Echinocandin drugs such as caspofungin (CASP), micafungin, and anidulafungin inhibit fungal cell wall biogenesis by blocking Fks1-mediated β-glucan deposition into the cell surface. Candins have become suitable drugs to treat life-threatening diseases caused by several fungal species, including Candida albicans, that are pathogenic for humans. Here, we present the discovery of a novel CASP-induced flocculation phenotype of C. albicans, which formed large cell aggregates in the presence of CASP. High concentrations of sugars such as mannose or glucose inhibit CASP-induced flocculation and impro

OBJECTIVES: In view of the poor therapy outcomes of invasive aspergillosis, the objective of this study was to evaluate the efficacy of combination treatment consisting of the polyene amphotericin-B-intralipid, the echinocandin caspofungin and granulocyte-colony stimulating factor (G-CSF) in experimental murine systemic aspergillosis. With inhibition of synthesis of 1,3-beta-d-glucan in the fungal cell wall by caspofungin and an effect on the cell membrane by amphotericin-B-intralipid, this treatment may result in a synergic effect against Aspergillus fumigatus. Addition of G-CSF may further c

A clear understanding of the pharmacodynamic properties of antifungal agents is important for the adequate treatment of fungal infections like candidiasis. For certain antifungal agents, the determination of Minimal Fungicidal Concentration (MFC) and time kill curve could be clinically more relevant than the determination of the Minimal Inhibitory Concentration (MIC). In this study, MIC and MFC to fluconazole, amphotericin B and caspofungin against C. albicans isolates and the killing patterns obtained with caspofungin and amphotericin B against susceptible and resistant strains to fluconazole

The emergence of antifungal resistance in Candida albicans is a major worldwide public health threat, particularly for immunocompromised patients. Antifungal resistance may arise through uniform resistance mechanisms or through variable evolutionary routes. The aim of this study was to investigate whether antifungal resistance arises in C. albicans through shared or isolate-specific mechanisms, in other words, following convergent or divergent adaptive pathways and genetic mechanisms. Six laboratory-induced fluconazole-resistant and six caspofungin-resistant isolates were analyzed using whole-

INTRODUCTION: Caspofungin is an echinocandin antifungal agent widely used for the treatment of invasive candidiasis because of its broad antifungal activity. Although ocular adverse events are listed among uncommon reactions in the Summary of Product Characteristics, detailed clinical descriptions of acute visual impairment temporally associated with caspofungin therapy are scarce.
CASE PRESENTATION: We report the case of a 21-year-old man who developed candidemia due to Candida guilliermondii following complex spinal surgery. Antifungal therapy with caspofungin was initiated in accordance wi

We report the case of a 13-year-old with systemic lupus erythematosus who received caspofungin for disseminated invasive aspergillosis and underwent continuous renal replacement therapy (CRRT) for acute kidney failure and fluid overload. The RRT membrane was made with polyacrylonitrile. During two days, blood samples were collected before and after the filter and from the effluent line to measure caspofungin concentrations and estimate total and CRRT (CLRRT) clearances. We found a total clearance of 13 mL/min and a mean (range) CLRRT of 10.1 (5.9-14.9) mL/min. Compared to previous studies, the

AIMS: Low caspofungin exposure is frequently encountered in patients with invasive candidiasis caused by Candida albicans. This study aimed to investigate the effects of caspofungin on C. albicans at sub-inhibitory concentrations.
METHODS AND RESULTS: First, a comparative transcriptomics analysis was performed on C. albicans receiving caspofungin at sub-minimum inhibitory concentrations (sub-MICs). The results showed that caspofungin significantly changed the mRNA expression profile in DAY185, with DE-mRNAs enriched in the functions of cell wall biosynthesis, metabolism, etc. Subsequently, ce

The human fungal pathogen Candida albicans maintains pathogenic and commensal states primarily through cell wall functions. The echinocandin antifungal drug caspofungin inhibits cell wall synthesis and is widely used in treating disseminated candidiasis. Signaling pathways are critical in coordinating the adaptive response to cell wall damage (CWD). C. albicans executes a robust transcriptional program following caspofungin-induced CWD. A comprehensive analysis of signaling pathways at the transcriptional level facilitates the identification of prospective genes for functional characterization

The kinetics of caspofungin (CAS) in cerebrospinal fluid (CSF) following intravenous (i.v.) administration has been studied exclusively in animal models. Human data are missing so far. In this study, 13 CSF samples were obtained at different time points following i.v. infusion of CAS in ten paediatric haemato-/oncological patients (age range 1.0-14.2 years, median 8.6 years) without signs of central nervous system (CNS) infection (n = 10 samples) or with infectious meningitis (n = 3 samples). Serum samples were obtained concurrently. Liquid chromatography-tandem mass spectrometry was used for

INTRODUCTION: Caspofungin treatment is frequently initiated in shock patients. In the present study, we investigated the influence of hypovolaemic shock requiring fluid loading on the plasma and pulmonary pharmacokinetic parameters of caspofungin in the pig.
METHODS: After being anaesthetised and mechanically ventilated, 12 pigs were bled to induce a two-hour deep shock and resuscitated using normal saline based on haemodynamic goals. A one-hour infusion of 70 mg of caspofungin was started at the beginning of the resuscitation period. The lungs were removed four hours after caspofungin admini

A high-dose-step-down strategy for caspofungin treatment was evaluated in an experimental model of advanced-stage invasive pulmonary aspergillosis. The therapeutic efficacy of caspofungin in relation to the severity of invasive pulmonary infection caused by Aspergillus fumigatus in transiently neutropenic rats was investigated by using rat survival and the decrease in the fungal burden as the parameters of efficacy. When treatment was started at either 16 h or 24 h after fungal inoculation, caspofungin administered intraperitoneally at 4 mg/kg of body weight/day for 10 days was highly effectiv

BACKGROUND AND OBJECTIVES: The ability of Candida albicans to produce biofilm is considered an important pathogenic factor. In addition, the low sensitivity of biofilms to antifungal drugs is a challenge for patients, clinicians, and laboratory workers. We aimed to investigate the effectiveness of luliconazole and caspofungin on the planktonic and biofilm types of C. albicans strains.
MATERIALS AND METHODS: Fifty C. albicans from vaginitis, candiduria, gastrointestinal candidiasis, and saliva were examined for antifungal susceptibility against caspofungin and luliconazole using the CLSI M27 g

Caspofungin (CAS) is a potent antifungal agent belonging to the echinocandin family. It is a water-soluble dication at physiological pH, making it a good candidate for iontophoresis. Intracorneal iontophoretic delivery and biodistribution of CAS and its electrically assisted transport into the sclera were investigated as a function of experimental conditions, including donor concentration (1, 5 and 10 mg/mL) and application time (5 and 20 min) using an in-house set-up (with Ag/AgCl electrodes) and a marketed iontophoretic applicator (Iontofor CXL®) that used inert (stainless steel) electrodes.

BACKGROUND: Severe fungal infection known as invasive candidiasis primarily impacts critically ill alongside immunocompromised patients with high mortality and long hospital stays and creates substantial healthcare expenses. Echinocandins constitute the current standard first-line treatment for invasive candidiasis, with caspofungin being among the most widely used agents, caspofungin is used to treat invasive candidiasis but requires daily intravenous administration, and its pharmacokinetics may vary between patients. A new generation echinocandin antifungal drug called Rezafungin demonstrate

Aspergillus fumigatus can cause invasive pulmonary aspergillosis (IPA). Fungicidal azoles and fungistatic caspofungin (CAS) are the first- and second-line therapies, respectively, used to treat IPA. Treatment of A. fumigatus with CAS or micafungin induces the production of the oxylipin 5,8-diHODE by the fungal oxygenase PpoA. For this article, we investigated the influence of ppo genes, which encode the fatty acid oxygenases responsible for oxylipin biosynthesis, on CAS tolerance. The influence of PpoA and PpoC on CAS tolerance is mediated by MpkA phosphorylation and protein kinase A (PKA) act

The fungal pathogen Cryptococcus neoformans causes up to 278 000 infections each year globally, resulting in up to 180,000 deaths annually, mostly impacting immunocompromised people. Therapeutic options for C. neoformans infections are very limited. Caspofungin, a member of the echinocandin class of antifungals, is generally well tolerated but clinically ineffective against C. neoformans. We sought to identify biological processes that can be targeted to render the cell more susceptible to echinocandins by screening the available libraries of gene deletion mutants made in the KN99α background

Aspergillus fumigatus is a deadly opportunistic fungal pathogen responsible for ~100,000 annual deaths. Azoles are the first line antifungal agent used against A. fumigatus, but azole resistance has rapidly evolved making treatment challenging. Caspofungin is an important second-line therapy against invasive pulmonary aspergillosis, a severe A. fumigatus infection. Caspofungin functions by inhibiting β-1,3-glucan synthesis, a primary and essential component of the fungal cell wall. A phenomenon termed the caspofungin paradoxical effect (CPE) has been observed in several fungal species where at
