Experimental microsporidiosis in immunocompetent and immunodeficient mice and monkeys.

Microsporidia cause opportunistic infections in AIDS patients and commonly infect laboratory animals, as well. Euthymic C57B1/6 mice experimentally infected with intraperitoneal injections of 1 x 10(6) Encephalitozoon cuniculi Levaditi, Nicolau et Schoen, 1923, Encephalitozoon hellem Didier et al., 1991, or Nosema corneum Shadduck et al., 1990 displayed no clinical signs of disease. Athymic mice, however, developed ascites and died 8-16 days after inoculation with N. corneum, 21-25 days after inoculation with E. cuniculi, and 34-37 days after inoculation with E. hellem. All athymic mice displa

Experimental encephalitozoonosis in the blue fox. Clinical and serological examinations of affected pups.

Two groups of blue fox pups about 1½— 2 and 2½—3 months old, respectively, suffering from experimental encephalitozoonosis, were examined clinically and serologically. Antibodies to Encephalitozoon cuniouli were detected in all pups, the titres varying within the range 10–12,800. In addition to unspecific signs of disease the pups showed various neurological disturbances including ataxia, posterior weakness, lameness and circling behaviour, terminaiting in recumbency, paralysis or convulsions. Reduced sight or blindness was observed occasionally. Some of the pups appeared thirsty. Haematologic

Microsporidiosis in HIV positive patients: current methods for diagnosis using biopsy, cytologic, ultrastructural, immunological, and tissue culture techniques.

Microsporidiosis is an increasingly important opportunistic infection in HIV-positive patients. Five species of microsporidia (Enterocytozoon bieneusi, Encephalitozoon hellem and E. cuniculi, Septata intestinalis, and Pleistophora sp.) have been reported to occur in AIDS, with each agent producing a different clinicopathologic spectrum of disease. This communication reviews routine and specialized methods for diagnosis of these important pathogenic protozoa, including biopsy, cytology, ultrastructural and immunologic examination, and tissue culture, and describes the current knowledge of organ

Recovery, bioaccumulation, and inactivation of human waterborne pathogens by the Chesapeake Bay nonnative oyster, Crassostrea ariakensis.

The introduction of nonnative oysters (i.e., Crassostrea ariakensis) into the Chesapeake Bay has been proposed as necessary for the restoration of the oyster industry; however, nothing is known about the public health risks related to contamination of these oysters with human pathogens. Commercial market-size C. ariakensis triploids were maintained in large marine tanks with water of low (8-ppt), medium (12-ppt), and high (20-ppt) salinities spiked with 1.0 x 10(5) transmissive stages of the following human pathogens: Cryptosporidium parvum oocysts, Giardia lamblia cysts, and microsporidian sp

Encephalitozoonosis in squirrel monkeys (Saimiri sciureus).

Twenty-two cases of naturally occurring encephalitozoonosis in squirrel monkeys are reported from breeding colonies of the Delta Regional Primate Research Center, Covington, La. Characteristic foci of granulomatous inflammation and organisms were demonstrated in brains, kidneys, lungs, adrenals, and livers. Vasculitis and perivasculitis were also common lesions in several organs. At least seven cases were congenital while ten others occurred in monkeys less than nine months old. Granulomatous placentitis, previously unreported in any species due to Encephalitozoon cuniculi, was present in one

Corneal microsporidioses: characterization and identification.

Two ocular infectious disorders attributed to Microsporidia have been observed. They differ in that one infection involves the corneal stroma leading to corneal ulceration and suppurative keratitis whereas the other infection involves the conjunctival and corneal epithelium. The corneal stromal infection is caused by a binucleated oval spore that is Nosema-like in character. The conjunctival and corneal epithelial infection occurs in HIV-sero-positive individuals and is caused by a spore containing a single nucleus that is a member of the genus Encephalitozoon. Characteristics of these genera

Resolution of microsporidial epithelial keratopathy in a patient with AIDS.

A patient with a positive human immunodeficiency virus (HIV) titer and cryptococcal meningitis suffered bilateral epithelial keratopathy caused by Encephalitozoon, which did not respond to sulfas, erythromycin, bacitracin, tobramycin, neomycin, polymyxin B, or fluconazole. Eventual administration of itraconazole for the meningitis apparently produced resolution of the long-lasting (2-month) ocular infection. This new oral triazole antifungal may be valuable against the increasingly prevalent microsporidial infections in patients with acquired immune deficiency syndrome. Debulking of the infect

New aspects of parasitic zoonoses.

Selected parasitic zoonoses are discussed with emphasis on epidemiological, diagnostic and some chemotherapeutic aspects. Pneumocystosis, toxoplasmosis, cryptosporidiosis and microsporidiosis are briefly reviewed as "AIDS-related zoonoses". Up to now 5 genera of Microsporidia have been identified as causative agents of human infections, including Encephalitozoon cuniculi, Enterocytozoon bieneusi and Pleistophora sp. which were found in AIDS patients. From the many "other parasitic zoonoses", giardiasis, echinococcosis and taeniosis/cysticercosis are discussed as examples. Significant advances

Molecular Prevalence of Microsporidia Infection in Patients with Lung Cancer.

Infections are still among the most important causes of morbidity and mortality in patients with lung cancer, which has the highest rate of cancer-related deaths in the world. Microsporidia, which are opportunistic parasitic fungi, primarily localize to the intestine by ingestion but can disseminate to the respiratory tract or can be acquired by spore inhalation. Cancer patients are at higher risk for microsporidia, a life-threatening infection, than the normal population is. We aimed to characterize the prevalence of microsporidia infection for the first time by evaluating the intestinal and

Catecholamines and encephalitozoonosis in rabbits.

Twenty four rabbits (Oryctolagus cuniculus f. domestica) were used to detect specific anti-Encephalitozoon cuniculi antibodies. To identify microsporidian infection, a haemolytic test in agar gel was carried out. Blood samples of animals with and without spontaneous encephalitozoonosis were evaluated, and compared for the presence of epinephrine (EPI), norepinephrine (NE), and dopamine (DA). Rabbits infected spontaneously with E. cuniculi had significantly lower levels of catecholamines than healthy animals. This decrease in catecholamines is of special interest because of their role as factor

Human microsporidial infections.

Microsporidia are obligate intracellular spore-forming protozoal parasites belonging to the phylum Microspora. Their host range is extensive, including most invertebrates and all classes of vertebrates. More than 100 microsporidial genera and almost 1,000 species have now been identified. Five genera (Enterocytozoon spp., Encephalitozoon spp., Septata spp., Pleistophora sp., and Nosema spp.) and unclassified microsporidia (referred to by the collective term Microsporidium) have been associated with human disease, which appears to manifest primarily in immunocompromised persons. Treatment of microsporidial keratoconjunctivitis with topical fumagillin.

Encephalitozoon hellem is a newly described cause of microsporidial keratoconjunctivitis, occurring chiefly in patients with significantly diminished CD4+ T-lymphocyte levels. This disorder is symptomatically disabling and generally recalcitrant to topical antimicrobial therapy. Two homosexual men with E. hellem keratoconjunctivitis diagnosed by Gram stain, transmission electron microscopy, and specific indirect immunofluorescent assay were treated with topical fumagillin (Fumidil B). Both patients had marked symptomatic improvement with reduction of clinical findings. Symptoms and signs recur

Changing patterns of disease and treatment of opportunistic parasitic infections in patients with AIDS.

In the past 18 months, the significant effect of highly active antiretroviral therapy and immune reconstitution on the incidence of opportunistic protozoan infections, mainly cryptosporidiosis and microsporidiosis, has been demonstrated in HIV-infected patients. The major therapeutic advances of the past 18 months concern microsporidiosis, for which the efficacies of fumagillin and albendazole have been assessed against Enterocytozoon bieneusi and Encephalitozoon infections, respectively. The efficacy of macrolides is still uncertain for the treatment of cryptosporidiosis; however, promising r

Retrospective study of diseases in a captive Lemming Colony.

Fifty-four ill or nonproductive lemmings (Dicrostonyx spp.) were evaluated for signs, lesions and causes of disease for 5 yr in a domestic colony. Parasitic granulomas caused by Encephalitozoon cuniculi were the most common finding and were seen in 22 lemmings. The disease was characterized by circling and torticollis with granulomas in many tissues, especially the central nervous system. Suppurative otitis occurred in 12 lemmings and was associated with Klebsiella pneumonia infection; circling was the common sign. Hepatic microabscesses were present in seven lemmings but a cause was not ident

Microsporidia and Cryptosporidium in horses and donkeys in Algeria: detection of a novel Cryptosporidium hominis subtype family (Ik) in a horse.

A total of 219 and 124 individual fecal samples of horses and donkeys, respectively, were screened for the presence of Cryptosporidium spp., Encephalitozoon spp., and Enterocytozoon bieneusi DNA by genus-specific nested PCR. Isolates were genotyped by sequence analysis of SSU rRNA, GP60, TRAP-C1, COWP, and HSP70 loci in Cryptosporidium, and the ITS region in microsporidia. Cryptosporidium spp. was detected on 3/18 horse farms and 1/15 farms where donkeys were kept. Overall, five (2.3%) horse and two (1.6%) donkey specimens were PCR positive for Cryptosporidium. Genotyping at SSU and GP60 loci

Iberian wild leporidae as hosts of zoonotic enteroparasites in Mediterranean ecosystems of Southern Spain.

Wild lagomorphs including rabbits and hares can act as natural carriers or reservoirs of bacterial and parasitic zoonotic diseases. However, little is known on the epidemiology and potential public health significance of intestinal eukaryotes in wild leporids. We examined faecal samples from European wild rabbits (Oryctolagus cuniculus, n = 438) and Iberian hares (Lepus granatensis, n = 111) collected in the Autonomous Region of Andalusia in southern Spain during 2012-2021. We searched for the presence of DNA from the main intestinal protist and microsporidial pathogens of veterinary and publi

Detection of microsporidia by indirect immunofluorescence antibody test using polyclonal and monoclonal antibodies.

During a screening for monoclonal antibodies (MAbs) to the microsporidian Encephalitozoon hellem, three murine hybridoma cell lines producing strong enzyme-linked immunosorbent assay (ELISA) reactivities were cloned twice, were designated C12, E9, and E11, and were found to secrete MAbs to the immunoglobulin M isotype. On subsequent ELISAs, the three MAbs reacted most strongly to E. hellem, and they reacted somewhat less to Encephalitozoon cuniculi and least to Nosema corneum, two other microsporidian species. Encephalitozoonosis was identified for the first time in arctic lemmings (Dicrostonyx stevensoni and crosses of D. stevensoni and Dicrostonyx rubricatus). The most common clinical findings were circling, torticollis, posterior paralysis, depression, blindness and death related to parasitic granulomas in many tissues. Granulomas were most frequent in the central nervous system, but were seen in many other tissues throughout the body. Granulomas were characterized by collections of macrophages and varying numbers of lymphocytes and neutrophils with or without necrosis of parenchymal tissue. Most
