Phylogeography of Lassa Virus in Nigeria.

Lassa virus is genetically diverse with several lineages circulating in West Africa. This study aimed at describing the sequence variability of Lassa virus across Nigeria and inferring its spatiotemporal evolution. We sequenced and isolated 77 Lassa virus strains from 16 Nigerian states. The final data set, including previous works, comprised metadata and sequences of 219 unique strains sampled between 1969 and 2018 in 22 states. Most of this data originated from Lassa fever patients diagnosed at Irrua Specialist Teaching Hospital, Edo State, Nigeria. The majority of sequences clustered with t

Cascading network failure across the Alzheimer's disease spectrum.

Complex biological systems are organized across various spatiotemporal scales with particular scientific disciplines dedicated to the study of each scale (e.g. genetics, molecular biology and cognitive neuroscience). When considering disease pathophysiology, one must contemplate the scale at which the disease process is being observed and how these processes impact other levels of organization. Historically Alzheimer's disease has been viewed as a disease of abnormally aggregated proteins by pathologists and molecular biologists and a disease of clinical symptoms by neurologists and psychologi

Emergence of dual β-lactam and colistin resistance via blaFRI-8 and mcr-10.2 co-carriage on an IncFII-family plasmid in Enterobacter vonholyi.

OBJECTIVES: Enterobacter vonholyi isolate E323169 represents a rare case of co-carriage of the antimicrobial resistance genes (ARGs) blaFRI-8 and mcr-10.2, recovered from a clinical rectal swab. E323169 represents one of only 11 known E. vonholyi genomes. To date, within the NCBI Pathogen Detection resource, blaFRI-8 is present in three assemblies and mcr-10.2 in two. In that dataset, blaFRI-8 co-occurred with an mcr-10 variant only once (Enterobacterasburiae). This study analyses the genomic, phenotypic and epidemiological importance of this rare co-occurrence. METHODS: Species identificatio

Streamlined extraction of nucleic acids and metabolites from low- and high-biomass samples using isopropanol and matrix tubes.

UNLABELLED: An essential aspect of population-based research is collecting samples outside of a clinical setting. This is crucial because microbial populations are highly dynamic, varying significantly across hosts, environments, and time points, a variability that clinical sample collection alone cannot fully capture. At-home sample collection enables the inclusion of a larger and more diverse group of participants, accounting for differences in ethnicity, age, and other factors. However, managing large studies is challenging due to the complexities involved in sample acquisition, processing,

AI-Generated "Slop" in Online Biomedical Science Educational Videos: Mixed Methods Study of Prevalence, Characteristics, and Hazards to Learners and Teachers.

BACKGROUND: Video-sharing sites such as YouTube (Google) and TikTok (ByteDance) have become indispensable resources for learners and educators. The recent growth in generative artificial intelligence (AI) tools, however, has resulted in low-quality, AI-generated material (commonly called "slop") cluttering these platforms and competing with authoritative educational materials. The extent to which slop has polluted science education video content is unknown, as are the specific hazards to learning from purportedly educational videos made by AI without the use of human discretion. OBJECTIVE: Th

Gastric Cancer Pre-Stage Detection and Early Diagnosis of Gastritis Using Serum Protein Signatures.

BACKGROUND: A gastric cancer (GC) diagnosis relies on histopathology. Endoscopy rates are increasing. Helicobacter pylori infection is a major GC risk factor. In an effort to elucidate abundant blood biomarkers, and potentially reduce the number of diagnostic surgical interventions, we investigated sera and biopsies from a cohort of 219 H. pylori positive and negative patients diagnosed with GC, gastritis, and ulcers. This allowed the comparative investigation of the different gastroduodenal diseases, and the exclusion of protein changes resulting from bacterial infection or inflammation of th

Associations between sexual habits, menstrual hygiene practices, demographics and the vaginal microbiome as revealed by Bayesian network analysis.

The vaginal microbiome plays an influential role in several disease states in reproductive age women, including bacterial vaginosis (BV). While demographic characteristics are associated with differences in vaginal microbiome community structure, little is known about the influence of sexual and hygiene habits. Furthermore, associations between the vaginal microbiome and risk symptoms of bacterial vaginosis have not been fully elucidated. Using Bayesian network (BN) analysis of 16S rRNA gene sequence results, demographic and extensive questionnaire data, we describe both novel and previously d

Synthetic Medical Document Generation Pipeline (MedDocSynth) for Optical Character Recognition: Development and Validation Study on Korean Clinical Documents.

BACKGROUND: The development of structured optical character recognition (OCR) systems for Korean medical documents is hindered by strict privacy regulations that limit access to real clinical data. Existing synthetic document frameworks are primarily designed for generic layouts and Western languages, and fail to capture the complex formatting, bilingual terminology, and hierarchical field semantics required for clinical OCR and document-understanding tasks in hospital environments. OBJECTIVE: This study aimed to develop and validate a privacy-preserving, template-driven pipeline for generati

Identifying Strategies for Home Management of Ostomy Care: Content Analysis of YouTube.

BACKGROUND: The social media platform YouTube is a recognized educational resource for health information, but few studies have explored its value for conveying the lived experience of individuals managing chronic health conditions and end users' interactions with medical device technology. Our study explores self-care strategies and end user needs of people living with a stoma because patient education and engagement in ostomy self-care are essential for avoiding ostomy-related complications. Ostomy surgery creates a stoma (an opening) in the abdomen to alter the route of excreta from digesti

MINMON-J: a hybrid implementation pilot study evaluating a low-barrier hepatitis C treatment model in a jail setting.

INTRODUCTION: Hepatitis C virus (HCV) remains a leading cause of infectious disease-related morbidity in the USA, disproportionately affecting people who inject drugs and people who are incarcerated. Despite the availability of highly effective, highly tolerated direct-acting antivirals, treatment uptake in jails remains limited due to short stays, unpredictable release dates and system-level barriers. The original MINMON trial demonstrated that a low barrier 'minimal monitoring"' model can achieve high cure rates in community settings. This study, MINMON-J, aims to adapt and evaluate a modifi

Expression Profiles of Protease in Onychomycosis-Related Pathogenic Trichophyton rubrum and Tinea Capitis-Related Pathogenic Trichophyton violaceum.

The two fungal species Trichophyton rubrum and Trichophyton violaceum are common pathogens on human, infecting keratinized tissue of the outer body parts. Both species are belonging to the "Trichophyton rubrum complex" and share very high similarity in the genome. Secreted proteinases, key factors for keratin degradation, are nearly identical. Contrary, the ecological niches are differing. Trichophyton rubrum preferably infects skin and nails, whereas T. violaceum preferably infects the scalp. We postulate, that differences in the protease expression contribute to differences in ecological pre

Carnivorous Nepenthes Pitchers with Less Acidic Fluid House Nitrogen-Fixing Bacteria.

Carnivorous pitcher plants are uniquely adapted to nitrogen limitation, using pitfall traps to acquire nutrients from insect prey. Pitcher plants in the genus Sarracenia may also use nitrogen fixed by bacteria inhabiting the aquatic microcosms of their pitchers. Here, we investigated whether species of a convergently evolved pitcher plant genus, Nepenthes, might also use bacterial nitrogen fixation as an alternative strategy for nitrogen capture. First, we constructed predicted metagenomes of pitcher organisms from three species of Singaporean Nepenthes using 16S rRNA sequence data and correla

Reducing Clinician Annotation Fatigue in Open-Set Federated Learning: A Client-Adaptive Vision-Language Gatekeeper.

PURPOSE: Federated learning enables breast-imaging sites to jointly train mammography artificial intelligence (AI) without sharing images, but radiologists at each site must still annotate selected images during active-learning rounds. We developed and evaluated a client-adaptive vision-language gatekeeper that withholds radiopaque artifact-containing images considered unsuitable for a breast-density annotation objective and quantified its effect on annotation time, queue quality, and label reliability. METHODS: In this retrospective, scanner-partitioned study, we used FedEMBED, derived from

Multi-habitat microbiome profiling identifies habitat-dependent alterations and complementary discriminatory information in urolithiasis.

BACKGROUND: Urolithiasis has been associated with microbial alterations in individual anatomical niches; however, whether microbial signatures across urinary, intestinal, and oral habitats represent shared, site-specific, or complementary disease-associated information remains unclear. This study aimed to characterize multi-habitat microbiome alterations associated with urolithiasis and to evaluate whether integrated multi-site profiling captures internally cross-validated disease-associated microbial information. METHODS: Salivary, clean-catch midstream urinary, and fecal samples were collec

Sex-, Age-, and Projection-Related Variability in Pneumothorax and Pulmonary Mass Detection Frequencies on Chest Radiographs: A Retrospective Cross-Sectional Study.

BACKGROUND: Chest radiography remains one of the most widely used imaging modalities for the detection of thoracic abnormalities, including pneumothorax and pulmonary masses. However, the frequency with which these findings are identified may vary according to demographic characteristics and radiographic acquisition techniques. Understanding such variability is important for interpreting large imaging datasets and may improve diagnostic assessment and epidemiological research. Therefore, this study aimed to evaluate sex-, age-, and projection-related differences in the dataset-derived detectio

Beyond the Inner Cell Mass: Evaluating the Impact of Inner Cell Mass Visibility on Implantation Prediction in Static Blastocyst Images - A Retrospective Cohort Study.

BACKGROUND: Embryo selection in IVF often relies on inner cell mass (ICM) morphology as a predictor of implantation. However, in static two-dimensional images, ICM visibility may be limited by technical factors like orientation or focus, rather than actual quality. Since most IVF labs still rely on static images for embryo evaluation, it is important to reconsider the emphasis placed on ICM and investigate other morphological features that may support clinical decision-making. AIM: To assess whether the visibility of the inner cell mass (ICM) in static blastocyst images affects the accuracy o
