Optimizing blood culture diagnostics through laboratory automation: reducing turnaround time and improving clinical outcomes.

Bloodstream infections pose significant clinical challenges, requiring rapid and accurate diagnostics to guide timely antimicrobial therapy. Although efforts to reduce turnaround time (TAT) have focused on emerging molecular techniques and rapid antimicrobial susceptibility testing (AST) methods, these innovations currently complement rather than replace standard protocols. To balance innovation with reliability, we investigated the impact of integrating laboratory automation into the blood culture diagnostic workflow while preserving conventional pathogen identification via mass spectrometry

Perioperative Fluid and Vasopressor Therapy in 2050: From Experimental Medicine to Personalization Through Automation.

Intravenous (IV) fluids and vasopressor agents are key components of hemodynamic management. Since their introduction, their use in the perioperative setting has continued to evolve, and we are now on the brink of automated administration. IV fluid therapy was first described in Scotland during the 1832 cholera epidemic, when pioneers in medicine saved critically ill patients dying from hypovolemic shock. However, widespread use of IV fluids only began in the 20th century. Epinephrine was discovered and purified in the United States at the end of the 19th century, but its short half-life limit

Automation protocol for high-efficiency and high-quality genomic DNA extraction from Saccharomyces cerevisiae.

Although many protocols have been previously developed for genomic DNA (gDNA) extraction from S. cerevisiae, to take advantage of recent advances in laboratory automation and DNA-barcode sequencing, there is a need for automated methods that can provide high-quality gDNA at high efficiency. Here, we describe and demonstrate a fully automated protocol that includes five basic steps: cell wall and RNA digestion, cell lysis, DNA binding to magnetic beads, washing with ethanol, and elution. Our protocol avoids the use of hazardous reagents (e.g., phenol, chloroform), glass beads for mechanical cel

Automation of On-Resin Enrichment of S-Nitrosylated Proteins for Oxidized Cysteine-Selective cPILOT.

S-Nitrosylation (SNO) is a cysteine post-translational modification that increases with normal aging and is present in Alzheimer's disease and other aging-related illnesses. Detection of SNO-modified proteins can be challenging; however, we previously developed a robust quantitative proteomics approach termed "Oxidized Cysteine-Selective combined precursor isobaric labeling and isobaric tagging (OxcyscPILOT)" that allows for detection of endogenous SNO-modified proteins. OxcyscPILOT involves enrichment of SNO-modified proteins using a thiol-based resin. This enrichment is performed manually, a

Impact of automation on the quantitation of low molecular weight markers of hemostatic defects.

Through in depth studies, the biochemical pathways of hemostasis-related systems have been elucidated in terms of well-defined molecular mechanisms. The interrelationships of coagulation, fibrinolytic, kallikrein-kinin, platelets, prostaglandins, blood vessel, and complement systems are now well understood. Methods are currently developed to quantitate the molecular markers of each of these systems and define the involvement of each in disease and drug-related aberrations. Molecular markers allow for very early detection of disease states well before clinical manifestations are seen or current

Artificial intelligence (AI)-assisted ultrasound in clinical trials: Endpoint automation, decentralized monitoring, and regulatory readiness.

Ultrasound is among the most widely used imaging modalities in clinical trials, and yet its dependence on operator skill and equipment settings has historically limited the reproducibility of ultrasound-based endpoints in multi-center studies. Artificial intelligence (AI) now addresses this limitation across two complementary dimensions: automated measurement algorithms that quantify cardiac function, organ volume, and vascular parameters with reproducibility approaching or, in select settings, exceeding that of trained human readers and real-time acquisition guidance systems that enable clini

Automation, Artificial Intelligence (AI), and Digital Management in IVF Laboratories: Current Status, Challenges and Potential.

Current in vitro fertilization (IVF) laboratory procedures are limited by labor-intensive, multi-step processes and technical specificity. Manual manipulation introduces variability in outcomes due to differences in operator skill and experience, and increases the risk of errors such as mix-ups or accidental loss of gametes or embryos. Additionally, subjective judgment on embryo scoring can lead to inter-observer discrepancy, compromising the reliability and consistency of assessments. However, automation, AI and digital technologies offer the solutions by standardizing IVF processes, reducing

Interface access control graph neural network method for flexible automation production of mine ventilation duct.

Interface authorization in flexible automated mine ventilation duct production lines is difficult because heterogeneous users and services access safety-critical resources under time-varying cyber-physical risk that static RBAC/ACL rules cannot capture. This study presents HGRAD, a heterogeneous-graph risk-adaptive access control framework for industrial cyber-physical systems. HGRAD models each access event as a dynamic graph with four node types: user nodes encode operator identity, role, and history; interface nodes represent exposed PLC/MES/service access points and protocol/load states; r

Centrifugal microfluidic automation of the protein aggregation capture workflow for robust mass spectrometry-based proteomics.

Proteomic sample preparation for liquid chromatography-tandem mass spectrometry (LC-MS/MS) is increasingly addressed by automated approaches. However, in clinical settings for precision medicine, where a limited number of samples must be processed in a standardized and reproducible manner with minimal user interaction, fully automized workflows remain scarce. Here, we present the AutoPAC-disk, a centrifugal microfluidic implementation of a protein aggregation capture (PAC) sample preparation workflow for bottom-up proteomics that automates all necessary steps for on-bead proteolysis including

Optimization, automation and validation of NOP receptor PET radiopharmaceutical, [11C]NOP-1A for human use.

BACKGROUND: In vivo human studies of the nociceptin/orphanin FQ peptide (NOP) receptor have been limited by the availability and reliable production of suitable PET radioligands. [11C]NOP-1A is a high-affinity PET radioligand for imaging NOP receptors; however, reproducible production suitable for routine clinical use has been challenging. The aim of this study was to optimize, automate, and validate the radiosynthesis of [11C]NOP-1A for cGMP-compliant human PET imaging. RESULTS: We developed a fully automated loop radiomethylation method for the production of [11C]NOP-1A using the commercial

Machine learning and automation methods for the segmentation, classification and quantification of testicular tissue sections.

ABSTRACT: Analysis of immunofluorescent images can be time-consuming and observer-dependant. We aimed to develop a standardised method for analysing immunofluorescent images of human and mouse testicular tissues using an existing software package. We used QuPath to train and apply an artificial neural network with multilayer perception (ANN-MLP) on images. We were able to automate the segmentation of cells in regions of interest (ROIs) using both StarDist (a convolution neural network) and Watershed image transformation. Segmented cells were classified using QuPath object classification system

Optimizing the Experimental Method for Stomata-Profiling Automation of Soybean Leaves Based on Deep Learning.

Stomatal observation and automatic stomatal detection are useful analyses of stomata for taxonomic, biological, physiological, and eco-physiological studies. We present a new clearing method for improved microscopic imaging of stomata in soybean followed by automated stomatal detection by deep learning. We tested eight clearing agent formulations based upon different ethanol and sodium hypochlorite (NaOCl) concentrations in order to improve the transparency in leaves. An optimal formulation-a 1:1 (v/v) mixture of 95% ethanol and NaOCl (6-14%)-produced better quality images of soybean stomata.
