Assessment of testicular biopsy after cessation of maintenance chemotherapy in childhood acute lymphoblastic leukemia: a report from the Children's Cancer and Leukemia Study Group.

Among 484 male patients with acute lymphoblastic leukemia (ALL) registered into the protocols CCLSG 811, 841 and 874, from 1981 through 1990, 246 patients completed their protocols and were in continuous complete remission (CCR) for more than 3 years. One hundred and seven patients received bilateral testicular biopsies at the time of cessation of maintenance chemotherapy. Eight patients (7.5%) were found to have occult testicular leukemia (TL). Three of them did not receive any additional therapy and all suffered subsequent relapses; one bone marrow relapse and two testicular relapses. The ot

Adjusted Comparison of First-line Enfortumab Vedotin Plus Pembrolizumab Versus Platinum-based Chemotherapy ± Avelumab Maintenance for the Treatment of Locally Advanced/Metastatic Urothelial Carcinoma.

BACKGROUND AND OBJECTIVE: First-line (1L) treatment options for locally advanced or metastatic urothelial carcinoma (la/mUC) include enfortumab vedotin plus pembrolizumab (EV+P), based on EV-302, or platinum-based chemotherapy (PBC) with avelumab maintenance for patients without progression, based on JAVELIN-Bladder 100 (JB-100). Clinicians must choose between EV+P and PBC without knowing whether patients will respond to PBC and qualify for avelumab. In EV-302, avelumab maintenance was approved after trial initiation and therefore, was potentially underutilized among patients without progressi

Dynamic Loss of Psoas Muscle Volume During Induction Chemotherapy Predicts Survival in Patients with Advanced Urothelial Carcinoma Receiving Avelumab Maintenance: A Multicenter Retrospective Study.

BACKGROUND: Sarcopenia is increasingly recognized as a prognostic factor in oncology; however, most studies have focused on static baseline muscle mass in heterogeneous populations. Whether dynamic skeletal muscle loss during systemic therapy carries independent prognostic value, particularly in clinically stable patients with controlled disease, remains unclear. We investigated the prognostic relevance of three-dimensional volumetric muscle assessment in advanced urothelial carcinoma (UC) patients who achieved disease control with first-line platinum-based chemotherapy and subsequently receiv

Lenalidomide maintenance fails to overcome the unfavourable prognosis of low NK-cell counts in rituximab-chemotherapy responsive elderly DLBCL patients: A LYSA group study.

Low baseline NK-cell counts (NKCCs) in patients with diffuse large B-cell lymphoma (DLBCL) are associated with a poor prognosis. The REMARC phase III trial (NCT01122472) showed that lenalidomide maintenance prolonged PFS in rituximab-chemotherapy responders. We conducted a REMARC ancillary study analysing the impact of lenalidomide maintenance on the prognostic value of low NKCCs. Blood samples from 335 elderly French patients enrolled in the REMARC trial were analysed by flow cytometry to obtain NKCCs at diagnosis (n = 220), at randomization (n = 186) and/or six months after randomization (n

Amlodipine with letrozole as maintenance post-chemotherapy: targeting calcium calcineurin calmodulin pathway and aromatase inhibitor in disseminated endometrioid adenocarcinoma of the endometrium-a case report.

World is now clinging to immune checkpoint inhibitor in almost every type of cancer. PD-1 and PD-L1 inhibitor are taking over the treatment algorithm. But cost is an issue; we have a perspective to target energy mechanism of cytotoxic T cells to modulate the representation of PD-1 receptor so as to increase immunogenicity of cytotoxic T cells. Cellular energy mechanism is largely dependent on calcium channels and pharmacological modulation of it may help in modulating response of cytotoxic T cell. We hereby present a case of endometroid adenocarcinoma of endometrium successfully treated with C

Outcomes of allogeneic or autologous stem cell transplantation followed by maintenance chemotherapy in adult patient with B-ALL in CR1 with no detectable minimal residual disease.

Autologous haematopoietic stem cell transplantation (auto-HSCT) as a treatment for B-cell acute lymphoblastic leukaemia (B-ALL) has been rigorously debated in recent years. We retrospectively analysed the outcomes of 355 adult patients with B-ALL in first complete remission who had received auto-HSCT or allogeneic HSCT (allo-HSCT) in our centre. The treatment efficacy was evaluated from a model stratified on the risk classification and minimal residue disease (MRD) status after three chemotherapy cycles. Auto-HSCT demonstrated comparable 3-year overall survival (OS) (72.7% vs. 68.5%, p = 0.441

Novel Strategy Involving High-Dose Chemotherapy with Stem Cell Rescue Followed by Intrathecal Topotecan Maintenance Therapy without Whole-Brain Irradiation for Atypical Teratoid/Rhabdoid Tumors.

Atypical teratoid/rhabdoid tumor (AT/RT) is a rare aggressive central nervous system tumor that typically affects children under three years old and has poor survival with a high risk for neurologic deficits. The primary purpose of this study was to successfully treat the disease and delay or avoid whole-brain radiotherapy for children with AT/RT. A retrospective analysis was performed for six children diagnosed with AT/RT and treated with multimodal treatment at a single institute between 2014 and 2020. Furthermore, germline SMARCB1 aberrations and MGMT methylation status of the tumors were a
