Prenatal stress modifies hippocampal synaptic plasticity and spatial learning in young rat offspring.

Clinical studies demonstrate that prenatal stress causes cognitive deficits and increases vulnerability to affective disorders in children and adolescents. The underlying mechanisms are not yet fully understood. Here, we reported that prenatal stress (10 unpredictable, 1 s, 0.8 mA foot shocks per day during gestational days 13-19) impaired long-term potentiation (LTP) but facilitated long-term depression (LTD) in hippocampal CA1 region in slices of the prenatal stressed offspring (5 weeks old). Cross-fostering neonate offspring by the prenatal stressed or control mothers did not change the eff

A single exposure to the tremorgenic mycotoxin lolitrem B inhibits voluntary motor activity and spatial orientation but not spatial learning or memory in mice.

The indole diterpenoid toxin lolitrem B is a tremorgenic agent found in the common grass species, perennial ryegrass (Lolium perenne). The toxin is produced by a symbiotic fungus Epichloë festucae (var. lolii) and ingestion of infested grass with sufficient toxin levels causes a movement disorder in grazing herbivores known as 'ryegrass staggers'. Beside ataxia, lolitrem B intoxicated animals frequently show indicators of cognitive dysfunction or exhibition of erratic and unpredictable behaviours during handling. Evidence from field cases in livestock and controlled feeding studies in horses h

Comparative analysis of the spatial distribution of brain metastases across several primary cancers using machine learning and deep learning models.

OBJECTIVE: Brain metastases (BM) are associated with poor prognosis and increased mortality rates, making them a significant clinical challenge. Studying BMs can aid in improving early detection and monitoring. Systematic comparisons of anatomical distributions of BM from different primary cancers, however, remain largely unavailable. METHODS: To test the hypothesis that anatomical BM distributions differ based on primary cancer type, we analyze the spatial coordinates of BMs for five different primary cancer types along principal component (PC) axes. The dataset includes 3949 intracranial me

Genomic and spatial epidemiology: lessons learned from SARS-CoV-2 pandemic.

PURPOSE OF REVIEW: The SARS-CoV-2 pandemic presented unprecedented challenges, particularly in understanding its complex spatial transmission patterns. The high transmissibility of the virus led to frequent super-spreading events. These events demonstrated clear spatial clustering patterns, often tied to specific events that facilitated transmission. The uneven geographic distribution of medical resources and varying access to care amplified the impact of SARS-CoV-2. Asymptomatic cases further complicated the situation, as infected individuals could silently spread the virus before being ident

Characterization and classification of chronic kidney disease by spatial MIST and deep learning algorithm.

Chronic kidney disease (CKD) is characterized by disruption of the native kidney architecture at the cellular and molecular levels, leading to eventual kidney fibrosis. To better resolve the spatial complexity of fibrotic remodeling, we applied spatial multiplexed immunostaining with signal tagging (Spatial MIST), a high-dimensional proteomic platform capable of quantifying protein expression at single-cell resolution across intact human kidney tissue specimens. Using kidney biopsies from control/low-grade and high-grade fibrosis, we profiled 22 protein markers to assess structural alterations

Effects of FB1 and HFB1 on Autonomous Exploratory and Spatial Memory and Learning Abilities in Mice.

Fumonisin B1 (FB1) is a representative form of fumonisin and is widely present in food and feed. Hydrolyzed fumonisin B1 (HFB1) emerges as a breakdown product of FB1, which is accompanied by FB1 alterations. While previous studies have primarily focused on the liver or kidney toxicity of FB1, with limited studies existing on its neurotoxicity and even fewer on the toxicity of HFB1, this study focuses on the neurotoxicity of FB1 and HFB1 exposure in mice investigated by the open field test, Morris water maze test, histopathological analysis, and nontargeted metabolomics. Further, the levels of

Finding food in the dark: how trajectories of a gymnotiform fish change with spatial learning.

We analyzed the trajectories of freely foraging Gymnotus sp., a pulse-type gymnotiform weakly electric fish, swimming in a dark arena. For each fish, we compared the its initial behavior as it learned the relative location of landmarks and food with its behavior after learning was complete, i.e. after time/distance to locate food had reached a minimal asymptotic level. During initial exploration when the fish did not know the arena layout, trajectories included many sharp angle head turns that occurred at nearly completely random intervals. After spatial learning was complete, head turns becam

A bedside application-based assessment of spatial orientation and memory: approaches and lessons learned.

Spatial orientation and memory deficits are an often overlooked and potentially powerful early marker for pathological cognitive decline. Pen-and-paper tests for spatial abilities often do not coincide with actual navigational performance due to differences in spatial perspective and scale. Mobile devices are becoming increasingly useful in a clinical setting, for patient monitoring, clinical decision-making, and information management. The same devices have positional information that may be useful for a scale appropriate point-of-care test for spatial ability. We created a test for spatial o

Impaired insulin signaling and spatial learning in middle-aged rats: The role of PTP1B.

The insulin and Brain-Derived Neurotrophic Factor (BDNF) signaling in the hippocampus promotes synaptic plasticity and memory formation. On the other hand, aging is related to the cognitive decline and is the main risk factor for Alzheimer's Disease (AD). The Protein-Tyrosine Phosphatase 1B (PTP1B) is related to several deleterious processes in neurons and emerges as a promising target for new therapies. In this context, our study aims to investigate the age-related changes in PTP1B content, insulin signaling, β-amyloid content, and Tau phosphorylation in the hippocampus of middle-aged rats. Y

Prolame ameliorates anxiety and spatial learning and memory impairment induced by ovariectomy in rats.

N-(3-hydroxy-1, 3, 5 (10) estratrien-17beta-yl)-3-hydroxypropylamine (17β aminoestrogen, prolame) is a steroidal compound with weak estrogen-related trophic-proliferative effects in uterus. Contrasting with 17β-estradiol (E2) pro-coagulant effects, this compound has high anticoagulant and antiplatelet effects. It has been extensively demonstrated that E2 plays important roles in brain function. However, prolame's influence on central nervous system has not been documented. In this study, we evaluated the effects of prolame replacement in young ovariectomized rats on spatial learning and memory

Chronic impairments in spatial learning and memory in rats previously exposed to chlorpyrfos or diisopropylfluorophosphate.

The acute toxicity of organophosphates (OPs) has been studied extensively; however, much less attention has been given to the subject of repeated exposures that are not associated with overt signs of toxicity (i.e., subthreshold exposures). The objective of this study was to determine if the protracted spatial learning impairments we have observed previously after repeated subthreshold exposures to the insecticide chlorpyrifos (CPF) or the alkylphosphate OP, diisopropylfluorophosphate (DFP) persisted for longer periods after exposure. Male Wistar rats (beginning at two months of age) were init

The effects of chronic nicotine on spatial learning and bromodeoxyuridine incorporation into the dentate gyrus of the rat.

RATIONALE: Nicotine is reported to improve learning and memory in experimental animals. Improved learning and memory has also been related to increased neurogenesis in the dentate gyrus (DG) of the hippocampal formation. Surprisingly, recent studies suggest that self-administered nicotine depresses cell proliferation in the DG. OBJECTIVE: To test the hypothesis that the effects of nicotine on cell proliferation in the DG and learning and memory depend upon the nicotine dose administered. METHODS: Rats were chronically infused from subcutaneous osmotic mini pumps with nicotine (0.25 or 4 mg k

ProxiNet transfers spatially learned cellular proximity to dissociated single-cell transcriptomes.

Spatial transcriptomics reveals cellular organization within intact tissues, whereas dissociated single-cell RNA sequencing provides broad transcriptomic coverage but loses information about cellular proximity and neighborhood structure. Here, we developed ProxiNet , a spatially supervised framework that learns transcriptomic signatures of pairwise cellular proximity from spatial reference datasets and transfers these relationships to dissociated single-cell transcriptomes. ProxiNet predicted cellular proximity across brain regions and spatial technologies, including zero-shot cross-technology

Temporal dynamics of adapting to novel contexts during the generalization of learned spatial suppression.

The spatial suppression of a high-probability distractor location (HPDL) acquired through statistical learning critically reduces its attentional priority. The present study conducted four experiments to systematically investigate the mechanisms underlying how this learned suppression generalizes to novel tasks. Specifically, we focused on how generalization is influenced by the priority-enhancing attentional capture of a salient target and the competing demands of a newly introduced suppression process. Following training, Experiments 1 and 2 employed a salient color singleton target to test

Spatially decoupled reliable mutual learning for semi-supervised medical image segmentation.

BACKGROUND: Semi-Supervised Learning reduces the annotation burden for medical image segmentation but currently suffers from spatial context overfitting and confirmation bias due to noisy pseudo-labels. This study aims to propose a robust framework to overcome these limitations. METHODS: We introduce a novel framework termed Spatially Decoupled Reliable Mutual Learning (SDRML). To address context overfitting, we propose a Spatial Decoupling strategy that utilizes translation consistency, compelling the model to focus on intrinsic anatomical features rather than fixed background contexts.
