The behavioral profile of severe mental retardation in a genetic mouse model of phenylketonuria.

Pah(enu2) mice, created by chemically induced genetic mutation, are characterized by biochemical phenotypes closely resembling untreated human phenylketonuria (PKU). However, studies conducted in adult Pah(enu2) mice have shown no indices of the severe mental retardation that characterizes untreated PKU. The present experiments explored recognition of novel spatial and non-spatial information in Pah(enu2) mice by two nonassociative tests that do not use explicit reinforcement and avoid lengthy training. Moreover, we evaluated emotional reactivity by the Elevated Plus Maze. Finally, the perform

Spontaneous circling behavior and dopamine neuron loss in a genetically hypothyroid mouse.

The genetically hypothyroid mouse, Tshr(hyt), has a single point mutation resulting in a defective thyroid-stimulating hormone receptor, and therefore a non-functional thyroid gland. This is an autosomal recessive disorder and affected mice have been reported to have a number of somatic and behavioral deficits. This study reports a pronounced, spontaneous, asymmetrical circling behavior in the Tshr(hyt) mouse. The spontaneous circling behavior appeared in about 25% of the homozygous animals, in both males and females. The circling usually appeared by postnatal day 35 and continued throughout t

Longitudinal genetic analysis of problem behaviors in biologically related and unrelated adoptees.

The genetic and environmental influences on problem behaviors at two assessment points, three years apart, and their stability were studied in a sample of international adoptees, initially aged 10 to 15 years. Parents of 111 pairs of adopted biological siblings, 221 pairs of adopted nonbiological siblings and 1484 adopted singletons completed the Child Behavior Checklist (75 pairs, 154 pairs and 1080 singletons respectively at second assessment). At first assessment, genetic factors accounted for more than 50% of the variance in the Externalizing, Aggressive Behavior, Attention Problems and So

Genetic, phenotypic, and behavioral variation in North American sylvatic isolates of Trichinella.

Two restriction endonucleases (Cla I and Hpa II) produced polymorphic repetitive DNA profiles which were used in a clustering analysis to quantify the level of genetic variation among 14 North American sylvatic isolates of T5 Trichinella. Differences in genetic profiles reflected phenotypic differences in parasite reproductive success as measured by an isolate's reproductive capacity index in natural hosts. Two genetically distinct isolates of the T5 genotype and T. spiralis were used to infect white-footed mice Peromyscus leucopus for a behavioral analysis. Three behavioral characteristics we

[The characteristics of a change in the congenital and acquired forms of behavior in rats with genetic catalepsy].

In two groups of rats--outbred control rats and GC line ones the behaviour in the open field was studied, as well as alimentary situational instrumental conditioned reflex was elaborated. Rats of GC line were less mobile in the open field as compared with the control ones; their investigating reflex was low; several parameters showed a higher level of agitation and fear. Handling of rats of GC line improves these behavioural parameters and brings them closer to the norm. Elaboration of alimentary situational conditioned reflex in rats of GC line is delayed as compared with the control ones. Pa

Genetic Models in Applied Physiology. HXB/BXH rat recombinant inbred strain platform: a newly enhanced tool for cardiovascular, behavioral, and developmental genetics and genomics.

This review deals with the largest set of rat recombinant inbred (RI) strains and summarizes past and recent accomplishments with this platform for genetic mapping and analyses of divergent and complex traits. This strain, derived by crossing the spontaneously hypertensive rat, SHR/Ola, with a Brown Norway congenic, BN-Lx, carrying polydactyly-luxate syndrome, is referred to as HXB/BXH. The RI strain set has been used for linkage and association studies to identify quantitative trait loci for numerous cardiovascular phenotypes, including arterial pressure, stress-elicited heart rate, and press

Sedentary Behavior, Physical Activity, and Hypertension: Genetic Liability and the Role of Metabolic Factors.

BACKGROUND: Whether sedentary behavior and physical activity causally influence hypertension remains uncertain. We used Mendelian randomization (MR) to examine whether genetically predicted domain-specific sedentary behaviors and leisure-time physical activity are associated with hypertension risk and to explore potential cardiometabolic pathways. METHODS: We performed a two-sample MR using genome-wide significant, LD-independent SNPs as instruments for domain-specific sedentary traits and physical activity. Hypertension was defined by the FinnGen R9 endpoint I9_HYPTENSESS. Pleiotropy-robust

Neurotoxic effects of 2,4-DCP in adult zebrafish: A behavioral, biochemical, and genetic approach.

2,4-Dichlorophenol (2,4-DCP) is a persistent and toxic metabolite derived from the degradation of the herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) and other chlorinated compounds, representing an emerging environmental concern. Despite evidence of its toxicity, its neurotoxic effects in adult organisms remain poorly understood. This study aimed to evaluate the behavioral, biochemical, and molecular responses of adult zebrafish (Danio rerio) following 14-day exposure to environmentally relevant (30 μg L-1) and high (300 μg L-1) concentrations of 2,4-DCP. Behavioral analyses revealed decreas

Nonlinearities and Switch-Like Behavior in Gene Expression: From Genetics to Biochemistry and Back.

Many developmental and physiological outcomes depend on transcriptional thresholds or switches. The transcriptional output, ultimately measured as mature RNA, is determined by a series of molecular steps that can introduce thresholds, bottlenecks, or amplification mechanisms. Their combination can convert quantitative changes in regulator abundance into more-than-proportional changes in mature RNA production. Such nonlinear, S-shaped/sigmoidal input-output relationships may account, at least in part, for the haploinsufficiency (HI) of transcription factors (TFs) and chromatin regulators. Here,

Challenging Behavior Domains in Individuals With Neurodevelopmental Genetic Syndromes: The Role of Psychological Features.

Problem behaviors (PB) commonly co-occur in individuals with neurodevelopmental genetic syndromes (NGDs) and increase the risk of injury to oneself and others. Despite the prevalence and clinical impact of these behaviors, knowledge regarding the psychological risk markers for PB among individuals with NGDs is currently lacking. To fill this gap, we explored the relative contributions of key developmental (age, speech production) and clinical (emotion regulation, anxiety, sensory sensitivity, social communication) characteristics as predictors of unique PB subdomains in a sample of 255 individ

Genetic tuning of retinal ganglion cell subtype identity to drive visual behavior.

The distinct blend of molecular and cellular features that define neuronal subtype identity are central to shaping how individual subtypes impact animal behavior. The diversity of the mammalian nervous system is vast - the retina alone contains over 100 neuronal subtypes. Yet, the genetic processes giving rise to this stunning structural and functional diversity remain poorly understood. Here, we uncover a graded expression pattern of the transcription factor BRN3B that tunes and maintains multiple, subtype-defining transcriptional and morphophysiological features of the melanopsin-expressing,

Suicidal behavior in ADHD: the role of comorbidity, psychosocial adversity, personality and genetic factors.

Adult ADHD is associated with increased risk for suicide attempts, as indicated by investigations of population- and community-based cohorts. However, there is little data regarding suicide attempts in a clinical setting. To address this, we used a comprehensively phenotyped clinical adult ADHD (aADHD) cohort to assess to which extent comorbidity, psychosocial adversity, personality, and ADHD symptoms contribute to suicidal behavior in ADHD. Furthermore, we investigated a triallelic variation in the serotonin transporter-linked polymorphic region (5-HTTLPR), which has previously been associate

Frequency and Longitudinal Course of Behavioral and Neuropsychiatric Symptoms in Participants With Genetic Frontotemporal Dementia.

BACKGROUND AND OBJECTIVES: Behavioral and neuropsychiatric symptoms are frequent in patients with genetic frontotemporal dementia (FTD). We aimed to describe behavioral and neuropsychiatric phenotypes in genetic FTD, quantify their temporal association, and investigate their regional association with brain atrophy. METHODS: We analyzed data of pathogenic variant carriers in the chromosome 9 open reading frame 72 (c9orf72), progranulin (GRN), or microtubule-associated protein tau (MAPT) gene from the Genetic Frontotemporal dementia Initiative cohort study that enrolls both symptomatic pathogen

Psychotropic medication usage in sporadic versus genetic behavioral-variant frontotemporal dementia.

INTRODUCTION: Psychotropic medication (PM) use in behavioral-variant frontotemporal dementia (bvFTD) is higher than in other dementias. However, no information exists on whether PM use differs between sporadic and genetic bvFTD. METHODS: We analyzed data from sporadic and genetic bvFTD participants with PM prescriptions in the Advancing Research and Treatment in Frontotemporal Lobar Degeneration/Longitudinal Evaluation of Familial Frontotemporal Dementia Subjects study. We estimated ordinal odds ratio (OOR) of having more PM comparing sporadic and genetic bvFTD. Finally, we explored the neuro

Triangulating causality between childhood obesity and neurobehavior: Behavioral genetic and longitudinal evidence.

Childhood obesity is a serious health concern that is not yet fully understood. Previous research has linked obesity with neurobehavioral factors such as behavior, cognition, and brain morphology. The causal directions of these relationships remain mostly untested. We filled this gap by using the Adolescent Brain Cognitive Development study cohort comprising 11,875 children aged 9-10. First, correlations between the age- and sex-specific 95th BMI percentile (%BMIp95) and neurobehavioral measures were cross-sectionally analyzed. Effects were then aggregated by neurobehavioral domain for causal

Genetically influenced tobacco and alcohol use behaviors impact erythroid trait variation.

Genome wide association studies (GWAS) have associated thousands of loci with quantitative human blood trait variation. Blood trait associated loci and related genes may regulate blood cell-intrinsic biological processes, or alternatively impact blood cell development and function via systemic factors and disease processes. Clinical observations linking behaviors like tobacco or alcohol use with altered blood traits can be subject to bias, and these trait relationships have not been systematically explored at the genetic level. Using a Mendelian randomization (MR) framework, we confirmed causa

The Genetic, Environmental, and Cultural Forces Influencing Youth Antisocial Behavior Are Tightly Intertwined.

The aggressive and rule-breaking behaviors that constitute youth antisocial behavior (ASB) are shaped by intertwined genetic, developmental, familial, spatial, temporal, cultural, interpersonal, and contextual influences operating across multiple levels of analysis. Genetic influences on ASB, for example, manifest in different ways during different developmental periods, and do so in part as a function of exposure to harsh parenting, delinquent peers, and disadvantaged neighborhoods. There is also clear evidence documenting societal effects, time-period effects, sex-assigned-at-birth effects,

FOXN3 and GDNF Polymorphisms as Common Genetic Factors of Substance Use and Addictive Behaviors.

Epidemiological and phenomenological studies suggest shared underpinnings between multiple addictive behaviors. The present genetic association study was conducted as part of the Psychological and Genetic Factors of Addictions study (n = 3003) and aimed to investigate genetic overlaps between different substance use, addictive, and other compulsive behaviors. Association analyses targeted 32 single-nucleotide polymorphisms, potentially addictive substances (alcohol, tobacco, cannabis, and other drugs), and potentially addictive or compulsive behaviors (internet use, gaming, social networking s

Electrophysiological and Behavioral Evidence for Hyper- and Hyposensitivity in Rare Genetic Syndromes Associated with Autism.

Our study reviewed abnormalities in spontaneous, as well as event-related, brain activity in syndromes with a known genetic underpinning that are associated with autistic symptomatology. Based on behavioral and neurophysiological evidence, we tentatively subdivided the syndromes on primarily hyper-sensitive (Fragile X, Angelman) and hypo-sensitive (Phelan-McDermid, Rett, Tuberous Sclerosis, Neurofibromatosis 1), pointing to the way of segregation of heterogeneous idiopathic ASD, that includes both hyper-sensitive and hypo-sensitive individuals. This segmentation links abnormalities in differen
