Novel autophagy-promoted cancer therapy: Discovery of matrine-based selective TBC1D2 inhibitors by drug-target complex purification.

Autophagy strategies have seen clinical or preclinical studies underway to combat cancer therapeutic resistance, but the on-target effects in normal tissues necessitate new selective approaches. Based on the antitumor efficacy and low toxicity of matrine, a traditional Chinese medicine component, we designed and synthesized 24 matrine derivatives with higher in vitro activities. Among these compounds, A1 demonstrated the highest activity and selectivity against the HeLa cell line, with a half maximal inhibitory concentration of 0.43 μM and a selective index of 3.5. Subsequently, a label-free t

Matrine alleviates Staphylococcus aureus-induced acute lung injury in mice by inhibiting MLKL and NLRP3-mediated inflammatory activity.

Acute lung injury (ALI) is a serious clinical condition with high incidence and mortality. The inflammatory response induced by gram-positive bacteria, especially Staphylococcus aureus (S. aureus), is a key factor contributing to ALI progression and other infectious diseases. Matrine, known for its diverse biological and pharmacological properties, has not been fully explored for its potential to prevent or treat S. aureus-induced ALI. Our study demonstrated that matrine exerts a protective effect against lung injury in mice infected with S. aureus. Specifically, matrine reduced pulmonary edem

Self-Assembled Matrine-PROTAC Encapsulating Zinc(II) Phthalocyanine with GSH-Depletion-Enhanced ROS Generation for Cancer Therapy.

The integration of a multidimensional treatment dominated by active ingredients of traditional Chinese medicine (TCM), including enhanced chemotherapy and synergistically amplification of oxidative damage, into a nanoplatform would be of great significance for furthering accurate and effective cancer treatment with the active ingredients of TCM. Herein, in this study, we designed and synthesized four matrine-proteolysis-targeting chimeras (PROTACs) (depending on different lengths of the chains named LST-1, LST-2, LST-3, and LST-4) based on PROTAC technology to overcome the limitations of matri

Matrine alleviates Staphylococcus aureus lipoteichoic acid-induced endometritis via suppression of TLR2-mediated NF-κB activation.

Endometritis is one of the main diseases that causes great economic losses in the dairy industry. Recent studies have shown that matrine extracted from the traditional Chinese herb Sophora flavescens is an alkaloid with a broad range of bioactivities. Here, we aimed to investigate the protective effects of matrine on Staphylococcus aureus lipoteichoic acid (LTA)-induced endometritis in mice and elucidate the possible molecular mechanisms in vitro. Histopathological changes showed that matrine remarkably attenuated the uterus injury in a mouse model of LTA-induced endometritis. qPCR and ELISA r

A Novel Matrine Derivative WM130 Inhibits Activation of Hepatic Stellate Cells and Attenuates Dimethylnitrosamine-Induced Liver Fibrosis in Rats.

Activation of hepatic stellate cells (HSCs) is a critical event in process of hepatic fibrogenesis and cirrhosis. Matrine, the active ingredient of Sophora, had been used for clinical treatment of acute/chronic liver disease. However, its potency was low. We prepared a high potency and low toxicity matrine derivate, WM130 (C30N4H40SO5F), which exhibited better pharmacological activities on antihepatic fibrosis. This study demonstrated that WM130 results in a decreased proliferative activity of HSC-T6 cells, with the half inhibitory concentration (IC50) of 68 μM. WM130 can inhibit the migration

Toxic Markers of Matrine Determined Using (1) H-NMR-Based Metabolomics in Cultured Cells In Vitro and Rats In Vivo.

Matrine is one of the main bioactive alkaloids of Sophora flavescens Aiton, which has been widely used to treat various diseases in China. These diseases include viral hepatitis, liver fibrosis, cardiac arrhythmia, skin diseases, and tumors. However, matrine is also the main toxic compound of this herb, and the available biomarkers are not reliable in detecting or quantifying matrine risk. Metabolomics is a powerful tool used to identify early toxicity biomarkers that are specific indicators of damage to biosystems. This study aimed to find the potential biomarkers of the matrine-induced toxic

Autosomal-dominant distal myopathy associated with a recurrent missense mutation in the gene encoding the nuclear matrix protein, matrin 3.

Distal myopathies represent a heterogeneous group of inherited skeletal muscle disorders. One type of adult-onset, progressive autosomal-dominant distal myopathy, frequently associated with dysphagia and dysphonia (vocal cord and pharyngeal weakness with distal myopathy [VCPDM]), has been mapped to chromosome 5q31 in a North American pedigree. Here, we report the identification of a second large VCPDM family of Bulgarian descent and fine mapping of the critical interval. Sequencing of positional candidate genes revealed precisely the same nonconservative S85C missense mutation affecting an int

Rational design, synthesis and effective mechanism of sophoridine and matrine contains 2,3,4-triphenylpyridine derivatives as TopoⅠ inhibitors for the treatment of gastric carcinoma.

Sophoradine and its derivatives have demonstrated promising progress as topoisomerase I inhibitors in cancer therapy, yet their further development has been constrained by moderate activity. Meanwhile, the molecular target of its isomer, matrine, remains unclear. In this study, we designed and synthesized a series of sophoridine and matrine derivatives containing 2,3,4-triphenylpyridine moieties, aiming to develop more potent Topo I inhibitors. Their antiproliferative activities were evaluated in vitro against three cancer cell lines (C6, HepG2, HGC-27). Among them, the sophoridine derivative

Design, Synthesis, and Biological Evaluation of Indolin-2-One-Matrine Derivatives as Potential VEGFR-2-Targeting Agents for Hepatocellular Carcinoma.

Inhibition of the VEGF/VEGFR-2 pathway is a validated strategy to suppress tumor angiogenesis and progression; however, long-term use of several VEGFR-2 tyrosine kinase inhibitors is limited by resistance and systemic toxicity. Here, a series of novel indolinone-matrine hybrids were designed and synthesized via a molecular hybridization strategy. The antiproliferative activities were evaluated against human hepatocellular carcinoma (HCC) cell lines (HepG-2, HuH7, and MHCC97H). Among them, J9 showed the most potent activity with IC50 values of 5.81, 2.14, and 3.03 μM, respectively, and relative

A co-amorphous salt of ursodeoxycholic acid and matrine incorporated into dissolving microneedles for combined antibacterial and anti-inflammatory treatment of acne vulgaris.

Acne vulgaris is characterized by microbial dysbiosis and excessive inflammatory activation, whereas current topical therapies are often limited by poor skin penetration and increasing antibiotic resistance. Ursodeoxycholic acid (UDCA) has anti-inflammatory potential but suffers from low aqueous solubility and limited cutaneous permeability. In this study, a co-amorphous salt of UDCA and matrine (MAT) was developed to improve the pharmaceutical performance of UDCA and to combine the complementary activities of the two components. UDCA-MAT was further incorporated into hyaluronic acid-based dis

Matrine ameliorated myocardial ischemia-reperfusion injury through lipid accumulation reduction by activating JAK2-STAT3-CPT1 signaling pathway.

Matrine (MT), the most abundant active compound derived from Sophora flavescens Ait, has demonstrated promising cardioprotective effects against myocardial ischemia-reperfusion (MIR) injury, and is known to reduce excessive triglyceride (TG) level in hyperlipidemia. However, the specific effects and molecular mechanism of MT in regulating cardiac lipid metabolism have remained elusive. This study investigated the role and underlying mechanism of MT in restoring disrupted lipid metabolism in cardiomyocytes. Our results indicate that MT treatment significantly improved MIR-induced cardiac insuff

Matrine inhibits the proliferation, invasion and migration of castration-resistant prostate cancer cells through regulation of the NF-κB signaling pathway.

Matrine is a naturally occurring alkaloid extracted from the Chinese herb Sophora flavescens. It has been demonstrated to exhibit antiproliferative properties, promote apoptosis and inhibit cell invasion in a number of cancer cell lines. It has also been shown to improve the efficacy of chemotherapy when it is combined with other chemotherapy drugs. However, the therapeutic efficacy of matrine for prostate cancer remains poorly understood. In the present study, we showed that matrine inhibited the proliferation, migration and invasion of both DU145 and PC-3 cells in a dose- and time-dependent

Two new matrine-methyl adducts from Sophora alopecuriodes with anti-inflammatory activity via ROS/HIF-1α regulation.

Phytochemical investigation of the seeds of Sophora alopecuroides led to the isolation of two previously undescribed matrine-methyl adducts (1-2). These compounds represent the first naturally occurring methylated derivatives featuring two distinct types of newly constructed C16 matrine-type skeletons, arising from unique CC bond formations at the C-9 and C-3 positions, respectively. Their structures and absolute configurations were established by comprehensive spectroscopic analysis (HR-ESI-MS, 1D/2D NMR) and comparison of experimental electronic circular dichroism (ECD) spectra. 10-Hydroxy-2-decenoic acid /matrine deep eutectic solvent encapsulated in hyalurosomes for enhanced transdermal delivery and antioxidant efficacy.

As the primary functional constituent of royal jelly, 10-hydroxy-2-decenoic acid (10-HDA) exhibits potent anti-inflammatory, and antioxidant activities. However, its practical application in cosmetic formulations has been severely limited by poor aqueous and oily solubility. To overcome these drawbacks, a deep eutectic solvent composed of 10-HDA and matrine (designated HM-DES) was first prepared to improve its solubility. Furthermore, HM-DES was encapsulated into sodium hyaluronate-decorated liposomes named hyalurosomes (HA-HM-Lip) to further enhance its transdermal delivery efficiency. Fourie

Matrine ameliorates anxiety and depression-like behaviour by targeting hyperammonemia-induced neuroinflammation and oxidative stress in CCl4 model of liver injury.

Acute or chronic liver injury is associated with hyperammonemia which induced neuroinflammation and oxidative stress in the brain. Neuroinflammation, oxidative stress, reduced neurogenesis, and apoptosis are critical factors for the development of anxiety and depression. The present study was aimed to evaluate the anxiolytic and antidepressant properties of matrine against acute liver injury in the rodent model. Acute liver injury in mice was induced by administration of the acute hepatotoxic dose of carbon tetrachloride (CCl4) (1 ml/kg, i.p.). Pretreatment of mice with matrine (50 mg/kg i.p.)

The EV-A71 RNA-binding protein Matrin 3 facilitates viral replication by stabilizing viral RNA via ZDHHC20-mediated palmitoylation.

Enterovirus 71 (EV-A71) is a major causative agent of severe hand, foot, and mouth disease (HFMD) in young children. However, the molecular interplay between EV-A71 and host factors, particularly how these interactions govern viral replication, remains incompletely understood. To systematically identify host factors involved in EV-A71 infection, we performed comprehensive identification of RNA-binding proteins (RBPs) by mass spectrometry (ChIRP-MS) during viral infection and identified 374 host proteins associated with the EV-A71 genomic RNA.
