Host-gene control of C-type tumor virus-expression and tumorigenesis: relevance of studies in inbred mice to cancer in man and other species.

C-type RNA viruses are tumorigenic in mice and other species. They are widespread and transmitted in an intimate relationship with host-cell genetic material. Tumorigenesis is no doubt a complex biological phenomenon, and involves both genetic and environmental factors. Among the genetic factors, single-locus determination is probably exceptional; yet, a number of genes are known in inbred mice and other species, including man, that can exert a major influence. Clearly, studies with inbred mice apply outside the murine model and justify generalizations on the etiology of cancer. The types of c

A Natural Occurring Mouse Model with Adgrv1 Mutation of Usher Syndrome 2C and Characterization of its Recombinant Inbred Strains.

BACKGROUND/AIMS: Our laboratory discovered a Kunming mouse with enormous electroretinogram (ERG) defects. Its auditory brainstem response (ABR) threshold was significantly elevated and closely resembled the features of Usher syndrome (USH). This study sought to cross these USH-like mice (named KMush/ush mice) with CBA/CaJ mice to establish recombinant inbred strains and identify their phenotypes and genotypes. METHODS: KMush/ush mice were crossed with CBA/CaJ mice to establish inbred strains by sibling mating. ERG, ABR, ocular fundus morphology, histological examinations of the retina and inn

Carotid chemoreceptor development in mice.

Mice are the most suitable species for understanding genetic aspects of postnatal developments of the carotid body due to the availability of many inbred strains and knockout mice. Our study has shown that the carotid body grows differentially in different mouse strains, indicating the involvement of genes. However, the small size hampers investigating functional development of the carotid body. Hypoxic and/or hyperoxic ventilatory responses have been investigated in newborn mice, but these responses are indirect assessment of the carotid body function. Therefore, we need to develop techniques

Susceptibility of inbred mice to Paragonimus miyazakii infection.

Differences in the susceptibility among inbred strains of mice to Paragonimus miyazakii infection were examined. Recovery of worms varied among the strains used. More were recovered from BALB/c mice than from any of the other strains; whereas, the fewest were recovered from C57BL/6 and C57BL/10. No worm formed a cyst in the lung or matured in any of the strains.

Host defenses in experimental scrub typhus: genetics of natural resistance to infection.

Genetic resistance to lethal infection with Rickettsia tsutsugamushi was studied in over 30 inbred strains, inbred hybrids, and outbred stocks of mice. Inbred mice infected intraperitoneally with the Gilliam strain of R. tsutsugamushi showed three patterns of response: susceptible (A/HeJ, C3H/HeDub, C3H/HeJ, C3H/HeN, C3H/St, CBA/J, DBA/1J, DBA/2J, and SJL/J), resistant (AKR/J, BALB/cDub, BALB/cJ, C57BL/6J, C57L/J, and SWR/J), and selectively resistant (A/J). The selectively resistant pattern was characterized by random deaths occurring throughout the titration range and was also observed in th

Patterns of cerebellar foliation in recombinant inbred mice.

Morphometric analysis of cerebellar foliation patterns at the midsagittal plane was done in the inbred strains BALB/cByJ, C57BL/6ByJ and their 7 recombinant inbred strains in order to assess possible major gene influences. The cerebellum was dissected away from the brainstem and weighed prior to histology so that measures of the depth of fissures and sulci could be related to overall size of the brain and cerebellum. Results for 177 mice revealed that many brains had extra sulci present within the central lobe, the culmen, the declive and the uvula, and that patterns within a genetically unifo

Phenotypic characterization of the KK/HlJ inbred mouse strain.

Detailed histopathological diagnoses of inbred mouse strains are important for interpreting research results and defining novel models of human diseases. The aim of this study was to histologically detect lesions affecting the KK/HlJ inbred strain. Mice were examined at 6, 12, and 20 months of age and near natural death (ie, moribund mice). Histopathological lesions were quantified by percentage of affected mice per age group and sex. Predominant lesions were mineralization, hyperplasia, and fibro-osseous lesions. Mineralization was most frequently found in the connective tissue dermal sheath

Murine thyroiditis. 3. Influence of syngeneic and allogeneic thyroid antigen on induction of the disease.

Experimental autoimmune thyroiditis, characterized by thyroid antibodies (precipitating, haemagglutinating and complement fixing) and infiltration of the gland with mononuclear cells, was induced in inbred and randomly bred strains of mice, using thyroid extract from the same (syngeneic) or different (allogeneic) strains emulsified in sonicated Freund's complete adjuvant. The syngeneic antigen was much less efficacious than the allogeneic in inducing thyroid antibody and lesions. The subcutaneous route was more effective than intradermal immunization and pertussis vaccine decreased the inciden

The mouse as a model for human audition. A review of the literature.

The mouse has several distinct advantages as an experimental model for human audition. Mice and humans express a similar presbyacusic and ototraumatic pattern. Several genetic mouse models also exist for conditions resembling human auditory disorders, such as otosclerosis. This paper reviews the strains of inbred mice (e.g. the CBA/J) which have recently been used as models for the normal human auditory system, describing their deficiencies. It is suggested that the F1 offspring of CBA/CaJ and AU/SsJ inbred mice would have advantages over existing models.

Antigen-specific lymphocyte proliferative responses in inbred mice after Trichinella spiralis infection.

The in vitro antigen-specific lymphoproliferative response of spleen, mesenteric lymph node (MLN), and coeliac lymph node (CLN) cells taken from various strains of inbred mice infected with Trichinella spiralis was assessed. In most experiments cell populations were stimulated with excretory/secretory antigens (ESA) derived from adult and larval worms. Lymphoid cells collected 5-7 days postinfection were usually the most responsive to ESA as measured by [3H]thymidine uptake. Spleen cells were more responsive than either MLN or CLN cells. There was a correlation between in vitro ESA stimulation

The hypervirulent Type-1/Type-17 phenotype of Cryptococcus neoformans clinical isolates is specific to A/J mice.

Cryptococcus neoformans is a fungal pathogen that causes cryptococcal meningitis in immunocompromised individuals. Both host- and pathogen-specific factors are known to affect patient outcome, and recent studies showed that strain-specific differences in C. neoformans clinical isolates can influence virulence in A/J mice. However, it is unclear how the immunologic and genetic background of inbred mouse strains affects disease outcome during C. neoformans infection. In this study, we show that a hypervirulent phenotype is dependent on the host immune response and mouse genetic background. A/J m

The inheritance of factors controlling resistance in mice infected with Trypanosoma brucei rhodesiense.

In crosses between 2 recombinant inbred strains of mice (B x H-2 and B x H-14), resistance to infection with Trypanosoma brucei rhodesiense as measured by survival time is suggested to be controlled by a dominant gene(s). In prior studies using the same clone of trypanosomes, but a different set of inbred mouse strains, it was demonstrated that resistance in H-2 congenic mice was a recessive trait. This work suggests that in mouse trypanosomiasis, the number of genes involved in resistance and their dominant or recessive nature will vary between different inbred mouse strains. There was a stat

The new disease models from genetic polymorphisms of Japanese wild mice.

Personalized medicine offers a custom-made treatment for each patient directed by information of individual's genetic variation. Despite plenty of information about human single nucleotide polymorphisms (SNPs) and gene expression profile, predicting functions of genetic variations in humans is still a difficult task. Genetic analysis using experimental animals is possible to provide information about the functions of genetic polymorphisms over experimental invention in humans. In particular, inbred strains established from wild mice are valuable resources for analyzing functions of genetic pol

Qualitative effects of dioxin on molars vary among inbred mouse strains.

OBJECTIVE: We evaluated the effects of different levels of the potent environmental toxicant and teratogen, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), on molar development in mice in six inbred strains, all with TCDD responsive Ahr alleles. DESIGN: Pregnant females were exposed on gestation day 13 to 4 different levels of TCDD (control, 0.01, 0.1 and 1.0 microg/kg) and their offspring were examined for the frequency of missing third molars (M3s) and for differences in first mandibular molar (M1) cuspal morphology. RESULTS: Missing M3s were prevalent only in mice in two strains, C3H/HeJ and

Whole-genome sequencing reveals the genetic mechanisms of domestication in classical inbred mice.

BACKGROUND: The laboratory mouse was domesticated from the wild house mouse. Understanding the genetics underlying domestication in laboratory mice, especially in the widely used classical inbred mice, is vital for studies using mouse models. However, the genetic mechanism of laboratory mouse domestication remains unknown due to lack of adequate genomic sequences of wild mice. RESULTS: We analyze the genetic relationships by whole-genome resequencing of 36 wild mice and 36 inbred strains. All classical inbred mice cluster together distinctly from wild and wild-derived inbred mice. Using nucle

Genetics of responsiveness to high-fat and high-cholesterol diets in the mouse.

The natural variation among inbred strains of mice was used to elucidate the genetic factors underlying the responsiveness to high-fat and high-cholesterol diets. The nine strains examined are the progenitors of recombinant inbred strain sets: C57BL/6J, C57L/J, SWR/J, SJL/J, SM/J, A/J, AKR/J, C3H/HeJ, and DBA/2J. Plasma lipids, liver lipids, the prevalence of cholesterol gallstones, and the size of aortic fatty streak lesions were examined after 18 wk of consumption of the diet containing 15% fat and 1% cholesterol. The variation in aortic lesions found among inbred strains provided the basis

Resistance to Leishmania major infection correlates with the induction of nitric oxide synthase in murine macrophages.

Inbred strains of mice differ considerably in their innate resistance to leishmanial infection. BALB/c mice are highly susceptible to cutaneous leishmaniasis caused by Leishmania major, whereas CBA mice are resistant. We now show that this resistance correlates with the ability of macrophages to synthesize nitric oxide (NO) following activation with interferon-gamma or tumor necrosis factor alpha. Furthermore, the larger amounts of NO generated by resistant macrophages are related to higher levels of NO synthase activity, a difference which is not attributable to the number or the affinity of

Efficacy of BCG Vaccination Depends on Host Genetics.

We studied the effectiveness of anti-tuberculosis vaccination with BCG in mice of inbred strains and F1 hybrids (highly resistant to tuberculosis infection) that represent a wide range of genetically determined differences in susceptibility to infection with virulent Mycobacterium tuberculosis. The greatest relative effect was found in susceptible mice, with the exception of highly susceptible I/St mice that were practically not protected by vaccination. Despite significant effect of vaccination in inbred mice, their resistance to M. tuberculosis infection did not exceed that of non-vaccinated

Anticonvulsive effect of p-chlorophenylalanine in mice on a low magnesium diet.

We have reported previously that para-chlorophenylalanine prevented seizure manifestations in an inbred strain of mice genetically predisposed to audiogenic seizures. We now find that this protective effect is not limited to a particular inbred strain of susceptible animals but can be demonstrated in Swiss-Webster and CF No.1 strains which were not originally subject to audiogenic seizures but in which we have induced audiosusceptibility through maintenance on a low magnesium diet.

Adaptation in the vertebral column: a comparative study of patterns of metameric variation in mice and men.

In this paper we examine metamerism in the vertebral column of certain mammals from the perspectives of development and adaptation. To this end we examine the patterns of metameric variation of dimensions of the neural (vertebral) canal, vertebral body and spinous process in man and inbred strains of mice. The data from inbred strains of mice indicate that variability in dimensions within a strain reflects the temporal ordering and nature of developmental influences on vertebral morphogenesis. Differences between strains parallel the within-strain findings. These findings are attributed to som
