokainic acid (DKA) was used as
