Critical illness with hyponatraemia and impaired cell membrane integrity--the "sick cell syndrome" revisited.

OBJECTIVE: To determine whether impaired cell membrane permeability exists in critically ill patients with "sick cell" type hyponatraemia. DESIGN AND METHODS: A 36 year old male patient was identified in an intensive care unit (ICU) with liver disease and multi-organ failure. His initial serum sodium (Na) was 101 mmol/L and osmolar gap + 35 mmol/L. A flow cytometric system was used to assess lymphocyte membrane integrity using fluorescein diacetate (FDA) and propidium iodide (PI). Following this, similar studies were carried out in 17 hyponatraemic (Na < 130 mmol/L) and 19 normonatraemic (Na

Importance of an 85 kDa membrane glycoprotein for a variety of cell-cell interactions.

The membrane molecule termed "7F7-antigen" has been found to be involved in several examples of cell-cell interactions. This 85 kDa glycoprotein with a protein core of about 55 kDa contains N-linked and O-linked carbohydrates. It has an isoelectric point of 8.0-8.5 and is expressed on 20% of peripheral blood mononuclear cells, 35% of peripheral blood B-cells, follicular dendritic cells and vascular endothelium. It is also expressed on activated T-cells and its expression on B-cells, fibroblasts and monocytes increases after treatment with PWM, interferon-gamma and after three days culture, res

A cell membrane theory of endothelial repair and vertical cell loss after cataract surgery.

Endothelial cell counts were taken of the superior, central and inferior corneas of controls, immediate post-operative cases and a four year post-operative case. A vertical mean cell density was found to correlate well with central densities, and mean vertical cell loss (42%) compared well with central cell loss (44%). There was no trend for vertical cell disparity (VCD) in control eye, but a definite disparity was seen in all post-operative eyes, including the one four years after surgery. A cell membrane stimulus theory is proposed for endothelial cell reparation after endothelial damage des

Phthalate monoesters affect membrane fluidity and cell-cell contacts in endometrial stromal adherent cell lines and spheroids.

Phthalate monoesters have been identified as endocrine disruptors in a variety of models, yet understanding of their exact mechanisms of action and molecular targets in cells remains incomplete. Here, we set to determine whether epidemiologically relevant mono(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP) can affect biological processes by altering cell plasma membrane fluidity or formation of cell-cell contacts. As a model system, we chose endometrial stromal cell lines, one of which was previously used in a transcriptomic study with MEHHP or MEHHP-containing mixtures. A short-term exposure (1 h)

Single-cell analysis reveals the diversity of human fetal membrane and adjacent placental cells with preterm premature rupture of membranes.

The occurrence of preterm premature rupture of membranes (PPROM) significantly impacts maternal and fetal health due to its association with the cellular composition and genetic changes of the fetal membrane. However, the specific cell type responsible for triggering PPROM and the underlying mechanisms are still largely unexplored. We employed single-cell RNA sequencing (scRNA-seq) analysis on fetal membrane along with adjacent placental tissues about two centimeters from the umbilical cord obtained from women who delivered full-term in labor (FTIL), preterm premature without rupture of membra

Cell Membrane-Engineered FePDA Nanoparticles Integrate Ferroptosis and Antitumor Immunity.

Ferroptosis-based cancer therapy shows promise in tumor suppression but is often limited by insufficient activation of antitumor immunity. Here, a hepatocellular carcinoma (HCC) targeted nanoplatform was constructed by coating FePDA nanozymes with Hepa1-6 cell membranes overexpressing ovalbumin (OVA) and CD40 ligand (CD40L), generating tumor membrane-derived OVA/CD40L-functionalized FePDA nanoparticles (FePDA-TMOC). This hybrid architecture integrates homologous tumor targeting with iron homeostasis disruption and co-stimulatory immune activation within a single nanosystem. Mechanistically, th

Intravitreal injection of cell membrane-coated nanoparticles for retinoblastoma treatment.

Chemotherapy is a widely adopted treatment for malignant intraocular tumors such as retinoblastoma. However, achieving high delivery efficiency remains challenging due to rapid clearance and lack of targeting specificity. Cancer cell membrane-coated nanoparticles have been extensively reported to exhibit specific affinity to homotypic cancer cells. Here we designed poly (lactic-co-glycolic acid) (PLGA)-based polymeric nanoparticles coated with the plasma membrane of WERI-Rb-1 cells, derived from human retinoblastoma. These nanoparticles showed remarkable affinity to WERI-Rb-1 cells in vitro. L

Application of Biomimetic Cell Membrane-Coated Nanocarriers in Cardiovascular Diseases.

Biomimetic cell membrane-coated nanocarriers have gained attention as an innovative therapeutic strategy for cardiovascular diseases (CVDs) due to their capacity to mimic natural cellular architectures. This distinctive characteristic improves biocompatibility, enables evasion of immune surveillance, and promotes targeted drug delivery to specific disease sites. By harnessing cell membrane components from sources such as red blood cells, platelets, and immune cells, these nanocarriers can transport therapeutic agents directly to pathological areas, including atherosclerotic lesions, ischemic m

Advances on the genetic basis of red cell membrane disorders.

PURPOSE OF REVIEW: The development and widespread use of genetic testing for diagnosis of red blood cell (RBC) membrane disorders and other hereditary hemolytic anemias (HHA) have expanded our understanding of these diseases and revealed additional complexities. We describe here such complexities and make suggestions regarding genotypic/phenotypic evaluation with the goal of early diagnosis of patients with HHA before inappropriate treatment or complications occur. RECENT FINDINGS: Widespread use of clinical genetic diagnosis in patients, for more than a decade now, has revealed the heterogen

Cell Membrane-Coated Nanoparticles for Dental, Oral, and Craniofacial Diseases.

Dental, oral, and craniofacial diseases can substantially impact the quality of human life, thereby posing a serious public health concern. Although conventional therapies such as surgery have solved these problems largely, the prognosis of patients is not always satisfactory. Cell membrane-coated nanoparticles (CMCNPs) carry nanodrugs with the help of natural cell membranes, therefore utilizing their remarkable ability to interface and interact with their surrounding environment. These nanoparticles have demonstrated substantial advantages in drug targeting, prolonging blood circulation time,

Filter-exchange spectroscopy is sensitive to gradual cell membrane degradation.

Transmembrane water permeability changes occur after initialization of necrosis and are a mechanism for early detection of cell death. Filter-exchange spectroscopy (FEXSY) is sensitive to transmembrane water permeability and enables its quantification by magnetic resonance via the apparent exchange rate (AXR). In this study, we investigate AXR changes during necrotic cell death. FEXSY measurements of yeast cells in different necrotic stages were performed and compared with established fluorescence cell death markers and pulsed gradient spin echo measurements. Furthermore, the influence of T2 r

Cell Membrane-Coated Oncolytic Adenovirus for Targeted Treatment of Glioblastoma.

An oncolytic virus is a promising strategy for glioblastoma (GBM) therapy. However, there are still some challenges such as the blood-brain barrier (BBB) and preexisting immunity for targeted treatment of GBM with an oncolytic virus. In this study, two kinds of cell membrane-coated oncolytic adenoviruses (NCM-Ad and GCM-Ad) were prepared using neural stem cells (NSCs) and GBM cells as sources of membranes, respectively, and were shown to improve the targeted infectivity on GBM cells and avoid the immune clearance of preexisting neutralizing antibodies in vitro and in vivo. Specifically, NCM-Ad

Cell-Membrane Biomimetic Indocyanine Green Liposomes for Phototheranostics of Echinococcosis.

Echinococcosis is an important zoonotic infectious disease that seriously affects human health. Conventional diagnosis of echinococcosis relies on the application of large-scale imaging equipment, which is difficult to promote in remote areas. Meanwhile, surgery and chemotherapy for echinococcosis can cause serious trauma and side effects. Thus, the development of simple and effective treatment strategies is of great significance for the diagnosis and treatment of echinococcosis. Herein, we designed a phototheranostic system utilizing neutrophil-membrane-camouflaged indocyanine green liposomes

Ultrastructural characteristics of finger-like membrane protrusions in cell competition.

A small number of oncogenic mutated cells sporadically arise within the epithelial monolayer. Newly emerging Ras- or Src-transformed epithelial cells are often apically eliminated during competitive interactions between normal and transformed cells. Our recent electron microscopy (EM) analyses revealed that characteristic finger-like membrane protrusions are formed at the interface between normal and RasV12-transformed cells via the cdc42-formin-binding protein 17 (FBP17) pathway, potentially playing a positive role in intercellular recognition during apical extrusion. However, the spatial dis

Annexin A2 Mediates Dysferlin Accumulation and Muscle Cell Membrane Repair.

Muscle cell plasma membrane is frequently damaged by mechanical activity, and its repair requires the membrane protein dysferlin. We previously identified that, similar to dysferlin deficit, lack of annexin A2 (AnxA2) also impairs repair of skeletal myofibers. Here, we have studied the mechanism of AnxA2-mediated muscle cell membrane repair in cultured muscle cells. We find that injury-triggered increase in cytosolic calcium causes AnxA2 to bind dysferlin and accumulate on dysferlin-containing vesicles as well as with dysferlin at the site of membrane injury. AnxA2 accumulates on the injured p

Peripheral Endothelial Cell Density in Descemet Membrane Endothelial Keratoplasty Grafts.

PURPOSE: To compare the variation in corneal endothelial cell density (ECD) from the center to the periphery in unpeeled and peeled donor corneas and to determine the impact of eccentric trephining on total endothelial cells in Descemet membrane endothelial keratoplasty (DMEK) grafts. METHODS: Mated donor cornea pairs were obtained. One cornea from each pair was peeled for DMEK, whereas the other was left unpeeled. Alizarin Red was used to stain the endothelial cells. High-resolution images at fixed magnification were obtained for the center, midperiphery (2.5 mm from the center), and the per

Interaction of injectable neurotropic drugs with the red cell membrane.

The normal red blood cell (RBC) shape is a biconcave discocyte. An intercalation of a drug in the outer half of the membrane lipid bilayer leads to echinocytosis, an intercalation in the inner half to stomatocytosis. We have used the shape transforming capacity of RBCs as a model to analyse the membrane interaction potential of various neurotropic drugs. Chlorpromazine, clomipramine, citalopram, clonazepam, and diazepam induced a reversible stomatocytosis, phenytoin induced echinocytosis, while the anticonvulsants levetiracetam, valproic acid and phenobarbital had no effect. This diversity of

Alkamides from Echinacea disrupt the fungal cell wall-membrane complex.

We tested the hypothesis that alkamides from Echinacea exert antifungal activity by disrupting the fungal cell wall/membrane complex. Saccharomyces cerevisiae cells were treated separately with each of seven synthetic alkamides found in Echinacea extracts. The resulting cell wall damage and cell viability were assessed by fluorescence microscopy after mild sonication. Membrane disrupting properties of test compounds were studied using liposomes encapsulating carboxyfluorescein. Negative controls included hygromycin and nourseothricin (aminoglycosides that inhibit protein synthesis), and the po

Red blood cell membrane grafting of multi-functional hyperbranched polyglycerols.

The covalent attachment of hydrophilic polymers or biopharmaceuticals to the surface of red blood cells (RBCs) has previously been shown as a relatively compatible and effective method for a range of applications. Here, the first example of cell-surface grafting with a hyperbranched and multi-functional macromolecule is described. A range (3 kDa-101 kDa) of dense, globular, and blood compatible hyperbranched polyglycerols (HPG) were synthesized and functionalized with cell-surface reactive, succinimidyl succinate groups (1-12 groups per polymer). Subsequently, HPG was grafted to the RBCs, whic

Bipolar disorder and cell membrane dysfunction. Progress toward integrative management.

Bipolar disorder (BD) is characterized by periods of abnormally elevated mood (mania) that cycle with abnormally lowered mood (depression). Multiple structural, metabolic, and biochemical abnormalities are evident in the brain's cortex, subcortex, and deeper regions. This disorder is highly genetically conditioned but also highly susceptible to environmental stressors: prenatal or perinatal insults, childhood sexual or physical abuse, challenging life events, substance abuse, and other toxic chemical exposures. Its high morbidity, lost productivity, and suicide risk place a great toll on socie
