Compensatory ion transport buffers daily protein rhythms to regulate osmotic balance and cellular physiology.

Between 6-20% of the cellular proteome is under circadian control and tunes mammalian cell function with daily environmental cycles. For cell viability, and to maintain volume within narrow limits, the daily variation in osmotic potential exerted by changes in the soluble proteome must be counterbalanced. The mechanisms and consequences of this osmotic compensation have not been investigated before. In cultured cells and in tissue we find that compensation involves electroneutral active transport of Na+, K+, and Cl- through differential activity of SLC12A family cotransporters. In cardiomyocyt

Mindfulness-Based Stress Reduction Buffers Glucocorticoid Resistance Among Older Adults: A Randomized Controlled Trial.

OBJECTIVE: Mindfulness interventions have been effective for improving a range of health outcomes; however, pathways underlying these effects remain unclear. Inflammatory processes may play a role, possibly through increased resistance of immune cells to the anti-inflammatory effects of glucocorticoids (i.e., glucocorticoid resistance, or GCR). Here, we conducted an initial examination of whether mindfulness training mitigates GCR among lonely older adults. METHODS: Lonely older adults (65-85 years; n = 190) were randomly assigned to an 8-week Mindfulness-Based Stress Reduction (MBSR) or a ma

Fluorescence Stability of Mercaptopropionic Acid Capped Cadmium Telluride Quantum Dots in Various Biochemical Buffers.

Quantum dots are the semiconductor nanocrystals having unique optical and electronic properties. Quantum dots are category of fluorescent labels utilized for biological tagging, biosensing, bioassays, bioimaging and in vivo imaging as they exhibit very small size, signal brightness, photostability, tuning of light emission range, longer photoluminescence decay time as compared to organic dyes. In this work, we have synthesized and characterized mercaptopropionic acid capped cadmium telluride quantum dots (MPA-CdTe QDs) using hydrothermal method. The study further reports fluorescence intensity

Presentation matters: Buffers, packaging, and delivery devices for new, oral enteric vaccines for infants.

Oral administration of vaccines is simpler and more acceptable than injection via needle and syringe, particularly for infants (Fig. 1) This route is promising for new vaccines in development against enterotoxigenic Escherichia coli (ETEC) and Shigella that cause childhood diarrhea with devastating consequences in low-resource countries. However, vaccine antigens and adjuvants given orally need buffering against the degradative effects of low stomach pH, and the type and volume of antacid buffer require special attention for infants. In addition, container/closure systems must be compatible wi

EF-hand protein Ca2+ buffers regulate Ca2+ influx and exocytosis in sensory hair cells.

EF-hand Ca(2+)-binding proteins are thought to shape the spatiotemporal properties of cellular Ca(2+) signaling and are prominently expressed in sensory hair cells in the ear. Here, we combined genetic disruption of parvalbumin-α, calbindin-D28k, and calretinin in mice with patch-clamp recording, in vivo physiology, and mathematical modeling to study their role in Ca(2+) signaling, exocytosis, and sound encoding at the synapses of inner hair cells (IHCs). IHCs lacking all three proteins showed excessive exocytosis during prolonged depolarizations, despite enhanced Ca(2+)-dependent inactivation

Maternal sensitivity buffers the adrenocortical implications of intimate partner violence exposure during early childhood.

This study prospectively examined the effect of intimate partner violence (IPV) on adrenocortical reactivity and recovery during early childhood. The sample (n = 1102 mother-infant dyads; 49.2% male) was racially diverse and from predominantly low-income, rural communities. To measure IPV exposure mothers completed the Conflicts Tactics Scale, and her caretaking behaviors were observed when her child was approximately 7, 15, and 24 months of age. Children's saliva samples, later assayed for cortisol, were collected around challenge tasks designed to elicit emotional reactivity. IPV was related

Endogenous calcium buffers regulate fast exocytosis in the synaptic terminal of retinal bipolar cells.

Calcium-triggered exocytosis at the synapse is suppressed by addition of calcium chelators, but the effects of endogenous Ca(2+) buffers have not been tested. We find that 80% of Ca(2+) binding sites in the synaptic terminal of retinal bipolar cells were associated with mobile molecules that suppressed activation of Ca(2+)-sensitive K(+) channels with an efficiency equivalent to approximately 1.2 mM BAPTA. Removing these buffers caused a 30-fold increase in the number of vesicles released by Ca(2+) tail currents lasting approximately 0.5 ms and a 2-fold increase in the rapidly releasable pool

Total weak acid concentration and effective dissociation constant of nonvolatile buffers in human plasma.

The strong ion approach provides a quantitative physicochemical method for describing the mechanism for an acid-base disturbance. The approach requires species-specific values for the total concentration of plasma nonvolatile buffers (A(tot)) and the effective dissociation constant for plasma nonvolatile buffers (K(a)), but these values have not been determined for human plasma. Accordingly, the purpose of this study was to calculate accurate A(tot) and K(a) values using data obtained from in vitro strong ion titration and CO(2) tonometry. The calculated values for A(tot) (24.1 mmol/l) and K(a

[The broncholytic effect of buffers applied by inhalation. A new pharmacological concept (author's transl)].

A new method was developed by which the tone of bronchial smooth muscle can be measured breath by breath in man. Thereby state and reactions of bronchial smooth muscle can be studied. Former investigations had shown that carbonic anhydrase inhibitors have a broncholytic effect. The mechanism of action presumably is the prevention by carbonic anhydrase inhibitors of CO2 induced changes of pH within the bronchial epithelium occurring breath by breath with different partial pressures of CO2. Principally a stabilisation of pH can also be achieved by buffers. Therefore the broncholytic effect of bu

Two new non-barbiturate buffers for electrophoresis of serum proteins on cellulose acetate membranes.

Two new non-barbiturate buffers have been formulated for serum protein electrophoresis on cellulose acetate membranes. Both buffers give five distrinct fractions and are suitable for all systems, but have been primarily designed to meet the needs of the Gelman "Sepratek" system. One buffer is a tris(hydroxymethyl)aminomethane (Tris)/hippurate formulation; the other is composed of Tris/N-[tris(hydroxymethyl)methyl]glycine (Tricine)/sodium chloride/sodium salicylate. Both buffers allow reliable quantitation of protein fractions, giving results that correlated well with those obtained from barbit

Intracellular pH regulation and buffer capacity in CO2/HCO3-buffered media in cultured epithelial cells from rainbow trout gills.

The influence of a CO2/HCO3(-)-buffered medium on intracellular pH regulation of gill pavement cells from freshwater rainbow trout was examined in monolayers grown in primary culture on glass coverslips; intracellular pH (pHi) was monitored by continuous spectrofluorometric recording from cells loaded with 2',7'-bis(2-carboxyethyl)-5(6)-carboxy-fluoroscein. When cells in HEPES-buffered medium at normal pH = 7.70 were transferred to normal CO2/HCO3(-)-buffered medium ¿PCO2 = 3.71 mmHg, [HCO3-] = 6.1 mmol l(-1), extracellular pH (pHe) = 7.70¿, they exhibited a brief acidosis but subsequently reg

Creating a replicable, valid cross-platform buffering technique: the sausage network buffer for measuring food and physical activity built environments.

BACKGROUND: Obesity researchers increasingly use geographic information systems to measure exposure and access in neighborhood food and physical activity environments. This paper proposes a network buffering approach, the "sausage" buffer. This method can be consistently and easily replicated across software versions and platforms, avoiding problems with proprietary systems that use different approaches in creating such buffers. METHODS: In this paper, we describe how the sausage buffering approach was developed to be repeatable across platforms and places. We also examine how the sausage buf

Sleep duration in adolescence buffers the impact of childhood trauma on anxiety and depressive symptoms.

BACKGROUND: Childhood trauma is a pervasive issue contributing to adverse mental health outcomes. Obtaining optimal sleep supports healthy development and protects against mood-related disorders. Whether sleep serves as a potential buffer between trauma and adverse mental health outcomes holds promise for informing targeted interventions and prevention for adolescents. METHODS: Data were drawn from the baseline assessment of a randomised controlled trial of a mental health prevention program. A total sample of 752 adolescents completed an online, self-report survey in 2023. Participants were

Social capital buffers divergent prenatal depression trajectories: A latent class growth analysis in Chinese women.

BACKGROUND: The manifestation of prenatal depression symptoms demonstrates heterogeneous trajectories that may be mediated by maternal social capital reserves. OBJECTIVES: This longitudinal study aimed to (1) characterize distinct developmental trajectories of prenatal depressive symptoms and (2) examine the predictive capacity of early pregnancy social capital in determining trajectory subgroup membership. METHODS: Longitudinal data were collected from 455 pregnant women in Chengdu assessed at three gestational phases: first trimester (<14 weeks), second trimester (14-28 weeks), and third t

Negative emotion differentiation buffers against intergenerational risk for social anxiety in at-risk adolescent girls.

Social anxiety disorder (SAD) tends to emerge during adolescence and is more prevalent among those assigned female at birth. Parental social anxiety confers risk for adolescent SAD but less is known regarding protective factors. Research suggests that emotion differentiation (the ability to discriminate between similarly valenced emotions, e.g., fear vs. sadness vs. anger) may be protective, as it is associated with adaptive psychosocial outcomes. However, little work has examined how emotion differentiation influences the development of SAD, particularly during periods of higher risk such as

Negative density-dependence buffers against mismatch-induced population decline in the Sinai baton blue butterfly.

Phenological mismatches caused by climate change pose a major threat to global biodiversity, yet relatively few studies have reported population declines resulting from mismatch. It has been hypothesised that density effects may underlie this lack of observed responses by buffering against mismatch-induced population decline. We developed an individual-based model of the critically endangered Sinai baton blue butterfly (Pseudophilotes sinaicus) and its hostplant Sinai thyme (Thymus decussatus), parameterised using real field data, to test this hypothesis. Our model showed that the baton blue e

Relationship Quality Buffers Association Between Co-rumination and Depressive Symptoms Among First Year College Students.

Co-rumination, the tendency to dwell on negative events and feelings with a relationship partner, is an aspect of relationships that has been associated with socioemotional adjustment tradeoffs and is found to be associated with depressive symptoms. However, depending on the context in which it occurs, co-rumination is not necessarily associated with detriments to mental well-being. Differences in relationship quality within certain relationships may explain why co-rumination is not always associated with depressive symptoms. In the current study, we utilized self-report measures in an ethnica
