Application of unfolding transformation in the random matrix theory to analyze in vivo neuronal spike firing during awake and anesthetized conditions.

General anesthetics decrease the frequency and density of spike firing. This effect makes it difficult to detect spike regularity. To overcome this problem, we developed a method utilizing the unfolding transformation which analyzes the energy level statistics in the random matrix theory. We regarded the energy axis as time axis of neuron spike and analyzed the time series of cortical neural firing in vivo. Unfolding transformation detected regularities of neural firing while changes in firing densities were associated with pentobarbital. We found that unfolding transformation enables us to co

Fgfr1 conditional-knockout in neural crest cells induces heterotopic chondrogenesis and osteogenesis in mouse frontal bones.

Most facial bones, including frontal bones, are derived from neural crest cells through intramembranous ossification. Fibroblast growth factor receptor 1 (Fgfr1) plays a pivotal role in craniofacial bone development, and loss of Fgfr1 leads to cleft palate and facial cleft defects in newborn mice. However, the potential role of the Fgfr1 gene in neural crest cell-mediated craniofacial development remains unclear. To investigate the role of Fgfr1 in neural crest cells, we analyzed Wnt1-Cre;Fgfr1flox/flox mice. Our results show that specific knockout of Fgfr1 in neural crest cells induced hetero

Advanced biomaterial technologies for neural regeneration in central nervous system diseases: mechanisms and applications.

Effective repair of central nervous system (CNS) injuries remains highly challenging due to several formidable barriers, including severe structural disruption, hostile microenvironments, and limited intrinsic regenerative capacity. In response, biomaterials incorporated with therapeutic agents provide advanced interventional strategies for neural regeneration. This review systematically summarizes recent progress in biomaterial-based CNS repair, focusing on three core strategies: physical remodeling of the neural structure, targeted drug delivery systems for biochemical regulation, and cellul

Spike Analysis of the Neural Activities Across the Rats' Auditory Brain Structures.

Tinnitus is a health condition that affects a large population. Clinical diagnosis and treatment have been developed for treating tinnitus for years. However, there are still limitations because researchers have yet to elucidate the mechanisms underlying how tinnitus neural signals develop in brain structures. Abnormal neural interactions among the brain areas are considered to play an important role in tinnitus generation. Researchers have been studying neural activities in the auditory brain structures, including the dorsal cochlear nucleus (DCN), inferior colliculus (IC), and auditory corte

Adaptation of retinal discriminability to natural scenes.

Sensory systems discriminate between stimuli to direct behavioral choices, a process governed by two distinct properties - neural sensitivity to specific stimuli, and neural variability that importantly includes correlations between neurons. Two questions that have received extensive investigation and debate are whether visual systems are optimized for natural scenes, and whether correlated neural variability contributes to this optimization. However, the lack of sufficient computational models has made these questions inaccessible in the context of the normal function of the visual system, wh

Computational design and nonlinear dynamics of a recurrent network model of the primary visual cortex.

Recurrent interactions in the primary visual cortex make its output a complex nonlinear transform of its input. This transform serves preattentive visual segmentation, that is, autonomously processing visual inputs to give outputs that selectively emphasize certain features for segmentation. An analytical understanding of the nonlinear dynamics of the recurrent neural circuit is essential to harness its computational power. We derive requirements on the neural architecture, components, and connection weights of a biologically plausible model of the cortex such that region segmentation, figure-

Reduced expression of MECP2 affects cell commitment and maintenance in neurons by triggering senescence: new perspective for Rett syndrome.

MECP2 protein binds preferentially to methylated CpGs and regulates gene expression by causing changes in chromatin structure. The mechanism by which impaired MECP2 activity can induce pathological abnormalities in the nervous system of patients with Rett syndrome (RTT) is not clearly understood. To gain further insight into the role of MECP2 in human neurogenesis, we compared the neural differentiation process in mesenchymal stem cells (MSCs) obtained from a RTT patient and from healthy donors. We further analyzed neural differentiation in a human neuroblastoma cell line carrying a partially

The role and mechanism of 5-HTDRN-BNST neural circuit in anxiety and fear lesions.

Central 5-hydroxytryptaminergic dorsal raphe nucleus (5-HTDRN)-bed nucleus of stria terminalis (BNST) neural circuit dysfunction is one of the important neurobiological basis of anxiety and fear disorders. Under stress, 5-hydroxytryptamine (5-HT) neurons act on BNST receptors to attenuate anxiety and fear responses or enhance anxiety and fear. In BNST, corticotropin releasing factor neurons play a role in regulating emotions by reversely regulating excitatory or inhibitory 5-HT neurons. The composition of 5-HTDRN-BNST neural circuit, the pathological changes of 5-HTDRN-BNST neural circuit func

A scale out approach towards neural induction of human induced pluripotent stem cells for neurodevelopmental toxicity studies.

Stem cell's unique properties confer them a multitude of potential applications in the fields of cellular therapy, disease modelling and drug screening fields. In particular, the ability to differentiate neural progenitors (NP) from human induced pluripotent stem cells (hiPSCs) using chemically-defined conditions provides an opportunity to create a simple and straightforward culture platform for application in these fields. Here, we demonstrated that hiPSCs are capable of undergoing neural commitment inside microwells, forming characteristic neural structures resembling neural rosettes and fur

Regional differences in neural crest morphogenesis.

Neural crest cells are pluripotent cells that emerge from the neural epithelium, migrate extensively, and differentiate into numerous derivatives, including neurons, glial cells, pigment cells and connective tissue. Major questions concerning their morphogenesis include: 1) what establishes the pathways of migration and 2) what controls the final destination and differentiation of various neural crest subpopulations. These questions will be addressed in this review. Neural crest cells from the trunk level have been explored most extensively. Studies show that melanoblasts are specified shortly

[Neural network abnormalities caused by alcohol: approach for repair using neural stem cells].

Recent clinical neuroimaging studies have revealed the possible relation between morphological brain changes, and memory, cognitive impairment in the course of alcoholism and depression. In the previous studies, we have been analyzing the mechanism of neural network disruption by ethanol using postmortem human brain and cultured cells, and identified the sensitive effect of ethanol on the neural stem cell (NSC) differentiation rather than the influence on neuronal cell survival. Furthermore, to develop a novel method for reconstruction of the neural network damaged by ethanol, we tried to anal

Optimizing S1 nerve root block using three-dimensional computed tomography imaging: Identifying the ideal fluoroscopic angle and predicting S1 neural foramen position.

BACKGROUND: S1 nerve root block (S1NRB) is frequently used to diagnose and treat lumbosacral and lower limb pain. However, the visibility of the S1 neural foramen can be obscured by factors including intestinal gas, increasing the procedure's technical difficulty. This study identified the optimal fluoroscopic angle for S1NRB and standardized the technique using three-dimensional (3D) computed tomography (CT) images. METHODS: We analyzed 3D CT images of 101 patients with lumbar degenerative diseases (lumbar disc herniation and lumbar spinal canal stenosis). The reference position angle (RPA)

Epidemiology of neural tube defects in Utah, 1940-1979.

The prevalence and distribution of 991 cases of neural tube defects who were born in Utah in 1940-1979 are analyzed. The average prevalence at birth is 1.00 per 1000 births (live and stillbirths), which is similar to other states in the western United States. A decline in prevalence is observed during the past 10 years, although there is not a long-term secular trend for the whole time period. Examination of hospital records shows that 17.9% of neural tube defects were not reported on birth or fetal death certificates. The male/female sex ratio is 0.67. No seasonality is found for spina bifida

Discovering the unexpected: Insights into the dynamics of mouse neural tube closure revealed by time-lapse imaging.

The use of time-lapse imaging to study neural tube closure in mouse embryos has provided unexpected insights into the complex morphogenetic processes involved. When neural tube closure is disrupted, it leads to neural tube defects (NTDs), which are among the most common structural birth defects in humans, associated with long-term disabilities and death. This review explores the growing body of research on time-lapse imaging experiments conducted in mice, emphasizing discoveries of the dynamic cellular movements and changes that enable neural tube formation. Advances in mouse embryo culture an

Distinct modes of functional neural organization in autism: Insights from dynamical systems analysis of resting-state EEG.

While differences in patterns of functional connectivity and neural synchronization have been reported between individuals on the autism spectrum and neurotypical peers at various age stages, these differences appear to be subtle and may not be captured by typical quantitative measures of EEG. We used the dynamical systems approach to analyze resting-state EEG to investigate fine-grained spatiotemporal organization of brain networks in autistic and neurotypical young adults. While power spectra showed minimal group differences, autistic participants exhibited higher Lyapunov exponents (indicat
