The rise of antimicrobial resistance and the persistence of difficult-to-treat infections have stimulated interest in new strategies to overcome these problems. Among these strategies, dendrimers, which are highly branched monodisperse macromolecules, have emerged as innovative antimicrobial and anti-infective platforms. Dendrimers can act as intrinsic antimicrobial agents through multivalent interactions or membrane disruption or serve as nanocarriers for antibiotics, antiviral agents, antibiofilm compounds, gas-releasing active molecules, or photosensitizers. This review analyzes dendrimer-b

BACKGROUND: There is widespread consensus that adjuvant local use of antimicrobial agents in combination with their systemic administration can better prevent and treat implant-associated musculoskeletal infections. The advantage of local antibiotics lies in their particular pharmacokinetics with initially high antibiotic concentrations at the implant site with only low systemic uptake.
AIM OF TREATMENT: The aim of local application is to protect the foreign bodies directly at the implantation site from bacterial colonization and biofilm formation (prophylaxis) and to support the eradication

Drugs induced respiratory disorders include bronchospasm, extrinsic alveolitis, and diffuse interstitial pulmonary fibrosis. Bronchospasm may be due to the action of pharmacodynamic agents on autonomic control of bronchial tone (beta-blockers, beta-agonists...) or to a hypersensitivity reaction (penicillins, sulfonamides, anti-inflammatory compounds...). Extrinsic alveolitis usually follows administration of anti-infective medication but other drugs may be implicated. Prolonged use of a wide variety of drugs, particularly cytotoxic agents, is necessary to provoke the onset of a diffuse interst

Peptoids, a class of peptide mimetics, have emerged as promising anti-infective agents against a range of bacterial infections. Herein we present the first study of the antiparasitic and specifically the anti-leishmanial properties of linear peptoids. Peptoids were identified as having promising activity against Leishmania mexicana axenic amastigotes, a causative agent of cutaneous leishmaniasis.

Postoperative fever represents a common clinical challenge in orthopedics, and the differential diagnosis of its etiology directly influences anti-infective decision-making and patient outcomes. In the context of anti-infective therapy, "drug fever" induced by antimicrobial agents themselves can easily be mistaken for uncontrolled or recurrent infection, leading to antibiotic overuse. This article reports the case of a 49-year-old male with a comminuted fracture of the right femur. Preoperatively, he presented with post-traumatic absorption fever. Postoperatively, due to intraoperative observa

Antimicrobial agents are undoubtedly one of the key advances in the history of modern medicine and infectious diseases, improving the clinical outcomes of infection owing to their inhibitory effects on microbial growth. However, many antimicrobial agents also have biological activities stemming from their interactions with host receptors and effects on host inflammatory responses and other human or bacterial cellular biological pathways. These result in clinical uses of antimicrobial drugs that are distinct from their direct bacteriostatic or bactericidal properties. We reviewed the published

INTRODUCTION: Bronchiectasis is a chronic respiratory condition characterised by cough, sputum production and recurrent chest infections. There are multiple aetiologies; but in up to 50% of patients, the aetiology is unknown. The treatment is largely symptomatic with regular chest physiotherapy and antibiotics for infective exacerbations. Research is being directed towards breaking the 'vicious circle' of bronchiectasis with therapies directed at improving mucociliary clearance, treating chronic infection and reducing inflammation in the airways.
AREAS COVERED: This review highlights the curr

BACKGROUND/AIM: Probiotics help to provide an optimum balance in the intestines. Probiotics species competitive block toxic substances and growth of unwanted bacteria and yeast species while they compete for the space and food. Lactogyn is the first oral probiotics on Serbian market dedicated to maintaining a normal vaginal flora. Lactogyn contains two well studied probiotics strains - Lactobacillus rhamnosus GR-1 and Lactobacillus reuteri RC-14. Both of them are considered as probiotic agents with therapeutic properties increase the population of beneficial lactobacillus organisms within the

ETHNOPHARMACOLOGICAL RELEVANCE: C.A. Meyer (ginseng) has been esteemed for centuries in traditional Chinese, Korean, and indigenous medicine as a vital tonic and therapeutic agent for enhancing resilience and treating infectious diseases. Historically, used for respiratory or skin infections, gastrointestinal disorders, etc. These applications align with modern evidence supporting its broad-spectrum anti-infective activity. Ginsenosides, the principal bioactive constituents, have been shown to exert targeted effects against diverse infectious pathogens. This review integrates ethnomedical insi

Timely provision of adequate antimicrobial coverage in an initial anti-infective treatment regimen results in optimal outcomes for bacterial and fungal infections. However, selection of appropriate antimicrobial regimens for treatment of infections in the intensive care unit (ICU) can be challenging due to expansion of resistance, which typically requires use of multidrug anti-infective regimens to provide adequate coverage of important pathogens commonly seen in the ICU setting. Indeed, a recent additional call to action by the Infectious Diseases Society of America (IDSA) has enforced the im

Background: Therapeutic drug monitoring (TDM) is essential for ensuring safe, effective, and individualized anti-infective therapy, particularly in patients with complex clinical needs. Variability in pharmacokinetics, challenges in drug administration, and high-dose regimens can compromise adherence and increase the risk of therapeutic failure or resistance. Swallowing difficulties, a common barrier to oral therapy, often necessitate alternative administration routes or customized formulations. However, interventions such as pharmaceutical compounding or manipulation of solid dosage forms may

This study aimed to evaluate the intrinsic in vitro performance of naïve porcine prepubertal Sertoli cells (SCs) and SCs loaded with blank poly(lactic acid) microparticles (MP) or amphotericin B poly(lactic acid) microparticles (AmB-MP) against Candida tropicalis, a prevalent pathogenic non-albicans species. The objective was to assess their impact on biofilm formation and the cellular response mechanisms involved, building on previous findings that highlight SCs' potential as anti-infective agents and drug carriers. Our results demonstrated that SCs successfully internalized Candida tropicali

Periodontal diseases are multi-factorial in etiology, and bacteria are one among these etiologic agents. Thus, an essential component of therapy is to eliminate or control these pathogens. This has been traditionally accomplished through mechanical means (scaling and root planing (SRP)), which is time-consuming, difficult, and, sometimes, ineffective. From about the past 30 years, locally delivered, anti-infective pharmacological agents, most recently employing sustained-release vehicles, have been introduced to achieve this goal. This systematic review is an effort to determine the efficacy o

Bacterial invasion into eukaryotic cells and the establishment of intracellular infection has proven to be an effective means of resisting antibiotic action, as anti-infective agents commonly exhibit a poor permeability across the host cell membrane. Encapsulation of anti-infectives into nanoscaled delivery systems, such as liposomes, is shown to result in an enhancement of intracellular delivery. The aim of the current work is, therefore, to formulate colistin, a poorly permeable anti-infective, into liposomes suitable for oral delivery, and to functionalize these carriers with a bacteria-der

Omadacycline is a novel tetracycline antibiotic that exhibits good in vitro antibacterial activity against atypical pathogens such as Mycoplasma pneumoniae. It is approved for the treatment of adults with community-acquired bacterial pneumonia. However, the safety and efficacy of omadacycline in pediatric patients under 18 years of age have not yet been established. In the present paper, we report a case of pediatric community-acquired pneumonia in which initial empirical anti-infective therapy had failed. The patient received empirical anti-infective therapy with azithromycin and other antimi

The escalating crisis of antimicrobial resistance and biofilm-associated infections has fundamentally compromised conventional anti-infective therapy. Current antimicrobial agents for refractory infections face critical limitations, including systemic toxicity, inadequate tissue penetration, and the rapid emergence of multidrug resistance (MDR). As a promising strategy, cell-based drug delivery systems (CDDS) leverage living cells or their bioactive derivatives as sophisticated carriers to overcome these barriers. By harnessing innate biological functions, such as prolonged circulation, immune

Cinnamaldehyde (CNM) is a cyclic terpene alcohol found as the major compound of essential oils from some plants of the genus Cinnamomum (Lauraceae). CNM has several reported pharmacological activities, including antimicrobial, antivirulence, antioxidant, and immunomodulatory effects. These properties make CNM an attractive lead molecule for the development of anti-infective agents. In this descriptive review, we discuss the application of CNM in experimental models of microbial infection using invertebrate and vertebrate organisms. CNM (pure or in formulations) has been successfully applied in
