Delirium due to scopolamine patch in a 4-year-old boy.

The scopolamine patch is usually used to reduce postoperative nausea and vomiting associated with anesthesia and/or surgery. It is also commonly used for the prevention of motion sickness. Transdermal scopolamine patches have been used for decades and there are few reports in the literature of toxic psychosis associated with the product. Most documented cases of acute psychosis following administration of scopolamine or other anticholinergic agents have been from the adult population. Here we present a 4-year-old boy with deteriorated cognitive function and changed mental status acutely. Besid

An investigation of the effects of benzodiazepine receptor ligands and of scopolamine on conceptual priming.

Scopolamine and lorazepam both produce anterograde impairments of explicit memory but only lorazepam impairs implicit memory as assessed by perceptual priming tasks. The main aim of the two experiments reported in this article was to determine the effects of these drugs on conceptual priming. Experiment 1 compared the effects of lorazepam (1,2 mg PO) with scopolamine (0.3,0.6 mg SC) and placebo in a study with 60 healthy volunteers. Experiment 2 compared the separate and combined effects of lorazepam (2 mg PO) and flumazenil (2 mg IV) with placebo in a study with 48 healthy volunteers. We foun

Induction of cognitive impairment by scopolamine and noncholinergic agents in rhesus monkeys.

In primates, treatment with scopolamine impairs performance of a spatial delayed response task in a way which mimics deficits seen spontaneously in aged primates and demented patients. Despite their efficacy in reversing scopolamine induced disruption, the effects of cholinergic agonists on cognition in aged primates and dements are unimpressive, suggesting that other neurotransmitter systems are also involved in this type of deficit. We have induced a scopolamine-like impairment of spatial delayed response performance in rhesus monkeys using phencyclidine (0.1-0.2 mg/kg i.m.), lorazepam (0.4-

Differential effects of scopolamine and amphetamine on microcomputer-based performance tests.

Scopolamine (1.0 mg) and d-amphetamine (10 mg) were administered alone and in combination to 16 subjects (medical students), randomly assigned to testing sessions in a fully crossed-over (Latin square) within-subjects design. After being practiced to stability, 9 performance tests from a menu of portable microcomputer-based tests were administered double-blind over 4 weekly treatments (including a placebo). Differential effects of drugs on performance were found. Motor and perceptual speed tests appeared enhanced by d-amphetamine and not degraded by scopolamine. Two of the five cognitive tests

Effects of scopolamine on antipredator defense reactions in wild and laboratory rats.

Two experiments were designed to investigate the effects of scopolamine hydrobromide (0.25-1.0 mg/kg), and its methyl derivative, on the defensive reactions of rats to nonpainful threat stimuli. In the first experiment, over the dose range studied neither compound significantly altered avoidance, freezing, defensive threat or attack in wild Rattus rattus confronted by the experimenter and other predator-related stimuli. Scopolamine hydrobromide did, however, produce a dose-dependent increase in flight distance; this effect was not seen with the methyl compound, confirming central cholinergic m

Antihistaminics enhance morphine-, but not amphetamine- and scopolamine-induced hyperactivity in mice.

Three histamine H1-receptor antagonists, chlorpheniramine, diphenhydramine and tripelennamine, were tested alone or in combination with morphine, amphetamine and scopolamine on locomotor activity in mice. All three antihistaminics, at some dosage levels, enhanced morphine-induced hyperactivity, but did not change or even reduce locomotor stimulation induced by amphetamine and scopolamine. The results suggest that H1-blocking agents may specifically interact, though not necessarily directly, with opiate mechanisms in producing behavioural effects.

Alleviation of induced vertigo. Therapy with transdermal scopolamine and oral meclizine.

Twelve healthy subjects received seven-day treatments on a randomized, double-blind, crossover basis, of a transdermal scopolamine system, oral meclizine, and placebo, separated by one-week intervals. Just prior to each treatment, and on days 1 and 7 of each treatment, subjects received two warm (44 degrees C) caloric irrigations of each external auditory canal. Following each irrigation, subjects rated their vertigo symptoms. Subjects reported on their side effects daily throughout each treatment period. Vertigo symptoms on day 1 of treatment were significantly less with transdermal scopolami

Psychomotor, physiological and cognitive effects of scopolamine and ephedrine in healthy man.

Three placebo-controlled double-blind studies in healthy volunteers were performed to reveal the psychophysiological effects of scopolamine, ephedrine and their combination. Single intravenous dose of scopolamine 6 micrograms/kg (scopolamine hydrobromide 7.4 micrograms/kg) impaired various psychomotor functions both subjectively and objectively. It caused sedation, impairment of coordinative and reactive skills, visual disturbances and impairment of shortterm memory. Oral scopolamine hydrobromide in single doses of 0.3 mg and 0.9 mg, or 0.9 mg b.d. for 3 days, had few effects. A slight impairm

Differential reversal by scopolamine and THIP of the antistereotypic and cataleptic effects of neuroleptics.

Scopolamine reversed the neuroleptic-induced catalepsy and neuroleptic blockade of amphetamine-induced stereotypies in rats. The effect on catalepsy was the most pronounced. Butyrophenones were mainly sensitive to scopolamine reversal whereas the thioxanthenes cis-Z-flupentixol and piflutixol were not significantly influenced. THIP did not change the effects of cis-Z-flupentixol, fluphenazine and haloperidol. The relevance of these findings for the use of stereotypy and catalepsy tests as screening for antiscizophrenic and extrapyramidal side-effects, respectively, is discussed.

Accidental foodborne poisoning by atropine and scopolamine: a fatal case report.

Tropane alkaloids such as atropine and scopolamine are naturally occurring compounds found in various plants of the Solanaceae family, notably Datura species. Although widely used in clinical settings, their potent anticholinergic properties can lead to severe toxic effects. Fatalities involving these substances remain rare, particularly in accidental contexts. Here, the authors report the case of a 58-year-old male who died following the accidental consumption of a meal suspected to have been contaminated with plant material containing tropane alkaloids. At autopsy, blackish vomitus containin

Transdermal scopolamine for the treatment of recurrent parotid sialocele: a case report.

Recurrent parotid sialocele is rare and challenging to treat. Treatment options are limited for cases of parotid sialocele that recur despite ductal ligation. This case study presents a patient who underwent wide excision of the right buccal mucosa due to squamous cell carcinoma. During the wide excision, a segment of the parotid duct was excised, and ductal ligation was performed to prevent the occurrence of a sialocele, followed by reconstruction using a folded anterolateral thigh free flap. Twenty-two days after surgery, parotid sialocele occurred despite the initial ductal ligation and sub

The Ameliorating Effect of Myrrh on Scopolamine-Induced Memory Impairments in Mice.

Myrrh has been used since ancient times for the treatment of various diseases such as inflammatory diseases, gynecological diseases, and hemiplegia. In the present study, we investigated the effects of aqueous extracts of myrrh resin (AEM) on scopolamine-induced memory impairments in mice. AEM was estimated with (2E,5E)-6-hydroxy-2,6-dimethylhepta-2,4-dienal as a representative constituent by HPLC. The oral administration of AEM for 7 days significantly reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze test. In the passive avoidance task, AEM also restored the dec

Antidepressant treatment history as a predictor of response to scopolamine: clinical implications.

BACKGROUND: The intravenous administration of scopolamine produces rapid antidepressant effects. Generally, failing multiple previous antidepressant trials is associated with a poor prognosis for response to future medications. This study evaluated whether treatment history predicts antidepressant response to scopolamine. METHODS: Treatment resistant patients (2 failed medication trials) (n=31) and treatment naïve patients (no exposure to psychotropic medication) (n=31) with recurrent major depressive or bipolar disorder participated in a double-blind, placebo-controlled, crossover clinical t

Scopolamine selectively disrupts the acquisition of contextual fear conditioning in rats.

Muscarinic cholinergic antagonism produces learning and memory deficits in a variety of hippocampal-dependent tasks. Hippocampal lesions produce both acquisition deficits and retrograde amnesia for contextual fear conditioning, but do not impact fear conditioning to discrete cues. In order to examine the effects of muscarinic antagonism in this paradigm, rats were given scopolamine (1 mg/kg) either before or for 3 days after a Pavlovian fear-conditioning session in which tones were paired with aversive footshocks. Fear to the context and the tone was assessed by measuring freezing in separate

Reduction of salivary flow with transdermal scopolamine: a four-year experience.

Scopoderm transdermal therapeutic system (TTS) is applied to prevent nausea and vomiting associated with motion sickness. Dry mouth is the most common side effect, appearing in up to two thirds of the patients treated. We have used this side effect to the benefit of patients with sialorrhea or with difficulties swallowing normally secreted amounts of saliva. More than 100 patients with tumors of the aerodigestive tract before and after surgery, and patients after parotidectomies, after tracheotomies, with peritonsillar abscesses, tonsillitis and pharyngitis, and neurologic disorders were thus

Dissociative effects of scopolamine on working memory in healthy young volunteers.

Performance on tasks of digit span, mental rotation and immediate free recall of supraspan word lists was measured before and after oral administration of 1.2 mg scopolamine or placebo to healthy young volunteers. Digit span and mental rotation were sensitive to task-specific interference from articulatory suppression and spatial tapping tasks, respectively. Neither task was affected by scopolamine when completed alone or in combination with a secondary task. A concurrent secondary task reduced immediate free recall in a nonspecific fashion (i.e., spatial tapping or articulatory suppression im

Effects of scopolamine and nicotine on human rapid information processing performance.

In the first experiment, after a 10-min baseline test on a rapid information processing task, subjects received oral doses of either placebo, methscopolamine 1.2 mg, scopolamine 0.6 mg or scopolamine 1.2 mg, and 1 h later performed the task again for a 20-min period. Following scopolamine 1.2 mg, correct detections were significantly lower over the 20-min period, whereas no such decrement was observed in the other three conditions. In the second experiment a similar design was used to study the effects of nicotine 0.5 mg, 1.0 mg and 1.5 mg and placebo, except that post-drug testing was carried

Prevention of experimental motion sickness by scopolamine absorbed through the skin.

A double-blind placebo-controlled study compared the efficacy of the antimotion sickness drug scopolamine when administered by oral or transdermal routes. A secondary purpose was to extend our bioassay involving fixed-dose combinations of the homergic drugs promethazine and ephedrine. After receiving 12 apparently identical drug-placebo treatments, eight normal male students were exposed to a slow rotation room to stressful accelerations generated by their execution of 40 head movements out of the plane of the room's rotation of 1 rpm and at 1-rpm increments until either symptoms were experien
