A novel ANK1 gene mutation associated with hereditary spherocytosis: a case report.

BACKGROUND: Hereditary spherocytosis (HS) is an inherited form of hemolytic anemia resulting from defects in the red blood cell membrane skeleton. Its classic clinical presentation includes anemia, jaundice, and splenomegaly. Key laboratory findings that support the diagnosis are the presence of spherocytes and an elevated reticulocyte count on peripheral blood smear. CASE PRESENTATION: An 8-year-old girl presented with a longstanding history of jaundice-noted for four years-affecting both her skin and sclera. She also reported intermittent episodes of tea-colored urine. Physical examination

[Morphological characteristic of erythrocytes in experimental hypervitaminosis A].

This investigation was aimed at the analysis of the shape and morpho-densitometric parameters of the erythrocytes in rats with experimental hypervitaminosis A. Male Wistar rats received 0.64 mg/g (1167 IU/g) of retinol palmitate (RP) in oil solution orally for 11 consecutive days. Rats fed oil alone and intact animals were used as control groups. At days 5 and 6 of the experiment, the first manifestations of hypervitaminosis A were observed (body mass loss, localized erythema and hemorrhages). In contrast to control groups, in rats with hypervitaminosis A, the area of erythrocyte cytoplasm dec

Herediatary spherocytosis. A review.

Studies of the clinical features of hereditary spherocytosis since 1871 and laboratory investigation of the cellular abnormalities since 1940 have led to the characterization of hereditary spherocytosis as a prime example of a Mendelian dominant, genetically determined disorder of the erythrocyte membrane. This review of hereditary spherocytosis emphasizes the contributions of Dr. Lawrence Young and many others of out present understanding of the disease and discusses current studies of the protein abnormality in the membrane of hereditary spherocytes.

Atypical intraerythrocytic pyruvate kinase in some haematological diseases.

An atypical pyruvate kinase (PK) in erythrocytes was studied in a family with several different haematological diseases: non-spherocytic haemolysis, thrombocytopenia, myelofibrosis and polycythaemic traits. Atypical intraerythrocytic PK was also found in a group of non-related patients with myelofibrosis and polycythaemia vera. The atypical PK was characterised by abnormal urea inhibition and other relevant biochemical parameters.

Erythrocyte morphology as a clinical disease indicator in hospitalized COVID-19 patients.

INTRODUCTION: Most COVID-19 cases are mild, but some require hospitalization due to pneumonia, with symptoms like hypoxia and dyspnea. This has led to speculation about erythrocyte involvement in the infection. This study aimed to examine the differences in the presence of dysmorphic erythrocytes in COVID-19 patients upon admission compared to healthy subjects, as well as to assess these differences in relation to the severity of the clinical presentation of COVID-19. METHODOLOGY: This study included 150 participants: 100 COVID-19 patients and 50 healthy subjects who formed the control group.

Novel compound heterozygous mutations in the SPTA1 gene, causing hereditary spherocytosis in a neonate with Coombs‑negative hemolytic jaundice.

Hereditary spherocytosis (HS) is a common heterogeneous type of inherited hemolytic anemia characterized by jaundice and splenomegaly. Diagnosis of HS in neonates is considered unreliable, and is generally based on positive family history, spherocytes in peripheral smears, increased osmotic fragility, and jaundice. In the present study, routine laboratory tests, next‑generation sequencing, and Sanger sequencing were applied to diagnose a neonatal patient with Coombs‑negative hemolytic jaundice. The neonate had no family history of HS; however, spherocytes were observed in peripheral smears, an

Pitting function of the spleen in malaria: ultrastructural observations.

Ultrastructural studies of spleens from monkeys infected with Plasmodium knowlesi suggest that the spleen removes or "pits" malaria parasites from red cells. This function may explain the presence of nonparasitized spherocytic erythrocytes in the peripheral blood and may in part account for the discrepancy between the excessive hemolysis and the number of parasitized erythrocytes in animals with experimentally induced malaria.

Novel α-spectrin mutation in trans with α-spectrin causing severe neonatal jaundice from hereditary spherocytosis.

We evaluated a neonate with severe jaundice but a negative family history. Spherocytes were present and suspected hereditary spherocytosis was confirmed by osmotic fragility and eosin-5-maleimide erythrocyte staining. We found he was a compound heterozygote for two pathogenic mutations in the gene encoding α-spectrin: a previously reported α(LEPRA) inherited from his asymptomatic mother, and a novel α-spectrin mutation in intron 45 +1 disrupting the consensus splice site, from his asymptomatic father.

[A family with hereditary spherocytosis discovered after an infection with human parvovirus B19].

Hereditary spherocytosis was diagnosed in five of six children in one family after a human parvovirus B19 (B19-virus) infection. The diagnosis was made on the basis of severe anaemia, demonstrable haemolysis, decreased osmotic fragility and an increased number of spherocytes. Since in the serum of the patients an increased level of IgG and IgM antibodies against B19 virus was detectable two weeks after the crisis it was concluded that the B19-virus infection caused the severe anaemia.

Targeted next-generation sequencing identifies a novel nonsense mutation in SPTB for hereditary spherocytosis: A case report of a Korean family.

RATIONALE: Hereditary spherocytosis (HS) is an inherited disorder characterized by the presence of spherical-shaped red blood cells (RBCs) on the peripheral blood (PB) smear. To date, a number of mutations in 5 genes have been identified and the mutations in SPTB gene account for about 20% patients. PATIENT CONCERNS: A 65-year-old female had been diagnosed as hemolytic anemia 30 years ago, based on a history of persistent anemia and hyperbilirubinemia for several years. She received RBC transfusion several times and a cholecystectomy roughly 20 years ago before. Round, densely staining spheri

[Phospho-hexo-isomerase deficiency].

Hematologic, genetic and biochemical data of two patients suffering from glucosephosphate isomerase deficiency are described. In one of them a generalized deficiency could be demonstrated. The molecular instability of the deficient enzyme results in early inactivation. Its consequences for carbohydrate metabolism are exposed. Besides a non-spherocytic hemolytic anemia, a hepatic glycogenosis could be observed. Our patients are compared to litterature.

Coexistence of hereditary spherocytosis and beta-thalassemia: case report of severe hemolytic anemia in an American black.

The first reported case of hereditary spherocytosis (HS) and beta-thalassemia in an American black is presented. The diagnosis rested on clinical presentation, family history and specialized laboratory findings. The tendency toward hemolysis of spherocytes and thalassemia red cells may explain the severity of anemia in this patient. The clinical course of these coexistent diseases and the role of splenectomy in relieving the hemolytic component caused by hereditary spherocytosis are described. A detailed review of the literature on HS and beta-thalassemia is also included.

The spleen and splenectomy.

After 1940, the number of splenectomies performed in the United States and elsewhere increased rapidly. Splenectomy for Banti's disease and malaria decreased gradually into disrepute. Removal of the spleen for idiopathic thrombocytopenic purpura, congenital spherocytic anemia and acquired hemolytic anemia became accepted practice. However, debate still continues regarding the proper indications for splenectomy in Gaucher's disease, Felty's syndrome and leukemia.

Abnormal pattern of erythrocyte ageing in hereditary spherocytosis as shown by Percoll density gradient centrifugation.

Erythrocytes from individuals with Hereditary Spherocytosis (HS) were fractionated on Percoll density gradients. The osmotic fragility, mean cell volume, mean cell haemoglobin concentration, reticulocyte count and cell morphology of the fractions showed that the gradients separate the erythrocytes according to age, the older small spherocytes appearing in the dense fractions. Comparison of the well established parameters of erythrocyte ageing, pyruvate kinase activity, acetylcholinesterase activity and cell potassium content in normal and HS erythrocyte fractions of similar age revealed abnorm

G-6-PD Poznań, variant with severe enzyme deficiency.

A new variant of G-6-PD with severe enzyme dificiency without chronic hemolysis is characterized. The biochemical properties of the variant closely resemble those of the G-6-PD Ramat-Gan described in a case of chronic non-spherocytic hemolytic anemia. As the family data on linking of chronic hemolysis with Ramat-Gan variant are lacking, the differentiation of the variants on the basis of clinical manifestations is not well-founded.

High-throughput biomimetic cycling of red blood cells: elucidating the morpho-mechanical determinants of fatigue and clearance.

During their 120-day circulatory lifespan, red blood cells (RBCs) undergo repeated mechanical deformation as they traverse microcapillaries and splenic inter-endothelial slits (IES). This cyclic mechanical loading gradually impairs RBC deformability, ultimately leading to their clearance by the spleen. However, current platforms for investigating RBC fatigue often couple mechanical loading with real-time observation, which obscures the cumulative impact of cyclic strain. To address this limitation, we developed an integrated microfluidic chip equipped with a dedicated "S"-shaped fatigue zone-e

Haemagglutination assay for HBsAg -- a new approach to reagent preparation.

Human group O rhesus negative erythrocytes were spherocyted and fixed in glutaraldehyde, then coated initially with HBsAg and subsequently with anti-HBs. The resulting reagent was found to agglutinate in the presence of HBsAg positive serum, but not in HBsAg negative serum. The assay was evaluated for blood donor screening (39,962 donations) over a 6-month period, and was compared with Hepatest (RPHA) and AUSRIA-2 (RIA) for the study of two established HBsAg panels.

Inherited erythrocyte pyruvate kinase deficiency: Studies on 15 members of two related families.

The case of a haemolytic non-spherocytic anaemia with pyruvate kinase (PK) deficiency is reported. The investigation concerns two families with a low level of PK. In the propositus and in the members of his family we have also examined the behaviour of some enzymes and the concentration of red cell metabolites. We confirm the heterogeneity of the manifestation and agree that anaemias due to PK deficiency are complex forms in which the enzyme defect is only one of the symptoms.
