Engineered gut symbionts mediate cross-phylum antagonism to suppress uropathogenic Escherichia coli colonization.

Urinary tract infections (UTIs) are among the most common bacterial infections globally and create a large burden on the healthcare system. Uropathogenic Escherichia coli (UPEC) account for the majority of UTIs and increase the risk of recurrence. The standard treatment is antibiotics and, with the rise of multi-drug resistant UPEC lineages, there is a need for alternative treatments and prevention. Colicins, bacteriocins targeting and produced by E. coli, have previously been shown to inhibit the growth of pathogenic E. coli and are a promising alternative. Here, we engineer commensal Bactero

Bovine colostrum-derived exosomes alleviate muscle degeneration by modulating gut microbiota and metabolic homeostasis in atrophy models.

Sarcopenia, a condition characterized by chronic systemic inflammation and a significant decline in the quality of life, is primarily associated with aging and degenerative diseases. Several studies have shown that milk exosomes contain a substantial number of miRNAs that are involved in immunity, inflammation, osteoporosis, and gut microbiota regulation. This study aimed to evaluate the potential functional role of bovine colostrum-derived exosomes (BCE) in reducing muscle atrophy. BCE treatment enhanced the viability of C2C12 myotube and stimulated myogenic differentiation, while inhibiting

Gut Dysbiosis and Hemodynamic Changes as Links of the Pathogenesis of Complications of Cirrhosis.

The aim was to evaluate the relationship between gut dysbiosis and hemodynamic changes (hyperdynamic circulation) in cirrhosis, and between hemodynamic changes and complications of this disease. This study included 47 patients with cirrhosis. Stool microbiome was assessed using 16S rRNA gene sequencing. Echocardiography with a simultaneous assessment of blood pressure and heart rate was performed to assess systemic hemodynamics. Patients with hyperdynamic circulation had more severe cirrhosis, lower albumin, sodium and prothrombin levels, higher C-reactive protein, aspartate aminotransferase a

Role of the Gut Microbiome in Modulating Arthritis Progression in Mice.

Genetics alone cannot explain most cases of rheumatoid arthritis (RA). Thus, investigating environmental factors such as the gut microbiota may provide new insights into the initiation and progression of RA. In this study, we performed 16S rRNA sequencing to characterise the gut microbiota of DBA1 mice that did or did not develop arthritis after induction with collagen. We found that divergence in the distribution of microbiota after induction was pronounced and significant. Mice susceptible to collagen-induced arthritis (CIA) showed enriched operational taxonomic units (OTUs) affiliated with

From Periodontal Pocket to Systemic Diseases: Mechanistic Insights Into the Role of Porphyromonas gingivalis in Host Pathology.

Porphyromonas gingivalis (P. gingivalis), a keystone pathogen in periodontitis, has been increasingly recognised as a mechanistic bridge linking periodontal infection to pathological destruction in distant organs. P. gingivalis virulence factors, including gingipains, lipopolysaccharides (LPS), and outer membrane vesicles (OMVs), mediate complex host-pathogen interactions. In this review, we critically evaluated recent experimental studies which demonstrate the effects of P. gingivalis oral infection on systemic diseases, including cardiovascular disease (CVD), diabetes mellitus (DM), adverse

Intraluminal gas in deep vein thrombosis associated with Rhodotorula mucilaginosa fungemia and Prevotella intermedia bacteremia: a unique clinical case report and literature overview.

Intravenous drug use (IVDU) is associated with multiple complications, including thrombus and infection with rare pathogens. Here, we present a unique case of septic thrombophlebitis with gas present within the lumen of the thrombus that was identified from an otherwise unassuming wound via point-of-care ultrasound (POCUS). In conjunction with past literature, this finding raises the possibility of POCUS detection of intraluminal gas as a sensitive indicator of septic thrombophlebitis. Furthermore, the patient's polymicrobial infection with Rhodotorula mucilaginosa and Prevotella intermedia ex

From a better knowledge of periodontal disease to Porphyromonas gingivalis target for rheumatoid arthritis disease activity.

Periodontal disease (PD) and rheumatoid arthritis (RA) are both inflammatory diseases affecting the tooth and joint, with local inflammation associated with bone loss. Bacterial infections by oral bacteria are involved in periodontal inflammation, and the best known to be associated with PD is Porphyromonas gingivalis (Pg). A large body of recent data suggests a strong involvement of this specific bacteria, Pg, in PD outcomes, but also in RA. The aim of this review is to discuss the association between PD and Pg, RA and its mechanisms, and to determine whether targeting Pg bacteria could impro

Porphyromonas gingivalis and adverse pregnancy outcomes: a review on its intricate pathogenic mechanisms.

Porphyromonas gingivalis (P. gingivalis), a Gram-negative facultative anaerobe of the oral cavity, is associated with the onset of various adverse pregnancy outcomes. P. gingivalis is linked with the development of preeclampsia, preterm labour, spontaneous abortion, gestational diabetes, foetal growth restriction, and misconception. The unique virulence factors, surface adhesions, enzymes of P. gingivalis can directly injure and alter the morphology, microbiome the foetal and maternal tissues. P. gingivalis can even exaggerate the production of cytokines, free radicals and acute-phase proteins

A mouse model for pathogen-induced chronic inflammation at local and systemic sites.

Chronic inflammation is a major driver of pathological tissue damage and a unifying characteristic of many chronic diseases in humans including neoplastic, autoimmune, and chronic inflammatory diseases. Emerging evidence implicates pathogen-induced chronic inflammation in the development and progression of chronic diseases with a wide variety of clinical manifestations. Due to the complex and multifactorial etiology of chronic disease, designing experiments for proof of causality and the establishment of mechanistic links is nearly impossible in humans. An advantage of using animal models is t

Anaerobic orbital abscess/cellulitis in a Yorkshire Terrier dog.

A retrobulbar abscess/cellulitis occurred in a Yorkshire Terrier dog. The clinical signs were exophthalmos, prolapsed nictitating membrane and purulent ocular discharge. Ultrasonography showed a marked soft tissue swelling of the retrobulbar tissues as well as echogenic parallel lines between the globe and the medial orbital rim. Surgical exploration of the orbit was performed and no foreign body was found. The pterygopalatine fossa was incised and therapeutic retrobulbar drainage attempted. A drain was placed to encourage ventral drainage of the abscess. Anaerobic cultures revealed heavy grow

Production and sensitivity of bacteriocin-like activity among Porphyromonas gingivalis, Prevotella intermedia and Pr. nigrescens strains isolated from periodontal sites.

The production of and sensitivity to bacteriocin-like activity among 44 strains of black-pigmented anaerobes isolated from periodontal sites were evaluated by both an overlay and an agar diffusion method. The species studied were Porphyromonas gingivalis, Prevotella intermedia and the closely related species Pr. nigrescens. Pr. intermedia strains (90%) produced bacteriocin-like activity against Pr. nigrescens and all Pr. nigrescens were active against Pr. intermedia. Both species showed a high degree of activity against P. gingivalis, whereas only one P. gingivalis strain produced bacteriocin-

Life below the gum line: pathogenic mechanisms of Porphyromonas gingivalis.

Porphyromonas gingivalis, a gram-negative anaerobe, is a major etiological agent in the initiation and progression of severe forms of periodontal disease. An opportunistic pathogen, P. gingivalis can also exist in commensal harmony with the host, with disease episodes ensuing from a shift in the ecological balance within the complex periodontal microenvironment. Colonization of the subgingival region is facilitated by the ability to adhere to available substrates such as adsorbed salivary molecules, matrix proteins, epithelial cells, and bacteria that are already established as a biofilm on to

Genomic characterization and preclinical evaluation of the candidate probiotic strain Lactococcus cremoris FBMS_5810.

This study presents a comprehensive genomic and preclinical evaluation of Lactococcus cremoris FBMS_5810, establishing its taxonomic identity, genomic uniqueness, and safety profile. Genomic analyses identified strain-specific genes linked to adhesion, colonization, and pathogen exclusion, aligning with previously observed in vitro probiotic properties. In vivo studies in healthy mice demonstrated that Lactococcus cremoris FBMS_5810 modulates gut microbiota composition. Specifically, the relative abundance of Muribaculaceae, Erysipelotrichaceae, and Streptococcaceae was significantly increased

An in vitro analysis of how lactose modifies the gut microbiota structure and function of adults in a donor-independent manner.

INTRODUCTION: Following consumption of milk, lactose, a disaccharide of glucose and galactose, is hydrolyzed and absorbed in the upper gastrointestinal tract. However, hydrolysis and absorption are not always absolute, and some lactose will enter the colon where the gut microbiota is able to hydrolyze lactose and produce metabolic byproducts. METHODS: Here, the impact of lactose on the gut microbiota of healthy adults was examined, using a short-term, in vitro strategy where fecal samples harvested from 18 donors were cultured anaerobically with and without lactose. The data were compiled to

Impact of Triacetin- and Tributyrin-Based Formulations on IBS Microbiome Composition and SCFA Profiles: A Preclinical Study.

Irritable bowel syndrome (IBS) is a prevalent yet poorly understood functional gut disorder frequently associated with microbial dysbiosis, making microbiome-targeting interventions a promising management strategy. In this study, we evaluated the microbiome-modulating potential of three doses of triacetin (TA; ReBiome™; 100, 200 and 300 mg/day) and two finished formulations containing TA and/or tributyrin (TB): Active Biome™ (AB) and Bio Complete 3™ (BC3). Inulin served as a benchmark prebiotic. The validated ex vivo Systemic Intestinal Fermentation Research (SIFR®) technology pipeline was app

The Therapeutic Effect and Mechanism of Vismodegib on COPD: Focusing on NETs and Macrophage Polarization.

The Hedgehog pathway may be involved in chronic obstructive pulmonary disease (COPD). Its inhibitor, Vismodegib, has therapeutic potential, but the underlying mechanisms require further investigation. A COPD mouse model was established using lipopolysaccharide (LPS) and cigarette smoke exposure, with concurrent Vismodegib intervention. Assessments included histopathology, pulmonary function, inflammatory cytokines, neutrophil extracellular traps (NETs) markers, macrophage polarization, and integrated transcriptomic and gut microbiota (16S rRNA) analysis. The results show that Vismodegib allevi

Gut microbiome dysbiosis and functional alterations in Campylobacter-associated gastroenteritis using metagenomic approaches.

Campylobacter species are a major cause of bacterial gastroenteritis worldwide. Using shotgun metagenomic sequencing of stool samples from PCR-confirmed Campylobacter-positive patients and symptomatic PCR-negative controls, we reveal dysbiosis marked by reduced species richness (median Shannon diversity was significantly lower in the Campylobacter-positive group [3.24] vs. Campylobacter-negative group [3.63], P = 0.038), taxonomic shifts toward inflammation-associated taxa (Campylobacteriaceae, Enterobacteriaceae, Pasteurellaceae), and depletion of key commensals involved in short-chain fatty

Distinct trajectory of gut microbiota driven by a human oral commensal: insights from a murine study.

BACKGROUND: Oral microbes modulate the gut microbiota. Haemophilus parainfluenzae, a core human oral commensal with immunomodulatory properties, is reduced in autoimmune diseases, while mitigating Sjögren's syndrome-like disease with improved oral microbiota in female NOD mice. However, whether it modulates the gut microbiota remains unknown. OBJECTIVE: To study the modulatory effect of oral H. parainfluenzae inoculation on the gut microbiota. DESIGN: Female NOD mice were orally inoculated with H. parainfluenzae following antibiotic treatment. Fecal samples were collected pre- and post-inocu

Distinguishing bipolar depression, bipolar mania, and major depressive disorder by gut microbial characteristics.

BACKGROUND: Gut microbial disturbance has been widely confirmed in mood disorders. However, little is known about whether gut microbial characteristics can distinguish major depressive disorder (MDD), bipolar depression (BP-D), and bipolar mania (BP-M). METHODS: This was a prospective case-control study. The composition of gut microbiota was profiled using 16S ribosomal RNA (rRNA) gene sequencing of fecal samples and compared between healthy controls (HC; n = 46), MDD (n = 51), BP-D (n = 44), and patients with BP-M (n = 45). RESULTS: Gut microbial compositions were remarkably changed in the
