Iron-Catalyzed Photoinduced Decarboxylative Cascade Process for the Synthesis of Functionalized Pyrazolones and Evaluation of Biological Activity.

In this work, we established a decarboxylative cascade process to achieve functionalized pyrazolones. This approach directly employs carboxylic acids as alkylating reagents. An inexpensive iron photocatalyst enables this process when irradiated by visible light. The reaction demonstrated a broad substrate scope, proving compatible with various carboxylic acids and N-acrylohydrazones. Furthermore, biological activity evaluation revealed that compound 3ak acted as a potent inhibitor of NO production, while compound 3al exhibited potent antiproliferative activity against the PANC-1 cell line, wit

Conversion of Phenyliodine(III) Dicarboxylates to Carbonyl Compounds via Hantzsch Ester-Enabled Decarboxylative Oxygenation.

A series of phenyliodine(III) dicarboxylates, readily formed in situ from the reaction of carboxylic acids with PhIO, could undergo Hantzsch ester (HE)-enabled decarboxylative oxygenation with O2 from air as the oxidant to give a wide variety of the corresponding aldehydes and ketones. Mechanistic studies provide strong evidence for hydroperoxides as key intermediates in this transformation.

Decarboxylative alkylation of unactivated olefins via photoinduced Fe-LMCT: access to alkylated dihydropyrazoles/tetrahydropyridazines.

Herein, we disclose a photoinduced FeCl3-mediated ligand-to-metal charge transfer (LMCT) approach for the decarboxylative alkylation/cyclization of N-allyl and N-homoallyl aldehyde hydrazones. Readily available tertiary and secondary aliphatic carboxylic acids serve as alkyl radical precursors, enabling functionalization of unactivated olefins to furnish alkylated dihydropyrazoles and tetrahydropyridazines in good to excellent yields under mild, oxidant-free conditions, without requiring precious-metal photocatalysts. The protocol exhibits broad substrate scope and functional group tolerance,

One-Pot Synthesis of Sulfonamides from Unactivated Acids and Amines via Aromatic Decarboxylative Halosulfonylation.

The coupling of carboxylic acids and amines to form amide linkages is the most commonly performed reaction in the pharmaceutical industry. Herein, we report a new strategy that merges these traditional amide coupling partners to generate sulfonamides, important amide bioisosteres. This method leverages copper ligand-to-metal charge transfer (LMCT) to convert aromatic acids to sulfonyl chlorides, followed by one-pot amination to form the corresponding sulfonamide. This process requires no prefunctionalization of the native acid or amine and extends to a diverse set of aryl, heteroaryl, and s-ri

A Practically Unified Electrochemical Strategy for Ni-Catalyzed Decarboxylative Cross-Coupling of Aryl Trimethylammonium Salts.

By merging electrocatalysis and nickel catalysis, a unified strategy has been successfully applied to achieve the decarboxylative cross-coupling of four types of α-oxocarboxylic acids and their derivatives with aryl trimethylammonium salts under mild conditions. Our strategy provides a practical way for preparing aryl ketones, amides, esters, or aldehydes.

Carbene and photocatalyst-catalyzed decarboxylative radical coupling of carboxylic acids and acyl imidazoles to form ketones.

The carbene and photocatalyst co-catalyzed radical coupling of acyl electrophile and a radical precursor is emerging as attractive method for ketone synthesis. However, previous reports mainly limited to prefunctionalized radical precursors and two-component coupling. Herein, an N-heterocyclic carbene and photocatalyst catalyzed decarboxylative radical coupling of carboxylic acids and acyl imidazoles is disclosed, in which the carboxylic acids are directly used as radical precursors. The acyl imidazoles could also be generated in situ by reaction of a carboxylic acid with CDI thus furnishing a

Diversification of Unprotected Alicyclic Amines by C-H Bond Functionalization: Decarboxylative Alkylation of Transient Imines.

Despite extensive efforts by many practitioners in the field, methods for the direct α-C-H bond functionalization of unprotected alicyclic amines remain rare. A new advance in this area utilizes N-lithiated alicyclic amines. These readily accessible intermediates are converted to transient imines through the action of a simple ketone oxidant, followed by alkylation with a β-ketoacid under mild conditions to provide valuable β-amino ketones with unprecedented ease. Regioselective α'-alkylation is achieved for substrates with existing α-substituents. The method is further applicable to the conve

Potent inhibitors of furin and furin-like proprotein convertases containing decarboxylated P1 arginine mimetics.

Furin belongs to the family of proprotein convertases (PCs) and is involved in numerous normal physiological and pathogenic processes, such as viral propagation, bacterial toxin activation, cancer, and metastasis. Furin and related furin-like PCs cleave their substrates at characteristic multibasic consensus sequences, preferentially after an arginine residue. By incorporating decarboxylated arginine mimetics in the P1 position of substrate analogue peptidic inhibitors, we could identify highly potent furin inhibitors. The most potent compound, phenylacetyl-Arg-Val-Arg-4-amidinobenzylamide (15

Catalytic role of serum albumin in decarboxylation of acetoacetate: A biochemical insight into the study by Yoshida et al.

This correspondence provides a biochemical commentary and supplementary hypothesis regarding the article by Yoshida et al. (2025) published in this journal on the relationship between blood acetone levels and gastric contents. While the original study demonstrated that blood acetone levels increase in the absence of food residue, we highlighted the catalytic role of human serum albumin (HSA) in the decarboxylation of acetoacetate (AcAc) to acetone, a non-redox process distinct from β-hydroxybutyrate formation. We hypothesized that under hypoglycaemic conditions (i.e., empty stomach), the lack

Molecular characterization of histidine and tyrosine decarboxylating Enterococcus species isolated from some milk products.

BACKGROUND: Fermented foods can cause adverse effects on human health because of the biogenic amines (BAs) accumulating through amino acid decarboxylation. This study investigated the presence of BAs including tyramine and histamine in 240 samples of some cheese and fermented milk samples using high-performance liquid chromatography. Another aim of this study is to isolate and identify Enterococcus spp. as the most important and frequent BA producer in the examined samples. The isolated Enterococcus spp. was investigated phenotypically for their capacity to produce amino acid decarboxylase enz

Modular access to saturated bioisosteres of anilines via photoelectrochemical decarboxylative C(sp3)-N coupling.

In drug development, the substitution of benzene rings in aniline-based drug candidates with saturated bridged bicyclic ring systems often enhances pharmacokinetic properties while preserving biological activity. However, current efforts predominantly focuses on bicyclo[1.1.1]pentylamines, accessing analogs capable of mimicking ortho- and meta-substituted anilines remains challenging due to the lack of a versatile and modular synthetic methods. Herein, we present a modular approach to access a diverse array of saturated bioisosteres of anilines via photoelectrochemical-induced decarboxylative

Copper(ii)-catalyzed enantioselective decarboxylative Mannich reaction coordinated by supramolecular organic amine cages.

Using supramolecular chiral cages to create a favorable chiral environment can effectively address the limitations of traditional metal asymmetric catalysis in controlling chiral catalytically active centers. However, achieving harmonious interactions among the molecular cage, the metal, and the substrate within the cavity remains a significant challenge. To overcome this, we have designed a pyridinium-modified, chiral-diamine-functionalized cage with a distinct bowl-shaped geometry. This structure features three quaternary ammonium linkers at the base and three chiral cyclohexanediamine units

Decarboxylative Ketonization of Aliphatic Carboxylic Acids in a Continuous Flow Reactor Catalysed by Manganese Oxide on Silica.

The sustainable synthesis of long carbon chain molecules from carbon dioxide, water and electricity relies on the development of waste-free, highly selective C-C bond forming reactions. An example for such a power-to-chemicals process is the industrial-scale fermentation for the production of hexanoic acid. Herein, we describe how this product is transformed into 6-undecanone via decarboxylative ketonization using a heterogeneous manganese oxide/silica catalyst. The reaction reaches full conversion with near-complete selectivity when carried out in a continuous flow reactor, requires no solven

Integrated Approach for Allyl Amine Synthesis Combining the Decarboxylative Coupling of Arylacrylic Acids with the Petasis Reaction.

A metal-free multicomponent reaction strategy for synthesizing allyl amines through decarboxylative coupling combined with the Petasis reaction is described. Utilizing (E)-3-(2,4,6-trimethoxyphenyl)acrylic acid, various boronic acids, a formaldehyde solution, and primary amines, the method operates efficiently without metal catalysts. Investigations reveal that electron-donating substituents in cinnamic acid derivatives significantly enhance reactivity, which is crucial for effective transformations. Under the optimized conditions, yields of ≤78% were obtained across a diverse range of 30 ally

Copper-Catalyzed Enantioselective Decarboxylative Cyanation of Benzylic Acids Promoted by Hypervalent Iodine(III) Reagents.

Copper-catalyzed asymmetric radical cyanation reactions have emerged as a powerful strategy for rapid construction of α-chiral nitriles. However, the directly decarboxylative cyanation reactions of common alkyl carboxylic acids remain largely elusive. Herein, we report a protocol for copper-catalyzed direct and enantioselective decarboxylative cyanation of benzylic acids. The in situ activation of acid substrates by a commercially inexpensive hypervalent iodine(III) reagent promoted the yield of the alkyl radicals under mild reaction conditions without prefunctionalization. The structurally di

Copper-Catalyzed Aerobic Oxidative/Decarboxylative Phosphorylation of Aryl Acrylic Acids with P(III)-Nucleophiles.

A copper-catalyzed aerobic oxidative/decarboxylative phosphorylation of aryl acrylic acids with P(III)-nucleophiles via the Michaelis-Arbuzov rearrangement for the synthesis of β-ketophosphine oxides, β-ketophosphinates, and β-ketophosphonates is reported. The present reaction could be conducted effectively without the use of a ligand and a base. Various kinds of aryl acrylic acids and P(III)-nucleophiles are tolerated in the transformation, generating the desired β-keto-organophosphorus compounds as a valuable class of phosphorus-containing intermediates with good to excellent yields. Copper(I)-catalyzed asymmetric decarboxylative Mannich reaction enabled by acidic activation of 2H-azirines.

Chiral aziridines are structure units found in many biologically active compounds and are important building blocks in organic synthesis. Herein, by merging nucleophilic generation through copper(I)-catalyzed decarboxylation and activation of poorly electrophilic 2H-azirines through protonation with carboxylic acids, an asymmetric decarboxylative Mannich reaction between α,α-disubstituted cyanoacetic acids and 2H-azirines is uncovered, which leads to generation of chiral aziridines containing vicinal tetrasubstituted and acyclic quaternary stereogenic carbon centers in good to excellent diaste
