Isolation, screening, and molecular identification of endopytic fungus producing cellulose and cyanide degrading enzyme its application for waste cassava.

OBJECTIVE: This research aims to isolate, screen, and identify some candidates for endophytic fungus-producing cellulase and cyanidase. MATERIALS AND METHODS: Fungi were isolated from cassava leaves that had undergone surface sterilization. The fungal isolates were qualitatively tested for their ability to produce cellulase and cyanidase enzymes by adding carboxy methyl cellulose (CMC) and KCN to the media. Enzyme production was indicated by the formation of clear zones around the growing colonies. Isolates that tested positive for cellulase and cyanidase production underwent further quantita

Xy-31 and Lat-1 regulate the induction of polysaccharide disintegrating genes by arabinose in Neurospora crassa OR74A.

BACKGROUND: Arabinose has been identified as a potent inducer of the cellulase regulon in filamentous fungi. However, the precise molecular mechanisms that drive this induction remain largely unknown. This study aimed to elucidate the mechanisms underlying the arabinose-induced activation of cellulase and hemicellulase enzyme clusters in the model filamentous fungus Neurospora crassa OR74A. RESULTS: Arabinose stimulates the secretion of cellulases and hemicellulases in N. crassa. While genome-wide scrutiny predicted several sugar transporters in N. crassa, expression studies under avicel and

Bioconversion of lignocellulosic biomass: biochemical and molecular perspectives.

In view of rising prices of crude oil due to increasing fuel demands, the need for alternative sources of bioenergy is expected to increase sharply in the coming years. Among potential alternative bioenergy resources, lignocellulosics have been identified as the prime source of biofuels and other value-added products. Lignocelluloses as agricultural, industrial and forest residuals account for the majority of the total biomass present in the world. To initiate the production of industrially important products from cellulosic biomass, bioconversion of the cellulosic components into fermentable

Cellulolytic Potential of Newly Isolated Alcohol-Tolerant Bacillus methylotrophicus.

Reprocessing lignocellulosic waste to obtain new products for industrial purposes is a vital part of circular economy. This paper reports the cellulase production by newly isolated Bacillus methylotrophicus cultured on lignocellulosic agro-industrial by-products, out of which brewer's spent grain (BSG) was selected as most beneficial. Plackett-Burman design was used for screening medium components, while Box-Behnken design was further applied to model the impact of the three most influential variables. The maximum approximated cellulase activity was 0.469 U/mL (1 U = 1 µmol of reducing sugars/

Environmental pH signals the release of monosaccharides from cell wall in coral symbiotic alga.

Reef-building corals thrive in oligotrophic environments due to their possession of endosymbiotic algae. Confined to the low pH interior of the symbiosome within the cell, the algal symbiont provides the coral host with photosynthetically fixed carbon. However, it remains unknown how carbon is released from the algal symbiont for uptake by the host. Here we show, using cultured symbiotic dinoflagellate, Breviolum sp., that decreases in pH directly accelerates the release of monosaccharides, that is, glucose and galactose, into the ambient environment. Under low pH conditions, the cell surface

[The synthesis of 4'-(2-carboxyethyl)phenyl trans-4-aminomethyl cyclohexane carboxylate hydrochloride (cetraxate hydrochloride) by means of enzymatic debenzylation].

Cetraxate hydrochloride (1) (antiulcer agent) has been industrially produced by the chemical protective method of p-hydroxy propionic acid derivatives. Screening of enzymes which quantitatively hydrolyzed cetraxate benzyl ester hydrochloride (2) into 1 was undertaken to establish a novel enzymatic method of production of 1. It was found that the enzyme activity for debenzylation of 2 is contained in cellulase enzymes originated from Aspergillus sp. Lower alkyl groups or phenyl groups of p-hydroxy propionic acid derivatives are likewise selectively hydrolyzed by the cellulase enzyme. This enzym

Effect of using a nitrogen atmosphere on enzyme hydrolysis at high corn stover loadings in an agitated reactor.

A comprehensive review of the literature shows that enzyme hydrolysis efficiency decreases with increased solids loadings at constant enzyme:cellulose ratios for pretreated lignocellulosic substrates. In seeking a mechanistic explanation for this phenomenon, we found that a nitrogen atmosphere enhances enzyme hydrolysis and minimizes the decrease in glucose yields as solids loadings are increased in an agitated bioreactor. For liquid hot water pretreated corn stover, at solids loadings of both 100 and 200 g/L and hydrolyzed for 72 hr in a 1 L bioreactor at pH 5.0 with 3.6 mg protein per g biom

Inhibition of cellulose enzymatic hydrolysis by laccase-derived compounds from phenols.

The presence of inhibitors compounds after pretreatment of lignocellulosic materials affects the saccharification and fermentation steps in bioethanol production processes. Even though, external addition of laccases selectively removes the phenolic compounds from lignocellulosic prehydrolysates, when it is coupled to saccharification step, lower hydrolysis yields are attained. Vanillin, syringaldehyde and ferulic acid are phenolic compounds commonly found in wheat-straw prehydrolysate after steam-explosion pretreatment. These three phenolic compounds were used in this study to elucidate the in

Interaction Between Liver Metabolism and Gut Short-Chain Fatty Acids via Liver-Gut Axis Affects Body Weight in Lambs.

The gut-liver axis and its interactions are essential for host physiology. Thus, we examined the jejunal microbiota, fermentation parameters, digestive enzymes, morphology, and liver metabolic profiles in different growth development lambs to investigate the liver-gut axis's role in their development. One hundred male Hu lambs of similar birth weight and age were raised under the same conditions until they reached 180 days of age. Subsequently, the eight lambs with the highest (HADG) and lowest (LADG) average daily weight gains were slaughtered for index assessment. The study indicates that th

Growth of toxic-shock-syndrome strain of Staphylococcus aureus after enzymic degradation of 'Rely' tampon component.

beta-glucosidase, cellulase, alpha-mannosidase, beta-galactosidase, and N-acetyl-beta-glucosaminidase were tested for their ability to hydrolyse the carboxymethylcellulose contained in 'Rely' tampons (R-CMC). The end-products of the hydrolysis were determined by chromatography. Only beta-glucosidase and cellulase hydrolysed R-CMC and the major product detectable after enzymic degradation was glucose, as confirmed chromatographically and by the glucose oxidase test. The enzymic-degradation products of R-CMC were able to support the growth of a toxic-shock-syndrome strain of Staphylococcus aureu

Mixed submerged fermentation with two filamentous fungi for cellulolytic and xylanolytic enzyme production.

The efficient saccharification of lignocellulosic materials requires the cooperative actions of different cellulase enzyme activities: exoglucanase, endoglucanase, beta-glucosidase, and xylanase. Previous studies with the fungi strains Aureobasidium sp. CHTE-18, Penicillium sp. CH-TE-001, and Aspergillus terreus CH-TE-013, selected mainly because of their different cellulolytic and xylanolytic activities, have demonstrated the capacity of culture filtrates of cross-synergistic action in the saccharification of native sugarcane bagasse pith. Improving cellulosic ethanol production by an engineered yeast consortium displaying a pentafunctional mini-cellulosome.

As a traditional ethanol-producing microorganism, Saccharomyces cerevisiae is an ideal host for consolidated bioprocessing. However, when overloaded cellulase genes are expressed in yeast, the metabolic burden on cells may greatly affect cell growth and cellulosic ethanol production. In this study, we developed a yeast consortium system that secretes and assembles five types of cellulases on the yeast cell surface to improve cellulosic ethanol production. This system involves one display strain, which provides the scaffoldin on the surface and several secretion strains that secrete each cellul
