Resistance to pyrethroids in Aedes aegypti: Insights into transcriptomic response to different insecticide concentrations.

Insecticide spraying is a common strategy for controlling dengue outbreaks, but its effectiveness is compromised by the development of resistance in mosquito populations. In this study, we subjected a strain of Aedes aegypti known for its exceptional ability to develop resistance to controlled permethrin and lambda-cyhalothrin insecticides pressure using two different concentrations. We analyzed resistance mechanisms that are enhanced at each concentration and used RNA sequencing to identify transcripts specifically associated with these exposure levels. Our objective was to uncover the molecu

Policy-practice gaps and barriers to the effective management of insecticide resistance in Tanzania.

BACKGROUND: Malaria control gains are stagnating or reversing, in part due to widespread insecticide and drug resistance, as well as increasing financial and implementation challenges. This study assessed the national strategies for insecticide resistance monitoring and management in Tanzania to identify key barriers and misalignments between policy and actual practice. METHODS: We initially reviewed technical documents from Tanzania's National Malaria Control Programme (NMCP) produced between 2014 and 2024, along with World Health Organization (WHO) guidelines and reports on insecticide resi

A multi-omic meta-analysis reveals novel mechanisms of insecticide resistance in malaria vectors.

Malaria control faces challenges from widespread insecticide resistance in major Anopheles species. This study, employing a cross-species approach, integrates RNA-Sequencing, whole-genome sequencing, and microarray data to elucidate drivers of insecticide resistance in Anopheles gambiae complex and An. funestus. Here we show an inverse relationship between genetic diversity and gene expression, with highly expressed genes experiencing stronger purifying selection. Gene expression clusters physically in the genome, revealing potential coordinated regulation, and we find that highly over-express

Mathematical modelling and optimal control of malaria transmission with antimalarial drug and insecticide resistance.

This study presents a mathematical model to explore malaria transmission dynamics in the presence of antimalarial drug-resistant parasites and insecticide-resistant mosquitoes. The analytical findings demonstrate a stable disease-free equilibrium when the effective reproduction number is below one. For single-strain malaria infections, the endemic equilibrium may exhibit one, two or no solutions. The model is extended to incorporate three time-dependent controls: long-lasting insecticidal nets, antimalarial treatment and mosquito adulticides. Simulation results indicate that interventions excl

Identification of Novel Gene Cluster Potentially Associated with Insecticide Resistance in Anopheles gambiae s.l.

BACKGROUND/OBJECTIVES: Despite the increasing emergence of resistance, insecticide-based tools remain the primary method for malaria vector control in Africa. To maintain the effectiveness of these interventions, continuous monitoring and identification of novel resistance mechanisms is essential. This study aimed to investigate potential new insecticide resistance genes in the Anopheles gambiae complex. METHODS: We analyzed whole-genome sequencing data from the An. gambiae 1000 Genomes Project. A broad range of genomic analysis techniques and tools were used to identify and explore genetic v

Trends in insecticide resistance in Anopheles mosquitoes (Diptera: Culicidae) in Ghana: a systematic review.

Malaria continues to be a major public health issue in Ghana, contributing significantly to hospital outpatient visits. Vector control remains central to malaria prevention; however, the growing resistance of malaria vectors to insecticides presents a major obstacle to control and elimination efforts. This review examined the evolution of insecticide resistance in Ghana from 2001 to 2024, summarising resistance mechanisms across the country's bioclimatic zones to inform evidence-based vector control strategies aligned with Ghana's malaria elimination goals. A systematic literature search was c

Genetic isolation drives divergent insecticide resistance between coastal and western Kenya Anopheles funestus populations.

Escalating levels of insecticide resistance and adaptive changes in mosquito populations threaten recent gains in malaria control. Understanding the molecular, ecological, and evolutionary processes driving these transformations is important for extending the effectiveness of current insecticides and informing the development of effective strategies and tools for vector control and insecticide resistance management. This study analyzed 103 whole-genome sequences from Anopheles funestus s.s. mosquitoes collected before the implementation of non-pyrethroid-based vector control tools in Kenya in

Copy number variants underlie major selective sweeps in insecticide resistance genes in Anopheles arabiensis.

To keep ahead of the evolution of resistance to insecticides in mosquitoes, national malaria control programmes must make use of a range of insecticides, both old and new, while monitoring resistance mechanisms. The outdoor-biting malaria vector Anopheles arabiensis is of increasing concern for malaria transmission because it is apparently less susceptible to many indoor control interventions, yet knowledge of its mechanisms of resistance remains limited. Furthermore, comparatively little is known in general about resistance to non-pyrethroid insecticides such as pirimiphos-methyl (PM), which

Uncovering the genetic diversity in Aedes aegypti insecticide resistance genes through global comparative genomics.

Aedes aegypti is vector of many arboviruses including Zika, dengue, yellow fever, West Nile, and Chikungunya. Its control efforts are hampered by widespread insecticide resistance reported in the Americas and Asia, while data from Africa is more limited. Here we use publicly available 729 Ae. aegypti whole-genome sequencing samples from 15 countries, including nine in Africa, to investigate the genetic diversity in four insecticide resistance linked genes: ace-1, GSTe2, rdl and vgsc. Apart from vgsc, the other genes have been less investigated in Ae. aegypti, and almost no genetic diversity in

Review of dengue vectors in Cambodia: distribution, bionomics, vector competence, control and insecticide resistance.

BACKGROUND: Dengue fever is one of the most prevalent mosquito-borne diseases in Cambodia. Until now, no specific vaccine nor antiviral treatment exists the virus causing Dengue fever. Consequently, its prevention relies only on vector control strategies. However, efficient vector control in turn relies on a good knowledge of the biology of the vector species. Therefore, this study aims to provide the first review of the distribution, ecology, meteorological impacts, trophic behavior, vector competence, vector control and insecticide resistance of dengue vector species in Cambodia. Evidence of population expansion and insecticide resistance mechanism in invasive fall armyworm (Spodoptera frugiperda).

BACKGROUND: The invasive and calamitous polyphagous pest Spodoptera frugiperda or commonly known as fall armyworm (FAW) poses serious menace to the global agricultural production. Owing to the revamped invasion of FAW in 2018 in India, present study was undertaken for precise assessment of its genetic identity and pesticide resistance to aid in pest-management strategies. RESULTS: To evaluate the diversity in FAW population across Eastern India, mitochondrial COI sequences were used which revealed a low nucleotide diversity. Analysis of molecular variance indicated significant genetic variati

Trends in insecticide resistance in Culex pipiens pallens over 20 years in Shandong, China.

BACKGROUND: Culex pipiens pallens is the most abundant Culex mosquito species in northern China and is an important vector of bancroftian filariasis and, potentially, West Nile virus. Insecticides, particularly pyrethroids, are widely used for adult mosquito control. Insecticide resistance has become common in several mosquito species, and vector control is the main method currently available to prevent disease transmission. The voltage-gated sodium channel (Vgsc) gene is the target site of pyrethroids, and mutations in this gene cause knockdown resistance (kdr). METHODS: Culex pipiens pallen

The evolution of multiple-insecticide resistance in UK populations of tomato leafminer, Tuta absoluta.

BACKGROUND: The tomato leafminer, Tuta absoluta, is an economically important pest of tomatoes in Europe, Africa, Asia and South America. In the UK this species is controlled using an integrated pest management (IPM) programme which incorporates the insecticides spinosad and chlorantraniliprole. In response to UK grower concerns of loss of efficacy of these compounds at certain sites, insecticide bioassays were performed on five populations collected from four commercial glasshouses and potential mechanisms of resistance investigated. RESULTS: We observed high levels of resistance to spinosad

Management of insecticide resistance in the major Aedes vectors of arboviruses: Advances and challenges.

BACKGROUND: The landscape of mosquito-borne disease risk has changed dramatically in recent decades, due to the emergence and reemergence of urban transmission cycles driven by invasive Aedes aegypti and Ae. albopictus. Insecticide resistance is already widespread in the yellow fever mosquito, Ae. Aegypti; is emerging in the Asian tiger mosquito Ae. Albopictus; and is now threatening the global fight against human arboviral diseases such as dengue, yellow fever, chikungunya, and Zika. Because the panel of insecticides available for public health is limited, it is of primary importance to prese

Insecticide resistance in Culex pipiens quinquefasciatus and Aedes albopictus mosquitoes from La Réunion Island.

Resistance to insecticides was monitored on Culex pipiens quinquefasciatus mosquitoes collected in twelve localities of La Réunion, a geographically isolated island of the Indian Ocean. This mosquito is of medical concern in the region as a known vector for filariasis and a potential vector for West Nile and Rift Valley Fever viruses. Our bioassays indicated the presence of resistance to all tested insecticides, i.e. organochlorides, organophosphates and pyrethroids. A molecular investigation revealed a higher frequency of resistance genes in the coastal areas compared to elevated rural sites,

Filariasis vector in China: insecticide resistance and population structure of mosquito Culex pipiens complex.

Seven field populations of mosquito Culex pipiens complex (Diptera: Culicidae) were collected from four provinces of China. The resistance status of larvae to dichlorvos, parathion, chlorpyrifos, fenobucarb (BPMC) and propoxur were determined by bioassays, disclosing that they were more resistant to organophosphate (moderate or low resistance) than to carbamate (low or no significant resistance) insecticides. Starch gel electrophoresis confirmed the presence and distribution of overproduced esterases B1, A2-B2, A8-B8 and A9-B9, the frequencies of which varied according to their regional origin

Spatiotemporal distribution of insecticide resistance in Anopheles culicifacies and Anopheles subpictus in Sri Lanka.

The malaria situation in Sri Lanka worsened during the 1990s with the emergence and spread of resistance to the drugs and insecticides used for control. Chloroquine resistance has increased rapidly over this period, but adverse changes in malaria transmission are more closely associated with insecticide use rather than drug resistance. Insecticide susceptibility tests were routinely carried out in key anopheline vectors across the country for more than a decade. These sentinel data were combined with data collected by other research programmes and used to map the spatial and temporal trends of
