Becoming in-formed: genetic counselling, ambiguity and choice.

The paper presents findings from an ethnography of dysmorphology, a specialism in genetic medicine, to explore genetic counselling as a process through which parents 'become informed.' Current professional and policy debate over the use of genetic technology in medicine emphasises the need for informed choice making, and for genetic services that provide parents with what is referred to as 'non-directive genetic counselling.' In the paper the process of becoming informed is shown to be very specific and to have its own effects. Specifically, genetics is performed in dysmorphology as a space of

Prenatal detection of copy number variants.

Prenatal detection of copy number variants (CNVs) plays an important role in the diagnosis of fetal genetic abnormalities. Understanding the methods used for prenatal CNV detection and their clinical significance contributes to the implementation of advanced genetic screening techniques in prenatal care; facilitating early identification and management of genetic disorders in fetuses. Some CNVs impose significant genetic counselling challenges; especially those which are associated with uncertain clinical significance, in the context of variable penetrance and/or expressivity or when identifie

Calcium Chloride-Mediated Transformation of Chlamydia trachomatis and Other Chlamydia spp.

Despite a high level of conservation within its core genome, the Chlamydia genus comprises various species that exhibit diverse pathogenic profiles. Genetic engineering techniques employing shuttle vectors are crucial tools for elucidating the pathogenesis of these obligate intracellular bacteria. In recent years, the introduction of calcium chloride-mediated transformation methods has significantly advanced genetic manipulation techniques for Chlamydia trachomatis. However, the efficiency of transformation using shuttle vectors, as well as plasmid maintenance, varies not only between Chlamydi

Preimplantation genetic diagnosis.

Preimplantation genetic diagnosis now represents an alternative reproductive option for parents at high risk of having offspring affected with certain genetic diseases. Progress in the past year has included increasing reliability in embryo sexing by both polymerase chain reaction and fluorescent in situ hybridization techniques; delivery of babies free of specific diseases such as cystic fibrosis, Lesch-Nyhan syndrome, and Tay-Sachs disease; and successful development of molecular techniques for detecting common diseases such as fragile-X syndrome. In addition, sperm separation in combination

[Psychological implications of family studies by molecular biology. An example--muscular dystrophy].

A dialogue between a geneticist and a M.D. trained in medical psychology has been established about problems arising in genetic counselling. A better knowledge of themselves allows them to increase the quality of genetic counselling by improving the communication techniques. The whole personality and not only the genetic disease of the individuals seeking genetic counselling is taken into account during the counselling process.

[The role of molecular genetics in childhood cancer].

In the last few years molecular genetic studies of childhood cancer have acquired great importance. Advances in these techniques have increased knowledge of the various genes involved in tumoral development. Genetic alterations can occur in three large groups of genes: oncogenes, tumor suppressor genes, and DNA repair genes. Cytogenetic analyses (karyotyping) are complemented by various molecular techniques, such as fluorescence in situ hybridization (FISH), reverse transcriptase-polymerase chain reaction (RT-PCR) and spectral karyotyping (SKY). These are the most reliable techniques and impro

Molecular Diagnostics and Genetic Counseling in Primary Congenital Glaucoma.

Primary congenital glaucoma (PCG) is a childhood irreversible blinding disorder with onset at birth or in the first year of life. It is characterized by the classical traid of symptoms viz. epiphora (excessive tearing), photophobia (hypersensitivity to light) and blepharospasm (inflammation of eyelids). The only anatomical defect seen in PCG is trabecular meshwork dysgenesis. PCG shows autosomal recessive mode of inheritance with considerable number of sporadic cases. The etiology of this disease has not been fully understood but some genes like CYP1B1, MYOC, FOXC1, LTBP2 have been implicated.

[The use of biotechnology in animal breeding].

Biotechnological techniques are extensively used in dairy breeding programs. Thus, artificial insemination and embryo transfer (and associated techniques) constitute an integral part of modern breeding programs. In pig breeding, embryo transfer is mostly restricted to special problem areas because of its high costs. Currently this technique is used for the exchange of genetic material on an international level, for the creation of specific pathogene free herds, and in connection with cryoconservation for the setup of embryobanks in the context of the preservation of genetic resources.

Genetic analysis of capsule and its associated economic traits in opium poppy (Papaver somniferum L.).

Four single crosses (VG20 x SGE48, SGE48 x SG35II, VG26 x SG35II, and SG35II x VG20) in opium poppy (Papaver somniferum L.) were analyzed to study the gene actions involved in the inheritance of quantitative traits, namely plant height, branches/plant, capsules/plant, peduncle length, capsule index, stigmatic rays, straw yield/plant, and morphine content. Simple additive, dominance, and epistatic genetic components were found to be significant for inheritance pattern. Dominance effect (h) was higher than additive effect (d). Digenic interaction indicated the prevalence of dominance x dominance

Multifactor dimensionality reduction: an analysis strategy for modelling and detecting gene-gene interactions in human genetics and pharmacogenomics studies.

The detection of gene-gene and gene-environment interactions associated with complex human disease or pharmacogenomic endpoints is a difficult challenge for human geneticists. Unlike rare, Mendelian diseases that are associated with a single gene, most common diseases are caused by the non-linear interaction of numerous genetic and environmental variables. The dimensionality involved in the evaluation of combinations of many such variables quickly diminishes the usefulness of traditional, parametric statistical methods. Multifactor dimensionality reduction (MDR) is a novel and powerful statist

[Cooperation between genetic counseling centers, cytogenetic laboratories and in vitro fertilization centers in the treatment of reproductive disorders].

The techniques of assisted reproduction have recently become the most effective methods of treatment of infertility; namely ICSI (intra-cytoplasmic sperm injection), MESA (microepididymal sperm aspiration), TESA (testicular sperm aspiration) and TESE (testicular sperm extraction). The techniques have been increasingly successful, even above the average efficiency of classical IVF (in vitro fertilization). It can be demonstrated by the percentage of ICSI-aided births percentage per 100% of embryotransfers in ISCARE; 1996 = 21.3%, 1997 = 26.15%, 1998 = 29.7%. The successful use of these techniqu

Genetic counselling in the era of next generation sequencing.

Advances in next-generation sequencing (NGS) technologies have made the detection of the molecular causes of paediatric diseases increasingly affordable, accessible and rapid. While exome sequencing and genome sequencing were until recently only available for research, they are now used in health care practice. The clinical application of NGS has raised many challenges in genetic counselling for families in terms of the interpretation of test results and incidental findings, as well as technical limitations in the event of inconclusive results. Given the impact of genetic results in clinical d

Detection of Aneuploidies in Products of Conception and Neonatal Deaths in Iranian Patients Using the Multiplex Ligation-Dependent Probe Amplification (MLPA).

BACKGROUND: Around 70% of all pregnancies (Including 15% of clinically-recognized ones) are lost due to various fetal or maternal disorders. Chromosomal aneuploidies are among the most common causes of pregnancy loss. Standard chromosome analysis using G-banding technique (Karyotype) is the technique of choice in studying such abnormalities; however, this technique is time-consuming and sensitive, and limited by vulnerabilities such as cell culture failure. The use of molecular cytogenetic techniques, including array-based techniques and Multiplex Ligation-Dependent Probe Amplification (MLPA),

The genetics of keratoconus.

Keratoconus is a relatively common, bilateral, non-inflammatory corneal ectasia. The aetiology of this condition is probably multifactorial, or it represents the final common pathway for a variety of different pathological processes. Although a familial history is present only in a minority of cases, one of the major aetiological factors is certainly genetic. This is evidenced by: the condition's familial inheritance; its discordance between monozygotic and dizygotic twins; and its association with other known genetic disorders such as Down's and Marfan's syndromes. In the keratoconic cornea,
